Shape Optimization for Multi-Connected Structures (다연결체 구조물에 대한 형상 최적화)

  • The growth-strain method was used for shape optimization of multi-connected structures. It was verified that the growth-strain method is very effective for shape optimization of structures with only one free surface to be deformed. But it could not provide reasonable optimized shape for multi-connected structures, when the growth-strain method is applied as it is. The purpose of this study is to improve the growth-strain method for shape optimization of multi-connected two- and three- dimensional structures. In order to improve, the problems that occurred as the growth-strain method was applied to multi-connected structures were examined, and then the improved method was suggested. The effectiveness and practicality of the developed shape optimization system was verified by numerical examples.

Isolation and Identification of Antagonistic Microorganisms for Biological Control to Major Diseases of Apple Tree(Malus domestica Borkh) (사과 주요 병해 방제를 위한 길항미생물 분리 및 동정)

  • For the purpose of acquiring microbial agents that can be utilized to biologically control the major airborne diseases to apple trees, such as canker(Botryosphaeria dothidea), bitter rot(Glomerella cingulata), alternaria leaf spot(Alternaria mali), root rot(rosellinia necatrix), canker(Valsa ceratosperma) and gray mold rot(Botrytis cinerea), the effective microorgaisms were isolated, tested for antagonistic activity to the pathogens causing major diseases to apple trees and identifed. Screening of more than 5,000 species of microorganisms collected in nature for them antagonistic action to the pathogens causing 5 major diseases to apple trees resulted in selection of effective species. Out of the 11 species, one species designated as CAP134 demonstrated outstanding activity. The bacterial strain, CAP134 exerted antagonistic efficiency of 57% on an isolated strain and 40% on a donated strain of Botryosphaeria dothidea., 52% on an isolated strain and 46% on a purchased strain of Alternaria mali, 60% on Valsa ceratosperma 25% on Glomerella cingulata, and 64% Rosellinia necatrix. The CAP134 was identified as a bacterial strain to Bacillus subtilis ATCC 6633 based on morephology, culture conditions, and physio-biochemical characteristics.

Increase of Low Cycle Fatigue Life at 300℃ for Type 304 Stainless Steel (304 스테인리스강의 300℃에서 저주기 피로수명 증가)

  • Tensile, low cycle fatigue, and fatigue crack growth rate tests were conducted at RT and $300^{\circ}C$ for type 304 stainless steel. Tensile was tested under displacement control and low cycle fatigue was tested under strain control. Fatigue crack growth rate test was conducted under load control and crack was measured by DCPD method. Yield strength and elongation decreased at $300^{\circ}C$. Dynamic strain aging was not detected at $300^{\circ}C$. Low cycle fatigue life increased but fatigue strength decreased at $300^{\circ}C$. Fatigue crack growth rate increased at $300^{\circ}C$. Dislocation structures were mixed with cell and planar and did not change with temperature. Grain size did not change but plastic strain increased at $300^{\circ}C$. Strain induced martensite after low cycle fatigue test increased at RT but decreased at $300^{\circ}C$. It was concluded that the increase of low cycle fatigue life at $300^{\circ}C$ was due to the decrease of strain induced martensite at which crack was initiated.

Biocontrol of Blue Stain in Pine Wood with Lyophilized Mycelium of Ophiostoma quercus Albino Strain

  • Mycelium of Ophiostoma quercus albino strain cultured in liquid culture media was harvested, lyophilized, and stored for examining biocontrol efficacy against wood discoloration by staining fungi in the laboratory and field conditions. Dry weight of mycelium grown in brown sugar yeast extract broth(BYB) showed 3.8 times higher than that grown in potato dextrose broth(PDB). The optimum culture period in BYB was 4 weeks. In vitality test of the albino strain, the lyophilized mycelium stored in liquid nitrogen($-196^{\circ}C$) or in a refrigerator($4^{\circ}C$) kept the vitality until 13 months after storage; however, the mycelium stored at room temperature lost the vitality completely after 13 months. The mycelium stored in liquid nitrogen or in a refrigerator protected wood chips from the discoloration by pretreating mycelial suspension on pine wood chips. The mycelium stored at room temperature for 7 months also showed complete protection. These results suggest that the lyophilized mycelium have a biocontrol efficacy only if it keeps the least vitality. In the field conditions, both albino strain and $Woodguard^{(R)}$(commercial chemical protectant) showed significant differences(p=0.05) in discoloration rate as compared to the non-treated control when these were treated on the wood logs of Pinus rigida. The albino strain showed better protection than $Woodguard^{(R)}$. Isolation frequency of blue stain fungi from the chips of wood logs treated with the albino strain was 0% at three months after treatment, while that treated with $Woodguard^{(R)}$ was 76.7%. In another experiment, pre-treatment of mycelial suspension on the cut surface of wood logs also showed significant protection from wood discoloration. Spraying of both albino strain on the cut surface and insecticides on the bark also showed relatively good control effects as compared to insecticide alone on the bark or nontreated control.

Nematocidal Effect of B. thuringiensis subsp.indiana Strain BtTH109 on Root-Knot Nematode of Tomato (Bacillus thuringiensis subsp. indiana BtTH109의 토마토 뿌리혹선충에 대한 치사효과)

  • For a biological control of root-knot nematode (Meloidogyne incognita) in tomato, efficiency of Bacillus thuringiensis subsp. indiana strain TH109 (BtTH109) on the nematode control was investigated. After the mixture of strain BtTH109 and wheat bran was treated into rhizosphere of the tomato plants with nematode eggs, the stem height and root growth of plants increased. And the juveniles and eggs of nematode are not found in both roots of tomatoes and pot soil after cultured broth of the strain BtTH109 treated 4 times at 3 day-interval into rhizosphere of the infected tomatoes.

Microbial Control of Fly Maggots with Entomopathogenic Nematodes and Fungus in Outhouses of Farmhouses (곤충병원선충과 곰팡이를 이용한 농가화장실 파리의 미생물적 방제)

  • Infectivity of entomopathogenic nematodes and fungus was evaluated against fly larvae in the laboratory and outhouses. Mortalities of Muscina stabulans larvae were 96.7f 2.8% in Steinernema glaseri Dongrae strain, 90.0+0.0% in S. carpocapsae All strain, 86.7f 2.7% in Heterorhabditis bacteriophora Hamyang strain, and 70.0+9.4% in S. carpocapsae Pocheon strain on the filter paper. When 260, 000 nem\ulcornertodes were sprayed into the outhouses, H. bacterwphora Hamyang strain killed 100%, S. glaseri Dongrae strain killed 76.9+3.9%, and S. carpocapsae Pocheon strain killed 58.5+6.1% of maggots. When 130, 000 nematodes and 7.0X lo9 cfu of entomopathogenic fungus, Beauveria brongniartii were sprayed alone or combined into outhouses, mortalities of maggots were 73.6+0.1% in B. brongniartii alone, 77.8+3.9% in S. carpocapsae Pocheon strain plus B. brongniartii, and 77.7f 5.1% in H. bacteriophora Hamyang strain plus B. brongniartii. Entomopathogenic nematodes and fungus were potential biological control agents in this study.

Enhanced antibacterial activity of tilmicosin against Staphylococcus aureus small colony variants by chitosan oligosaccharide-sodium carboxymethyl cellulose composite nanogels

  • Background: The poor bioadhesion capacity of tilmicosin resulting in treatment failure for Staphylococcus aureus small colony variants (SASCVs) mastitis. Objectives: This study aimed to increase the bioadhesion capacity of tilmicosin for the SASCVs strain and improve the antibacterial effect of tilmicosin against cow mastitis caused by the SASCVs strain. Methods: Tilmicosin-loaded chitosan oligosaccharide (COS)-sodium carboxymethyl cellulose (CMC) composite nanogels were formulated by an electrostatic interaction between COS (positive charge) and CMC (negative charge) using sodium tripolyphosphate (TPP) (ionic crosslinkers). The formation mechanism, structural characteristics, bioadhesion, and antibacterial activity of tilmicosin composite nanogels were studied systematically. Results: The optimized formulation was comprised of 50 mg/mL (COS), 32 mg/mL (CMC), and 0.25 mg/mL (TPP). The size, encapsulation efficiency, loading capacity, polydispersity index, and zeta potential of the optimized tilmicosin composite nanogels were 357.4 ± 2.6 nm, 65.4 ± 0.4%, 21.9 ± 0.4%, 0.11 ± 0.01, and -37.1 ± 0.4 mV, respectively; the sedimentation rate was one. Scanning electron microscopy showed that tilmicosin might be incorporated in nano-sized crosslinked polymeric networks. Moreover, adhesive studies suggested that tilmicosin composite nanogels could enhance the bioadhesion capacity of tilmicosin for the SASCVs strain. The inhibition zone of native tilmicosin, tilmicosin standard, and tilmicosin composite nanogels were 2.13 ± 0.07, 3.35 ± 0.11, and 1.46 ± 0.04 cm, respectively. The minimum inhibitory concentration of native tilmicosin, tilmicosin standard, and tilmicosin composite nanogels against the SASCVs strain were 2, 1, and 1 ㎍/mL, respectively. The in vitro time-killing curves showed that the tilmicosin composite nanogels increased the antibacterial activity against the SASCVs strain. Conclusions: This study provides a potential strategy for developing tilmicosin composite nanogels to treat cow mastitis caused by the SASCVs strain.

  • A single w& optical fiber strain gauge subjected to the excited PZT-plate is presented which was obtained using a Mach-Zehnder interferometer. This paper has been performed the considerations to the experimental situation In which the dynamical behavior of a optical fiber strain gauge is illustrated. A comparison is reported between the dynamic response of a optical fiber strain gauge and the semiconductor strain gauge in the frequency range 5-50Hz. This result is shown in very good usage as the dynamical measurement of the low strain below l.mu..epsilon. by this system.

Development of Shape Optimization System using Stress Control (응력 제어를 통한 형상 최적화 시스템 개발)

  • In this study, the growth-strain method was used for shape optimization. The adequate value of growth ratio in the method was used the value obtained by volume control. And the linear PID control theory was applied to control internal stresses by stresses required by a designer. The effect of the values of $K_{P}$, $K_{I}$, and $K_{D}$ was investigated and the adequate values of $K_{P}$, $K_{I}$, and $K_{D}$ were determined empirically. Finally, a shape optimal design system was built up by the improved the growth-strain method with a commercial software I-DEAS. The effectiveness and practicality of the developed shape optimal design system was verified by some examples.les.les.les.

Simulation and experimental analysis of active vibration control of smart beams under harmonic excitation

  • In the present study, active control of a smart beam under forced vibration is analyzed. The aluminum smart beam is composed of two piezoelectric patches and strain gauge. One of the piezoelectric patches is used as controlling actuator while the other piezoelectric patch is used as vibration generating shaker. The smart beam is harmonically excited by the piezoelectric shaker at its fundamental frequency. The strain gauge is utilized to sense the vibration level. Active vibration reduction under harmonic excitation is achieved using both strain and displacement feedback control. Control actions, the finite element (FE) modeling and analyses are directly carried out by using ANSYS parametric design language (APDL). Experimental applications are performed with LabVIEW. Dynamic behavior at the tip of the beam is evaluated for the uncontrolled and controlled responses. The simulation and experimental results are compared. Good agreement is observed between simulation and experimental results under harmonic excitation.
