INTERPRETATION OF BACTERIAL CONTAMINATION OF ALLOGENEIC TISSUES OBTAINED FROM CADAVERIC AND LIVING DONORS (조직은행에서 채취한 동종조직의 세균 배양 평가)

  • Thorough screening of donors medical and social history, extensive serological and bacterial screening combined with developed processing and sterilization methods have improved the safety of the allogeneic tissues in recent decades. The risk of bacterial infection through allogenic tissue transplantation is one of the major problems facing tissue banks. The purpose study is to report the contamination rate in 358 retrieved tissues obtained strictly aseptic conditions, between 2001 and 2002 in Korea Tissue Bank. Samples from 9 donors(total 13 donors) were used in blood culture, and in 7 donors the blood culture were negative. Of the 358 tissues cultured in their entirety, 186(52%) were initially culture negative and 177(48%) were positive. Organism low pathogenicity were cultures from 20.2% of the tissues. To minimize the bacterial load, donors should be obtain in operating rooms, using aseptic techniques with only a few personnel for procurement. The procurement cultures from donors and retrieved tissues with multiple should be carefully interpreted. Blood cultures should be taken account, since these can help to find contamination not detect swab culture. A prospective cohort study is needed to determine which of the varied processing and sterilization methodologies gives the best quality.

Effects of Chlorine Water and Plasma Gas Treatments on the Quality and Microbial Control of Latuca indica L. Baby Leaf Vegetable during MA Storage (염소수와 플라즈마 가스 처리가 왕고들빼기 어린잎채소의 MA저장 중 품질과 미생물 제어에 미치는 영향)

  • This study was carried out to investigate the effect of chlorine water and plasma gas treatment on the quality and microbial control of Latuca indica L. baby Leaf during storage. Latuca indica L. baby leaves were harvested from a plant height of 10cm. They were sterilized with $100{\mu}L{\cdot}L^{-1}$ chlorine water and plasma-gas (1, 3, and 6hours), and packaged with $1,300cc{\cdot}m^{-2}{\cdot}day^{-1}{\cdot}atm^{-1}$ films and then stored at $8{\pm}1^{\circ}C$ and RH $85{\pm}5%$ for 25days. During storage, the fresh weight loss of all treatments were less than 1.0%, and the carbon dioxide and oxygen concentrations in packages were 6-8% and 16-17%, respectively for all treatments in the final storage day. The concentration of ethylene in the packages fluctuated between $1-3{\mu}L{\cdot}L^{-1}$ during the storage and the highest concentration of ethylene was observed at 6 hours plasma treatment in the final storage day. The off-odor of all treatments were almost odorless, the treatments of chlorine water and 1 hour plasma maintained the marketable visual quality until the end of storage. Chlorophyll content and Hue angle value measured at the final storage day were similar to those measured before storage in chlorine water and 1 hour of plasma treatments. E. coli was not detected immediately after sterilization in all sterilization treatments. After 6 hours of plasma treatment, the total bacteria fungus counts were lower than the domestic microbial standard for agricultural product in all sterilization treatments. The total aerobic counts in the end storage day increased compared to before storage, whereas E. coli was not detected in all sterilization treatments. The sterilization effect against bacteria and fungi was the best in chlorine water treatment. Plasma treatment showed sterilization effects, but within a prolonged period of time. In addition, the sterilization effect decreased gradually. These results suggest that chlorine water and plasma treatment were effective in maintaining Latuca indica L. baby Leaf commerciality and controlling microorganisms during postharvest storage.

Effects of UV Sterilization on Quality of Acer mono Sap (자외선 살균이 고로쇠 수액의 품질에 미치는 영향)

  • An ultraviolet (UV) sterilization system was developed to decrease the number of microorganisms in filtered Acer mono sap (FAS). The Acer mono sap (AS) was passed through 18 strips of PTA fluoroplastic tubing with 30 UV lamps (total 1,170 W). During passage, the AS was effectively exposed to the UV rays. The total bacteria, coliform group and fungi were sterilized at a flow rate ranging from 852 to 1,358 mL/min and a UV power higher than 156 W. Although the sensory score of the UV-sterilized AS was significantly lower than that of the untreated AS, the sensory score was better under the condition of 390 W and 852~1,358 mL/min than under the other conditions examined. ANOVA did not reveal a significant difference in pH, total acidity, sugar content and color characteristic under all conditions tested (p<0.05). Considering the overall attributes of the AS quality, the optimum sterilization condition was determined to be 390 W and 852~1,358 mL/min.

Effect of Microwave Plasma on Sterilization of Acidovorax citrulli Infected Watermelon Seeds (저온플라즈마(Microwave Plasma)를 활용한 Acidovorax citrulli 감염 수박종자의 살균 효과 검정)

  • This study was conducted to check the sterilization efficacy of microwave plasma (MWP) against the watermelon seeds infected with Acidovorax citrulli 11-251. Watermelon seeds were artificially vacuum inoculated to produce A. citrulli 11-251 infected seeds. Aac ImmunoStrip and scanning electron microscope (SEM) results suggests that, seeds (coat and endosperm) were infected under the concentration of 1×107/30 min. MWP sterlization process was carried out at 50 W (3 min, 5 min, and 10 min), 80 W (3 min, 5 min, and 10 min), and 100 W (3 min, 5 min, and 10 min). According to the results, MWP sterilized the artificially inoculated seed coats by 95.96% at 80 W/10 min and seed endosperms by 100% at 100 W/10 min respectively. Although, seeds were sterlized by MWP, germination rate of seeds were low as compared to non treated (negative control) seeds. Moreover, cell membrane of A. citrulli 11-251 was damaged while observed in SEM after sterilized with MWP. Further studies regarding the appropriate sterilization condition by MWP against A. citrulli infected seeds for germination will be conducted in our next study.

Concentration- and Time-Dependent Effect of Disinfectant Treatment on Sorghum Seeds (소독제의 침지시간 및 희석농도가 수수 발아 및 오염율에 미치는 영향)

  • Sorghum (Sorghum bicolor L.) is an annual crop belonging to Poaceae, and is the fifth-largest crop after maize, wheat, rice, and barley. This study was conducted to establish an efficient seed sterilization method to manage fungal or bacterial infections of germinating sorghum seeds. Two varieties of sorghum seeds (BTx623 and SAP317) were treated with benomyl-thiram and thiophanate-methyl triflumizole which are known to be effective disinfectants for sorghum seeds. For SAP317, the highest germination rate was accomplished with 24-hour treatment of both chemicals at a 200× dilution rate. For BTx623, the highest germination rate was observed after 24-hour treatment at a 200×/400× dilution rate for benomyl-thiram and control/200× for thiophanate-methyl triflumizole. Consequently, the optimal treatment for the seed disinfection in sorghum seeds may be at the dilution rate of 200× or 400× for 24 hours.

Disinfection of harmful organisms for sea water using electrolytic treatment system (전해처리를 통한 해수의 유해생물 살균처리)

  • The treated ballast water from previous treatment contains microorganisms and pathogenic organisms in an electrolytic treatment system. The experimental methods included using a peristaltic flow pump placed upward on an electrode pole. Due to the reaction time, the hydraulic retention time indicated unlike microorganisms on the flow rate. In electrolysis, dioxide iridium-coated titanium (Ti/Ir02) and stainless steel plates were used for the anode and cathode, respectively. Current density controls make use of a DC power supply on 250V, 100Amper. Experimental use of a current density between 0.1 and 1.0A/dm2 was able to disinfect the microorganism (E. coli, Bacteria, Bacillus sp.) in seawater for 5 seconds of reaction time. The removal rate was approximately $90\%,$ while the current density was 2.0A/dm2 and the electrode distance was 75mm. This study shows that the electrolytic treatment system has a potential for the sterilization of ballast water.

Comparison of Ventral Midline and Right Flank Approaches of Ovariohysterectomy in Bitches

  • The ventral midline approach (VMA) and right flank approach (RFA) are common procedures for the sterilization of bitches. This study compared the different parameters viz. total duration of surgery, recovery time, and length of the incision as well as body temperature, heart rate, respiration rate, and SpO2 in each approach. Twenty (20) bitches were divided randomly for the RFA and VMA. Meloxicam (0.2 mg/kg) was administered subcutaneously half an hour before the induction to provide preemptive analgesia. Diazepam and ketamine were administered intravenously at dose rates of 0.25 mg/kg and 2.5 mg/kg, and 0.17 mg/kg and 3.33 mg/kg, respectively to produce and maintain anesthesia. Each parameter was recorded at the pre-operative, operative and post-operative times. The average duration of surgery and length of incision of RFA (16.1 ± 5.13 min and 2.44 ± 0.83 cm) were significantly lower (p < 0.05) than the VMA (21.3 ± 5.48 min and 3.53 ± 0.7 cm). The operated bitches showed hypothermia (p < 0.05) at 1 hour compared to baseline and 24 hours of surgery. Heart and respiration rates increased significantly (p < 0.05) during traction and severing of ovarian ligaments in bitches within the RFA group, but there was no significant difference within VMA approaches. The sedation score was significantly higher (p < 0.05) at 1 hour after surgery in both approaches. Based on the duration of surgery and length of incision RFA approach was quick and minimal skin wound. Further studies on bitches considering molecular investigations of surgical stress are imperative.

Characteristics of Electrolyzed Water Manufactured from Various Electrolytic Diaphragm and Electrolyte (전기분해 격막 및 전해액에 따른 전해수의 특성 비교)

  • This study was carried out to investigate the efficacy of surface sterilization and the physicochemical properties of electrolyzed water manufactured from various electrolytic diaphragm and electrolyte. Physical properties of electrolyzed water manufactured from the diaphragm system were the most effective at the following conditions; the distance between diaphragms was 1.0 mm and the supplying rate of 20% NaCl was 6 mL/min. The ORP, HClO content and pH at above conditions were 1,170 mV, 100 ppm and 2.5, respectively. And two stage electrolyzed system was more effective than one stage electrolyzed water manufactured from non-diaphragm system, with 4 mL/min supplying rate of 20% NaCl, was similar to the most effective diaphragm system. But ORP, HClO content and pH were abut 800 mV, 200 ppm and 9 level, respectively. Sealed electrolyzed water could be preserved more than one month at room temperature with ORP of 750 mV in non-diaphragm system and 1,150 mV in diaphragm system, and HClO content of 100 ppm. Twelve kinds of microorganism tested were sterilized within 30 seconds from initial total counts of 10$\^$5/∼10$\^$6/cfu/ml by electrolyzed water. Though the ORP and HCIO contents of electrolyzed water manufactured from various electrolyte were higher in order of NaCl>KCl>CaCl$_2$, there were no difference between the electyolytes in the efficacy of sterilization. Salmonella typhi and Vibrio Proteolyticus were also sterilized by electrolyzed water manufactured from elctrolytes of NaCl CaC1$_2$and KCl

A Study on the Ralstonia Solanacearum Inactivation using Improved Plasma Process (개선된 플라즈마 공정을 이용한 Ralstonia Solanacearum 불활성화에 관한 연구)

  • Effect of improvement of the dielectric barrier discharge (DBD) plasma system on the inactivation performance of bacteria were investigated. The improvement of plasma reactor was performed by combination with the basic plasma reactor and UV process or combination with the basic plasma reactor and circulation system which was equipped with gas-liquid mixer. Experimental results showed that tailing effect was appeared after the exponential decrease in basic plasma reactor. There was no enhancement effect on the Ralstonia Solanacearum inactivation with combination of basic plasma process and UV process. The application of gas-liquid mixing device on the basic plasma reactor reduced inactivation time and led to complete sterilization. The effect existence of gas-liquid mixing device, voltage, air flow rate (1 ~ 5 L/min), water circulation rate (2.8 ~ 9.4 L/min) in gas-liquid mixing plasma, plasma voltage and UV power of gas-liquid mixing plasma+UV process were evaluated. The optimum air flow rate, water circulation rate, voltage of gas-liquid mixing system were 3 L/min, 3.5 L/min and 60 V, respectively. There was no enhancement effect on the Ralstonia Solanacearum inactivation with combination of gas-liquid mixing plasma and UV process.

Optimization of in vitro seed germination of Taraxacum platycarpum

  • Dandelion (Taraxacum platycarpum) has been widely utilized for medicinal purposes. However, the dandelion seeds are relatively difficult to germinate under cultivation conditions, which hampers seedling propagation of dandelion plants and reduces the opportunity of usage of such a useful medicinal plant. Thus, in this study, in vitro conditions for the dandelion seed germination were optimized to enhance the germination rate. In seed washing steps, the sequential treatments with 20% of ethanol, 20% of NaOCl, and distilled water avoided microbial contamination with the highest in vitro germination rate (67.5%) from seeds sown in germination media. The media supplemented with 1.4 g/L of MS salts and 1% of sucrose significantly enhanced the germination rate compared to the media with 4.4 g/L of MS and 3% of sucrose. Sowing the seeds vertically in the optimized media supplement conditions, 1.4 g/L of MS salts and 1% of sucrose, gave the maximum in vitro germination rate (61%), which was almost three times higher than sowing seeds on a soil pot (23%). Our results indicate that the seed washing and sowing methods including germination medium supplements can be optimized to enhance in vitro seed germination of dandelion.
