Association Study Analysis of Cluster-of-Differentiation Antigen 9 (CD9) Gene Polymorphism (g.358A>T) for Duroc Boar Post-thawed Semen Motility and Kinematic Characteristics

  • Cryopreservation of boar semen is continually researched in reproductive technologies and genetic resource banking in breed conservation. For evaluating the boar semen quality, sperm motility (MOT) is an important parameter because the movement of spermatozoa indicates active metabolism, membrane integrity and fertilizing capacity. Various researches have been trying to improve the quality of semen post-thawed in boar. Recently, polymorphism (g.358A>T) of cluster-of-differentiation antigen 9 (CD9) gene reported to be significant association with MOT. Also, CD9 gene was expressed in the male germ line stem cells is crucial for sperm-egg fusion, and was therefore selected as candidate gene for boar semen. This study was conducted to evaluate the pig SNP (g.358A>T) of CD9 gene as a positional controlling for semen parameters of post-thawed boar semen. To results, the g.358A>T SNP of the CD9 gene was significantly associated with the traits such as MOT, curve linear velocity, straight line velocity, average path velocity and amplitude of lateral head displacement. Particularly, the g.358A>T SNP significantly has the highest association with MOT and animals with AA genotype (p<0.001). Therefore, we suggest that the g.358A>T in the intron 6 region of the porcine CD9 may be used as a molecular marker for Duroc boar Post-thawed semen quality, although its functional effect was not defined yet.

A Study on Characteristics of Crassulacean Acid Metablism in Leaves and Stems of Portulaca oleracea L. (쇠비름(Portulaca oleracea L.)에 있어서 잎과 줄기의 돌나물 과형 유기산대사(CAM)의 비교)

  • Diurnal acid fluctuations, stomatal resistance, and morphology and anatomy were investigated in leaves and stems P. oleracea L. growing under the natural environmental condition. A CAM-like pattern of acid fluctuation was exhibited not in leaves of the young purslane but in its stems. Defoliated stems showed a decreased in diurnal changes of total acidity as compared with normal stems. Excised stems stored in continuous darkness exhibited diurnal acid rhythms, and they showed light deacidifications for three days. Kranz-type arrangement was observed in leaves, but not in stems. Micrography of cross sections of stems showed cells with relatively large vaculoles and a few chloroplasts. The number of stomata was 3,275cm-2 in leaves, while the stomata could not be observed in stems. Stomatal resistance was high at night and low in daytime in leaves of the young purslane, and the range of its value was 5~40 sec.$\textrm{cm}^{-1}$. But stomatal resistance in leaves of the water-stressed plant was comparratively high in day time, and its value was 30 sec.$\textrm{cm}^{-1}$. The result of these studies showed the possibility that the stem of P.oleracea L. possesses CAM under certain stressed conditions.

Nutritional Properties and Biological Activities of Artemisia annua L. (개똥쑥의 영양적 특성 및 생리활성)

  • The nutritional properties and biological activities of leaves and stems of Gaeddongssuk (Artemisia annua L.) were investigated. Contents of moisture, crude lipid and crude protein were significantly higher in the leaves, and then ash, crude fiber and mineral were significantly higher in the stems. Contents of total phenols and flavonoids of leaves were about 2 fold higher than those of stems. Antioxidant activity was significantly increased in a dose-dependent manner; also, water and ethanol extracts of leaves were stronger than those of stems. Especially, DPPH radical scavenging activity, reducing power and tyrosinase inhibition activity were significantly higher in leaves extracts than stems extracts of Gaeddongssuk. But, ${\alpha}$-glucosidase inhibition activity was higher in stem than its leaves extract. In MTT assay by human breast adenocarcinoma cell line MCF-7 and MDA-MB-231, ethanol extracts of leaves showed the highest anticancer activity; the rates of growth inhibition were 76.26% and 52.59% on MCF-7 and MDA-MB-231 cells, at the concentration of $250\;{\mu}g$/mL, respectively. In conclusion, biological activities of extracts from Gaeddongssuk were dependent on the fiber, phenolic and flavonoid content.

Germinability, Morphological Development during Germination and Anatomical Observation Gentiana scabra Bunge var. buergeri Max. (자생 용담의 발아성 및 발아과정 중의 형태 형성과 조직의 해부학적 관찰)

  • Gentiana scabra Bunge var buergeri Max, can be widely used such as cut-flower, ornamental and medicinal plant among the native plants. Microscopic and Scanning electron microscopic observation plant were carried out to estimates the characteristics correlated with germinability and germination process, and the results were as follows. The seeds were germinated just after harvest, and it means that there is no or little dormancy of the seeds. Matured seeds showed higher germinability than those of immatured. As the storage period was longer, immatured seeds showed shorter longevity than those of matured. The average longevity was about 190 days. According to the observation results of morphological changes during the germination, seed coat was expanded with water absorption immediately after seeding, and radicals were observed 2 days after seeding. Root hairs were formed behind the growing tip of the root on 4 days after seeding and cotyledon were unfolded on 6 days after seeding. In the microscopic observation of leaf tissues, it could be easily distinguished between midrib, and leaf blade, and also observed upper epidermis, palisade parenchyma and spongy parenchyma consisted as a sing1e layer. Palisade parenchyma was consisted of a single layer of cell. Stomata were restricted lower surface, arranged as anamocytic type, and surrounded by guard cells. In the microscopic anatomy of stem were consisted of cortical layer, forming a single layer of epidermis and parenchyma, and vascular bundle which was consisted of continuous cylinder of vascular tissues.

Effects of Red Ginseng Extracts on Hydrogen Peroxide Production of Murine Prtitoneal Macrophages (홍삼추출물이 마우스 복강 대식세포 Hydrogen Peroxide 생산에 미치는 영향)

  • This experiment has conducted to evaluate whether single injection of red ginseng extract including 50% ethanol extract, crude saponin, and lipid soluble fraction can induce oxidative burst of mouse peritoneal macrophages with use of fluorescence spectrophotometer. To optimize conditions of fluorescent spectrophotometry, concentrations of DCFH-DA(2', 7' -dichlorofluorescin diacetate) was 1.6 ${\mu}{\textrm}{m}$ and control oxidative burst by Zymosan A and PMA(phorbol myristate acetate) were 100$\mu\textrm{g}$, 250ng, respectively. Though in vitro macrophages failed to induce increment of H2O2 production, but 50% ethanol extract group induced significant enhancement of H2O2 production when zymosan A triggered oxidative burst. On the other hand, lipid soluble fraction enhanced significantly H2O2 production than that of control group. These findings consisted with the other reports which showed ginsenosides inhibited nitric oxide production and lipid soluble fraction activated colony stimulating factor(granulocyte - monocyte) activity in bone marrow stem cells. As is well known, lipid soluble fraction contains phenol compound, polyacetylene compound and alkaloids. Further study would unravel which component of it can induce H2O2 production of macrophages.   • Objectives: Esthesioneuroblastoma is a rare malignant neoplasm that originates from the olfactory sensory cells. This tumor grows from the upper nasal cavity and ethmoid sinus and invades surrounding structures through the cribriform plate into intracranium or orbit in advanced stage. Even though there has been some controversies in determining standard treatment due to rarity of this tumor, the combination treatment of surgery and adjuvant radiation has been recommended for the locally advanced esthesioneuroblastomas. However, the recent clinical experiences of advanced cases showed that combination chemotherapy is highly effective to reduce tumor mass and improve clinical outcomes. Materials and Methods: The authors conducted a retrospective analysis of 6 esthesioneuroblastoma patients who were treated in our hospital from 1986. Results: The age of these patients was between 19 and 86 year-old. Among the 6 cases, 2 were diagnosed at stage B and 4 at stage C, according to Kadish classification. Anti-tumor treatments were performed in 5 patients. One patient refused active treatment and was lost to follow-up. Better survival outcome were observed in 3 patients who were treated with combination chemotherapy alone or combined modality treatment including chemotherapy. Conclusion: Based on our retrospective study, the combined treatment consisting of surgery, radiotherapy, and combination chemotherapy should be used to improve treatment results. And furthermore, innovative clinical approaches such as neoadjuvant chemotherapy, high-dose chemotherapy and autologous peripheral stem cell transplantation, which have been reported to have good therapeutic results, should be considered and applied actively.

The Effect of Antibacteria and Antioxidantion Activities from Needle Leaf Tree (침엽수 추출물의 항균.항산화 효과)

  • We studied anti-microbial and anti-oxidant activities of bio-active components in stem of Juniperus chinensis L. and leaf of Abies koreana Wilson. Those plants of needle leaf tree family were reported to contain anti-cancer compounds. The anti-bacterial activity was tested by Broth dilution method against Escherichia coli and Staphylococcus aureus. As results, Juniperus chinensis L. and Abies koreana Wilson extracts showed 17.0% and 8.5% higher anti-bacterial activity than methyl paraben, respectively. The free radical scavenging activity of Juniperus chinensis L. and Abies koreana Wilson extracts showed 45 % and 44 % at 5,000 ppm. We measured polyphenol (catechin equivalent) and flavonoids quantity. The Juniperus chinensis L. extract contained 312 mg/g of polyphenol and 105 mg/g of flavonoids. The Abies koreana Wilson extract contained 280 mg/g of polyphenol and 103.8 mg/g of flavonoids. The cytotoxicity of extracts was measured by neutral red assay. Extracts did not affect the viability of CCK-986sk cells up to a concentration of 1,250 ppm. In conclusion, these data suggest that extracts of needle leaf trees would be usefull as antiseptic agents and anti-oxidants for cosmetic products.

Association of KIR (Killer Cell Immunoglobulin-like Receptor) Genotype with Psoriasis in Korean Population (한국인에서 건선과 KIR (Killer Cell Immunoglobulin-like Receptor) 유전자형 사이의 연관성)

  • Background: Psoriasis is a multifactorial autoimmune skin disease with a pathogenesis that has remained obscure. Recently, T cells bearing natural killer receptors (NKRs) were precisely and strongly targeted as new putative pathogenic immunocytes in psoriasis. Among NKRs, killer cell immunoglobulin-like receptor (KIR) is the major molecule recognizing HLA class I allotypes and might be closely related to psoriasis. Methods: To investigate the association of KIR genotype and patients with psoriasis in Korean, we defined the 14 KIR genotypes in 96 patients with psoriasis and 86 healthy controls using PCR-SSP methods. Results: The frequencies of KIR2DS4 and KIR3DL1 were significantly decreased in psoriasis compared with controls (RR=0.21, p<0.02). When patients were divided into two subgroups at the age of onset, type I (<30 years) and type II ($({\geq}30$ years) respectively, these phenomena were similarly observed independent of groups divided (type I: RR=0.26, p<0.005; type II: RR=0.14, p<0.0006). When the patients were divided into subgroups according to the age of onset and family history, the frequencies of KIR2DS4, KIR3DL1, and KIR2DS3 were significantly decreased in type I compared with type II psoriasis (3DL1, 2DS4: p<0.004; 2DS3: p<0.04) and were significantly decreased in psoriasis without family history compared to with family history (3DL1, 2DS4: p<0.007; 2DS3: p<0.05). The frequency of haplotype combination BB was significantly increased in psoriasis compared with controls (RR=2.74, p<0.009). Conclusion: These results suggest that KIR genotype is a factor for the occurrence and development of psoriasis and in future how combinations of HLA and KIR genes influence psoriasis needs to be defined.

Hwang Woo-Suk, Pasteur and ANT (황우석과 파스퇴르 그리고 ANT)

  • Could STS throw another-colored light on the Hwang's Affair, the scientific fraud committed by Hwang Woo-Suk and his research team in Korea? And could analytic tools of STS unfold another meanings which have been overlooked in most of the traditionally social-sciences-oriented analyses? In this essay, I try to answer these questions by analyzing the Hwang's Affair in the view of STS, especially by using some concepts of actor-network theory(ANT): movement, translation and displacement. I want to say that the Hwang's Affair seems to be a part of normal scientific activity, not an abnormal phenomenon, and as an evidence, focus on the similarities of their life styles between "pure/real scientist" Louis Pasteur and "impure/political scientist" Hwang Woo-Suk. I try to mobilize some concepts of ANT, especially movement, and find out why scientists came to move toward the opposed direction on the pure/real-impure/political line. I suggest that there exists "laboratory politics" as the key factor in this bifurcation. My tentative conclusion is that Pasteur can take a position to make his great world, so-called the Pasteurian world, owing to the success of "double movement" in which he treated his laboratory as a fulcrum to lift up the world, but Hwang degrades himself to "ugly scientific politician" due to the loss of the momentum of his movement; Hwang treated his laboratory only as the symbolic resources and in turn failed to solidify material entities, his real political resources, even though he knew the importance of laboratory.

Silk Fibroin and Substance P Combination Graft for the Reconstruction of a Bone Defect (실크 피브로인 지지체와 Substance P를 이용한 골 이식재)

  • Purpose: Substance P is a well known neurotransmitter and has been known to mediate pain. Recently, it has been unveiled that substance P is involved in the recruitment of mesenchymal stem cells to wound sites. The purpose of this study was to exam bone formation when a combination of substance P and silk fibroin was used in a bone defect model. Methods: Twenty rabbits were used and 40 calvarial defects were formed. They were divided as 4 groups (unfilled control, silk only, silk+$10{\mu}g$/ml substance P; Sub10, and silk+$100{\mu}g$/ml substance P; Sub100). All animals were humanely sacrificed 4 or 8 weeks after grafting. The specimens were analyzed by micro-computerized tomography and histological analysis. Results: When compared to the unfilled control to silk only group, there was significant difference in bone mineral density (BMD) and the attenuation coefficient (AC) at 4 weeks ($p$=0.037 and 0.038, respectively). When compared Sub10 group to Sub100 group, there was significant difference in BMD and AC at 8 weeks ($p$=0.004 for all). Residual graft amounts were $52.1{\pm}15.8$%, $15.2{\pm}9.2$% and $9.0{\pm}3.3$% for silk only, Sub10, and Sub100 groups, respectively. When comparing the residual graft amount of silk only to sub10 or sub100, the differences were statistically significant ($p$ <0.001). Conclusion: The silk fibroin scaffold showed higher BMD and AC than the unfilled control. The combination graft with substance P and silk fibroin scaffold showed a faster graft degradation than with a silk fibroin scaffold only.
