당귀(當歸) 향기액(香氣液)이 Rat의 뇌신경줄기세포의 분화(分化)와 증식(增殖)에 미치는 영향 (The Effect of an Essential Oil Fragrance from Radix Angelica Sinesis on Differentiation and Proliferation of Neural Stem Cells of Rat)

  • Objectives : In this study, an essential oil fragrance from Danggwi was administrated into the neural stem cell and the effect of the essential oil on the differentiation and proliferation of the neural stem cells were observed. Methods : The establishment of the neural stem cell was identified via Nestin, DAPI dye. An essential oil fragrance from Danggwi was administrated with a proved optimum level for the survival of the cell through MTT assay. Also, according to the analysis of Western blot, the essential oil fragrance from Danggwi promotes the phosphorylating of Akt, Erk, ERM protein. Results : MTT assay showed increased in GFAP. The result indicates that the differentiation to astrocyte is promoted. The phosphorylation levels of ERM, Erk and Akt were increased at 60 min after addition of 5 ug/ml of essential oil fragrance from Danggwi and sustained to 48 hours. These imply that essential oil fragrance from Danggwi may induce the survival and the proliferation of the differentiated cells. Conclusions : These results suggest that the essential oil fragrance from Danggwi can be effective for the in vivo study of degenerative neuronal disease using neural stem cell.

  • Mechanical stimulation is known to play a vital role on the differentiation of mesenchymal stem cells (MSCs) to pre-osteoblasts. In this research, we developed a tensile cell stimulator, composed of a DC motor-driven actuator and LVDT sensor for measuring linear displacement, to study the effects of tensile stress on osteogenic differentiation of MSCs. First, we demonstrated the reliability of this device by showing the uniform strain field in the silicon substrate. Secondly, we investigated the effects of tensile stretching on osteogenic differentiation. We imposed a pre-set cyclic strain at a fixed frequency on cell monolayer cultured on a flexible silicon substrate while varying its amplitude and duration. 60 min of resting period was allowed between 30 min of cyclic stretching and this cycle is repeated up to 7 days. Under the combined stimulation with osteogenic media and mechanical stretching, the osteogenic markers such as alkaline phosphatase (ALP), osterix, and osteopontin began to get expressed as early as 4 days of stimulation, which is much shorter than what is typically required for osteogenic media induced differentiation. Moreover, different markers were induced at different magnitudes of the applied strains. Lastly, for the case of ALP, we observed the antagonistic effects of osteogenic media when combined with mechanical stretching.

Role of gangliosides in the differentiation of human mesenchymal-derived stem cells into osteoblasts and neuronal cells

  • Gangliosides are complex glycosphingolipids that are the major component of cytoplasmic cell membranes, and play a role in the control of biological processes. Human mesenchymal stem cells (hMSCs) have received considerable attention as alternative sources of adult stem cells because of their potential to differentiate into multiple cell lineages. In this study, we focus on various functional roles of gangliosides in the differentiation of hMSCs into osteoblasts or neuronal cells. A relationship between gangliosides and epidermal growth factor receptor (EGFR) activation during osteoblastic differentiation of hMSCs was observed, and the gangliosides may play a major role in the regulation of the differentiation. The roles of gangliosides in osteoblast differentiation are dependent on the origin of hMSCs. The reduction of ganglioside biosynthesis inhibited the neuronal differentiation of hMSCs during an early stage of the differentiation process, and the ganglioside expression can be used as a marker for the identification of neuronal differentiation from hMSCs.

Comparison of Expression Profiles of HOX Gene Family in Human Embryonic Stem Cells and Selected Human Fetal Tissues

  • The HOX genes coding homeodomain proteins have been suggested as a candidate molecular switch that determines the fates of cells during embryonic development and patterning. It is believed that a set of differentiation-specific HOX genes enter into a turn-on state during tissue differentiation, in contrast to stem cell-specific HOX genes that enter into a turn-off state. However, comprehensive data of expression profiles of HOX genes in human embryonic stem cells (hESC) and differentiated embryonic tissues are not available. In this study, we investigated the expression patterns of all 39 HOX genes in hESC and human fetal tissues and analyzed the relationships between hESC and each tissue. Of the 39 genes, 18 HOX genes were expressed in stem cells, and diverse expression patterning was observed in human fetal tissues when compared with stem cells. These results indicate that HOX genes could be main targets for switching of stem cell differentiation into tissues.

Toxicity and Biomedical Imaging of Fluorescence-Conjugated Nanoparticles in Hematopoietic Progenitor Cells

  • Cellular uptake of nanoparticles for stem cell labeling and tracking is a critical technique for biomedical therapeutic applications. However, current techniques suffer from low intracellular labeling efficiency and cytotoxic effects, which has led to great interest in the development of a new labeling strategy. Using silica-coated nanoparticles conjugated with rhodamine B isothiocyanate (RITC) (SR), we tested the cellular uptake efficiency, biocompatibility, proliferation or differentiation ability with murine bone marrow derived hematopoietic stem/progenitor cells. The bone marrow hematopoietic cells showed efficient uptake with SR with dose or time dependent manner and also provided a higher uptake on hematopoietic stem/progenitor cells. Biocompatibility tests revealed that the SR had no deleterious effects on cell cytotoxicity, proliferation, or multi-differentiation capacities in vitro and in vivo. SR nanoparticles are advantageous over traditional labeling techniques as they possess a high level of cellular internalization without limiting the biofunctionality of the cells. Therefore, SR provides a useful alternative for gene or drug delivery into hematopoietic stem/progenitor cells for basic research and clinical applications.

Biology of Glioma Cancer Stem Cells

  • Gliomas, much like other cancers, are composed of a heterogeneous mix of neoplastic and non-neoplastic cells that include both native and recruited cells. There is extensive diversity among the tumor cells, with differing capacity for In vitro and in vivo growth, a property intimately linked to the cell's differentiation status. Those cells that are undifferentiated, self-renewing, with the capacity for developing tumors (tumorigenic) cells are designated by some as cancer stem cells, because of the stem-like properties. These cells may be a critical therapeutic target. However the exact identity and cell(s) of origin of the socalled glioma cancer stem cell remain elusive. Here we review the current understanding of glioma cancer stem cell biology.

심근세포로의 분화에 관여하는 새로운 생리활성 단백질 SPP2의 발굴 (Identification and Characterization of Secreted Phosphoprotein 2 as a Novel Bioactive Protein for Myocardial Differentiation)

  • 심장 발생과정에 관여하는 주요 전사인자들의 기능에 대한 규명 등의 발전에도 불구하고 줄기 세포에서 매우 효율적인 심근 세포로의 분화를 촉진하는 새로운 생체 활성 분자를 찾는 것이 여전히 필요하다. 마우스배아줄기세포(mESC) 유래 심근세포의 Illumina 발현 마이크로어레이 데이터를 분석하였다. 미분화 mESCs와 비교하여 mESC 유래 심근세포에서 4배 이상 유전자 발현이 증가한 276개 유전자가 스크리닝되었다. Secreted phosphoprotein 2 (Spp2)는 후보물질 중 하나이며 bone morphogenetic protein 2 (BMP2)에 대한 슈도수용체로서 BMP2 신호 전달을 억제하는 것으로 알려져 있다. 그러나 심근 형성과의 연관성은 알려진 바 없다. 우리는 mESC 세포주인 TC-1/Kh2와 E14를 이용하여 기능성 심근세포로 분화하는 동안 Spp2 발현이 증가함을 검증하였다. 흥미롭게도, Spp2 분비는 배아체(embryoid body, EBs) 형성 후 3일차에 일시적으로 증가했는데, 이는 Spp2의 분비가 ESCs의 심근세포로의 분화에 관여함을 시사한다. Spp2의 기능을 분석하기 위해, 우리는 BMP2를 처리하면 분화 경로를 근모세포에서 골모세포로 전환되는 특성을 가진 C2C12 마우스 근모세포 세포주를 사용하여 실험을 수행하였다. mESCs의 분화와 유사하게, Spp2의 전사는 C2C12 근모세포가 근관으로 분화됨에 따라 증가하였다. 특히, 분화 초기 단계에서 Spp2의 세포외 분비가 극적으로 증가하였다. 또한, Spp2-Flag 재조합 단백질로 처리하면 C2C12 근모세포의 근관으로의 분화가 촉진되었다. 종합하면, ESCs를 심근 세포로 분화시키는 새로운 생체 활성 단백질로 Spp2를 제안한다. 이것은 심근형성의 분자 경로를 이해하고 허혈성 심장질환에 대한 줄기세포 요법의 실험적 또는 임상적 발전을 촉진하는 역할을 할 것으로 기대한다.
