한국약용식물의 최종당화산물 생성저해활성 검색 (II) (Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (II))

  • Advanced glycation end products (AGEs) contribute to the progression of micro and macrovsacular complication of diabetes and therefore present a promising target for therapeutic agents. In this study, 40 Korean herbal medicines have been investigated with an in vitro evaluation system using AGEs inhibitory activity. Of these, 21 herbal medicines $(IC_{50}<50{\mu}g/ml)$ exhibited an inhibitory activity against AGEs formation compared with anminoguanidine $(IC_{50}=72.12{\mu}g/ml)$. Particularly, 7 herbal medicines, Actinidia arguta (root and stem), Crataegus pinnatifida (twig), Camellia japonica (whole), Kalopanax pictus (bark), Lagerstroemia indica (leaf-stem), Reynoutria sachalinensis (root) showed more potent inhibitory activity (approximately 3-10 fold) than the positive control aminoguanidine.

금강형 소나무림에 있어서 토양의 이화학적 성질 (Physicochemical Properties of Soil in Pine (Pinus densiflora for. erecta Uyeki) Forests)

  • Uyeki(1928) classified Pinus densiflora into six ecotypes(Northeastem type, Middle-southern flat type, middle-southern upland type, Wibong type, Ankang type, and Geumgang type) based on the pine tree type. The bark color of Geumgang type was ash-brown color on the lower parts of stem and yellowed color on the upper parts of stem. We investigated the physicochemical properties of soil forests to obtain basic data for preservation of exellent pine (Pinus densiflora for. erecta Uyeki). The results were as follows; The soil texture of the Pinus densiflora for. erecta Uyeki forests were showed nearly as sandy loam, that is, sand, silt and clay were consisted of 72%, 15% and 13%, respectively. Soil acidity(pH 4.6) was lower than Korea average forest soil acidity(pH 5.2). The average contents of available phosphate was 11.7ppm at Sokwang-ri, 26.8ppm at Mt. Eungbong, 24.2ppm at Mt. Kumma. It was the lowest at Uljin(4.6ppm). The contents of carbon was 6.2% at Mt. Chungok, 6.1% at Mt. Eungbong. This value was more than average of Korea forest soil.

붉나무 수피의 성분 (Chemical Components from the Stem Bark of Rhus javanica L.)

  • Seven compounds were isolated from the n-BuOH extract of the dried stem barks of Rhus javanica L. On the basis of physico-chemical, spectroscopic evidences and comparison with authentic samples, the compounds $1{\sim}7$ were identified as gallic acid (1), methyl gallate (2), scopoletin (3), scopolin (4), $1,\;2,\;3,\;4,\;6-penta-O-galloyl-{\beta}-D-glucose$ (5), orcinol (6) and $orcinol-{\beta}-D-glucoside$ (7). Among these compounds, scopolin, $orcinol-{\beta}-D-glucoside$ were isolated from this plant for the first time.

Occurrence of Root Rot caused by Fusarium fujikuroi on Adzuki Bean in Korea

  • In July 2020, wilting symptoms were observed among adzuki bean plants (Vigna angularis var. angularis L.) in the fields in Yeosu, Korea. Infected plants showed yellowing of leaves, browning inside the stems, splitting of stem bark, and wilting. When these plants were uprooted, their roots were found to be brown. The fungal pathogens NC20-737, NC20-738, and NC20-739 were isolated from symptomatic stem and root tissues. These pathogens were identified as a Fusarium fujikuroi species complex based on their morphological characteristics. Molecular identification was performed using the DNA sequence of translation elongation factor 1 alpha and the RNA polymerase II second largest subunit regions. The nucleotide sequences of all three isolates were similar to the F. fujikuroi reference isolates NRRL 13566 and NRRL 5538 of the National Centre for Biotechnology Information GenBank. A pathogenicity test was conducted by the soil inoculation method with cornmeal sand inoculum. Approximately 3 weeks after inoculation, symptoms were observed only in the inoculated adzuki bean seedlings. To the best of our knowledge, this is the first report of Fusarium root rot caused by F. fujikuroi in adzuki beans, both in Korea and worldwide.

당느릅나무로부터 Sesquiterpene o-Naphthoquinone류 화합물, Mansonone E, F 및 H의 분리와 구조결정 (Isolation and Identification of Sesquiterpene o-Naphthoquinones, Mansonones E, F and H, from the Root Bark of Ulmus davidiana Planch)

  • 한방에서 종기, 치질, 위암 및 염증의 치료목적으로 사용되어온 당느릅나무 (Ulmus davidiana Planch) 근피의 메탄을 추출물로부터 sesquiterpene o-naphthoquinone류 화합물 세가지를 분리 정제하였다. 이들의 구조를 UV-vis, IR, EIMS, HR-EIMS 및 여러 가지 NMR 스펙트럼, 특히 새로운 기법인 pulse field gradient (PFG)-HMQC 및 HMBC 스펙트럼의 분석에 의하여 결정하였다. 그 결과 이들은 2,3-dihydro-3,6,9-trimethylnaphtho(1,8-b,c)pyran-7,8-dione, 3,6,9-trimethylnaphtho(1,8-b,c)pyran-7,8-dione 및 2,3-dihydro-4-hydroxy-3,6,9-trimethylnaphtho(1,8-b,c)pyran-7,8-dione으로 동정 되었으며, 각각 mansonone E, F 및 H로 밝혀졌다. 이 화합물들은 당느릅나무에서는 처음 분리되었으며, 특히 mansonone H는 아프리카 열대식물인 Mansonia altissima Chev 및 Helicters angustifolia에서 분리된 후 자연계에서 처음으로 분리되었다. 문헌에 잘못 보고된 mansonone E 와 F의 C-3a, C-6a 및 C-9b의 세 사급탄소의 chemical shift를 최신 PFG-NMR기법에 의하여 동정하여 교정하였다.

Assessment of anatomical characteristics of the medicinal plant African cherry (Prunus africana) for its accurate taxonomic identification

  • The genus Prunus (family: Rosaceae) consists of over 400 plant species and exhibits vast biodiversity worldwide. Given the wide distribution of this genus, its taxonomic classification is important. Anatomical characteristics are conserved and stable and can therefore be used as an important tool for the taxonomic characterization of plants. Therefore, this study aimed to assess and document the anatomical characteristics of the leaf, stem, and seed of P. africana using micrographs and photographs for possible use in the identification, quality control, and phylogenetic analysis of the species. The anatomical sections of a young stem revealed a cortex consisting of isodiametric parenchyma cells, druse crystals, primary vascular bundles, and pith. The mature stem bark majorly consisted of the rhytidome, with the periderm densely arranged in multiple layers; a cluster of stone cells; and sclerenchyma. The leaf sections were hypostomatic, with stomata sizes ranging from 18.90-(22.34)-26.90 × 15.41-(18.40)-21.22 ㎛. The leaf sections showed the presence of characteristic druse crystals, vascular bundles, and mesophyll layers. The pericarp contained the epicarp, mesocarp, and endocarp, with their thickness being approximately 350-400, 300-350, and 30-50 ㎛, respectively. In addition, it contained a seed testa with a thickness of approximately 50-60 ㎛. The morphological and anatomical characteristics observed in P. africana leaves, stems, and seeds in this study could serve as useful data for the taxonomic identification of this species.

Antifungal Activity of Magnolol and Honokiol

  • Two neolignan compound, magnolol $(5,5^{l}-diallyl-2,2^{l}-dihydroxybiphenyl, 1)$ and honokiol $(5,5^{l}-diallyl-2,4^{l}-dihydroxybiphenyl, 2)$ were isolated from the stem bark of Magnolia obovata and evaluated for antifungal activity against various human pathogenic fungi. Compound 1 and 2 showed significant inhibitory activities against Trichophyton mentagrophytes, Microsporium gypseum, Epidermophyton floccosum, Aspergillus niger, Cryptococcus neoformans, and Candida albicans with minimum inhibitory concentrations (MIC) in a range of $25-100{\mu}g/ml$. Therefore, compound 1 and 2 could be used as lead compounds for the development of novel antifungal agents.

Structure-Activity Relationship of Xanthones from Mesua daphnifolia and Garcinia nitida towards Human Estrogen Receptor Nagative Breast Cancer Cell Line

  • Extensive chemical studies on the stem bark extracts of two Guttifereous plants namely Mesua daphnifolia and Garcinia nitida have led to the isolation of eight xanthones. Mesua daphnifolia gave cudraxanthone G (1), ananixanthone (2), 1,3,5-trihydroxy-4-methoxyxanthone (3) and euxanthone (4) while Garcinia nitida gave inophyllin B (5), 1,3,7-trihydroxy-2,4-bis (3-methylbut-2-enyl)xanthone (6), 3-isomangostin (7) and rubraxanthone (8). All these compounds were assayed against the MDA-MB-231 (human estrogen receptor negative breast cancer) cells. A structure-activity relationship study showed that structurally, all the 1, 3-oxygenated xanthones which carried unsaturated prenyl side chains (either 3-methylbut-2-enyl or 1,1-dimethyl-2-propenyl) at carbones C-2 and C-4 in the xanthone ring A are essential for the outstanding activities in the assay.
