Comparative Analysis of Host Insect Immunodepression Induced by Two Entomopathogenic Bacteria, Xenorhabdus nematophilus and Staphylococcus gallinarum, with Differential Pathogenicities (병원력 차이를 보이는 두 곤충병원세균(Xenorhabdus nematophilus와 Staphylococcus gallinarum)의 면역저하 능력 비교 분석)

  • Immunodepression can be required for entomopathogenic bacteria to induce their potent pathogenicities to the target insects. Here, we raise a hypothesis that the capacity of a pathogenic bacterium to induce the target insect immunodepression has positive relationship with the degree of pathogenicity. X. nematophilus had 1,200 times as potent as another entomopathogenic bacterium, Staphylococcus gallinarum against the fifth instar larvae of silkworm, Bombyx mori, when they were Injected into the hemocoel. Although both bacteria had significant cytotokic effect on the hemocytes of B. mori, X. nematophilus gave faster and greater cytotoxicity than did S. gallinarum. In cellular immune reactions, B. mori could form 20 hemocyte nodules against the bacterial injection with 5${\times}$10$\^$5/ cells. The number of the hemocyte nodules was significantly depressed when live X. nematophilus was inject-ed, but not in S. gallinarum. Activation of prophenoloxidase (proPO) was depressed in the bacterial injection. The depression of PO activation was significantly greater in X. nematophilus infection than in S. gallinarum injection. Lysozyme activity was induced by the injection of S. gallinarum at 4 h after the treatment, but not induced in X. nematophilus at all the time. These results showed that X. nemato-philus induced greater immunodepression against B. mori and resulted in higher pathogenicity than did S. gallinarum. Therefore, this study suggests that the immunodepression induced by entomopathogenic bacteria has positive relationship with their pathogenicity.

Antimicrobial Activity of Medicinal Herbs against Staphylococcus aureus and Salmonella gallinarum. (Staphylococcus aureus와 Salmonella gallinarum에 대한 한약재의 항균활성)

  • The extracts from approximately 40 different Korean traditional medicines were prepared to investigate the antimicrobial activities against poultry disease-related bacteria. Among tested, the extracts of Schizandra chinensis (SC), Melia azedarach (MA), Caesalpinia sappan (CS) and Rhus javanica (RJ) exhibited significant antimicrobial activities against Salmonella gallinarum, whereas the extracts of Elsholtzia ciliata (EC), Myristica fragrans (MF), Alpinia katsumadai (AK), Poncirus trifoliata (PT), Prunella vulgaris (PV), CS and RJ exhibited antimicrobial activities against Staphylococcus aureus. Minimum inhibitory concentrations (MIC) of MA, CS and RJ extracts against S. gallinarum were 1.2 mg/ml, whereas MIC of RJ extract for S. aureus was 0.6 ㎎/ml, which was the lowest among tested. The antimicrobial activities of SC and RJ extracts against S. gallinarum were reduced, but those of AK and CS extracts against S. aureus were not affected by heating treatment. The antimicrobial activities of SC extract against S. gallinarum and those of EC, PT and RJ extracts against S. aureus were stable by acid treatment but unstable by alkaline treatment. those of CS extract was not effected by either acid or alkaline treatment. The growth of all bacteria was significantly inhibited within 24 hours by the addition of at least 100 ppm and 300 ppm of RJ and CS extracts, respectively, compared with the control group. In conclusion, these findings suggest that RJ and CS extracts may play important roles for antimicrobial activities against poultry disease-related bacteria.

Identification of a Pathogenic Bacterium, Staphylococcus gallinarum, to Bombyx mori (누에(Bombyx mori) 무름병을 야기하는 병원성 세균 Staphylococcus gallinarum의 동정)

  • Flacherie symptom was found in the fifth instar larvae of silkworm, Bombyx mori. The bacterial pathogen was isolated from the hemolymph of the infected silkworm and identified. The isolated bacteria caused a significant flacherie pathogenicity to the fifth instar larvae of B. mori when $5{\times}10^{6}$ cfu (colony-forming unit) of the bacteria was injected into each larva. The infected larvae began to die at 6 days after injection and resulted in complete mortality at 10 days. The bacterium was identified as Staphylococcus gallinarum based on the morphological and physiological characteristics described in Bergey's manual. This identification was further supported by the characters of carbohydrate utility analyzed from a bacterial identification system ($MicroLog^{\circledR}$) and also by the molecular structure of 165-23S rDNA internal transcribed spaces. As an insecticidal action, S. gallinarum caused hemolymph septicemia by its cytotoxic effect on the hemocytes of B. mori.

Screening of Antibacterial Activity Against to Staphylococcus aureus, Listeria monocytogens, Mannhemia haemolytica and Salmonella gallinarum using Different Plant Extracts (다양한 식물들을 이용한 Staphylococcus aureus, Listeria monocytogens, Mannhemia haemolytica 및 Salmonella gallinarum 항균 추출물 탐색)

  • Antibacterial activity is an important feature for the development of antibiotics alternatives. Plant extract is considered as a promising alternative for organic farming. In this study, a total of 11 plants were extracted using ethanol to determine their antibacterial activities against to Staphylococcus aureus, Listeria monocytogens, Mannheimia haemolytica and Salmonella gallinarum. The synergistic interaction among plant extracts was also investigated. Plants used in this study were Carthamus nctoricus L. (pA), Poncirus trifollata Raf. (pB), Scutellaria balcalensis Georgi (pC) Prunus sargentii (pD), Cucurbita moschata $D_{UCH}$ Leaf (pE), Allium cepa L. peel (pF) Portulaca oleracea L. (pG), Xanthium strumarium L. (pH), Duchesnea chrysantha (pI), Cudrania tricuspidata (pJ) and Juniperus chinensis L. (pK). The pB and pA had the most broad antibacterial spectrum and the highest activity against to Staph. aureus among plant extract, respectively. In the synergistic interaction, the mixtures of pA and pC as well as pA and pF had batter antibacterial activity against to Staph. Aureus compared with other mixtures.

  • The present study was conducted to investigate the antimicrobial activities of extracts from approximately 40 different traditional Korean medicinal herbs against S. gallinarum and S. epidermidis. The extracts from Schizandra chinensis Baill., Melia azedarach Linn$\acute{e}$, Caesalpinia sappan Linn$\acute{e}$. and Rhus javanica Linn$\acute{e}$. exhibited high antimicrobial activities against S. gallinarum, whereas the extracts from Melia azedarach Linn$\acute{e}$ and Rhus javanica Linn$\acute{e}$. exhibited high antimicrobial growth for S. epidermidis. Minimum inhibitory concentrations (MIC) of Melia azedarach Linn$\acute{e}$, Caesalpinia sappan Linn$\acute{e}$. and Rhus javanica Linn$\acute{e}$. for S. gallinarum were 1.2 mg/mL, whereas MIC of exracts from Rhus javanica Linn$\acute{e}$. extract for S. epidermidis were 0.6 mg/mL. Heat treatment of the extracts from Schizandra chinensis Baill. and Rhus javanica Linn$\acute{e}$. caused a significant reduction in antimicrobial activities against S. gallinarum. but didn't affect antimicrobial activities against S. edidermidis. Alkaline treatment of the extracts from Schizandra chinensis Baill. caused a significant reduction in antimicrobial activities against S. gallinarum, while similar treatment of the extracts from Rhus javanica Linn$\acute{e}$. caused a significant increase in antimicrobial activities against S. edidermidis. Since extracts from Rhus javanica Linn$\acute{e}$. and Caesalpinia sappan Linn$\acute{e}$. exhibited the highest antimicrobial activities, these extracts at the concentrations of 100, 300 or 500 ppm were added and then bacterial growth-inhibiting activities for S. gallinarum and S. epidermidis by these two extracts were further examined. Optical density at 620 nm ($OD_{620}$) after 24 hours incubation in the absence of Rhus javanica Linn$\acute{e}$. extract ranged from 0.30 to 0.45 compared with $OD_{620}$ value ranging from 0.06 to 0.18 in the presence of 100, 300 or 500 ppm of the extract, indicating that growth of all bacteria was significantly inhibited within 24 hours by the addition of at least 100 ppm of Rhus javanica Linn$\acute{e}$ extract. Value of $OD_{620}$ after 24 hours incubation in the absence of Caesalpinia sappan Linn$\acute{e}$. extract ranged from 0.30 to 0.55 compared with $OD_{620}$ value ranging from 0.05 to 0.15 in the presence of 300 or 500 ppm of the extract, indicating that growth of all bacteria was also significantly inhibited within 24 hours by the addition of at least 300 ppm of Caesalpinia sappan Linn$\acute{e}$. extract. In conclusion, these findings suggest that extracts from Rhus javanica Linn$\acute{e}$. and Caesalpinia sappan Linn$\acute{e}$. may play important roles in antimicrobial activities against S. gallinarum and S. epidermidis.

Characteristics of Antimicrobial Organic Acids Produced by lactobacillus pentosus K34 isolated from Small Intestines of Korean Native Chickens (한국토종닭 소장에서 분리한 Lactobacillus pentosus K34가 생산하는 항균성 유기산의 특성)

  • Seven lactic acid bacteria showing highly inhibitory activities against Salmonella gallinarum, Staphylococcus aureus, and Escherichia coli were isolated from Korean native chickens. The inhibitory activities were insensitive to various pretenses indicating that the inhibitory substance is not proteinaceous. The culture broths seem to contain other inhibitory substances in addition to lactic acid. The metabolic profile of organic acids produced by Lactobacillus pentosus K34 was investigated by GC-MSD and 28 different organic acids were detected in the culture broth. Compared with the prominent lactic acid, acetic acid, formic acid, the thirdly most abundant phenyllactic acid showed high inhibitory activity against S. gallinarum. After pHs of the acids were adjusted to S, the inhibitory activities of lactic acid, acetic acid, formic acid against S. gallinarum were greatly reduced while the inhibitory activity of phenyllactic acid was unchanged. The inhibitory activity of the phenyllactic acid was specifically high against S. gallinarum and S. aureus but very low against yeast and mold.

Inhibiton Activity and Charaterization of Lactic Acid Bacteria from Pig Feces (돼지분변으로부터 분리한 유산균주들의 헬리코박터 저해력과 항균활성 및 배양특성)

  • Lactic acid bacteria were isolated from pig feces for probiotics. The six isolated strains were identified as Lactobacillus paracasei (Lp), Lactobacillus fermentum (Lf), Lactobacillus brevis (Lb), Lactobacillus plantarum (P1 , P2), and Pediococcus pentosaceus (P3) by its sugar utilization, morphological and physiological characteristics. Pl was showed largest antibacterial inhibition zone among the isolated strains. It was against Salmonella gallinarum 25mm, E. coli 20.5mm, Staphylococcus aures 24mm, and Pseudomonas aeruginosa 28mm by inhibitory zone, respectively. Lf was showed hyper acid tolerance, $80\%$ survival rate for 40 minutes, and P1, Lb showed hyper bile tolerance, $408\%,\;283\%$ survival rate for 9 hrs, respectively. Therefore the Lf, P1, and P2 strains were expected to probiotics.

Isolation and Identification of Lactic Acid Bacteria Inhibiting Gastro-intestinal Pathogenic Bacteria of Domestic Animal. (가축 소화기 병원성 세균을 저해하는 유산균의 분리 및 동정)

  • To isolate probiotic lactic acid bacteria having superior inhibitory activities against animal gastro-intestinal pathogenic bacteria such as Salmonella gallinarum, Staphylococcus aureus and Escherichia coli, 130 strains were initially isolated from the small intestines of Korean native chickens and 7 lactic acid bacteria were finally selected. By using API CHL kit and 16S rRNA sequencing method, the selected lactic acid bacteria were found to be belonged to genus Lactobacillus except BD14 identified as Pediococcus pentosaceus. Especially, Lactobacillus pentosus K34 showed the highest resistancy to both of HCl and bile salt, as well as the highest inhibitory activities against S. gallinarum, S. aureus and E. coli. All the selected strains were sensitive to various antibiotics such as neomycin, erythromycin, cephalosporin, amoxicillin/clavulanic acid, ampicillin, oxytetracycline, but resistant to ciprofloxacin. All the selected strains except BL strain were resistant to colistin and streptomycin, and BD14, BD16, K34 strains were resistant to gentamicin.

Effects of Different Starter Cultures on the Antibacterial and Antioxidant Activity of Ethanol Extract from Fermented Chelidonium majus var. asiaticum (발효 애기똥풀 주정추출물의 항균 및 항산화활성에 있어 발효 균주의 효과)

  • This study was conducted to investigate the effect of fermentation on biological activity of Chelidonium majus var asiaticum and to screen effective starter culture strains. Antibacterial activity against to Staphylococcus aureus, Listeria monocytogenes and Salmonella gallinarum and antioxidant activity as free radical scavenging activity by using DPPH were tested. Total six starter culture strains, two of Lactobacillus brevis, one of Lactobacillus plantarum and three of Saccharomyces cerevisiae were used. Plant extract was prepared after fermentation by using ethanol. All strains showed normal growth in viable cell counts of fermented cultures and L. plantarum showed the highest cell growth significantly (p<0.05). In antibacterial activities of extracts, the activity was found only in the extract from the fermentation using L. plantarum. In antioxidant activity, the highest activity was shown in the fermentation using L. plantarum significantly (P<0.05). Newly produced spots in two of three elution systems on TLC-DPPH test were detected in the fermentation using L. plantarum.

Medium Optimization for Pediocin SA131 Production by Pediococcus pentosaceus SA131 against Bovine Mastitis Using Response Surface Methodology

  • Pediococcus pentosaceus SA131 was isolated from jeotgal, is the bacteriocin producer against bovine mastitis pathogens, Streptococcus uberis E290, Enterococcus gallinarum E362, and Staphylococcus epidermidis ATCC 12228. The medium composition for pediocin SA131 production by P. pentosaceus SA131 was optimized using response surface methodology. Component of medium was studied as carbon source (glucose, fructose, lactose, glycerol, sucrose, maltose, and mannitol), nitrogen source (beef extract, yeast extract, peptone, malt extract, and tryptone), mineral and surfactant ($MgSO_4$, $KH_2PO_4$, $(NH_4)_2SO_4$, $MnSO_4$, NaCl, sodium acetate, and Tween 80). Through one factor-at-a-time experiment, glucose, fructose, yeast extract, malt extract, NaCl, $MgSO_4$, and Tween 80 were determined as the good ingredient. The effects of major factors for pediocin SA131 production were investigated by two-level fractional factorial designs (FFD). By a $2^4$ FFD, fructose, yeast extract, and $MnSO_4$ were found to be the important factors for the bacteriocin production. Subsequently, a $2^3$ central composite design (CCD) was adopted to derive a statistical model for optimizing the composition of the fermentation medium. The estimated optimum composition for the production of pediocin SA131 by P. pentosaceus SA131 was as follows; 0.13% fructose, 1% glucose, 1.8% yeast extract, 2.58% $MnSO_4$, 0.2% NaCl, and 0.2% Tween 80. The pediocin production under optimized medium was increased to 1,000 AU/mL, compared to the 400 AU/mL in MRS medium.
