Intraorbital infection shows a low incidence, but it might cause blindness or even death. This case is unusual in that its origin from a craniofacial bone fracture prior to infection of the maxillary sinus. A 33-year-old female patient was referred for right cheek swelling. When she visited the emergency room, we removed right cheek hematoma and bacterial examination was done. In the past, she had craniofacial bone surgical history due to a traffic accident 6 years ago. Next day, the swelling had remained with proptosis and pus was recognized in the conjunctiva. We planned an emergency operation and removed the pus which was already spread inside the orbit. And the evaluation for sinusitis was consulted to the otorhinolaryngology department simultaneously. There were Prevotella oralis and methicillin-resistant Staphylococcus epidermidis bacterial infection in the intraorbital and sinus respectively. Afterwards, the vigorous dressing was done for over a month with intravenous antibiotics. Though the intraorbital infection was resolved, blindness and extraocular movement limitation were inevitable. In conclusion, close follow up of the maxillary sinus in facial bone fracture patients is important and aggressive treatment is needed when an infection is diagnosed.
As part of our attempt to discover novel active compounds against multi-drug resistant pathogens, we hereby report two new complexes of iron(III) with formulae: [Fe(L1)2(H2O)2]Cl3 and [Fe(L1)2(L2)(H2O)]Cl2 where L1 = 1,10-phenanthroline (C12H8N2) and L2 = guanide (C5H4N5O-). The synthesized complexes were characterized using spectroscopic analysis (ESI-MS, ICP-OES, FT-IR, and UV-Vis), cyclic voltammetry, CHN analysis, gravimetric chloride determination, melting point determination, and conductance measurement. Octahedral geometries are assigned to both complexes. In vitro antibacterial activity was tested on two Gram-positive (Staphylococcus aureus, Streptococcus epidermidis) and two Gram-negative (Escherichia coli and Klebsiella pneumoniae) bacteria using the disc diffusion method. The complexes demonstrated appreciable activity against these pathogens. Interestingly, the [Fe(L1)2(L2)(H2O)]Cl2 complex manifested a higher degree of inhibition against the drug-resistant Gram-negative bacteria than the commercially available drug, namely erythromycin.

Coagulase-negative staphylococci (CoNS) are a typical group of microorganisms, and the recent advances in laboratory technology and medicine has dramatically modified their significance in medical practice. CoNS, which were previously classified as normal bacterial flora, have recently been reported to be associated with serious infectious diseases, such as surgical wound infection or periprosthetic joint infection. Representative CoNS include Staphylococcus epidermidis, S. haemolyticus, and S. saprophyticus, which are known to cause serious problems in biomaterial-based and prosthetic device infections, as well as to cause simple urinary tract infections in sexually active women. Over the last decade, the clinical isolation rate of CoNS has been increasing, and antibiotic resistance has also been occurring. This review aimed to investigate the incidence of CoNS infection and to use the results as basic data for the management of CoNS, with a focus on the isolation rate and antibiotic resistance in clinical surgery.
Talipariti hamabo is a plant growing around salt marshes in the Lava Coast region of Jeju Island, Korea. In this study, the extract of T. hamabo leaves was investigated for the anti-inflammatory, anti-oxidative and anti-bacterial activities and their active constituents were identified. In the anti-inflammatory tests using lipopolysaccharide-stimulated RAW264.7 cells, the ethyl acetate (EtOAc) fraction inhibited the nitric oxide production without causing cell toxicity. Moreover, the EtOAc fraction reduced pro-inflammatory cytokines (tumor necrosis factor-α, interleukin-6) as well as prostaglandin E2. In the anti-oxidative studies with DPPH and ABTS+ radicals, potent scavenging activities were observed in the EtOAc and n-butanol fractions. Upon the anti-bacterial tests using Staphylococcus epidermidis, EtOAc and n-butanol fractions exhibited good activities. Through the phytochemical studies on EtOAc fraction, three components were isolated by repeated column chromatography; oleic acid (1), p-hydroxyphenethyl-trans-ferulate (2), nicotiflorine (3). Based on these results, the extract of T. hamabo leaves can be developed as natural resources for cosmetic applications.
Background: Ventilator-associated pneumonia (VAP) requires prompt and appropriate treatment. Since methicillin-resistant Staphylococcus aureus (MRSA) is a frequent pathogen in VAP, rapid identification of it, is pivotal. Our aim was to evaluate the utility of quantitative polymerase chain reaction (qPCR) as a useful method for etiologic diagnoses of MRSA pneumonia. Methods: We performed qPCR for mecA, S. aureus-specific femA-SA, and S. epidermidis-specific femA-SE genes from bronchoalveolar lavage or bronchial washing samples obtained from clinically-suspected VAP. Molecular identification of MRSA was based on the presence of the mecA and femA-SA gene, with the absence of the femA-SE gene. To compensate for the experimental and clinical conditions, we spiked an internal control in the course of DNA extraction. We estimated number of colony-forming units per mL (CFU/mL) of MRSA samples through a standard curve of a serially-diluted reference MRSA strain. We compared the threshold cycle (Ct) value with the microbiologic results of MRSA. Results: We obtained the mecA gene standard curve, which showed the detection limit of the mecA gene to be 100 fg, which corresponds to a copy number of 30. We chose cut-off Ct values of 27.94 (equivalent to $1{\times}10^4$ CFU/mL) and 21.78 (equivalent to $1{\times}10^5$ CFU/mL). The sensitivity and specificity of our assay were 88.9% and 88.9% respectively, when compared with quantitative cultures. Conclusion: Our results were valuable for diagnosing and identifying pathogens involved in VAP. We believe our modified qPCR is an appropriate tool for the rapid diagnosis of clinical pathogens regarding patients in the intensive care unit.
본 연구는 가임기 여성의 질로부터 젖산균인 Lactobacillus plantarum UK-3를 분리하여 다양한 생리화학적 특성조사와 여러 가지 병원균들에 대한 항균활성을 확인하고자 실시하였다. 균주 UK-3는 MRS 배지에서 배양되었으며, 형태학적 관찰 및 생화학적 특성을 조사하였으며, 16S rRNA 염기서열 분석을 통해 균주를 동정하여, Lactobacillus plantarum UK-3로 명명하였으며, GenBank에 [JK266589]로 등록하였다. 배양기간 동안에 L. plantarum UK-3의 생장과 유기산의 생성, pH 변화를 조사하였으며, HPLC를 사용하여 대사산물로서 생성된 젖산(lactic acid)과 아세트산(acetic acid)을 정량 및 정성분석 하였다. 이들 유기산의 생성은 L. plantarum UK-3의 생장과 비례하였고, 배양 48시간 경과 후에 젖산과 아세트산의 농도는 각각 약 684.11 mM과 174.26 mM이었으며, 초기 pH 7.0은 배양기간 동안 3.7로 감소하였다. 여성의 질에서 감염의 가능성이 있는 10가지의 그람양성 세균, 그람음성 세균, 그리고 효모에 대하여 25배로 농축된 배양상등액을 처리하여 plate diffusion assay 방법으로 항균활성을 측정하였다. 그 결과 본 연구에 사용된 10가지 미생물에 대해서 광범위하게 항균효과가 있는 것이 관찰되었으며, 정상균총으로서 다른 젖산세균인 Lactobacillus acidophilus에 대해서는 항균활성을 나타내지 않았다.
본 연구는 산돌배나무 잎에서 추출한 분획물의 항암 및 항균효과를 조사하여 화장품소재로 적용 가능성을 확인하기 위하여 수행하였다. 산돌배나무 잎을 채취하여 건조시킨 후 분쇄하여 60% acetone-$H_{2}O$(6:4, v/v)로 추출하고 아세톤 가용부를 분획하여 동결건조 시켰다. 아세톤 및 물 가용부를 Sephadex LH-20으로 충진한 칼럼에서 MeOH을 용출용매로 사용하여 칼럼크로마토그래피를 실시하고 또한 MeOH를 전개용매로 사용한 MIC-gel을 이용했다. 단리된 화합물들은 silica-gel TLC로 확인하였다. 항암효과는 마우스 피부암 세포(B16F10)를 이용한 MTT assay로 측정했다. 산돌배나무 잎으로부터 얻어진 분획물의 암세포 생장 저해율은 80% 이상의 저해률을 보였다. 항균활성 실험에서 산돌배나무 잎의 분획물은 그람양성균인 Staphylococcus aureus, Staphylococcus epidermidis에 대하여 0.25 mg/disc에서 1.3 cm, 2 cm의 생육 저해환을 나타내었다. 그람음성균인 Escherichia coli 에 대하여 0.5 mg/disc에서 각 분획물들은 1.1 cm~1.5 cm의 생육 저해환을 나타내었다. 이상의 결과로 미루어 보아 산돌배나무 잎의 아세톤 분획물은 항암 및 항균효과를 가진 천연소재임을 확인할 수 있었다.
본 연구에서는 개복숭아꽃과 흰색, 분홍색 매화 꽃의 항균 활성과 항산화, 미백 효과에 대해 알아보고자 하였다. 먼저, 3종의 꽃추출물들은 4종의 균주 중에 S. aureus에 대해 우수한 항균 활성을 나타냈다. 흰매화 클로로포름층은 0.5 mg/mL 농도에서 S. aureus에 대해 84%의 항균 활성을 보였고 개복숭아꽃과 분홍매화 클로로포름층도 0.5 mg/mL 농도에서 각각 49, 30%의 항균 효과를 나타냈다. 3종의 꽃추출물들은 P. aeruginosa에 대해서도 항균 효과를 보였다. 개복숭아꽃 클로로포름층과 분홍매화 클로로포름층은 0.5 mg/mL 농도에서 각각 36, 30%의 항균 활성을 보였다. 이 추출물들은 S. epidermidis와 E. coli에 대해서는 항균 활성을 보이지 않았다. 3종의 꽃추출물들은 HaCaT cell의 생존에 크게 영향을 미치지 않았다. 분홍매화 증류수층은 20, 40 ㎍/mL 농도에서 항산화 효과를 보였다. 분홍매화 에틸아세테이트층도 40 ㎍/mL에서 항산화 효과를 보였는데 이는 글루타치온 보다 우수하였다. 꽃추출물들은 B16F10 cell의 생존율에는 영향을 미치지 않았다. 꽃추출물들의 미백 효과는 개복숭아꽃 클로로포름층 40 ㎍/mL에서 18%가 나타났다. 이러한 결과들을 바탕으로 3종의 꽃추출물들은 항균 활성과 항산화, 미백 효과가 있는 원료로 판단할 수 있었다.
본 연구에서는 참외의 부위별 물 및 알코올 추출물의 항산화효과, ${\alpha}$-Glucosidase 저해활성 및 항균효과를 조사하였다. 총 phenol 함량을 측정한 결과 물 추출물에서는 껍질이 151.64 ${\mu}g/g$, 에탄올 추출물에서는 껍질이 224.77 ${\mu}g/g$로 가장 높은 페놀함량을 나타내었다. 총 flavonoid 함량은 껍질이 물 추출물의 경우 45.53 ${\mu}g/g$, 에탄올 추출은 338.37 67.16 ${\mu}g/g$로 가장 높게 나타내었다. 참외의 항산화 효과는 물 추출물의 경우 1%에서 껍질이 25%로 가장 높았으며, 에탄을 추출은 1%에서 껍질이 83.3로 가장 높은 전자공여능을 나타내었으며, 과육과 태좌는 20% 미만의 낮은 활성을 나타내었다. ABTS는 물 추출물은 1%에서 껍질이 79.2%로 가장 높았으며, 과육이 57.6%, 태좌가 74.0%로 50%이상의 활성을 나타내었고, 에탄올추출물은 99.9%로 껍질이 가장 높게 나타났고, 과육과 태좌는 52.1%, 41.2%의 활성을 보였으며, Xanthine oxidase의 저해활성의 경우 1%에서 껍질이 물 추출물이 29.3%, 에탄올추출물이 30.5%의 가장 높은 활성을 보였지만, 비교적 낮은 저해율을 나타내었다. ${\alpha}$-Glucosidase 저해활성을 측정한 결과 1%에서 열수 추출물은 25.9%, 에탄올 추출물은 37.5%의 활성을 보인 껍질이 가장 높은 활성을 보였다. 또한 Streptococcus agalactiae, Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Staphylococcus epidermidis 및 Salmonella typimimum에 대한 항균활성을 평가한 결과 참외의 에탄올 추출물에서 Streptococcus agalactia에서만 껍질, 과육, 태좌에서 각각 15, 13 및 12mm의 clear zone를 나타내었다.
