This study was performed to evaluate the effects of ocular bacteria and tear lactoferrin on the Tear Staining Syndrome(TSS) in poodle dogs. Staphylococcus, Klebsiella, Micrococcus and Pseudomonas were the most Prevalent microorganisms isolated in normal eyes of poodle dogs, whereas the predominant isolates in the poodle dogs with TSS were Micrococcus, Staptococcus and Staphylococcus. There were no significant differences between the normal and tear-stained poodle dogs in the quantity of tear lactoferrin. In vitro hair staining experiment, bacteria isolated from the eyes with TSS and lactoferrin didn't stain hair. The results have shown that either ocular bacteria or tear lactoferrins were not related to the TSS.

A fast and matrix-assisted laser desorption/ionization-mass spectrometry compatible protein staining method in one- and two-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis is described. It is based on the counterion dye staining method that employs oppositely charged two dyes, Zincon and Ethyl Violet to form an ion-pair complex. It is safe to use since the methanol used previously in staining solution was replaced with ethanol, which is not toxic. The protocol including fixing, staining and quick washing steps can be completed in 1 to 1.5 h depending upon gel thickness. (omitted)
Objective : In order to establish the index of degeneration, the authors performed a histochemical study with Safranin-O staining and investigated the occurrence of apoptosis in the human intervertebral disc. Methods : Eighteen intervertebral disc specimens surgically extracted from the patients and two additional specimens from the autopsied cases were stained with Safranin-O for proteoglycan according to a standard protocol. Terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate- biotin nick end labeling[TUNEL] was used to detect the fragmented DNA known to be associated with apoptotic cell death and classification scheme was formulated for categorization of the degree of Safranin-O staining [normal, moderate reduction, faint] by modification of Makin's histological-histochemical grading. The Kruskal-Wallis H test and Chi-square test were used for statistical analysis. Results : The statistical results showed a significant difference in the mean age between "normal" Safranin-O staining group and the others [19.3 versus 55, 43.4, p=0021]. However, there was no statistically significant correlation between Safranin-O staining and MR grading of disc degeneration. Only six of eighteen surgical specimens and none in autopsies showed positive apoptotic cells in TUNEL staining. Conclusion : The determination of the degree of degeneration in surgically obtained disc tissue per se by histochemical staining or by the degree of apoptosis that corresponds to its morphologic change was not feasible.

The acrosome cap allows sperm to penetrate the egg membrane and produce male pronuclei within female chicken eggs, facilitating successful fertilization. Given this, it is important to establish practical methods for evaluating the integrity of the acrosome cap and thus the quality of the rooster's sperm. There are several established methods for evaluating the acrosomes of mammalian sperm, but none of these methods are suitable for evaluating the acrosome status of rooster spermatozoa. Therefore, a simplified method for evaluating the rooster acrosome is needed. Here we evaluated the usefulness of CBB (coomassie brilliant blue) staining of the acrosome at concentrations of 0.04%, 0.08%, and 0.3% CBB solutions. Our data revealed a clear staining pattern for intact acrosome caps at 0.04% and 0.08% CBB but not at 0.3% CBB. This protocol revealed differences in acrosome integrity between fresh and frozen rooster sperm smears suggesting that CBB staining may facilitate easier semen evaluation in roosters. This protocol allows for the accurate differential staining of acrosome cap in rooster spermatozoa.
Background: Modified toluidine blue staining (MTBs) is a simple, inexpensive and time saving method to detect H. pylori in gastric biopsy specimens. As a metachromatic stain, it simultaneously highlights intestinal metaplasia, a gastric cancer precancerous lesion. The aim of this study was to assess the reliability of MTBs compared with hematoxylin-eosin (H&E) for H. pylori detection using immunoperoxidase staining as the gold standard. This technique would be beneficial for a routine diagnosis and confirmation of H. pylori eradication in developing countries where endoscopic-based approaches are dominant. Materials and Methods: Esophagogastroduodenoscopy with triple site gastric biopsies was undertaken in 207 dyspeptic patients at Thammasat University Hospital, Thailand between 1997 and 1999. H&E, MTBs and immunoperoxidase staining were applied to each specimen. The presence or absence of H. pylori with each stain was interpreted separately and the sensitivity, specificity, positive and negative predictive values of H&E and MTBs were calculated. Results: A total of 282 specimens from 207 patients were evaluated. Using immunoperoxidase staining, organisms were positive in 117 specimens (41%). MTBs proved almost equally sensitive as immunoperoxidase (99%) and significantly more sensitive than H&E (85%). It has comparable specificity (96% vs 96%), PPV (95% vs 94%), and NPV (99% vs 90%) to H&E, using immunoperoxidase staining as gold standard. MTBs compared with immunoperoxidase staining, is cheaper (2 USD vs 12 USD) and faster (20 min vs 16 hrs) compared to immunoperoxidase staining. Conclusions: MTBs is effective, economical and easy to use in daily practice for the detection of H. pylori in gastric biopsy specimens. In addition to saving time in evaluating H. pylori associated gastritis, with a high sensitivity and ability to demonstrate intestinal metaplasia, the technique may have a role in confirmation of H. pylori eradication for gastric cancer prevention in a developing country setting.

Background: Astrocytic tumors, the most common primary glial tumors of the central nervous system, are classified from low to high grade according to the degree of anaplasia and presence of necrosis. Despite advances in therapeutic management of high grade astrocytic tumors, prognosis remains poor. In the present study, the frequency and prognostic significance of c-erb-B2 in astrocytic tumors was investigated. Materials and Methods: Records of 72 patients with low- and high-grade astrocytic tumors were evaluated. The expression of C-erbB-2 was determined immunohistochemically and intensity was recorded as 0 to 3+. Tumors with weak staining (1+) or no staining (0) were considered Her-2 negative, while tumors with moderate (2+) and strong (3+) staining were considered Her-2 positive. Results: Of the 72 patients, 41 (56.9%) had glioblastoma (GBM), 10 (13.9%) had diffuse astrocytoma, 15 (20.8%) had anaplastic astrocytoma, 6 (8.3%) had pilocytic astrocytoma. C-erbB-2 overexpression was detected in the tumor specimens of 17 patients (23.6%). Six (8.3%) tumors, all GBMs, exhibited strong staining, 2 (2.7%) specimens, both GBMs, exhibited moderate staining, and 9 specimens, 5 of them GBMs (12.5%), exhibited weak staining. No staining was observed in diffuse astrocytoma and pilocytic astrocytoma specimens. Median overall survival of patients with C-erbB-2 negative and C-erbB-2 positive tumors were 30 months (95%CI: 22.5-37.4 months) and 16.9 months (95%CI: 4.3-29.5 months), respectively (p=0.244). Conclusions: Although there was no difference in survival, C-erbB-2 overexpression was observed only in the GBM subtype.
The object to this study is to identify the genus and species of the microfilariae which were recently found in the area of Youngju-Gun. THe identification of the microfilariae was made on the morphologicla aspects. 1. Blood samples were collected through vena punction from known microfilariae carriers living in the newly confirmed filaria endemic area of Youngju0Gun in Jyongsang Pukdo Province. Youngju-Gun is located in the mountainous central part of Korean Peninsula. 2. The following fixation and staining techniques were applied. (1) Fixation by drying in the air, followed by staining with Azeo or Giemsa. (2) Knott's fixation method (2% formalin), followed by staining with Azur II. 3. A comparative study of the body length of the microfilariae after different fixation and staining techniques were applied. (1) Knott's fixation method followed by staining with Azur II : average body length found was 28.4$\mu$. (2) Dry fixation followed by staining with Giemsa : average body length found was 209.4$\mu$. (3) Dry fixation followed by staining with Azeo : average body length found was 205.4$\mu$. 4. The locations of the different body cells were measured in 60 individuals of microfilariae in the wet preparation fixed by Knott's method and stained with Azur II. The distance of the different body cells to cephalic apex of microfilariae was measured and calculated as a percentage of the total body length. The average results are as follows : BNC , 3.04% ; N, 22.74%, EP, 31.4% ; EC, 37.77%, G1 cell , 67.94%; G2 cell, 73.54% : G3 cell , 75.55% ; G4 cell, 77.65% ; AP, 82.02%%. 5. As a result of the above findings the microfilariae found in the above mentioned area could be identified as Brugia malai(BRUG, 1927) BUCKLEY, 1960.
본 연구는 선박평형수처리장치 성능을 평가하기 위한 일환으로 Evan blue, Aniline blue 그리고 CMFDA 염색방법을 활용하여 해양부유생물의 생사판별을 이해하고자 하였다. Evans blue와 Aniline blue의 염색은 죽은 생물을 파란색으로 염색하여 죽은 생물을 평가하는 방법이고, CMFDA의 방법은 살아있는 생물을 녹색으로 염색하여 평가한다. 현장 동 식물플랑크톤 군집을 대상으로 Evans blue와 Aniline blue의 방법을 적용한 결과, 죽은 생물을 90%이상 염색시키는 것으로 나타났다. 하지만, 살아있는 일부 식물플랑크톤의 군집에서도 파란색으로 염색 되어, 실제 선박평형수 처리장장치의 성능을 평가하기에는 일정의 한계성을 나타내었다. 한편, 살아있는 생물을 염색하는CMFDA방법을 적용한 결과, 현장의 부유생물의 70%의 염색효과를 보였다. CMFDA방법은 FDA방법과 유사하게 살아있는 생물을 녹색으로 염색하여 생물 생사판별이 가능하게 함으로, 두 가지 방법을 중복염색(double staining)하는 방법을 고안해 보다 높은 효율의 생사판별방법을 찾고자 노력하였다. 그 결과, 두 가지의 염색시약을 중복으로 염색한 실험군에서 95%이상의 높은 효율을 보였고. 이는 단일 염색한 실험군보다 상대적으로 높은 염색효율을 얻을 수 있었다. 따라서 CMFDA+ FDA를 중복염색한 평가법이 해양생물의 생사를 판별하는데 보다 높은 효율을 보였고, 선박평형수처리장치의 성능을 평가하는 방법으로 활용이 가능할 것으로 판단된다.
This study was performed to investigate the best staining method for the somatic cell classification of dairy goat milk. Dairy goat milk samples, which were collected randomly from a dairy goat farm in Jeollanam-do, South Korea, were stained and analyzed with direct microscopic method, using 5 different staining methods; Wright's stain, Giemsa stain, Diff-quik stain, Newman's stain and Pyronin Y-Methyl Green stain, respectively. Among them, The Newman's staining was found to be the most rapid and effective method, for it required the shortest time for staining and provided the easiest way to classify somatic cells.
Small peptide tags such as c-myc, HA, or FLAG tag have facilitated efficient Western-blotting of proteins of interest especially when specific antibodies for the proteins are not available. However, the conventional Western-blotting requires the multi-steps process taking at least several hours up to two days. With examples of various applications, here we show a convenient and time-saving method for protein detection which employs a fluorescent chemical BDED and its binding peptide RC-tag. And we propose "Estern staining", as a standard term for protein detection method using fluorescent chemicals and their binding small peptide tags. Eastern staining may substitutes for the time-consuming "immuno-staining" in many versatile applications.
