Development of Acaricide Resistance and Cross-Resistance in Tetranychus urticae(Acarina: Tetranychidae) (점박이응애의 설비제정항성 발달과 교차저항성에 관한 연구)

  • These studies were conducted to investigate development of chemical resistance in two-spotted spider mite (Tetranychus urticae Koch) trough single selection for a given period of time with five acaricides including carbophenothion and ethion of organo-phosphate compound, dicofol of organo-chlorine compound, cyhexatin of organo-tin compound, biphenthrin of synthetic pyrethroid compound; and to examine cross-resistance among the acaricides to the selected populations. The development of chemical resistance of two-spotted spider mite was greatly varied under single selection of the each five acaricides. The carbophenothion- and the ethion-populations at the 22nd and the 24th selected generations showed 156- and 64.1-fold resistant levels, respectively, as compared with the susceptible population. However, the dicofol population at the 28th selected generation exhibited 39.7-fold resistant level, and the biphenthrin population at the 24th selected generation revealed 25.2-fold resistant level, while the cyhexatin population at the 20th selected generation showed 13-fold resistant level. The carbophenothion-selected population showed high cross-resistance to ethion, and low cross-resistance to dicofol and biphenthrin. The ethion-selected population revealed also high cross-resistance to carbophenothion. The dicofol-or the biphenthrin-selected populations, however, exhibited moderate cross-resistance to carbophenothion and ethion. Meantime, the cyhexatin-selected population showed high cross-resistance to ethion and carbophenothion.

Synthesis and Insecticidal Activities of N-Phosphinothioyl Carbofuran Derivatives (N-Phosphinothioyl carbofuran 유도체의 합성 및 살충 활성 검정)

  • N-Dimethoxyphosphinothionyl carbofuran, PSC, has a high insecticidal activity and low mammalian toxicity. Ten N-phosphinothionyl carbofuran derivatives were synthesized and their insecticidal activities were determined against brown plant hopper (Nilaparvata lugens), green peach aphid (Myzus persicae), diamondback moth (Plutella xylostella), and two-spotted spider mite (Tetranychus urticae). Green peach aphid, diamondback mea and brown plant hopper were controlled over 90% by application of 125 ppm, 125 ppm, and 63 ppm, respectively, of carbosulfan. Two hundred and fifty ppm of newly synthesized compounds could control most of brown plant hopper and diamondback moth. Especially, insecticidal activities of compound 10 against brown plant hopper, diamondback moth, and green peach aphid were similar to those of carbosulfan. Our results show that the newly synthesized derivatives of NV-phosphinothionyl carbofuran have a similar insecticidal activity to carbosulfan.

Occurrence of Fusarium wilt and Twospotted Spider Mite under Plastic Mulched and Non-Plastic Mulched Bed in Hydroponic Culture of Strawberry (딸기 수경재배에서 베드 피복유무에 따른 시들음병과 점박이응애 발생 양상)

  • Hydroponic strawberry culture system is increasing annually. Most of strawberry farmers use mulched bed in hydroponic culture and strawberry plants were transplanted in early September. After transplanting, Fusarium wilt caused by Fusarium oxysporum f. sp. fragariae and twospotted spider mite (TSSM), Tetranychus urticae, can increase their occurrence under high temperature condition. Therefore, we conducted for comparison occurrence of Fusarium wilt and TSSM on mulched with green polyethylene film and non-mulched bed. Occurrence of Fusarium wilt on mulched bed was started from early October and more increase than non-mulched bed. Damage rate of TSSM on mulched bed was shown higher than non-mulched bed. Temperature of substrate in mulched bed increased than non-mulched bed, but relative humidity near plants was decreased. As a result, use of non-mulched bed should be effective for reducing of Fusarium wilt and TSSM on strawberry plants.

Effect of Wolbachia Infection on Fitness of Resistance to Dicofol in Tetranychus urticae (Acarina: Tetranychidae) (Wolbachia 감염이 디코폴 감수성 및 저항성 점박이응애 계통의 적합도에 미치는 영향)

  • Contribution of Wolbachia infection to fitness of a species (developmental time, adult life span, fecundity and ovipositional period) was measured in the susceptible and dicofol-resistant strains of two spotted spider mite, Tetranychus urticae Koch, on miniature roses. Based on ftsZPCR assay, Wolbachia infection was confirmed only in the susceptible strain. The susceptible strain had significantly higher fecundity (eggs/female) and shorter developmental times than the resistant strain. Longer adult life span and ovipositional period were observed in the susceptible strain. Fitness differences were appeared to influence dicofol resistance development. Similar measurements were performed with progeny from two reciprocal $F_1$crosses. Similar to other examples of cytoplasmic incompatibility induced by Wolbachia, subsequent cross between uninfected female and infected male spider mites were different from the other combination: a high egg mortality and a male-biased sex ratio. When the intrinsic rate of natural increase was calculated, the cross between uninfected female and infected male spider mites had a significantly lower rate($0.09\pm$0.01) than did the other combination ($0.20\pm$0.01). These results suggest that the dynamic and evolution of the fitness are closely associated with dicofol resistance and Wolbachia infection in the two spotted spider mites.

Susceptibility of the Predatory Mite, Neoseiulus californicus (Acari: Phytoseiidae) to Acaricides (사막이리응애의 살응애제에 대한 감수성)

  • Effects of 9 acaricides to the predatory mite, Neoseiulus californicus were evaluated. Seven of the acaricides tested, cyenopyrafen. spirodiclofen, spiromesifen, acequinocyl, bifenazate, flufenoxuron and cyflumetofen exhibited low toxicity to adult females and nymphs of N. californicus and had little effect on the reproduction and eclosion of eggs deposited by treated predators. Moreover, hatch percentage of N. californicus eggs was unaffected by exposure to these seven acaricides. Etoxazole did not significantly affect the survival and reproduction of adult female predators but caused very low eclosion in eggs laid by treated females and high egg mortality. Pyraclofos was extremely toxic to adult female predators and caused 100% mortality. Adult female predators survived on a diet of spider mites treated with cyenopyrafen. spirodiclofen, spiromesifen, acequinocyl, bifenazate, flufenoxuron and cyflumetofen and their fecundity was not substantially affected. Based on the results, cyenopyrafen. spirodiclofen, spiromesifen, acequinocyl, bifenazate, flufenoxuron and cyflumetofen are appeared to be promising candidates for use in integrated mite management program where N. californicus is the major natural enemy.

Ventilation at Supra-Optimal Temperature Leading High Relative Humidity Controls Powdery Mildew, Silverleaf Whitefly, Mite and Inhibits the Flowering of Korean Melon in a Greenhouse Cultivation (참외 시설 재배 시 고온에서의 환기 처리에 의한 상대습도 상승과 흰가루병, 담배가루이, 응애 방제 및 개화 억제)

  • This study was conducted to investigate the effect of ventilation at high temperature on the control of powdery mildew, silverleaf whitefly two-spotted spider mite occurred at Korean melon cultivation greenhouse, and on leaf rolling and flowering of the plant in summer season. 'Alchanggul' grafted onto 'Hidden Power' rootstock was planted on soil bed with the distance of 40 cm. Three ventilation temperatures of 45℃, 40℃, and 35℃ as set points were compared. Ventilation treatment was done by control of side window operation from 18th June to 13th July when silverleaf whitefly, mite, and powdery mildew were occurred in all greenhouses. The temperature inside greenhouse was increased up to the set temperature point on sunny days and maintained for about 9 hours with high relative humidity at 45℃ condition. The differences of day maximum air temperature and day minimum RH were the highest at 45℃ treatment. After 11 days of treatments, the damage by powdery mildew and two-spotted spider mite was almost recovered at 45℃ treatment but not at 40 and 35℃. The population of silverleaf whitefly and two-spotted spider mite were significantly decreased at 45℃ treatment at 14 days after treatment, while powdery mildew symptom was not significantly decreased. Leaf rolling was observed at high temperature but not severe at 45℃ treatment. After 26 days of treatments, female flowers did not bloom at all at 45℃ treatment, and the number of male flowers was 1.2 among 15 nodes of newly grown shoots. As the result, it indicates that ventilation at the high temperature of 45℃ for about 2 to 3 weeks can be an applicable method to control above mentioned pests and disease, and to recover the vegetative growth of Korean melon by reducing flowering of the plant.

Insecticidal Activity of N'-phenvl-N-Methylformamidine Analogues against Two Spotted Spider Mite (Tetranychus urticae) and Design of New Potent Compounds (두 점박이 응애(Tetranychus urticae)에 대한 N'-phenyl-N-methylformamidine 유도체의 살충활성과 새로운 고활성 화합물들의 설계)

  • To predict and design of new potent insecticidal compounds, the two dimensional quantitative structure-activity relationships (2D-QSARs) and molecular hologram quantitative structure-activity relationships (HQSARs) between the various physicochemical parameters as descripters of N'-phenyl-N-methylformamidine analogues (1-22) and their insecticidal activity against the two spotted spider mite (Tetranychus urticae) were discussed quantitatively. From 2D-QSAR models (1 & 3), the width ($B_2$) of $R_3$-group as sterically factor and optimal total dipole moment (TDM=2.025D) of $R_4$-group were mainly influenced to increase the activity. Therefore, the activities were depend upon the $R_3$- and $R_4$-groups. Particularly, it is predicted that the activity of newly designed potent compound (PI; $EC_{50}$=0.516 ppm) by 2D-QSAR models (3) and HQSAR model F2 was about 34.3 fold higher than that of the commercialized insecticide, Amitraz ($EC_{50}$=17.7 ppm).

An Investigation of the Control of Two-Spotted Spider Mites (Tetranychus urticae Koch) Resistant to Organo-Phosphates (유기인제저항성 점박이응애(Tetranychus urticae Koch)에 관한 연구)

  • The study involved determination of resistance levels of spider mites ta argano-phosphates using topical application and slide dip techniques; laboratory serening tests of alternative acaricides using an O/P resistant strain and a field trial of the screened materials. 1. Strains of Tetranychus were from Timaru(TR), Havelock Narth (HNR), Lincaln (LN). Germany (GR, GN). Comparisons of the resistant strains and normal strains at the LD50 and LC50 levels were as follows : (a) Using the topical application tochnique; with Parathian. resistant levels of the GR. TR and HNR strains of T. urticae were respeativuly, 1035. 484 and 452 times as resistant' as the LN strain. (b) Using the slide dip technique; with Phosdrin, resistant of GR, TR and HNR strains of T. urticae were 635, 274 and 266 times greater respeativuly, than the GN strain. 2. The laboratory sereaning tests were carried out far their contact plus stomach and residual effect to assess the toxicities of eleven alternative materials which would be used far control of O/P resistant strain of T. urticae. The acaricide groups represented were 3 organo-chlorines (Spidex, Kelthane and C 8514), 2 nitrophenyls (UC 19786 and Morocide), 2 cyclic carbonates(Eradex and Morestan). I carbamate (UC2004 7A), 1 mixture of carbamate and orano-chlorine and 2 other chemicals (C 8677 and M2527). From all acaricide tested. Kelthane and Morocide were the most effective, folowed by Spidex and M2527. Morestan, C8514. C8677 and RS 143 were intermediate, but Eradex, UC 19786 and UC 20046A were poor. 3, The number of sapmles required for estimation of the population in the field evaluation of acaricidal effects was one giving the highest practical precision. It was decided, after preliminary sampling trials. to use samples of 30 leaves per replicate which gave a $5.7\%$ standard error. 4. In the field trials, Morocide applied at the $0.05\%\;and\; 0.04\%$ a. i. conc. to black currant trees gave excellent control of O/P resistant population of T. urticae for about 12 days, but Morocide 0.025 and Kel thane $0.02\%$ a. i. cone. gave efficient control for about 6 days. In other words. first applications of Kel thane ane Moroeide gave very high degrees of control of O/P resistant population of the two-spotted spider mite. However, the results indicate that secondary application would sometimes be necessary. There was no foliage damage of black Currants and strawberries by either acaricides at the concentrations used. Evaluation of Acaricidal Effect Against Two-spotted Spider Mite Collected from Strawberry in Greenhouse (딸기 시설 하우스 점박이응애에 대한 살비 효과 평가)

  • In the strawberry cultivation areas and domestic farms, two-spotted spider mites are the most serious pests. It decreases the product yield due to the direct feeding of chlorophyll by the mites resulting in reduced photosynthetic ability of host plant. In this study, a simplified acaricidal effect evaluation system (SAEES) was employed to choose the effective acaricidal products among 10 items based on leaf-dipping bioassay methods. SAEES had the advantage of being able to screen four commercial products with three replications at the same time in the recommended concentration. The susceptible strains (SL_YS) showed a high mortality rate of about 90-100%, whereas the mortality of local strains was differed by each acaricides. It suggests that the acaricide responses of field populations might differ due to spray frequencies and acaricide product. An efficacy index (0.8-1.0) was determined based on the mortality of susceptible strain, which would allow the most effective commercial products to be selected by the range of this index. In summary, SAEES will enable the selection of effective commercial products and contribute to increasing control against T. urticae in strawberries.

Analysis of Mitochondrial Gene Sequence in Etoxazole Resistant Two-Spotted Spider Mite, Tetranychus urticae (Etoxazole 저항성 점박이응애의 미토콘드리아 유전자 서열 분석)

  • The two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), is one of the most important pest species devastating many horticultural and ornamental crops and fruit trees. Difficulty in managing this mite is largely attributed to its ability to develop resistance to many important acaricides. Development of 3,700-folds resistance to etoxazole was found in the population of T. urticae collected from rose greenhouses in Buyeo, Chungnam Province in August 2000. This population has been selected for eleven years with etoxazole (over 500 times), and increased over 5,000,000-folds in resistance as compared with susceptible strain. Also, etoxazole-resistant strain was shown to be maternally inherited. The objective of this study was to determine whether resistance of T. urticae to etoxazole was linked with point mutations in the mitochondrial gene. DNA sequencing of cytochrome c oxidase subunit I (COX1), COX2, COX3, cytochrome b (CYTB), NADH dehydrogenase subunit 1 (ND1), ND2, ND3, ND4, ND5, and ND6 were analyzed by comparing two etoxazole-susceptible and etoxazole-resistant strains. As a result, differences were not detected between the nucleotide sequences of two strains within a mitochondrial gene.
