This study was conducted to elucidate microplastics detection of freshwater ecosystems in Geum river. Samples are collected at 6 points in freshwater, 5 points in fishes. Freshwater was sampled 100 L per each points and fish species were Opsariichthys uncirostris amurensis, Hemibarbus labeo, Pseudogobio esocinus, Zacco platypus, Micropterus salmoides and Cyprinus carpio. FTIR analyis was adopted to identify microplastic types. Extracted microplastics were PES (polyester), PE (polyethylene), PP (polypropylene), PET (polyethylene terephthalate), PVC(Polyvinyl chloride) in freshwater, and PES, PE, PP, PET, PVC in fishes. Our results were expected to be used basic research information for further study in microplastics of freshwater ecosystems.
병원성 원생동물인 지아디아 람블리아는 수인성 질병을 야기하는 주요 원인이 되고 있다. 본 연구는 PCR 및 RT-PCR 기법을 적용하여 한강본류 하천수 시료에서 사람감염성 종인 지아디아 람블리아를 동정하고, 활성 여부를 판별하여 현 표준시험방법을 보완하고자 하였다. PCR과 RT-PCR에는 지아디아의 복부 흡반 구성 유전자를 증폭하는 giardin primer를 사용하였으며, DNA/RNA 추출 및 PCR/RT-PCR 과정에서의 민감도 검사를 수행한 결과, 1포낭까지 검출가능한 것으로 나타났다. 또한 한강본류 및 유입지천 시료 48점에 적용하여 면역형광항체법과 PCR 및 RT-PCR 방법을 비교하였다. 면역형광항체법을 이용한 현미경관찰 결과 48개 시료의 지아디아 총포낭수의 평균 농도는 6.3 cysts/10 L이었고 양성율은 62.5%였으며, 속빈 포낭을 제외한 지아디아의 평균 농도는 4.5 cysts/10 L이었고 양성율은 52.1%였다. PCR 수행결과 48개 시료 중 24개(50%)의 시료에서 지아디아 람볼리아가 검출되었으며, RT-PCR 수행결과 10개(21%) 시료가 살아있는 G. lamblia를 포함한 것으로 나타났다. 본 연구를 통해 PCR/RT-PCR 기법이 지아디아 포낭을 저농도로 포함하고 있는 하천수 시료에 적용가능하며 원생동물 표준시험방법을 보완하여 종(species) 및 활성에 대한 정보를 제공할 수 있을 것으로 결과되었다.
영남지방 5개 농장에서 사육중인 돼지 251두의 폐렴병소로부터 P. multocida를 분리한 결과 91주의 P. multocida가 분리되어 분리율은 36.3%로 나타났다. 생화학적 특성을 조사한 후 PCR로 최종 확인한 결과 P. multocida species-specific 유전자 460 bp의 특이 증폭산물을 관찰하였다. 분리균 91주의 P. multocida에 대한 약제 감수성 검사 결과 amikacin (91.2%), cephalothin (87.9%), cefoxitin (84.6%), florofenicol (93.4%), norfloxacin (65.9%), ofloxacin (80.2%) 등의 약제에 비교적 높은 감수성을 나타내었다. 또한 대부분의 분리주는 5종 이상의 약제에 대해 다재 내성 양상을 나타내었다.
Co-infection with two virus species was previously reported in some cactus plants. Here, we showed that Notocactus leninghausii f. cristatus can be co-infected with six different viruses: cactus mild mottle virus (CMMoV)-Nl, cactus virus X (CVX)-Nl, pitaya virus X (PiVX)-Nl, rattail cactus necrosis-associated virus (RCNaV)-Nl, schlumbergera virus X (SchVX)-Nl, and zygocactus virus X (ZyVX)-Nl. The coat protein sequences of these viruses were compared with those of previously reported viruses. CMMoV-Nl, CVX-Nl, PiVX-Nl, RCNaV-Nl, SchVX-Nl, and ZyVX-Nl showed the greatest nucleotide sequence homology to CMMoV-Kr (99.8% identity, GenBank accession NC_011803), CVX-Jeju (77.5% identity, GenBank accession LC12841), PiVX-P37 (98.4% identity, GenBank accession NC_024458), RCNaV (99.4% identity, GenBank accession NC_016442), SchVX-K11 (95.7% identity, GenBank accession NC_011659), and ZyVX-B1 (97.9% identity, GenBank accession NC_006059), respectively. This study is the first report of co-infection with six virus species in N. leninghausii f. cristatus in South Korea.
Mastitis is a common and serious infection of the mammary gland in dairy cattle and has a major economic impact on the production of milk and dairy products. Bacterial mastitis is caused by several pathogens and is most frequently associated with coagulase-negative staphylococci (CNS). Although CNS are typically associated with subclinical or mild mastitis, the importance of CNS has increased as these pathogens have emerged as predominant mastitis-related pathogens in many countries. CNS can cause persistent infections, resulting in increased milk somatic cell counts and thereby affecting milk quality and decreasing milk production. Globally, Staphylococcus chromogenes, S. epidermidis, and S. simulans are the predominant CNS species in dairy cattle mastitis. Antibacterial resistance of CNS varies with species, and most CNS are susceptible to vancomycin and resistant to penicillin and tetracycline. As the most frequently isolated CNS species, some strains of S. chromogenes exhibit phenotypic resistance to ampicillin, erythromycin, oxacillin, penicillin, and tetracycline. Some strains of S. epidermidis and S. haemolyticus are only susceptible to vancomycin and rifampicin. Therefore, more studies are needed to achieve the control and prevention of CNS as environmental pathogens.
본 연구는 multiplex real-time PCR을 이용하여 Actinobacillus actinomycetemcomitans, Campylobacter rectus, Porphyromonas gingivalis, Tannerella forsythus, Treponema denticola, Prevotella intermedia 등과 같은 6종의 주요 치주 질환 원인 미생물들을 동시에 검출할 수 있는 분석 방법에 관한 것이다. 4개의 형광 염료를 사용하여 internal control과 함께 3개의 균종씩 나누어 분석하였으며, 분석 대상 균종 간 그리고 다른 종류의 구강 미생물 균종과의 간섭과 교차 반응이 없음을 확인하였다. 본 연구의 multiplex real-time PCR은 타액과 플라그 등의 다양한 샘플에 포함되어 있는 각 미생물들을 정성, 정량적으로 분석할 수 있었으며, 치주염 환자와 건강한 사람들에 대한 비교 분석 결과 분명한 차이를 발견 할 수 있었다.
This study assessed the characteristics of emission and cell toxicology of Methylethylketone(MEK) in ambient air of industrial area. MEK is produced by the oxidation of sec-butyl alcohol and used as the solvent for making ink, printing, coating of film, bonding material and drug extraction. The MEK concentrations in the ambient-air of industrial area in Gimhae City was detected in the range of $25.4{\sim}1,580{\mu}g/m^3$ with an average $297.4{\mu}g/m^3$. The concentration of MEK showed a descending tendency from April to August followed by its increased tendency since then. The effects of MEK on the human lung cancer A549 cells was examined by the generation of Reactive Oxygen Species(ROS) and cytotoxicity. The range of MEK concentration detected in the area induced ROS generation affecting the oxidation state with a little effects on the viability of the cells.

Next generation sequencing (NGS) technologies have led to the rapid accumulation of genome sequences through whole-genome sequencing and re-sequencing of crop species. Genomic resources provide the opportunity for a new revolution in plant breeding by facilitating the dissection of complex traits. Among vegetable crops, reference genomes have been sequenced and assembled for several species in the Solanaceae and Cucurbitaceae families, including tomato, pepper, cucumber, watermelon, and melon. These reference genomes have been leveraged for re-sequencing of diverse germplasm collections to explore genome-wide sequence variations, especially single nucleotide polymorphisms (SNPs). The use of genome-wide SNPs and high-throughput genotyping methods has led to the development of new strategies for dissecting complex quantitative traits, such as genome-wide association study (GWAS). In addition, the use of multi-parent populations, including nested association mapping (NAM) and multiparent advanced generation intercross (MAGIC) populations, has helped increase the accuracy of quantitative trait loci (QTL) detection. Consequently, a number of QTL have been discovered for agronomically important traits, such as disease resistance and fruit traits, with high mapping resolution. The molecular markers for these QTL represent a useful resource for enhancing selection efficiency via marker-assisted selection (MAS) in vegetable breeding programs. In this review, we discuss current genomic resources and marker-trait association analysis to facilitate genome-assisted breeding in vegetable species in the Solanaceae and Cucurbitaceae families.
In an attempt to clarify the epidemiological feature of distomiasis in Tongjin riverside area, the prevalence of distomiasis in the residents and infection rates of the metacercariae in fresh-water fishes were investigated at the upper, middle and lower reaches of the river from January to April, 1984. The results obtained were summarized as follows: 1. Out of a total of 931 fresh-water fishes which composed of 33 different species, 611 fishes(65.6%) of 31 species were found positive with digenetic trematode metacercariae of 16 different species, and there were some differences in infection rates of the metacercariae among the fishes in the 3 parts of the river; 53.8% in upper, 80.7% in middle, and 61.0% in lower reaches, respectively. 2. Infection rates of the metacercariae of Exorchis oviformis, Metagonimus yokogawai, Echinochasmus japenicus, Metorchis orientalis and Clonorchis sinensis in the fishes were 48%,29%, 115, 7.9% and 6.3oA, respectively. 3. The average number of the encysted larvae of Clonorchis found in fish body/gram showed 4.44 in Pseudorasbera larva, Gnathepegon coreanus (1.2), Microphysogoio yaluensis (0.76), Abbottina springeri (0.4), Acanthorhodeus asmussi (0.21) and Cultriculus eigenmanni (0.17), respectively. 4. The average number of the metacercariae of Metagonimus found in fish body/gram disclosed 34.01 in Zacco platypus, Zacco temmincki (16.46), Carassius carassius (5.35), Moroco oxycephalus (1.54) , Aphyocypris chinensis (1.5) and etc., respectively. 5. Detection rates of the eggs of Clonorchis and Metagonimus among residents were 1.1% and 0.8%, respectively, out of a total 923 Persons.
