The mycelial growth and ligninolytic enzyme activity of cauliflower mushroom (Sparassis latifolia)

  • We examined the activities of lignin-degrading enzymes of the mycelium of cauliflower mushroom (Sparassis latifolia). Three different strains of S. latifolia collected from several sites in Korea and one crossbred strain were cultured on potato dextrose broth (PDB) and Kirk's medium in order to study the activities of their ligninolytic enzymes. Mycelial growth reached maximum levels between 14 and 21 days after inoculation and pH increased by 0.12 units over 35 days. Laccase activity began increasing after 14 days on both types of media. Manganese peroxidase (MnP) activity followed a trend similar to that of laccase on Kirk's medium, but not on PDB. The activity of lignin peroxidase (LiP) differed from that of other enzymes; its activity decreased by half after 14 days on PDB but remained constant on Kirk's medium over 35 days. The total protein concentration increased considerably after 14 days and peaked at 21 days on PDB. A similar maximum was attained on Kirk's medium. In contrast, the residual glucose increased rapidly at 14 days on Kirk's medium, while increasing gradually up to 28 days on PDB. This study indicates that S. latifolia is more similar to white rot fungi than to other brown rot fungi.

In vitro anti-cancer activity of hydrophobic fractions of Sparassis latifolia extract using AGS, A529, and HepG2 cell lines (꽃송이버섯(Sparassis latifolia) 추출물 소수성 분획의 항암 활성)

  • The use of mushrooms has immense potential in many diverse applications. Until now, more than 3,000 species are consumed around the world, and more than 100 have shown promising clinical activity against cancer and other chronic diseases. Sparassis latifolia (formerly S. crispa) is an edible mushroom that harbors ${\beta}$-glucan reported to possess immunostimulatory and anticancer properties. However there have been no reports on the anticancer activity of hydrophobic fractions of S. latifolia. In this study, the anticancer activities of S. latifolia extract and hydrophobic fractions were investigated using AGS (stomach cancer), A529 (lung cancer), and HepG2 (liver cancer) cell lines. In cytotoxicity results of A529 cells, fractions of A2, A3, A4, A6, A7, A8, A9, and A10 in all 12 fractions show low $IC_{50}$ values. For HepG2 cells, A7 fraction results in the lowest $IC_{50}$ value while A7, A8, and A11 fractions show low $IC_{50}$ values in AGS cells. S. latifolia extract lead to low cell viability in cancer cells, compared to positive control of paclitaxel. A compound with molecular weight of 181 were detected using HPLC-MS but not identified yet. As a result, the hydrophobic fractions of S. latifolia EtOH extract would be a possible candidate as natural anticancer agents in the future.

Cultivation characteristics and yield of Sparassis latifolia depend on the substrate mixture (꽃송이버섯(Sparassis latifolia) 생육배지 조성에 따른 재배특성 및 수량)

  • We investigated the standard cultivation substrate for Sparassis latifolia "Neowul" bred in Jeollabuk-do Agricultural Research and Extension Services. Cultivation characteristics and yield were assessed by varying the kind of sawdust and additives, and the mixing ratio. The cultivation period in larch sawdust was the shortest at 97 days. The yield was excellent (143.6 g). The findings indicated that larch is a tree species appropriate for the cultivation of S. latifolia. When the additives were varied, the yield and productivity (53.1%) were highest (116.6 g) for the wheat bran treatment. Thus, wheat bran would be an additive appropriate for culturing S. latifolia. Concerning the use of different mixing ratios, larch sawdust:beet-pulp:wheat bran ratios of 80:15:5 and 85:10:5 resulted in yields of 114.4 g and 111.4 g, and productivity of 52.5% and 51.8%, respectively. These yield and productivity values were not statistically different. Thus, the standard cultivation substrate for S. latifolia can comprise larch sawdust, beet pulp, and wheat bran at a ratio of 80:15:5 or 85:10:5. The carbon/nitrogen (C/N) ratio assumed to be appropriate for the cultivation of S. latifolia was 184-223. Pinheading would be difficult below a C/N substrate ratio of 105. Thus, the C/N ratio of the media, as well as the pH, would be vital factors affecting pinheading during S. latifolia cultivation.

  • Sparassis latifolia is a useful medicinal mushroom that has recently gained popularity in Asia. It has a rich flavor and is a good source of nutrients contains a large number of polyphenols for a functional food or dietary supplement. In addition, S. latifolia is rich in beta-glucan and gamma-aminobutyric acid (GABA). These two compounds have been reported to show immune-stimulating and anticancer effects by numerous studies. In this study, four species of lactic acid bacteria (Lactobacillus plantarum subsp. plantarum, L. acidophilus, L. helveticus, and L. delbrueckii subsp. bulgaricus) were used to ferment the fruiting body of S. latifolia. Fermented S. latifolia extracts were found to have a higher polyphenol content and antioxidant activity following fermentation as well as increased protease activity.

The Quantities of Methyl Orsellinate and Sparassol of Sparassis latifolia by Host Plants (기주식물에 따른 꽃송이버섯의 Methyl orsellinate와 Sparassol의 함량)

  • It is known not only that antifungal compounds such as sparassol, methyl orsellinate (ScI) and methyl-dihydroxy-methoxy-methylbenzoate (ScII) were produced during submerged culture from Sparassis crispa, but also that ScI and ScII were appeared higher antifungal activity than sparassol. The aim of this study, antifungal compounds of Sparassis latifolia were purified from mycelial culture media and identified by using NMR and ESI-MS. Based on HPLC analysis, methyl orsellinate and sparassol were detected at 15 min and 31 min of retention time, respectively. The compounds derived from S. latifolia were classified into four production patterns according to their strains. The strains originated from host plant Larix kaempferi and Pinus koraiensis showed different patterns of compound production, whereas the strains originated from host plant P. densiflora and Abies holophylla showed almost same patterns. There was no correlation between mycelial biomass and compound production. KFRI 645 strain from L. kaempferi exhibited higher methyl orsellinate production (0.170 mg/ml). Sparassol was produced by KFRI 747 from P. densiflora (0.004 mg/ml). Thus, our result revealed the new fact that methyl orsellinate and sparassol have different patterns according to the strains originated from different host plants.

Effect of addition amino acids on the mycelial growth and the contents of β-glucan and γ-aminobutyric acid (GABA) in Sparassis latifolia (아미노산 첨가가 꽃송이버섯 균사체 성장 및 베타글루칸, GABA 함량 변화에 미치는 영향)

  • Sparassis latifolia (formerly S. crispa) is used in food and nutraceuticals or dietary supplements, as rich in flavor compounds and ${\beta}-glucan$. Some previous studies have reported the effects of mushroom on brain function, including its neuroprotective effect. Thus, for this mushroom to be used as an effective nutraceutical for brain function, it would be desirable for it to contain other compounds such as ${\gamma}-aminobutyric$ acid (GABA) in addition to ${\beta}-glucan$. In this study, the enhancement of growth and GABA production in the mycelium of medicinal and edible mushroom S. latifolia was investigated. Amino acids were added externally as the main source of nutrition, and the effects of amino acids were investigated using liquid medium, specifically amino acid-free potato dextrose broth (PDB). The amino acids added were L-glutamic acid (named PDBG medium) and L-ornithine (named PDBO medium). The growth of mycelia was determined to be $0.9{\pm}0.00g/L$, $2.2{\pm}0.16g/L$, and $1.93{\pm}0.34g/L$ PDBG respectively. The GABA content was $21.3{\pm}0.9mg/100g$ in PDB medium, and it in PDBG 1.4% medium, at $115.4{\pm}30.2mg/100g$. However, the PDBO medium was not effective in increasing the GABA content of mycelia. Amino acids had little effect on the ${\beta}-glucan$ content of mycelia. The ${\beta}-glucan$ content was $39.7{\pm}1.4mg/100mg$, $34.4{\pm}0.2mg/100mg$, and $35.2{\pm}9.2mg/100mg$ in PDB, PDBG 1.8% and PDBO 1.4% media, respectively. Addition of glutamic acid and ornithine positively affected the growth of S. latifolia mycelia, and glutamic acid positively affected GABA production; no degradation of GABA was observed with addition of glutamic acid.

Enhancement of ${\beta}$-Glucan Content in the Cultivation of Cauliflower Mushroom (Sparassis latifolia) by Elicitation

  • The effectiveness of three kinds of enzymes (chitinase, ${\beta}$-glucuronidase, and lysing enzyme complex), employed as elicitors to enhance the ${\beta}$-glucan content in the sawdust-based cultivation of cauliflower mushroom (Sparassis latifolia), was examined. The elicitors were applied to the cauliflower mushroom after primordium formation, by spraying the enzyme solutions at three different levels on the sawdust-based medium. Mycelial growth was fully accomplished by the treatments, but the metabolic process during the growth of fruiting bodies was affected. The application of a lysing enzyme resulted in an increase in the ${\beta}$-glucan concentration by up to 31% compared to that of the control. However, the treatment resulted in a decrease in mushroom yield, which necessitated the need to evaluate its economic efficiency. Although we still need to develop a more efficient way for using elicitors to enhance functional metabolites in mushroom cultivation, the results indicate that the elicitation technique can be applied in the cultivation of medicinal/edible mushrooms.

Characteristics of mycelial growth and fruit body of Sparassis latifolia strains and selection of suitable incubation conditions in liquid spawn

  • Sparassis latifolia is called "Cauliflower Mushroom" and is known as an edible mushroom that has high content of ${\beta}$-glucan. Recently, artificial cultivation of S. latifolia has been done by bottle, plastic bag and wood cultivation in Korea. However it is not widely used because there are low incubation ratio and yield. For the high efficiency of production, we aim to find the superior strains and media for better mycelial and fruit body growth. First, we analyzed the genetic relationship among 31 strains and divided five groups with three kinds of URP primers. And then ten strains were selected from five groups based on the experiment of mycelial growth. The suitability of media for mycelial growth was different according to media type. The suitable solid and liquid media for mycelial growth of S. latifolia isolates were PDA and M2, respectively. In addition, with regard to C/N ratio, the mycelial growth increased even until C/N 160. Second, we investigated the production of fruitbody of the strains by plastic bag cultivation. The substrate was mixed with larch sawdust, corn flour, and wheat flour (8:1:1, v/v). Moisture content of substrate was controlled by about 60% with 10% molasses solution. Out of 31 strains, 19 strains formed primordia. The eight strains produced more than 140g/1kg in fresh weight. Third, molasses culture media was selected for the mycelial growth. And molasses suitable sugar content and input aeration were around 8Brix% and 0.3~0.6vvm, respectively. The longer the incubation period is, the more dried weight of mycelia increased, but medium volume decreased. Therefore, the best incubation period was 9 to 11 days depending on strains. In the future, research project entitled development of culture system and new variety for stable production of S. latifolia will be considered as a new item.

  • Sparassis latifolia is one of the most expensive mushrooms in Korean market owing to its high ${\beta}$-glucan content and immunoactivity. However, because of the long cultivation period and high contamination rates, it has low production efficiency. Therefore, we first need to establish the optimum conditions for liquid spawn production to increase its production efficiency. As a result of experiments, molasses culture medium was selected for mycelial growth. Also, the optimum sugar content for molasses and amount of aeration used were approximately 8 Brix% and 0.3~0.6 vvm, respectively. Mycelial dry weight increases, while the medium decreases, as the incubation period increases. Therefore, to achieve maximum production efficiency, the incubation period of 9 to 11 days is appropriate.

Enhanced GABA content from sodium alginate-induced Sparassis latifolia influences dendrite development in primary cortical neurons (해조류 기반 엘리시터 처리에 의한 꽃송이버섯의 GABA 함량 증가 및 흥분성 신경세포의 수상돌기 발달 억제)

  • Sparassis latifolia is a fungus abundant in β-glucan and amino acids and is highly valued as a medicinal mushroom. Among amino acids, γ-aminobutyric acid (GABA) is a free amino acid and has biological effects, such as increase/decrease of hypertension, improvement of cerebral blood flow, and prevention of dementia. In this study, biological elicitors were used to increase bioactive substances as a biofortification method. Sodium alginate extracted from seaweed (Sargassum horneri, Sargassum fulvellum, Sargassum fusiforme) were used as the elicitor. The levels of β-glucan and GABA in the mycelium and fruiting body grown by adding the elicitor to the medium were investigated. Addition of sodium alginate positively affected GABA production and negatively affected the β-glucan production in these fungi. Sodium alginates extracted from S. fulvellum induced the highest increase in GABA in the mycelium and fruiting bodies. Moreover, we investigated the effects of the extracts from mycelium and fruiting bodies on dendrite development in primary cortical neurons. We found that the extract from the fruiting bodies of sodium alginate treated fungi with increased levels of GABA inhibited the dendrite outgrowth of excitatory neurons, but not inhibitory neurons.
