Studies on the Red Pigment Produced by Monascus sp. in Submerged Culture. Part I Isolation of Strain and Cultural Conditions of Pigment Produced (액침진탕 배양에 의한 Monascus sp..가 생산하는 적색색소에 관한 연구 제1보 균주의 분리 및 색소생산 배양 조건)

  • Fungi which produce red pigment were isolated from tapioca chips, Korean Koji, air, and plant leaves. Among the Fungi, T-1 strain was selected as test strain. This strain was identified as a Monatcus sp. by the morphological propeties. Various culture conditions, and physical and physiological characteristics of red pigment were studied. According to the studies of culture conditions, optimum condition was found to be pH 6.5; 4 days of incubation; temperature, 32~33 c: 3.5% of Tapioca chips powder as carbon source, 0.2% of sodium nitrate as nitrogen source and 100 ml of medium in the 500 ml Erlenmyer flask at a rotary shaker (rpm180) as aeration condition. Also effective levels of vitamins, amino acids and inorganic compounds was found to be l$\mu\textrm{g}$/ml of folic acid and niacin; 0.3% of L-arginine, L-glutamic acid and L-proline; and 0.001% of manganese dioxide giving good results.

  • Tetrahedral amorphous carbon(ta-C) films were deposited by the filtered vacuum arc(FVA) process. The FVA process has many advantages such as high ionization ratio and the ion energy, which is suitable for dense amorphous carbon film deposition. However, the energy of the carbon ion cannot be readily controlled by manipulating the arc source parameters. In order to control the film properties in wide range, we investigated the dependence of the film properties on the substrate bias voltage. The mechanical properties and the density of the film exhibit the maximum values at about -100 V of the bias voltage. The maximum values of hardness and density were respectively 54$\pm$3 GPa and 3.6$\pm$0.4 g/㎤, which are 3 to 5 times higher than those of the films deposited by RF PACVD or ion beam process. The details of the atomic bond structure were analysed by Raman and NEXAFS spectroscopy. The change in the film properties for various bias voltages could be understood in the view of the $sp^2$ and $sp^3$ bond fraction in the deposited films.

Saccharification and Ethanol Production from Chlorella sp. Through High Speed Extrusion Pretreatment (고속 압출 전처리 공정을 이용한 Chlorella sp. 당화 및 바이오에탄올 생산)

  • Among various pretreatment processes for bioethanol production, extrusion pretreatment, one of cheap and simple process was investigated to efficiently produce fermentable sugars from micro alga, Chlorella sp. The biomass was pretreated in a single screw extruder at five different barrel temperatures of 45, 50, 55, 60 and $65^{\circ}C$, respectively with five screw rotation speed of 10, 50, 100, 150 and 200 rpm. The pretreated biomass was reacted with two different hydrolyzing enzymes of cellulase and amyloglucosidase since the biomass contained different types of carbohydrates, compared to cellulose of agricultural by-products such wheat and corn stovers, etc. In general, higher glucose conversion yield was obtained as 13.24 (%, w/w) at $55^{\circ}C$ of barrel temperature and 100 rpm of screw speed conditions. In treating 5 FPU/glucan of cellulase and 150 Unit/mL of amyloglucosidase, ca. 64% of cellulose and 40% of polysaccharides in the micro alga were converted into glucose, which was higher yields than those from other reported data without applying an extrusion process. 84% of the fermentable sugars obtained from the hyrolyzing processes were fermented into ethanol in considering 50% of theoretical maximum fermentation yield of the yeast. These results implied that high speed extrusion could be suitable as a pretreatment process for the production of bioethanol from Chlorella sp.

Nitric Oxide Sensing Property of Gas Sensor Based on Activated Carbon Fiber Radiated by Electron-beam (전자빔이 조사된 활성탄소섬유 기반 가스센서의 일산화질소 감지 특성)

  • Activated carbon fibers (ACFs) were surface-modified by electron beam (E-beam) irradiation and used as a gas sensor electrode to investigate the effect of E-beam on nitric oxide (NO) gas sensing performance. XPS results showed that the oxygen component of ACFs surface treated by E-beam decreased and $sp^2$ bonded carbon of ACFs surface increased. These results were attributed to the structural transformation of ACFs surface irradiated by E-beam. NO gas sensitivity of the electrode composed of ACFs irradiated by100 kGy increased from about 4% to 8%, and the response time was also meaningfully enhanced from 360 s to 120 s. This is due to the fact that the $sp^2$ carbon bond increased by E-beam irradiation of activated carbon fibers, which significantly affects the resistance change of the electrode in NO gas sensing.

  • Pathogenicity and multiplication of entomopathogenic nematode, Steinernema carpocapsae Weiser, ere analyzed in two insect hosts, Spodoptera exiga (Hubner) and Sp. litura(Fabricius). The estimated ${LC}^{50}s$(lethal concentration of the infective juveniles to kill 50% of the host insect population) were not different between tow insect species or among their developmental stages on the filter paper assay, thought the actual numbers of the infected nematodes were varied among them. The significant variation, however occurred in nematode multiplication between tow inset hosts. Temperature also gave sosignificatn effect on nematode multiplication rate that it took 6 days after infection at $25^{\circ}C$, but did 12 days at $20^{\circ}C$ to show the maximal nematode population peak($\approx$500,000 infective juveniles(IJ) in a 5th instar larva of Sp. litura and $\approx$-100,000 IJ in a 5th instar larva of Sp. exigua).

Purification and Properties of Extracellular Inulinase of Pseudomouas sp. (Pseudomonas sp.가 생산하는 Inulinase에 관한 연구 -효소의 정제와 성질 -)

  • Two forms of extracellular inulinase, designated as PI and PII were detected in the crude enzyme preparation from n species of Pseudomonas isolated from soil. PI and PII were purified to homogeneity by ammonium sulfate fractionation, DEAE Sephadex A-50 chromatography, Sephadex G-100 and Sephadex G-200 gel filteration. Both isoenzymes catalyzed specifically and endowise the cleavage of the $\beta$-2,1-fructofranoside linkage of inulin, and displayed no action upon sucrose, raffinose and levan. The optimal pH values for the PI and PII enzyme were pH 5.5 and 6.0, respectively and the highest activity of the two enzymes was observed at 55$^{\circ}C$. The Km values of PI and PII were calculated to be 2$\times$10$^{-3}$M and 5$\times$10$^{-3}$M, respectively.

Bleaching of Lipids Extracted from Single Cell Oil Produced by Mortierella sp. (모르티에렐라(Mortierella)속 유래 단세포유지로부터 추출한 지방질의 탈색)

  • The deacidified oil obtained from the oleaginous fungus, Mortierella sp. (M-12) was bleached, after degumming, using activated clay under a 50-100 mmHg vacuum. The bleaching conditions were partially optimized as follows: activated clay, 1%, bleaching temperature $90^{\circ}C$, and treatment time 20 min. After bleaching, the color of bleached oil as determined by the Lovibond Tintometer, satisfied the specification for edible fats and oils. The bleaching process also decreased the contents of free fatty acids and phosphorus in the deacidified oil. The acid value of the bleached oil also satisfied the specification for edible fats and oils. It was early shown that the normal bleaching process can be used for the bleaching of heavily-colored microbial lipids for human consumption.

Characteristics of Mannitol Production by Leuconostoc sp. KY-002 (Leuconostoc sp. KY-002의 Mannitol 생산 특성)

  • The production of extracellular mannitol by a new mannitol-producing bacterium, Leuconostoc sp. KY-002 was studied in shake flask cultures. The new isolate has a capability of utilizing fructose and sucrose for mannitol formation. Maximum mannitol production was obtained with fructose as the sole carbon source. Under the optimal culture conditions, within 70 hours of incubation, a final concentration of 26 g/L of mannitol from 50 g/L fructose was obtained with an indicated yield of 52% based on fructose consumed. However, higher concentrations of fructose ranging from 100 to 250 g/L could not effectively be transformed to mannitol due to a lack of osmotolerance. The strain produced no other polyols such as glycerol and sorbitol as by-products. Yeast extract was the best nitrogen source and high levels of inorganic phosphate up to 10 g/L promoted mannitol formation. Any mineral ions and salts did not play important role in both cell growth and mannitol production. Nicotinic acid enhanced mannitol production by 16%. The optimum culture temperature and initial pH were $35^{\circ}C$ and 6, respectively.

  • A potent fungus producing milk clotting enzyme with fairly weak proteolytic activity was isolated from various soil and sewage, which the selected strain, SA-101, was identified as Mucor sp. with microbiological characteristics. Its milk clotting enzyme production was maximized when grown on 10g of wheat bran media added to 8ml of tap water containing 0.1M HCl for 60hrs at $30^{\circ}C$. This enzyme production was stimulated by addition of 6% lactose, 0.05% NaCl and reached a maximal level of 9810 unit/g wheat bran. The crude enzyme product could be produced effectively by salting out with ammonium sulfate fractionation and lyophilization. The ratio of milk clotting activity to proteolytic activity of crude enzyme product was lower than Hansen rennet, but resembled to Meito rennet. The optimal temperature of milk clotting activity of crude enzyme product was abound $60^{\circ}C$ on a substrate of 10% reconstituted skim milk containing 1/100M $CaCl_2$.

Characterization and Partial Purification of Storage Protein-i Receptor in the Fat Body of Hyphantria cunea (미국흰불나방 지방체에서 저장단백질-1의 수용체의 특성과 부분정제)

  • In vitro tissue culture of fat body of Hyphantria cunea in the medium containing [35S]-methionine reveaied that storage protein-i (SP-1) is taken up into fat body of prepupae and 1-day-old pupae. Using Western blotting and ligand binding method, we were able to identify the protein band of the SP-1 receptor protein. For the partial purification, the membrane proteins of fat body cells were solubilixed with 1% Triton X-1OO and applied to anion exchange chromatography. The results revealed the molecular weight of the receptor protein to be about 80 kl)a in SDSPAGE, and the P1 was estimated to be about 6.1. The mobility of the receptor protein in 8D8-PAGE was highly dependent on both temperature during electrophoresls and the condition of samples whether they were in reducing or nonreducing.
