Recruitment Potential of Cyanobacterial Harmful Algae (Genus Aphanizomenon) in the Winter Season in Boryeong Reservoir, Korea: Link to Water-level Drawdown (동계 보령호에서 수위 강하와 연계된 유해 남조류 Aphanizomenon sp.의 재입 잠재성)

  • Cyanobacteria Aphanizomenon population is widely distributed in the world, and well known as harmful algae by producing toxins and off-flavor materials, thus belonging to one of the taxa that became more interested in the field of limnoecology. In this study, the frequency, intensity, and duration of Aphanizomenon occurrence were increased with the abnormal drawdown of water level in the winter in Boryeong Reservoir, and the spatial and temporal characteristics of them are compared with each other in the perspective of hydrometeorology (1998 to 2017) and limnology (2010 to 2017). In Korea, Aphanizomenon flourished mainly in high temperature, and the appearance in the low temperature was rare in total five times. The harmful cyanobacteria Aphanizomenon was observed in the low temperature (December to February) in Boryeong Reservoir from 2014, and then reached a maximum value of $2,160cells\;mL^{-1}$ in January 2017. In addition, the period exceeding $1,000cells\;mL^{-1}$ at this time was more than 3 months. This was simultaneously associated with abnormal water level fluctuation in the low temperature ($<10^{\circ}C$). The large drawdown of water level in the winter season has the potential to promote or amplify the germination and development of harmful algae. Also, subsequent water quality and ecological impacts(e.g., algal toxins and off-flavor substances) need to be considered carefully.

Fluctuation of Bag-net Catches off Wando, Korea and the Effect of Sea Water Temperature (한국 완도해역 낭장망 어업의 어획량 변동과 수온의 영향)

  • Fluctuation of the yield of anchovy, Engraulis japonicus and species composition of by-catch were examined to clarify the effect of sea water temperature using samples by bag net off Wando, west southern of Korea from 1999 to 2001. In 1999 and 2001, sea water temperature were higher than 20$^{\circ}C$ from July to October, However that of 2000 was higher than 20$^{\circ}C$ from August to September owing to the strong tidal front formed in south western area of Korea from late spring to summer. Fish captured by bag net off Wando was composed of approximately 73 species which belonging to the juveniles and young stages. Among them, Gymnapogon urospilotus, Lagocephalus sp., Omobranchus elegans, Platycephalus indicus and Konosirus punctatus did not appeared in 2000 when tidal front were developed stronger than in 1999 and 2001. Yield of anchovy landed at fishery of Wando were 1,000 M/T in 1999, 620 M/T in 2000, 1,056 M/T in 2001 respectively and have a tendency to increase from July to August and reach to maximum from August to October.

Preservatory effect of stored 'Setoka'(Citrus sp.) using the noncontacted low temperature atmospheric pressure surface discharged plasma (비접촉식 저온 대기압 면방전 플라즈마를 이용한 저장 '세토카' 감귤의 선도유지 효과)

  • Activity of the noncontacted low temperature atmospheric pressure surface discharged plasma (LASDP) converts stable gas to ionized gas known as discharge or plasma. This ionized gas exhibits the antimicrobial activity. We examined the effects of 3 different storage treatments for 80 days on 'Setoka' : ambient storage (AS), low-tempperature storage (LTS), and low-temperature atmospheric pressure plasma+low-tempperature storage (PLTS). Total soluble solids showed no the significant differences between the 3 treatments. Acidity gradually decreased, and was 0.5% under AS after 30 days of storage. Fruit firmness increased by a few percent until 40 days of storage. Weight loss in AS was higher than for other treatments. After 80 days of storage, the decay ratio was significantly low in PLTS treatment: (AS, 50.5%; LTS, 5.6%; PLTS, 1.9%). In AS treatment, 73% of the rotten fruits were infected particularly with green and blue mold; however, only 1% of the rotten fruits were infected in case of PLTS treatment. In conclusion, LASDP treatment can prevent postharvest decay caused by fungi and is an efficacious alternative extending the shelf-life of citrus fruits.

Effect of Fermented Cudrania tricuspidata Fruit Extracts on the Generation of the Cytokines in Mouse Spleen Cells (발효 꾸지뽕(Cudrania tricuspidata) 열매 추출물이 마우스 비장세포의 cytokine 생성에 미치는 영향)

  • We investigated a physiological function by fermenting a medicinal mushroom, (Cudrania tricuspidata fruit). A fermentation using lactic acid bacteria and the extracts isolated from 70% ethanol fractionation was included in cultured mouse spleen cells for cytokine secretion. As a result, total polyphenol content improved by 47% by organic acid fermentation. This was regarded as immune activity in fermented C. tricuspidata fruits, as the levels of interleukin (IL)-2 and IL-4 secretion increased. In addition, when the extracts were treated with a stimulant lipopolysaccharide, the secretion of helper T (Th) 1 cytokines IL-2, IL-12, and tumor necrosis factor-${\alpha}$ was suppressed, while the secretion of Th2 cytokines IL-4, IL-5, IL-6, and IL-10 significantly increased. Therefore, this study suggests that fermentative C. tricuspidata fruit extracts can contribute to the suppression of cellular immune reactions induced by the expression of Th1 cells and activation of the expression of Th2 cells inducing humoral immune reactions associated with the antibody generation by B lymphocytes.

Inhibition of Browning in Yam Fresh-cut and Control of Yam-putrefactive Bacterium Using Acetic Acid or Maleic Acid. (초산 및 말레산을 이용한 생마 신선편이 갈변억제 및 생마 저온부패균의 제어)

  • To increase the consumer acceptability of yam and the shelf-life of fresh-cut yam, organic acid-treated fresh-cut yam was prepared. When uncontaminated fresh-cut yam was stored at $4^{\circ}C$ for 14 days after treatment with 1% (v/w) organic acids, the browning and microbial putrefaction of fresh-cut yam were inhibited by treatment of acetic acid or maleic acid, whereas treatment of citric acid and ascorbic acid, commonly used browning inhibitors in food industry, did not show apparent effects on the browning and putrefaction of yam. The Inhibitory effects of acetic acid or maleic acid were superior than those of NaOCl (100 ppm), hydrogen peroxide (100 ppm) or commercially available washing solution. Also, treatments of 1% acetic acid, or 1% maleic acid Into artificially-contaminated yam $(10^5\;CFU/g-yam)$ showed strong inhibition of browning and putrefaction during long term storage at $4^{\circ}C$. The growth inhibition test indicated that 0.1% is enough to inhibit the growth of psychrotrophic yam-putrefactive Pseudomonas sp., and treatment of 0.1% acetic acid, or 0.1% maleic acid inhibited the browning and microbial putrefaction of fresh-cut yam. Our results suggested long-term distribution of yam or other root crops products is possible by treatment of organic acid, such as acetic acid, combined with aseptic vacuum packaging technology.

Comparison of Plant Growth Promoting Methylobacterium spp. and Exogenous Indole-3-Acetic Acid Application on Red Pepper and Tomato Seedling Development (식물생장촉진 세균 Methylobacterium spp. 와 IAA 처리가 고추와 토마토 유묘의 생육에 미치는 영향)

  • A comparative study was performed in gnotobiotic and greenhouse conditions to evaluate the effect of exogenous application of indole-3-acetic acid (IAA) and inoculation of Methylobacterium spp. possessing 1-aminocyclopropane-1-carboxylate deaminase (ACCD) and IAA activity on red pepperand tomato seedling growth and development. Application of 1.0 ${\mu}g\;mL^{-1}$ IAA positively influenced root growth while high concentrations (>10.0 ${\mu}g\;mL^{-1}$) suppressed root growth of red pepper and tomato under gnotobiotic condition. On the other hand, inoculation of Methylobacterium strains with ACCD activity and IAA or without IAA enhanced root growth in both plants. Similarly, under greenhouse condition the inoculation of Methylobacterium sp. with ACCD activity and IAA enhanced plant fitness recorded as average nodal length and specific leaf weight (SLW) but the effect is comparable with the application of low concentrations of IAA. Seedling length was significantly increased by Methylobacterium strains while total biomass was enhanced by Methylobacterium spp. and exogenous applications of < 10.0 ${\mu}g\;mL^{-1}$ IAA. High concentrations of IAA retard biomass accumulation in red pepper and tomato. These results confirm that bacterial strains with plant growth promoting characters such as IAA and ACCD have characteristic effects on different aspects of growth of red pepper and tomato seedlings which is comparable or better than exogenous applications of synthetic IAA.

Quality Characteristics of Kimchi Fermented in Permeability-Controlled Polyethylene Containers (투과도 조절 플라스틱 용기에서 발효된 김치의 품질 특성)

  • Kimchi was fermented in permeability-controlled polyethylene containers, in glazed onggi (Korean ethnic earthenware) or glass bottles at $5^{\circ}C$ for 8 weeks. During 4 weeks of storage, kimchi fermented in the permeability-controlled container showed a stable fermentation pattern, in terms of changes in pH and acidity, compared with kimchi fermented in the other containers. With respect to changes in bacterial counts, kimchi fermented in polyethylene containers showed vigorous multiplication of lactic acid bacteria, especially Lactobacillus sp., but slow growth of total aerobic bacteria. The springiness of kimchi fermented in the polyethylene containers was optimal (about 10% more than that of glass bottle-fermented kimchi), and the overall acceptability and hardness of container-fermented kimchi were excellent upon sensory evaluation. The DPPH radical-scavenging activity of kimchi fermented in polyethylene containers was also greater (91%) than that of kimchi fermented in glazed onggi (73%) or glass bottles (63%). The $O_2$ and $CO_2$ permeabilities of the polyethylene containers were higher (458 and $357\;mmol\;h^{-1}\;m^{-2}\;atm^{-1}$, respectively) than were those of the other containers; the permeability ratio was 0.8. Glass bottles showed no permeance. The results indicate that permeability-controlled polyethylene containers may be used for kimchi fermentation.

Biological Activity and Biochemical Properties of Water Extracts from Bacillus subtilis-fermented Silkworm (Bombyx mori L.) Powder by Origin (산지별 고초균 발효누에의 이화학적 특성 및 생리활성)

  • The aim of this study was to investigate biological activity and biochemical properties of extracts from Bacillus subtilis-fermented silkworm (Bombyx mori L., SP) powder of different origin (Buan, Namwon, and Boeun). An additional aim was to determine the inhibition of cancer cell (B16-F10, HT-29, LNcaP, and MCF-7) proliferation and nitric oxide (NO) production from lipopolysaccharide (LPS)-induced RAW264.7 cells. Biological activities (${\alpha},{\alpha}^{\prime}$-diphenyl-${\beta}$-picrylhydrazyl [DPPH], free radical scavenging activity, fibrinolytic activity, antiproliferation activity, and anti-inflammatory activity) and biochemical properties (compositional amino acid contents, and mineral contents) were examined in water extracts from silkworm powder and B. subtilis-fermented silkworm powder. The highest amino acid contents were detected in Buan silkworm powder (BU). After fermented, the highest contents were found in B. subtilis-fermented Buan silkworm powder (BBO). The major minerals detected were K, Ca, and Mg. Rates of these minerals, especially those of Na increased after fermented. DPPH radical scavenging activity and fibrinolytic activity were stronger in the fermented group than non-fermented group. DPPH radical scavenging activity and fibrinolytic activity were highest in the extract from BBO. The inhibition activities of LNcaP and MCF-7 cells viability were significantly decreased in the BBO, and there was no inhibition activity in other cancer cells (B16-F10 and HT-29). An SRB assay of the cell viability of RAW 264.7 cells exposed to extracts of silkworm powder and B. subtilis-fermented silkworm powder revealed no toxicity in any of the groups. Compared with the LPS-treated group, the biggest reduction in NO production was detected in the BBO group. Based on these results, extracts from Boeun silkworm powder fermented with B. subtilis could be a candidate material as a dietary supplement for use in healthy functional foods.

Functional Properties of Soybean Curd Residue Fermented by Bacillus sp. (Bacillus firmus NA-1 균주와 Bacillus subtilis G7-D 균주를 이용한 발효비지의 기능성)

  • To convert the soybean curd residue (SCR) to functional food ingredient, alkaline fermentation of SCR was performed by Bacillus firmus NA-1 and Bacillus subtilis GT-D for 22 hr at $42^{\circ}C$. The micronized full-fat soy flour (MFS) was fortified to reduce the moisture content as well as to supply protein source. The mucilage and flavor productions in the fermented SCR were enhanced by the fortification of $20\%$ MFS. The peptide production from the SCR fermented with B. subtilis GT-D substantially increased when judged by the detectable amount of tyrosine $(480\;mg\%)$. The production of fibrinolytic enzyme was increased by the fermentation for 22 hr, indicating the relative activity of $62\%$ (B. firmus NA-1) and $47\%$ (B. subtilis GT-D), respectively. The SCR fermented by B. firmus NA-1 and B. subtilis GT-D indicated the consistency of $1.95\;Pa{\cdot}s^n\;and\;0.21\;Pa{\cdot}s^n$, respectively. After freeze-drying, the protease activity (615 unit/g) and a-amylase activity (180 unit/g) were obtained from SCR fermented by Bacillus firmus NA-1 and Bacillus subtilis GT-D, respectively.

  • The Polyvinyl alcohol(PVA) oxidase involved in PVA degradation by microorganism has been purified to homogeneity from culture broth of Xanthomonas campestris J2Y grown in a minimal medium containing PVA as a sole carbon source. The enzyme was purified by DEAE-cellulose chromatograpy and Sephadex G-150 gel filtration. The purified PVA oxidase was electrophoretically homogeneous both in the absence and presence of SDS. The molecular weight of the enzyme was estimated to be about 55,000 daltons by SDS-polyacrylamide gel electrophoresis and Sephadex G-150 gel filtration. The native enzyme existed as a monomer. The optimal pH and temperature was shown to be pH 7 and $37^{\circ}C$ respectively. The activity of enzyme was stable below $55^{\circ}C$ and between pH range of $5{\sim}11$. The enzyme activity was significantly inhibited by metal compounds such as $Ag^{2+},\;Hg^{2+}$. While, metal ions such as $Mn^{2+},\;and\;Cu^{2+}$ stimulated the reaction. Km value of the enzyme for PVA was $7.04{\times}10^{-2}mmol/{\ell}$.
