Antagonistic Assay of Bacillus spp. for Eco-friendly Biological Control of Melon Powdery Mildew (멜론 흰가루병 친환경 생물적 방제를 위한 Bacillus속 균의 길항력 평가)

  • Melon powdery mildew, caused by Podosphaera fusca, is one of the serious diseases of melon plant in Korea. In this study, we evaluated the effect of selected antagonistic bacteria on the inhibition of mycelial growth of various plant pathogens, and control of melon powdery mildew. Based on the 16S rDNA and gyrA gene sequences, the selected antagonistic bacteria, M09, M70, and M99-1, were identified as Bacillus velezensis. These bacteria not only inhibited the mycelial growth of 47~69% in various plant pathogens, but also significantly reduced the incidence of powdery mildew. The three strains selected in this study could be used as potential biological control agents for various plant diseases as well as melon powdery mildew.

  • The present study was undertaken to determine the inhibitory effects of orally administered ginseng saponins(SP), protopanaxadiol saponins (PD), and protopanaxatriol saponins(PT) on the development of morphine-induced tolerance and physical dependence in mice. The study also sought to determine the hepatic glutathione contents. which are closely related to the degree of detoxification of mine the effects of GS on morphine 6-dehydrogenase, which catalyzes the production of morphinone from morphine, and the roles of spinal descendign inhibitory systems in the production of antagonism. The results of the present study showed that GS, PD and PT administered orally inhibited the development of morphine induced tolerance and dependence. GS. PD and PT inhibited the reduction of hepatic glutathione concentration in mice treated chronically with morphine and the activity of morphine 6-dehydrogenase, and the activation of spinal descending inhibitory systems was inhibited by GS. So we hypothesized that the results were partially due to the dual action of the test drugs, the inhibition of morphinone production and the activated formation of morphinone-glutathinone conjugation, and the inhibition of the activatin of apinal descending inhibitory systems and the others.

Inhibitory effects of ginsenosides on basic fibroblast growth factor-induced melanocyte proliferation

  • Background: UV-B-exposed keratinocytes secrete various paracrine factors. Among these factors, basic fibroblast growth factor (bFGF) stimulates the proliferation of melanocytes. Ginsenosides, the major active compounds of ginseng, are known to have broad pharmacological effects. In this study, we examined the antiproliferative effects of ginsenosides on bFGF-induced melanocyte proliferation. Methods: We investigated the inhibitory effects of Korean Red Ginseng and ginsenosides from Panax ginseng on bFGF-induced proliferation of melan-a melanocytes. Results: When melan-a melanocytes were treated with UV-B-irradiated SP-1 keratinocytes media, cell proliferation increased. This increased proliferation of melanocytes decreased with a neutralizing anti-bFGF antibody. To elucidate the effects of ginsenosides on melanocyte proliferation induced by bFGF, we tested 15 types of ginsenoside compounds. Among them, Rh3, Rh1, F1, and CK demonstrated antiproliferative effects on bFGF-induced melanocyte proliferation after 72 h of treatment. bFGF stimulated cell proliferation via extracellular signal-regulated kinase (ERK) activation in various cell types. Western blot analysis found bFGF-induced ERK phosphorylation in melan-a. Treatment with Rh3 inhibited bFGF-induced maximum ERK phosphorylation and F1-delayed maximum ERK phosphorylation, whereas Rh1 and CK had no detectable effects. In addition, cotreatment with Rh3 and F1 significantly suppressed bFGF-induced ERK phosphorylation. Western blot analysis found that bFGF increased microphthalmia-associated transcription factor (MITF) protein levels in melan-a. Treatment with Rh3 or F1 had no detectable effects, whereas cotreatment with Rh3 and F1 inhibited bFGF-induced MITF expression levels more strongly than a single treatment. Conclusion: In summary, we found that ginsenosides Rh3 and F1 have a synergistic antiproliferative effect on bFGF-induced melan-a melanocyte proliferation via the inhibition of ERK-mediated upregulation of MITF.

  • The crystal structure of the title compounds with both coumarin and sulfonamide moieties were examined. These two groups have very special for their pharmaceutical and medicinal properties have been determined from single crystal X-ray diffraction data. In the title compound crystallizes in the monoclinic space group C2/c with unit cell dimension a = 28.633(3) ${\AA}$, b= 9.3215(7) ${\AA}$ and c= 24.590(2) ${\AA}$ [alpha & gamma=$90^{\circ}$ beta= $115.976(3)^{\circ}$]. In the structure The S1 atom shows a distorted tetrahedral geometry, with O1-S1-O2 [119.74 $(2)^{\circ}$] and N1-S1-C5 [$105.57(1)^{\circ}$] angles deviating from ideal tetrahedral values are attributed to the Thrope-Ingold effect. The sum of bond angles around N1 ($316.2(1)^{\circ}$) indicates that N1 is in sp2 hybridization. The Pyridine ring adopts boat conformation and pyran rings adopt a sofa conformation. The carboxylate group of atoms were disordered over two positions with site occupancy factors 0.598 (9):0.402 (9). Crystal structure and packing is stabilized by $C-H{\ldots}O$ intra and inter molecular hydrogen bond interactions.

  • The Xu-Tec process is also called the double glow surface alloying technology and is a new method of surface metallurgy which can produce an alloy layers with sp ecial phisical and chemical properties on the surface of common and inexpensive mater ials. Many super alloys and alloy steels, sueh as nickel base alloys, high speed steels and staiinless steels, have been produced by Xu-Tee Process on the surfaces of carbon steels. The depth of the alloy lasyers may vary from several microns up to 300 micr ons with alloying elements in a concentration of few percentage to 100%. World wide patents for Xu-Tec process have been granted in the United states, Canada, United Ki ngdom, Australia and Japan. High performance saw blades have been successfully produced by the Xu-Tee process with much simper processing steps and less cost than bimetal high speed saw blades. A comparison of the cutting times and wear rates of the Xu-Tee blades with the conventional bimetal blades has been made. The Xu-Tee bIases demonstrates sim ilar or better performance than bimetal blades. A Xu-Tec Unit for the commercial pr oduction of Xu-Tec saw blades has been designed and manufactured. This Unit can t reat 10,000 haek saw blades at one time. Three Xu-Tec hack saw blades production I ines have been set up in China. China.

Complete genome sequence of Salmonella Enteritidis MFDS1004839 isolated from food (식품에서 분리된 Salmonella Enteritidis MFDS1004839의 유전체 서열 분석)

  • Salmonella enterica subsp. enterica is a foodborne pathogen that has been detected throughout the world. Here, we present the complete genome sequence of Salmonella Enteritidis isolated from a commercial kimbap that caused foodborne illness in the Republic of Korea in 2014. Complete genome sequence analysis of Salmonella Enteritidis MFDS1004839 revealed a 4,679,649 bp chromosome and a 96,994 bp plasmid, with G + C contents of 52.2% and 49.3%, respectively. The chromosome and plasmid genome included 4,482 predicted protein-coding sequences, 84 tRNAs and 22 rRNAs genes.

  • Purpose: The purpose of this study was to evaluate the effects of standardized patient simulation on nursing students' communication skills, problem-solving skills, critical thinking dispositions, and clinical competency. Methods: A one-group pretest-posttest design was used. Data were collected from a convenient sample of 47 junior nursing students at W University. Scenarios to train SP and checklists to evaluate the students''competence were developed by our research team. The collected data were analyzed with descriptive analysis t-test, Pearson's Correlation Coefficient and Cronbach's ${\alpha}$ using SPSS WIN 21.0 Program. Results: The simulation practicum using a standardized patient was conducted based on four steps, namely, selections of scenario modules, standard patient training, implementation, and evaluation. The differences between the pre and post-test scores of problem-solving skills(t=-2.94, p=.005) and clinical competency(t=-2.84, p=.007) were statistically significant. But communication skill(t=-.32, p=.747) and critical thinking(t=-.68, p=.498) was not different in group. Conclusion: Standardized patient simulation in nursing education may be useful the improvement of problem-solving skills and clinical competency.

Geochemical characteristics and benthic faunal facies in the sediments around the Oenaro Island, southern part of Korea (외나로도 주변해역 퇴적물의 지화학적 특성과 저서 생물상)

  • In order to evaluate the relationship between geochemical characteristics and benthic facies of the sediments from the Oenaro Island where red tide proliferation is first observed every year including this year, surface and short multiple core sediment samples were analyzed in terms of geochemical and benthic facies variation. The contents of organic carbon, carbonate, and sulfide gas were relatively low. The variation in C/N ratios, which indicate nature of organic carbon, suggested that the organic carbon recorded in the study area is composed of mixtures of marine and terrigenous organic matters. The concentration of minor elements found at the surface and multiple core sediment samples were also low as well as the enrichment factors(Ef) for the seven heavy metals indicated that the sediment of this area is not polluted significantly. The macrobenthic faunal community comprised 61 species, and their mean density was 708 ind./m2. Polychaete worms were major taxa of this benthic community. A crustacean amphipod, Melita sp. was the most abundant species accounted for 20.7% of total abundance, and the small polychaetes such as Heteromastus filiformis, Paralacydonia poradoxa, Magelona japonica and Sigambra tentaculata were the next dominant species. The macrobenthos around the Oenaro Island were more diverse and abundant than that in Gamak Bay. The benthic communities in the study area sustained somewhat different species composition based on the cluster analysis and the MDS ordination. The benthic community health condition at three stations seemed to be unbalanced, and slightly polluted based on the biological index such as BPI and BC. There was no clear relationship between the geochemistry characteristic and the benthic faunal facies attributed by the micro-algal blooms in this coastal area.

Long-chain alcohols derived from the microalga Monoraphidium

  • This study was to investigate the composition and characteristics of long-chained alcohols extracted from the algal strain Monoraphidium 3s35. The production of biomass was optimized using different cultivation methods. Under the aerated growth condition, this strain yielded up to 37.26% extracts of dry weight and $576mgL^{-1}$ biomass. The major compounds of the extracts are mainly long-chained alcohols (89.24%), with carbon chain length ranging from 12 to 20. Interestingly, or the long-chained alcohols, 3-(2-Methoxyethyl)-1-nonanol, 3,7,11,15-Tetramethyl-2-hexadecen-1-ol and oleyl alcohol accounted for 53.68%, 23.45%, and 12.11%, respectively. Because of their amphipathic nature, these long-chained alcohols have been widely used in bioenergy production and cosmetics industry. Furthermore, Monoraphidium 3s35 produced 9.73% of $C_{17}$ and $C_{20}$ alkanes, which can be used as an important supplement for the petrodiesel-like fuel.

Buffered versus unbuffered local anesthesia for inferior alveolar nerve block injections in children: a systematic review

  • Background: The present study aimed to evaluate and compare the efficacy of buffered and unbuffered local anesthesia solutions during inferior alveolar nerve block (IANB) administration in children. Methods: PubMed, Ovid SP, and Cochrane databases were searched separately by two independent reviewers for potential papers published between 1980 and April 2020 using relevant MeSH terms and pre-specified inclusion and exclusion criteria. T Studies of IANB administration in children comparing buffered and unbuffered local anesthesia solutions were evaluated. The primary outcome evaluated was pain (perception and reaction), while the secondary outcome was the onset of anesthesia. Results: A total of five articles were included in a qualitative analysis; among them, four qualified for quantitative analysis of the primary outcome and three for quantitative analysis of the secondary outcome. A fixed-effects model was used to perform the meta-analysis. Pain perception (child-reported pain): Significantly lower pain scores were reported with buffered local anesthesia solution than with unbuffered solution (P = 0.006, MD: -0.32, 95% CI: -0.55 to -0.09). Pain reaction (observer-reported pain reaction in child): No significant difference was found between buffered and unbuffered solution in terms of observer-reported pain behavior in the child (P = 0.09, MD: -0.21, 95% CI: -0.46 to 0.04). Onset of anesthesia: A significantly lower duration of anesthesia onset was reported with buffered local anesthesia solution than with unbuffered solution (P = 0.00001, MD: -12.38, 95% CI: -17.64 to -7.13]. Conclusion: Buffering local anesthesia solution may reduce discomfort due to IANB injection administration and lower the initial onset time of anesthesia. More randomized control trials with adequate sample sizes should be carried out to validate the accuracy of these results.
