Hepatoprotective Effects of Potato Peptide against D-Galactosamine-induced Liver Injury in Rats

  • The effect of some peptides on hepatoprotection and cecal fermentation against D-galactosamine (GalN)-treated rats was studied. In acute hepatic injury tests, serum alanine aminotransferase (ALT), aspartate aminotranferase (AST), and lactic dehydrogenase (LDH) activities were remarkably increased after injection of GalN. However, potato and soybean peptides significantly decreased GalN-induced alterations of serum ALT and AST activities. Hepatic thiobarbituric acid-reactive substance (TBARS) concentration in GalN-treated groups fed potato and soybean peptides was significantly lower than that in GalN-treated control group. Hepatic glutathione level in the GalN-treated group fed potato peptide was significantly higher than that in GalN-treated control group. Furthermore, cecal Lactobacillus level in GalN-treated groups fed potato and soybean peptides was significantly higher than that in GalN-treated control group, and cecal short-chain fatty acid concentrations in GalN-treated group fed potato peptide were significantly higher than in GalN-treated control group. These results indicate that potato peptide may improve the cecal fermentation and prevent the GalN-induced liver damage in rats.

Preparation of Black Soybean Peptide and Its Effect on Weight Reduction in Rats

  • The large-scale preparation method for black soybean peptide (BSP; about 70% less than 10,000 Da; highly enriched with glutamic acid, aspartic acid, and arginine) was developed, and its effect on weight reduction and lipid profiles in rats was investigated. Sprague-Dawleymale rats were assigned to four dietary groups (high-fat diets containing 0, 2, 6, and 10% BSP) and fed four weeks to examine the effects of BSP. During the experiment, food intake was measured every two days and body weight was monitored two times a week. After the supplementation of BSP, liver and adipose tissues (epididymal, retroperitoneal and perirenal adipose tissue) in the rats were weighted and the lipid profiles in serum, liver, and feces were analyzed. At the results of body weight gain, liver and epididymal adipose tissue weight, BSP groups were more decreased than HF group (0% BSP), with greater decreases at higher BSP levels. The same patterns were shown in lipid profiles of serum, as BSP was increased, triglyceride and total cholesterol concentration decreased. The serum HDL-cholesterol level was increased with increasing at BSP levels. Total cholesterol concentration of liver and feces were decreased and increased, respectively, as BSP increased. The results confirm that BSP is involved in reducing the body weight and the improvement of lipid composition in serum and liver of rats and that BSP can be applied in weight reduction in the food products industry.

Effects of Popped Soybean on Concentration of Ruminal Peptide and Blood Amino Acids in Holstein Calves

  • This study conducted to evaluate effects of popped soybean on levels of ruminal peptides and blood amino acids in Holstein calves fed sudan grass hay as a forage source and popped (PSB) soybean as a concentrate supplement. At 0, 2, 4 and 6 h after feeding, rumen fluid and blood samples were collected from the rumen and jugular vein, respectively, and amino acids, peptides and other nitrogen-containing compounds in the rumen were analyzed. Ruminal pH tended to be higher in the RSB than in the PSB treatments, and declined upto 4 h after feeding, since then increased in both treatments. The concentrations of ammonia-N in all treatments increased upto 2 h after feeding, and then decreased gradually with time after feeding. The concentrations of ammonia N in the rumen were not significantly different between the treatments, however, those in RSB treatment appeared to be higher. Also, protein concentrations in the rumen were not significantly different between the treatments. Peptide productions were the highest at 2 h after feeding in the group fed RSB which is rapidly degradable in rumen, whereas those in the group fed PSB which is slowly degradable in rumen were maximized at 4 h after feeding. The concentration of total free essential amino acids in plasma was higher in the RSB treatment than in the PSB, but disappearance rates of these amino acids out of plasma was higher in the PSB treatment than in the RSB treatment. Disappearance rates of free non-essential amino acids in plasma were not significantly different between the treatments. Consequently, this study implies that the production of peptide and utilization of blood amino acid may be controlled by the modification of protein degradability.

된장으로부터 Angiotensin Converting Enzyme(ACE) 저해 Peptide의 분획 (Fractionation of Angiotensin Converting Enzyme(ACE) Inhibitory Peptides from Soybean Paste)

  • 식품 유래의 생리활성 peptide를 분리할 목적으로 전통발효식품인 된장으로부터 혈압강하기능을 가지는 ACE(angiotensin converting enzyme)저해활성 peptide를 분획하였다. 시판 된장의 동결건조분말을 냉수로 추출했을 때 총질소의 회수율은 추출시간 30분에 73.3%를 보였다. 용매추출액에서 고분자 polypeptide를 제거하고 저분자 peptide만을 얻기 위해 PM-10 membrane(Amicon)을 이용하여 3시간 동안 한외여과한 결과, 질소성분의 회수율은 80.8%, 염의 회수율은 99.2%에 달했고 투과액의 ACE $IC_{50}$$41.8{\mu}g/ml$이었다. 한외여과 투과액을 reverse phase prep-HPLC로 분획하여 7개의 획분을 얻었으며, 그 중 F5분획물의 ACE저해활성이 $IC_{50}=6.8{\mu}g/ml$로 비활성이 가장 높았다. 염은 F1분획물에서 모두 회수되었다. F5분획물을 ion exchange prep-HPLC로 다시 분획하여 얻은 5개의 모든 획분에서 높은 비활성을 보였다($IC_{50}=2.5{\sim}8.3{\mu}g/ml$). 그 중 F53분획물의 비활성이 가장 높았으며($IC_{50}=2.5{\mu}g/ml$), F53의 구성아미노산 분석 결과 histidine의 함량이 특징적으로 높았다.

Microplate법에 의한 된장유래의 항혈전 펩타이드 탐색 (Screening of Antithrombotic Peptides from Soybean Paste by the Microplate Method)

  • 전통식품인 된장으로부터 혈소판 응집 저해활성을 갖는 항혈전 펩타이드를 탐색하고자 하였다. 증류수로 추출한 된장액을 한외 여과하여 얻은 분자량 3,000 dalton 이하의 여액에 대하여, SD흰쥐의 세척혈소판 (washed platelet)을 이용하여 ADP 자극으로 유도되는 혈소판응집의 저해정도를 aggregometer에서의 탁도로 측정하였을 때, $96\;{\mu}g/ml$로 처리시 90% 가량의 응집 저해효과가 확인되었다. 추출물을 Dowex 50W X-2 강양이온 교환칼럼을 이용하여 pyridine-formic acid 완충액의 pH (2.8-5.0) 및 몰농도(0.1-3.0M)를 단계별로 증가시키면서 용출시켜 19개의 획분(No.B-18)으로 분리하였다. 이들 획분의 활성을 microplate를 이용한 방법으로 측정하였을 때, 전체적으로 앞의 방법보다는 감도가 낮게 나타났으나 모든 분획에서 응집억제 효과가 확인되었다. 이때 용량반응 곡선으로부터 구한 각 획분의 $IC_{50}$$10-1,000\;{\mu}g/ml)$의 범위를 보였으며, 대부분의 획분들은 양성대조구인 RGDS $(IC_{50},\;205\;{\mu}g/ml)$보다 높은 활성을 나타내었다. 또한, 후반부의 염기성 획분$(IC_{50},\;10-20\;{\mu}g/ml)$에서 된장추출물$(IC_{50},\;30\;{\mu}g/ml)$보다 활성이 높게 나타났다. 이들 중 활성이 가장 높은 16번 획분을 아미노산 분석한 결과, 펩타이드 부분에 histidine, arginine, alanine 등이 높은 비율로 존재하였다.

Effects of Gluten and Soybean Polypeptides on Textural, Rheological, and Rehydration Properties of Instant Fried Noodles

  • We investigated how the addition of polypeptides to instant fried noodle dough affects the dough properties, starch gelatinization, and textural properties of cup-type instant fried noodles. After comparing farinograph results of 100% wheat flour with 1% wheat flour substituted with gluten, there was a small difference in the mechanical dough properties. However, in the case of 1% wheat flour substituted with gluten peptides, the dough development time increased, dough stability decreased, and weakness increased. On the other hand, when gluten or gluten peptides were added, starch gelatinization did not change significantly. At the steaming stage, substitution with gluten peptides or soybean peptides markedly changed the molecular weight distributions of extractable polypeptides. Especially in the case of wheat flour substituted with 1% gluten peptides, the relative portion of low Mw extractable polypeptides (2.5-50 kDa) decreased more compared to a control. Also, the hardness and chewiness decreased in cooked cup-type instant fried noodles containing gluten peptides. This suggests that the addition of gluten peptides can reduce the rehydration time of cup-type instant fried noodles.

청국장으로부터 Angiotensin I 전환효소 저해 Peptide의 분리 (Isolation of Angiotensin I-Converting Enzyme Inhibitory Peptide from Chungkookjang)

  • 한국의 전통 대두 발효식품 청국장이 정장, 혈액순환 개선 둥의 기능성식품으로 부각되고 있다. 청국장은 발효가 되면서 미생물, 효소, 다양한 생리활성물질이 크게 증가하며, 그 중에는 peptide류도 포함된다. 청국장 Peptide류의 형성은 SDS-PAGE 분석을 통해 확인할 수 있었다. 청국장의 수용성 아미노산중, Tyr, Gln-Lys, Trp, Gln, Lys-Pro 등이 주요성분으로 발견되었고, Lys-Pro (0.083 mg/100 g 시료)가 HPLC에 의해 분리되었다. Lys-Pro은 angiotensin I-converting enzyme (ACE)저해효과 $(IC_{50}=32.1{\mu}m)$를 지니고 있었다. 고혈압군이 분말청국장 20 g을 복용하고 2시간 지났을 때, 수축기 혈압은 15 mmHg, 이완기 혈압은 8 mmHg떨어지는 강하효과가 있었다. 청국장은 ACE 저해제와 혈압강하효과를 갖고 있기 때문에 혈액순환개선에 상당한 도움을 주는 기능성 식품으로 개발될 수 있을 것이다.

단백질 분해 효소 처리가 청국장의 항산화 활성에 미치는 영향 (Effect of Enzymatic Hydrolysis by Proteases on Antioxidant Activity of Chungkukjang)

  • Alcalase, protamex 및 neutrase 효소처리가 청국장의 펩타이드 및 항산화 활성 증가에 미치는 영향을 동일한 조건으로 효소 처리한 대두분말과 비교하였다. 효소 20, 100 및 500 mAU를 $50^{\circ}C$에서 120분간 처리한 결과 청국장과 대두분말 모두에서 alcalase 및 protamex에 의한 펩타이드 생성이 neutrase보다 높게 나타났다. 항산화 활성은 청국장의 경우 alcalase 처리에 의해 DPPH, ABTS 및 hydroxyl radical 소거활성이 효소처리를 하지 않은 대조구와 비교하여 시료량에 따라 각각 37~57%, 59~106% 및 67~83%, protamex 처리에 의해 35~50%, 56~74% 및 52~75% 통계적으로 유의하게 증가하였다. 대두분말을 효소 처리한 시료에서도 유사한 결과를 보여 alcalase 처리에 의해 DPPH, ABTS 및 hydroxyl radical 소거활성이 각각 37~55%, 51~99% 및 67~83%, protamex 처리에 의해 36~44%, 44~66% 및 51~65% 증가하였다. 그러나 두 시료 모두에서 neutrase 처리에 의한 항산화 활성의 유의적 증가는 없었다. 이상의 결과는 alcalase와 protamex가 청국장에 특이적이지는 않으나 펩타이드 생성 및 항산화 활성 증가에 효율적임을 보여주고 있다.
