Effect of Egg White Lysozyme from Ogol Fowl on Preservation of Some Animal Foods (오골계 난백 lysozyme 이 몇가지 동물성 식품의 보존에 미치는 영향)

  • To elucidate the effect of egg white lysozyme from Ogol fowl on the preservation of milks, fishcurd and sausage, changes of pH, volatile base nitrogen content and viable cell count were investigated during the storage periods at $20^{\circ}C$, $30^{\circ}C$ and $37^{\circ}C$ after the addition of lysozyme in each foods. Volatile base nitrogen count of raw milk as marker of spoilage was lowest(63 mg%) in 0.05% lysozyme addition lot which was stored at $20^{\circ}C$ for 2 days, and its preservation effect by lysozyme at $30^{\circ}C$ was enhanced with addition of glycine(0.1%). Preservation effect by lysozyme in commercial milk at $37^{\circ}C$ and in fishcurd at $5^{\circ}C$ and $20^{\circ}C$ were also good, and when sausage was stored at $5^{\circ}C$ after treatment of lysozyme instead of sorbic acid, its preservation effect was acceptable.

Rearing Method for Ascotis selenaria (Lepidoptera: Geometridae) using an Artificial Diet (인공사료를 이용한 네눈쑥가지나방(Ascotis selenaria)(나비목: 자나방과) 실내 사육법)

  • This study was conducted to develop an artificial diet for the mugwort looper, Ascotis selenaria (Lepidoptera: Geometridae), which is an insect pest to leaves of citrus (Citrus unshiu). Corn and soybean powder were selected as main nutrient sources for larvae of A. selenaria after several diets consisted of wheat germ, corn, kidney bean and/or soybean were tested for larval development and survival. A higher amount of the main nutrients in the diet increased the larval survivorship. Addition of yeast and cholesterol in diet increased the larval survivorship. Finally the composition of diet was decided as followings; corn 100 g, soybean 100 g agar 25 g, Brewers' yeast 30 g, cholesterol 0.5 g, Vanderzant vitamin mixture 2 g, Wesson's salt mixture 2 g, sorbic acid 2 g, ascorbic acid 2 g, and methyl-4-hydroxybenzoate 2.5 g, and distilled water 1 liter. Development periods of larvae and pupae, survival rate and fecundity of A. selenaria reared on the diet were not significantly different with those on the host plant, citrus leaves. Larvae of early instars were reared in a group, while larvae of later instars (5-6th) were reared individually. Adult mating was conducted in a plastic cage and an oilpaper covered with a gauze was provided as an oviposition site.

Investigation of microbial contamination and use of food additives for pet foods in Gwangju, Korea (광주지역 내 유통 중인 반려동물 사료·간식의 미생물 오염도 및 식품첨가물 사용실태 조사)

  • This study was conducted for safety evaluation on 130 pet food products, which are distributed in Gwangju, South Korea. The microbial contamination part and the usage of food additives part were mainly investigated. The five microorganisms that we tested were total viable cell counts (TVC), Coliforms, Salmonella spp., Campylobacter spp., pathogenic Escherichia coli and there were 15 products that exceed the microbial criteria or detected food poisoning bacteria. Specifically, Coliforms (13 products, 10%), TVC (9 products, 6.9%), Salmonella spp. (2 products, 1.5%), and E. coli (2 products, 1.5%) were followed. On the other hand, food additives such as preservatives, antioxidants and sodium nitrite were detected in 61 products. Among the preservatives, sorbic acid and benzoic acid were detected in 58 (44.6%) products. In antioxidants, Butylated hydroxytoluene (BHT) was detected in 3 (2.3%) products. In addition, preservatives and antioxidants were detected in 8 of 20 products labeled as 'additive-free'. Microbial contamination tended to occur mainly in small-scale individual homemade feed stores, while food additives were all detected in pet shops and supermarkets. Currently, the criteria for microorganisms and food additives for pet foods are insufficient in Korea. So, it is necessary to establish detailed feed standards and specifications for companion animals.

Antifungal Activity of Polygodial against Food-Contaminants and Effects of Temperature and pH on the Action (Polygodial의 식품 오염 진균에 대한 항균 활성과 그 활성능에 미치는 온도 및 pH의 영향)

  • The antifungal activity of polygodial, which was isolated from Polygonum hydropiper used as a hot spice in Japan, was investigated through measuring its MICs and against food-contaminants including 4 strains of Saccharomyces cerevisiae, 3 strains of Zygosaccharomyces species, and 5 strains of Aspergillus species. All yeast-like fungi exhibited strong antifungal susceptivilities with MICs and MFCs below 3.13${\mu}g$/ml. Strains of Aspergillu species showed moderate with MIC of 25-50${\mu}g$/ml and MFC of 50-100${\mu}g$/ml. However these strains to sorbic acid, as already known, revealed very weak with MICs of 200-800${\mu}g$/ml and MFCs above 1600${\mu}g$/ml. In addition, effects of various pHs and temperatures on the antifungal activity of polygodial were tested against S. cerevisiae. At $4^{\circ}C$, the fungicidal activity of polygodial gradually increased with incubation time and reached the maximum(MFC of 3. 13${\mu}g$/ml) in 5 hours. At temperatures in the range of 30-$45^{\circ}C$, the activity of polygodial increased in proportion to temperature rise and in particular at $45^{\circ}C$ was possible with the concentration of 0.1${\mu}g$/ml. mOn the other hand, medium pH was also identified to affect the antifungal activity of polygodial. Namely, compared to neutral (pH7), acidic(pH 3) and basic(pH 9) medium synergized its activity (MIC and MFC) to 8- and 2- fold, respectively.

Antioxidant and Antimicrobial Activities of Smilax china Leaf Extracts (청미래 덩굴 잎 추출물의 항산화 및 항균 활성)

  • Hot-water and 70%-ethanol Smilax china leaf extracts were prepared, and their total polyphenol and flavonoid contents, DPPH-radical-scavenging ability, nitrite-scavenging ability, and antimicrobial activity were determined. The total polyphenol contents of the hot-water and ethanol extract were $5.433{\pm}0.171$ and $13.060{\pm}0.110mg/g$, respectively; their flavonoid contents were $1.599{\pm}0.017$ and $3.005{\pm}0.084mg/g$; their DPPH-radical-scavenging abilities, assayed at 1.0 mg/mL, were 33.6 and 92.3%; and their nitrite-scavenging abilities, assayed at 0.1-2.0 mg/mL, were 37.9-61.6 and 38.4-77.8%. The 70%-ethanol extract showed higher antimicrobial activity than the hot-water extract. The antimicrobial activities were high in Bacillus cereus, Vibrio parahaemolyticus, Salmonella typhymurium, and Staphylococcus aureus, in that order. The antimicrobial substances in the two extracts were maintained after heating at $65-125^{\circ}C$ for 30 min.

Studies on the Film forming Yeasts Isolated from Commercial Soy Sauce (제품(製品)간장에서 분리(分離)한 산막효모(産膜酵母)에 관(關)한 연구(硏究))

  • This study was conducted (1) to isolate the film-forming yeast from the commercially available soy sauce, (2) to identify the state of soy sauce fermentation by the use of yeasts, (3) to confirm characteristics of yeasts. The result were as follows. 1. These yeast strains in the soy sauce fermentation test showed full fermentation of whole sugar content, reduction of the pure extract and relative reduction in total nitrogen. Soy sauce color was somehow faded to lower the stability of soy sauce. 2. In anti-fungal activity test butylparaben at a higher level 60 ppm., sodium propionate 2,400 ppm, sodium benzoate 800 ppm., menadion 165 ppm, showed their anti-fungal effect, while alcohol did not show the effect in the 3% additive group. 3. The optimum sodium chloride concentration for these strains in the 2% G.Y.P. medium was 5% and optimum temperature was $30^{\circ}C$. The extinction temperature was $62^{\circ}C$ for strain No-1 and No-3, and was $65^{\circ}C$ for No-2 and No-4. 4. The film-forming soy sauce turned out in the gas chromatogram to possess much flavor of low boiling point as compared with the standard. These flavors were considered due to flavor spoilage of the soy sauce. 5. These isolated yeasts were identified Saccharomyces rouxii (film-forming yeast) in the Lodder's taxanomic study.

  • Potassium Sorbate (PS) is a potassium salt version of sorbic acid, which has antimicrobial and fungistatic features in foods. Therefore, PS is used as a food preservative against fungi and mold. PS has been found to be non-toxic even when taken in large quantities given its trait to be broken down in the body into water and carbon dioxide. Gap Junctional Intercellular Communication (GJIC) is essential in the maintenance of tissue homeostasis during development and differentiation. This study was made of the effects of PS on GJIC in WB-F344 rat liver epithelial (WB) cells. We found dramatic decrease of cell viability in time- and dose-dependent manners when WB cells were treated with PS. The effect of PS on GJIC is strong inhibition, and it took place in parallel with a hyperphosphorylation of connexin 43 expression. The finding that PS interferes with gap junction functionality should be considered with respect to the mechanism of PS-induced hepatotoxicity.

Determination of Preservatives in Raw Materials of Functional Foods by HPLC-PDA and GC-FID (HPLC 및 GC를 이용한 건강기능식품 원료 중 보존료 함유량 조사)

  • Preservatives, as food additives, are occasionally intrinsic to natural raw materials or sometimes generated during the fermentation process as reported in many research articles. Preservative compounds in raw food materials may persist in the final food product, which is not supposed to include such preservative compounds. In this study, we validated an analytical method for preservative compounds in raw materials of functional foods. Quantification of benzoic acid and sorbic acid was determined using HPLC-PDA analysis after distillation, whereas propionic acid was quantified with GC-FID. A significant set of validation data (accuracy, precision, linearity, recovery, etc) was acquired. A total of 212 samples were collected for analysis of naturally occurred preservatives, and preservatives were detected in 85 samples. Most of the detected samples showed less 10 mg/kg of preservatives. The results of this study provide fundamental data on naturally occurring preservatives in raw materials of functional foods. Moreover, building up a database of naturally occurring preservatives could solve problems in the current scientific data.

Studies on Distribution, Characterization and Detoxification of Shellfish in Korea 1. A Study on the Distribution of Paralytic Shellfish Poison (한국산 주요패류에 대한 독의 분포, 특성 및 제독에 관한 연구 1. 마비성패류독의 분포에 관하여)

  • Paralytic shellfish poison(PSP) accmulate in shellfish as a result of feeding toxic dinoflagellates. The shellfish do not seem to be harmed by the toxins, but become toxic to humans and other animals that feed on them. The purpose of this study was to investigate the distribution and changes of PSP by species of shellfish, collected area and collected month. Also, the correlation between PSP and toxic dinoflagellate, Protogonyaulax tamarensis, was investigated. Five hundred and six samples of 13 kinds of shellfish for PSP bioassay were collected at the shellfish growing area of Pusan, Masan, Chungmu, $Samch\check{o}npo, Y\check{o}su, Mokpo and Daech\check{o}n$ located in South Korea during the study period from May, 1985 to Octcber, 1987. Most of the samples submitted were free from PSP except sea mussel, short - necked clam and ark shell. Among the intoxicated samples, PSP was most often detected in sea mussel. PSP was detected mainly in spring$(February\~May)$ in the southern coast of Korea. In case of Pusan, exceptionally, toxic sea mussel have been found even June and July in 1987. The toxicity score of toxic shellfishes examined was ranged from 23.44 to $150.26{\mu}g/100g$ of edible meat and toxicity of sea mussel was higher than other toxic shellfishes. By the study of anatomical distribution of PSP in sea mussel collected at Masan in Febuary and March, 1986, the toxin accumulated in digestive gland was about $70\%$ of all. There was no significant correlation between toxicity of sea mussel and cell numbers of P. tamarensis that one of the causitive organism of PSP during the studying period in Masan area. There was almost no difference in toxicity of sea mussel by water depth of collection, but toxicity of surface shellfish was a little higher than those of 3.5, and 7.0m depth.

  • Collection of organisms for the fisheries resources community were obtained with a set net during 8 months between May, 1985 and May, 1986 on the coast of Pukchon, Cheju Island, Korea. Most of organisms, representing 36 species, were less than 20cm $(93\%)$ in length, and 130g $(91\%)$ in weight. The four most abundant species were jack mackerel, Trachurus japonicus ; squid, Todarodes pacificus : damsel fish, Chromis notatus : and rabbit fish, Siganus fuscescens, which comprised about $83\% in number and about $73\%$ of the total catch. The diversity index of the number of species and information indices for individual and catch were the highest in October and the lowest in Septemer, indicating a close relationship with the change between the species number and catch at that time. A cluster analysis of 17 species was illustrated from the similarity matrix. All of the 17 species were grouped at the 0.2 similarity level. Three groups were present at 0.60 level, whereas species in other groups were sporadic in occurrence.
