For breeding of a new rootstock for eggplant production, somatic hybrids between two species, Solanum integrifolium and S. sanitwongsei were obtained through protoplast fusion. The former species has been commonly used for rootstock for eggplant production in Japan. Eggplants on these rootstocks are more productive than ungrafted plants, but are susceptible to bacterial wilt caused Ralstonia solanacearum. While the latter species is resistant, the growth of eggplants on this rootstock is rather slow and low yield. Protoplast of both species were isolated from cotyledons, and inactivated with iodoacetamide or UV-irradiation, then fused electrically. The fused products were then cultured. Regenerated plantlets were then transplanted on soil then maintained in a green house. The plants were classified into four groups. Those in the first group showed morphological characters intermediate of the parentalspecies. The plants bore fruit with viable seeds. The plants showed a chromosome number of 2n=48, the sum of those of the parental species, and are suggested to be symmetric fusion products. While plants in the other groupswas less vigorous and showed chromosome number 2n= 68 to 72 suggesting asymmetric fusion products by genomic in situ hybridization(GISH). Isozyme pattern of shikimate dehydrogenase (SKDH; EC 1.1.1.25), isocitrate dehydrogenase (IDH; EC 1.1.1.41) and phosphoglucomutase (PGM; EC 2.7.5.1) showed that 24 regenerated plants in three groups were somatic hybrids. Analysis of random amplified polymorphic DNA (RAPD) showed that 43 S. integrifolium-specific and 57 S. sanitwongsei-specific bands were all found in 24 plants. Both somatic hybrids and its S1 plants were found to be resistant to bacterial wilt, and eggplant grafted these plants using for rootstocks were more productive than grafted mother plants. Now, S1 progenies are used for commercial eggplant production in Osaka Prefecture.

This study analyzed the distribution characteristics of invasive alien plants on the islands of the Korean Peninsula. Ten species, Rumex acetosella, Sicyos angulatus, Solanum carolinense, Ambrosia artemisiaefolia var. elatior, Ambrosia trifida, Aster pilosus, Eupatorium rugosum, Hypochaeris radicata, Lactuca scariola, and Paspalum distichum were present on 68 islands. These ecosystem-disturbing invasive alien plants appeared extensively on the islands of Gyeonggi province and Jeollanam province. The proportion of the plants on the inhabited islands (41 places, 2.15 species on average) was higher than that on the uninhabited islands (27 places, 1.07 species on average). This means that the distribution of invasive alien plants was closely related to human activity. The distribution of Ambrosia artemisiaefolia var. elatior(32 islands) and Rumex acetosella (31 islands) was the widest, and there were distinct distribution differences according to species. In the island area, the physical environment is poor compared to the mainland, and the native space is limited. Therefore, when invasive alien species enter and settle on the island, the native island plants can be damaged more than those on the mainland. In this regard, the discussion of the distribution of invasive alien plants in the island region can contribute to the conservation of biodiversity in the region.

This work aimed to evaluate 11 potato families for insect resistance horticultural traits. The families were derived from crosses between introduced insect resistant and adapted genotypes. A randomized complete block design, with three replications was used. The plot consisted of 25 genotypes of a family, summing up to 75 genotypes tested per family. Two susceptible cultivars were included in the experiment as checks. The genotypes were evaluated for insect resistance, tuber yield traits, tuber appearance and tuber skin smoothness. The genetic value for the intensity of insect attack in the leaves and in the tubers was lower(higher resistance) in the 11 families than in the checks. For yield traits, there was no predominant effect among the families regarding the origin of the resistant donor species(Solanum berthaultii and/or S. chacoense). However, there was a large contribution of the C-1485-16-87 recurrent parent in crosses with resistant genotypes for tuber yield and tuber number. In relation to tuber appearance, only the family derived form the C-1485-16-87/ ND140(S. berthaultii) cross did not differ from the checks. had genetic value tuber skin smoothness, it was identified that families derived from S. berthaultii resistant parents showed higher genetic values than the check cultivars. For tuber skin smoothness, the genetic values of the families neither differed significantly from each other nor from the checks.
The efficacy of plant breeding has been enhanced by application of molecular markers in population screening and selection. Pepper (Capsicum annuum L.) is a major staple crop that is economically important with worldwide distribution. It is valued for its spicy taste and medicinal effect. The aim of this study was to discover single nucleotide polymorphisms (SNPs), microsatellite markers information, and percentage sharing through orthologous analysis of pepper-specific pungency-related genes. Here, we report the results of transcriptome analysis and microsatellite markers for four pepper varieties that possess a pungency-related gene. Orthologous analyses was performed to identify species-specific pungency-related genes in pepper, Arabidopsis thaliana L., potato (Solanum tuberosum L.), and tomato (Solanum lycopersicum L.). Advancements in next-generation sequencing technologies enabled us to quickly and cost-effectively assemble and characterize genes to select molecular markers in various organisms, including pepper. We identified a total of 9762, 7302, 8596, and 6886 SNPs for the four pepper cultivars Blackcluster, Mandarine, Saengryeg 211, and Saengryeg 213, respectively. We used 454 GS-FLX pyrosequencing to identify microsatellite markers and tri-nucleotide repeats (54.4%), the most common repeats, followed by di-, hexa-, tetra-, and penta-nucleotide repeats. A total of 5156 (15.9%) pepper-specific pungency-related genes were discovered as a result of orthologous analysis.
Plant cells from which the cell walls have been enzymatically or mechanically removed are called protoplasts. The protoplasts are theoretically totipotent and can be used as sources of somatic cell fusion in practical breeding programs. Wild Solanum species have often been used as sources of important agricultural traits including diverse disease resistance. However, they cannot often be directly applied to breeding programs due to their sexual incompatibility with S. tuberosum. Somatic hybridization via protoplast fusion is one of the ideal methods to overcome this limitation and to introgress certain traits into S. tuberosum. This technique has still widely been used in potato since the first fusion was reported in 1970s. Therefore, this review highlights general perspectives of protoplast fusion and discusses the application of protoplast fusion in potato breeding.
Bacterial species isolated from common scab lesion on potato (Solanum tuberosum L. cv. Dejima) was identified as Streptomyces acidiscabies. This organism had flexuous spore chains and white spore mass color, produced melanin pigment on tyrosine agar medium but did not produce on peptone agar medium. S. acidiscabies grew on agar medium at pH 4.0, used L-arabinose, D-fructose, D-glucose, D-mannitol, rhamnose, sucrose, D-xylose and meso-inositol except reffinose as carbon sources. It was also susceptible to thallium acetate (10 $\mu\textrm{g}$/ml, 100$\mu\textrm{g}$/ml), phenol (0.1%, wt/vol), streptomycin (20 $\mu\textrm{g}$/ml), and was resistant to 7% NaCl, crystal violet (0.5 $\mu\textrm{g}$/ml), penicillin (10 IU/ml) and oleandomycin (25 $\mu\textrm{g}$/ml, 100 $\mu\textrm{g}$/ml).
This study attempts to examine farmers' preference and perception towards cropland agroforestry (CAF) and its economic benefits in Bangladesh. It surveys 84 farmers of two sub-districts named Manirampur and Bagherpara under Jessore district of Bangladesh to address the study objectives with the help of a questionnaire during the period of June to July 2013. We follow a multistage random sampling procedure for selecting respondents of the survey. A total of 27 plant species under 19 families are identified in the surveyed crop fields, among which 11 are tree species and 1 is shrub from 8 families and 15 species are agricultural crops from 11 families. According to the survey findings, most of the farmers prefer multipurpose tree species like Swietenia macrophylla (67 percent), Phoenix sylvestris (48 percent), Mangifera indica (48 percent) and Cocos nucifera (43 percent). We also find that Curcuma longa (92 percent), Oryza spp. (56 percent), Solanum melongena (43 percent) and Amorphophallus campanulatus (33 percent) are the available agriculture crops which are grown in association with trees in the study area. The surveyed farmers report that they practice CAF to get fuel wood, fodder, juice, fruit and food for family consumption and revenue earnings. About 76 percent of the surveyed farmers endorse the existence of a positive interaction between trees and agriculture crops, while the rest 24 percent endorse the existence of a negative interaction between trees and agriculture crops. This study finds that CAF farmers on an average earn US$ 1,410 per farm per year and the yearly average revenue difference between CAF and non-cropland agroforestry (NCAF) farmers is US$ 214. Overall, CAF needs to develop through scientific intervention in the study area to conserve the biodiversity and to enhance farmers' sustainable livelihood.
This report describes methods for selecting informative single nucleotide polymorphisms (SNPs), and the development of an online Solanaceae genome database, using 234 tomato resequencing data entries deposited in the NCBI SRA database. The 126 accessions of Solanum lycopersicum, 68 accessions of Solanum lycopersicum var. cerasiforme, and 33 accessions of Solanum pimpinellifolium, which are frequently used for breeding, and some wild-species tomato accessions were included in the analysis. To select tag-SNPs, we identified 29,504,960 SNPs in 234 tomatoes and then separated the SNPs in the genic and intergenic regions according to gene annotation. All tag-SNP were selected from non-synonymous SNPs among the SNPs present in the gene region and, as a result, we obtained tag-SNP from 13,845 genes. When there were no non-synonymous SNPs in the gene, the genes were selected from synonymous SNPs. The total number of tag-SNPs selected was 27,539. To increase the usefulness of the information, a Solanaceae genome database website, TGsol (http://tgsol. seeders.co.kr/), was constructed to allow users to search for detailed information on resources, SNPs, haplotype, and tag-SNPs. The user can search the tag-SNP and flanking sequences for each gene by searching for a gene name or gene position through the genome browser. This website can be used to efficiently search for genes related to traits or to develop molecular markers.
This study was conducted to investigate the influence of hydrophilic polymer (HP) mixture ratio (Control, 1.0%, 2.5%, 5.0%, and 10.0%) on growth of eggplant (Solanum melongena) for lower maintenance urban agriculture via green roofs. Although it was not statistically significant (p > 0.05), substrate temperature was decreased as hydrophilic polymer mixture ratio were increased. High substrate water content (95%) was found consistently in growing media under elevated hydrophilic polymer mixture ratio at over 5% during the entire growing period. Substrate electronic conductivity was increased as hydrophilic polymer mixture ratio were increased. Growth index was decreased as hydrophilic polymer mixture ratio was increased. It was reduced about 1/3 and 1/5 compared to that of Control in HP5.0 and HP10.0 treatment plants, respectively. Number of leaves, leaf length, and leaf width were decreased in following order: Control> HP1.0> HP2.5> HP5.0> HP10.0 treatments. There numbers were significantly lower in HP5.0 and HP10.0 treatment plants. Dry weight of shoot and root were decreased as hydrophilic polymer mixture ratio was increased. They were reduced by 1/4 compared to those of Control treatment plants. In addition, visual value was decreased as hydrophilic polymer mixture ratio was increased. Plants grown in HP1.0, HP2.5, and HP5.0 treatments all survived. However, plants grown in the HP10.0 treatment had the lowest survival rate (56%) after 3 months of growing. These results indicate that the advantage of adding hydrophilic polymer to green roof growing media may greater during dry periods. However, the proper mixture proportion of hydrophilic polymer should be determined according to different characteristics of growing media and plant species.
