Production of Antifungal Compost by Using Bacillus licheniformis KJ-9 (Bacillus licheniformis KJ-9를 이용한 항균발효퇴비의 생산)

  • In order to produce environmental-friendly fermented compost, a cattle manure-sawdust compost (antifungal compost) was developed by inoculation of B. licheniformis KJ-9 to cattle manure-sawdust. The thermal stability of the antifungal substance produced by B. licheniformis KJ-9 maintained more than 60% antifungal activity with heat treatment at $100^{\circ}C$ for 10 min, and the optimum pH of antifungal activity of the substance was 7.0. In a pot experiment with red pepper, the antifungal compost increased 1.5~2 times in leaf number and stem and root growth rate compared to those of commercial compost. Also, the diameter of stems increased 1.5-3 times in the antifungal compost treated group. The amount of microbes increased markedly in soil supplemented with antifungal compost compared to the control. In the field experiment for cultivation of garlic and Perilla japonica, the growth of both crops was significantly enhanced in the field treated with antifungal compost as compared to the commercial compost.

Effects of Pyroligneous Acid on Control of Large Patch in Zoysiagrass (목초액을 이용한 한국잔디 Large Patch병 방제효과)

  • This study was carried out to investigate control efficacy of fungicides mixed with pyroligneous acid on Large patch caused by Rhizoctonia solani AG2-2. R. solani AG2-2 was not inhibited by concentration of 500 times and 1,000 times diluents of pyroligneous acid. It is thought that R. solani AG2-2 was not inhibited by Pyroligneous acid alone. Pencycuron and tebuconazol mixed with pyroligneous acid had a good effect in prevention of R. solani AG2-2 in zoysiagrass. Concurrently, it was observed that those were reduced $50\%$ of usage of fungicides. 500 times and 1,000 times diluents of pyroligneous acid treated group had a significantly effect compared to control turf in the leaf length, leaf width and dry weight. In conclusion, usage of fungicides could be reduced about $50\%$ by a fungicides mixed with pyroligneous acid for protection of R. solani AG2-2. Therefore, It is thought that fungicides mixed with pyroligneous acids can be reduced an economic load and also an environmental pollution such as a tolerance to pathogenic bacteria and a destruction of an ecological system of soil microbes according to use fungicides.

  • This paper proposes "A Smart Farm Environment Optimization and Yield Prediction Platform based on IoT and Deep Learning" which gathers bio-sensor data from farms, diagnoses the diseases of growing crops, and predicts the year's harvest. The platform collects all the information currently available such as weather and soil microbes, optimizes the farm environment so that the crops can grow well, diagnoses the crop's diseases by using the leaves of the crops being grown on the farm, and predicts this year's harvest by using all the information on the farm. The result shows that the average accuracy of the AEOM is about 15% higher than that of the RF and about 8% higher than the GBD. Although data increases, the accuracy is reduced less than that of the RF or GBD. The linear regression shows that the slope of accuracy is -3.641E-4 for the ReLU, -4.0710E-4 for the Sigmoid, and -7.4534E-4 for the step function. Therefore, as the amount of test data increases, the ReLU is more accurate than the other two activation functions. This paper is a platform for managing the entire farm and, if introduced to actual farms, will greatly contribute to the development of smart farms in Korea.

Comparison of bacterial communities in leachate from decomposing bovine carcasses

  • Objective: Burial is associated with environmental effects such as the contamination of ground or surface water with biological materials generated during the decomposition process. Therefore, bacterial communities in leachates originating from the decomposing bovine carcasses were investigated. Methods: To understand the process of bovine (Hanwoo) carcass decomposition, we simulated burial using a lab-scale reactor with a volume of $5.15m^3$. Leachate samples from 3 carcasses were collected using a peristaltic pump once a month for a period of 5 months, and bacterial communities in samples were identified by pyrosequencing of the 16S rRNA gene. Results: We obtained a total of 110,442 reads from the triplicate samples of various sampling time points (total of 15 samples), and found that the phylum Firmicutes was dominant at most sampling times. Differences in the bacterial communities at the various time points were observed among the triplicate samples. The bacterial communities sampled at 4 months showed the most different compositions. The genera Pseudomonas and Psychrobacter in the phylum Proteobacteria were dominant in all of the samples obtained after 3 months. Bacillaceae, Clostridium, and Clostridiales were found to be predominant after 4 months in the leachate from one carcass, whereas Planococcaceae was found to be a dominant in samples obtained at the first and second months from the other two carcasses. The results showed that potentially pathogenic microbes such as Clostridium derived from bovine leachate could dominate the soil environment of a burial site. Conclusion: Our results indicated that the composition of bacterial communities in leachates of a decomposing bovine shifted continuously during the experimental period, with significant changes detected after 4 months of burial.

In vitro Antimicrobial Activity of a New Isolate Streptomyces sp. BCNU 1030 (신규 분리균주 Streptomyces sp. BCNU 1030의 in vitro 항균활성)

  • This work focused on screening and characterizing antibiotic-producing actinomycetes to develop new antibiotics that can overcome the growing resistance of disease-causing microbes. One-hundred actinomycetes strains were isolated from soil samples from Chungcheongbuk-do, Korea using various kinds of actinomycetes isolation media, including a starch casein agar medium and potato dextrose agar (PDA). Among them, strain BCNU 1030 was determined to show strong antimicrobial activity against methicillin-resistant Staphylococcus aureus (MRSA). Biochemical, physiological, and 16S rRNA sequence analyses indicated that strain BCNU 1030 belonged to the genus Streptomyces. Strain BCNU 1030 exhibited antibiotic activity against a wide range of bacteria, especially methicillin-resistant Staphylococcus aureus (MRSA). The minimum inhibitory concentration (MIC) of BCNU 1030 dichloromethane extract was determined to be $0.78\;{\mu}g/ml$ for MRSA CCARM 3090. Therefore, Streptomyces sp. BCNU 1030 has potential for anti-MRSA drug development.

Principle and Application of Composting for Soils Contaminated with Hazardous Organic Pollutants (오염토양 정화를 위한 콤포스팅 기술의 원리와 적용에 관한 고찰)

  • Composting is a cost-effective and environmentally-sound technology to treat soils contaminated with hazardous organic pollutants. Pollutants to be treated are as follows: explosives, phenolic compounds, PAHs, petroleum hydrocarbons, pesticides, and etc. Composting systems are windrow, static pile, and in-vessel. Design and operational parameters of composting are aeration modes, temperature, moisture content, nutrient supplement, amendment added, and etc. Appropriate oxygen concentration of composting for contaminated soils are 5~15%, while some compounds are degraded well at the low $O_2$ concentration of 2~5%. The most diverse microorganisms live in the temperature of $25{\sim}40^{\circ}$. 50~90% of the soil field capacity is the moisture content not to make a problem in composting. Assuming a bacterial chemical equation is $C_{60}H_{87}O_{23}N_{12}P$, theoretical C : N : P from bacterial chemical portion is approximately 20 : 5 : 1. It should be noted that the ratio does not apply to the total organic carbon measured in a waste because not all carbon metabolized by bacteria is synthesized to new cellular material. Initial C/N ratio of 25~40 is optimum. It is more economical to recycle soils or composts than to add commercial microbes.

Estimating Rainfall Interception Loss of Decomposed Floor in a Deciduous Forest Using Rainfall Simulation Experiments (인공강우실험에 의한 활엽수 부후낙엽층의 강우차단손실량 추정)

  • Forest floor is one of most distinctive features of forest ecosystem, which provides plants and soil microbes with nutrients, and controls hydrologic condition within the floor by intercepting water during a rainfall event and evaporates back into the atmosphere. In this study rainfall interception loss by decomposed forest floor of a deciduous forest has been experimentally estimated using rainfall simulation experiments. Litter-decomposing fungi were incubated on deciduous forest floor samples for the experiment purposes. On a deciduous floor, a $4.22mm{\cdot}kg^{-1}{\cdot}m^2$ of rain was intercepted immediately before rain ceased. Minimum values of interception loss ranged from 1.62 to $2.41mm{\cdot}kg^{-1}{\cdot}m^2$, with an average of $1.87mm{\cdot}kg^{-1}{\cdot}m^2$. Mann-Whitney test showed that decomposing fungi on the forest floor influenced on rainfall interception capacity.

An Evaluation of Plant Growth Promoting Activities and Salt Tolerance of Rhizobacteria Isolated from Plants Native to Coastal Sand Dunes (해안사구의 토착식물로부터 분리된 근권세균의 내염능과 식물성장촉진능 평가)

  • Coastal sand dunes are important for ecosystems due to the variety of rare species that can be found in this kind of habitat, and the beautiful landscapes they create. For environmental remediation, a potential strategy is phytoremediation using the symbiotic relationship of plants and microbes in the rhizosphere, which has proven ecologically sound, safe, and cost effective. Ninety-five colonies were isolated from the rhizosphere soil (RS) or rhizoplane (RP) of Rorippa islandica, Rumex crispus, Artemisia princeps var. orientalis, Lilium sp Stellaria media, and Gramineae. These colonies were then tested for plant growth promoting activities (PGPAs) such as 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity, and siderphores synthesis ability. In addition, salt tolerance was evaluated at 4% and 8% salt concentrations. It was observed that amongst the test subjects about 50% of the strains had a high resistance to salinity. Many of them could produce indole-3-acetic acid (IAA) IAA (in RS 13.9% and in RP 7.6%), exhibited ACC deaminase activity (55.8% in RS and 36.6% in RP), and could synthesize siderphores (62.7% in RS and 50% in RP). Correlation coefficient analyses were carried out for the three kinds of plant growth promoting abilities (PGPA) and salt tolerance. A positive correlation was found between an ability to synthesize siderphores and ACC deaminase activity (r=0.605, p<0.037). Similarly, positive correlations were noted between salt tolerance and ACC deaminase activity (r=0.762, p<0.004, r=0.771), and salt tolerance and an ability to synthesize siderphores (r=0.771, p<0.003).

Antimicrobial activities of Bacillus subtilis DS660 and Paenibacillus polymyxa DS842 (Bacillus subtilis DS660과 Paenibacillus polymyxa DS842의 항균활성)

  • This study was carried out to isolate soil bacteria with antimicrobial activity and evaluate antimicrobial substances produced by isolated bacteria. Among many isolates Bacillus subtilis DS660 and Paenibacillus polymyxa DS842 showed high antimicrobial activities against 6 species of microbial residents on human skin and 3 species of pathogenic bacteria. DS660 and DS842 showed 15.3~26.8 and 11.3~27.5 mm of inhibition zone diameter, respectively on nutrient agar medium against most target bacteria and fungi. DS660 and DS842 produced $57{\pm}8$ and $170{\pm}15{\mu}mol/ml$ of siderophore, respectively as an antimicrobial substance. Analysis of ethyl acetate extract of culture supernatants of DS660 and DS842 suggested production of glycolipid biosurfactant which reduced surface tension of culture supernatant of DS660 and DS842 from 60.0 to 40.3 and 30.3 mN/m, respectively. DS660 and DS842 also showed $169.2{\pm}9.9$ and $357.2{\pm}13.7nmol/min/mg$ protein of ${\beta}-1,3$-glucanase activity, respectively, and hydrolyzed cell wall components of 3 bacterial species. These results suggest that B. subtilis DS660 and P. polymyxa DS842 may be utilized as an environment-friendly biocontrol agent against some skin microbes and pathogenic bacteria.

Analysis of Soil Microbes in Continuous Cropping Fields of Gastrodia elata (천마 연작지의 토양 미생물상 분석)

  • 천마(天麻, Gastrodia elata Blume)는 난초과(蘭草科, Orchidaceae)에 속하는 식물로 잎과 뿌리가 없어 탄소동화능력이 없으며, 뽕나무버섯균과 공생하는 기생식물이다. 천마는 노지 재배에 따른 안정생산 문제가 지속적으로 발생하고 있다. 혹한, 폭우 등 기상환경에 따른 연차간 수량성 차가 673~1,175kg/10a로 크고, 연작에 따른 수량성이 연작 1회 시 29%, 연작 2회 시 68%가 감소하는 경향을 보이고 있다. 따라서, 본 연구는 천마의 연작에 따른 토양내 미생물상의 변화를 분석하기 위해 수행하였다. 본 실험은 초작지, 연작 1회지, 연작 2회지 및 자생지의 각각 3곳에 대한 토양을 Metagenome 분석법을 활용하여 미생물상을 분석하였다. metagenome 분석 결과, 초작지 3곳의 시료를 이용하여 확보한 총 read는 699,221개였으며, 이 중에서 Eukaryota로 분류되지 않은 read는 1,377개(0.2%), no hit, not assign된 read는 10,510개 (1.5%), Bacteria로 분류된 read는 342,916개(49.0%), Eukaryota로 분류된 read는 총 344,418개(49.3%)였다. 그리고 천마의 생육에 영향을 주는 Fusarium 속과 종은 3곳 포장에서 고르게 분포하고 있으며, 총 11,242 read로 전체의 3.3%를 차지하였다. 연작 1회지 3곳의 시료를 이용하여 확보한 총 read는 655,097개였으며, 이 중에서 Eukaryota로 분류되지 않은 read는 1,694개(0.3%), no hit, not assign된 read는 18,985개(2.9%), Bacteria로 분류된 read는 312,201개(47.7%), Eukaryota로 분류된 read는 총 322,217개 (49.2%)였다. 그리고 천마의 생육에 영향을 주는 Fusarium 속과 종은 3곳 포장에서 고르게 분포하고 있으며, 총 11,597 read로 전체의 3.6%를 차지하였다. 연작 2회지 3곳의 시료를 이용하여 확보한 총 read는 651,624개였으며, 이 중에서 Eukaryota로 분류되지 않은 read는 1,753개(0.3%), no hit, not assign된 read는 7,995개(1.2%), Bacteria로 분류된 read는 307,178개(47.1%), Eukaryota로 분류된 read는 총 334,698개(51.4%)였다. 그리고 천마의 생육에 영향을 주는 Fusarium 속과 종은 총 43,877 read로 전체의 13.1%를 차지하였다. 자생지 3곳의 시료를 이용하여 확보한 총 read는 731,719개였으며, 이 중에서 Eukaryota로 분류되지 않은 read는 2,828개(0.4%), no hit, not assign된 read는 585개(0.1%), Bacteria로 분류된 read는 356,690개(48.7%), Eukaryota로 분류된 read는 총 371,616개(50.8%)였다. 그리고 천마의 생육에 영향을 주는 Fusarium 속과 종은 MJH01 자생지에서만 전체의 0.2%(총 660 read)로 극히 일부가 분리되었다. 따라서, 천마의 생육에 영향을 미치는 Fusarium 속과 종은 초작지 3.3%, 연작 1회지 3.6%, 연작 2회지 13.1%, 자생지 0.2%로 나타났으며, 연작 2회 시 급격히 증가하는 것으로 판단되었다.
