  • For the development of new antitumor antibiotics produced by microorganisms, Streptomyces sp. YBE-316 was isolated from soil. The productivity of the antitumor antibiotic from Streptomyces sp. YBE-316 gradually increased after 60 hours, and was maximum after 100 hours after inoculation in growth medium (2.0% sucrose, 1.0% soybean meal, 0.1% K$_{2}$HPO$_{4}$, pH 7.0) at 30$\circ$C, 150 rpm, 5 NL/min by 30 l jar fermentor. This antitumor antibiotic was present only in mycelium, and stable in pH 5.0-10.0 for 20 minutes at 100$\circ$C. Antitumor and antibiotic activities were maintained at neutral pH, and heat stability was low. This antitumor antibiotic was soluble in methanol and ethanol, and insoluble in water, ethyl acetate, chloroform, and n-hexane. This antitumor antibiotic was sequentially purified by acetone extraction from mycelium, butanol extraction, and silica gel column chromatography. Antitumor activity was low against most tested cell lines, but antibiotic activity was high and low against yeasts and bacteria, respectivelv. The visualization test showed that this antitumor antibiotic had higher hydroxyl, ketone, amino, carboxyl groups, and sugar(s) in its structure. Instrumental analyses showed that this antitumor antibiotic was a pentaene in polyene class antibiotics. In pentaene class antibiotics, this was considered as an eurocidin or capacidin type antibiotics. The molecular weight of this antitumor antibiotic was higher than 683.0 daltons, and this antitumor antibiotic might be glycosylated by other sugar(s), instead of mycosamine or perosamine, an amino sugar.

  • The effects of various 2, 4-D-degradative plasmids on the axenic growth patterns, the degradation phenotypes, and the competitiveness of different host bacteria were evaluated in liquid cultures; the organisms and plasmids used were Alcaligenes eutrophus JMP134/pJP4, Alcaligenes paradoxus/p2811, Pseudomonas pickettii/p712, pJP4, and p712 or p 2811 exhibited very different restriction fragment profiles in restriction endonuclease digests. These plasmids were transferred to the recipients (P. cepacia and Alcaligenes JMP228) at relatively high frequencies ranging from 8.9 $\times$ 10$^3$ to 1.6 $\times$ 10$^5$ per donar cell. In the axenic liquid cultures the fast-growing strains, such as P. pseudomallei/p745 and P. cepacia/pJP4, exhibited short lag periods, high specific growth rates, and high relative fitness coefficients, while the slow-growing strains, such as P. pickettii/p712 and A. paradoxus/p2811, had long lag periods, low specific growth rates, and low relative fitness coefficients. Depending on the type of plasmid containing the genes for the 2, 4-D pathway, some transconjugants exhibited intermediate grwoth patterns between the fast-growing strains and the slow-growing strains. The plasmid and plasmid-host interactions determined specific growth rate and lag time, respectively, which were shown to be principal determinants of competitiveness among the strains, but relative fitness coefficient derived from the axenic culture was not always predictive for the mixed culture condition.

  • In this study, we isolated about 70 bacterial strains from terrestrial and marine environments in Jeju island, and finally, total 21 strains were obtained based on the 16S ribosomal RNA gene sequence analysis. These isolated strains were classified into 16 genera of 5 classes and were identified as an unrecorded species in the Republic of Korea. As a result of the substrate utilization and capability for polymer degradation, the physiological phenotypes for acid resistance and halophilic bacteria were observed to be distinct from each other, except for some acid resistance strains. This study might provide basic information on utilization for indigenous microorganisms.

  • BACKGROUND: Biological iron redox transformation alters iron minerals, which may act as effective adsorbents for arsenate [As(V)] in the environments. In the viewpoint of alleviating arsenate, microbial Fe(III) reduction was sought under high concentration of As(V). In this study, Fe(III)-reducing bacteria were isolated from the wild plant rhizosphere soils collected at abandoned mine areas, which showed tolerance to high concentration of As(V), in pursuit of potential agents for As(V) bioremediation. METHODS AND RESULTS: Bacterial isolation was performed by a series of enrichment, transfer, and dilutions. Among the isolated strains, two strains (JSAR-1 and JSAR-3) with abilities of tolerance to 10 mM As(V) and Fe(III) reduction were selected. Phylogenetic analysis using 16S rRNA genesequences indicated the closest members of Pseudomonas stutzeri DSM 5190 and Paenibacillus selenii W126, respectively for JSAR-1 and JSAR-3. Ferric and ferrous iron concentrations were measured by ferrozine assay, and arsenic concentration was analyzed by ICP-AES, suggesting inability of As(V) reduction whereas ability of Fe(III) reduction. CONCLUSION: Fe(III)-reducing bacteria isolated from the enrichments with arsenate and ferric iron were found to be resistant to a high concentration of As(III) at 10 mM. We suppose that those kinds of microorganisms may suggest good application potentials for As(V) bioremediation, since the bacteria can transform Fe while surviving under As-contaminated environments. The isolated Fe(III)-reducing bacterial strains could contribute to transformations of iron minerals which may act as effective adsorbents for arsenate, and therefore contribute to As(V) immobilization

  • BACKGROUND: Methane is a major greenhouse gas attributed to global warming partly contributed by agricultural activities from ruminant fermentation and rice paddy fields. Methanotrophs are microorganisms that utilize methane. Their unique metabolic lifestyle is enabled by enzymes known as methane monooxygenases (MMOs) catalyzing the oxidation of methane to methanol. Rice absorbs, transports, and releases methane directly from soil water to its stems and the micropores and stomata of the plant epidermis. Methylobacterium species associated with rice are dependent on their host for metabolic substrates including methane. METHODS AND RESULTS: Methylobacterium spp. isolated from rice were evaluated for methane oxidation activities and screened for the presence of sMMO mmoC genes. Qualitatively, the soluble methane monooxygenase (sMMO) activities of the selected strains of Methylobacterium spp. were confirmed by the naphthalene oxidation assay. Quantitatively, the sMMO activity ranged from 41.3 to 159.4 nmol min-1 mg of protein-1. PCR-based amplification and sequencing confirmed the presence and identity of 314 bp size fragment of the mmoC gene showing over 97% similarity to the CBMB27 mmoC gene indicating that Methylobacterium strains belong to a similar group. CONCLUSION(S): Selected Methylobacterium spp. contained the sMMO mmoC gene and possessed methane oxidation activity. As the putative methane oxidizing strains were isolated from rice and have PGP properties, they could be used to simultaneously reduce paddy field methane emission and promote rice growth.

  • PET (Polyethylene terephthalate), a representative plastic material, has useful physicochemical properties such as high durability and economic feasibility, and is used in various industrial fields such as bottles, fibers, and containers. Due to the recent increase in plastic usage including disposable products, eco-friendly strategy using microorganisms have drawn attention differentiated from conventional landfill and incineration methods. In this study, a soil-derived Streptomyces javensis Inha503 containing a PETase gene was selected and the ability to hydrolyze PU (Polyurethane) was confirmed through agar plate diffusion assay. This strain was cultured with PET for a month, and PET decomposition ability was also confirmed through a scanning electron microscope. Moreover, cloning and heterologous expression of S. javensis Inha503 PETase gene exhibited PET activity in the PETase non-containing S. coelicolor, confirming for the first time the presence of functional PETase gene in Streptomyces species.

  • The global problem of handling radioactive materials is facing limitations. Eco-friendly bioremediation methods using microorganisms are being studied. This study was conducted to screen cesium-resistant microbial strains. M1 strain was selected from the soil sample by enriched culture in R2A medium containing 100 mM CsCl. In liquid medium containing above 40 mM of CsCl, the growth of M1 was inhibited in a concentration-dependent manner. Otherwise, M1 can survive up to 80mM CsCl in solid medium although the growth rate was slow and colony size was small. M1 strain was genetically identified as a strain of the genus Acinetobacter through 16S rRNA sequencing, and radiation resistance (D10 value) of M1 was found to be 0.307 kGy. These results showed that M1 strain is highly resistant to cesium and can grow in radiation environment. It was considered that M1 strain is useful in the field of biological decontamination of cesium.

  • This study investigated the effects of two mycorrhizal fungi, Pisolithus tinctorius and Oidiodendron maius, on the growth and age-specific responses of Korean fir (Abies koreana E.H.Wilson) seedlings. The experiment was conducted from March 2020 to March 2023 at the National Institute of Forest Science in Suwon, South Korea to evaluate plant height, root length, root collar diameter, and biomass in the treatment and control groups. Significant growth increases were observed in all treatment groups compared to the control, with the OI (O. maius) treatment demonstratingthe highest growth increase (p < 0.05). In one-year-old seedlings, the PT (P. tinctorius) treatment demonstrated overall higher growth than the OI treatment and control, with PT demonstrating an approximately 1.3 times increase than OI. The PT and OI treatments both exhibited higher growth than the control in the three-year-old seedlings, with less difference between the two treatments than in the one-year-olds. Both the treatments showed approximately 1.8 times more growth than the control in the five-year-old seedlings, and the OI treatment showed approximately 0.5 times more growth than PT. These results confirm that the OI treatment is generally more effective for the growth of Korean fir seedlings. Although the PT treatment was more beneficial in the one-year-old seedlings, OI was more effective in three-year-old and older seedlings. This suggests that early mycorrhizal inoculation has a significant long-term effecton growth, emphasizing the importance of age-specific mycorrhizal applications in forest conservation and restoration strategies.

  • An antagonistic strain, Burkholderia MP-1, showed antimicrobial activity against various filamentous plant pathogenic fungi, yeasts and food borne bacteria (Gram-positive and Gram-negative). The nucleotide sequence of the 16S rRNA gene (1491 pb) of strain MP-1 exhibited close similarity (99-100%) with other Burkholderia 16S rRNA genes. Isolation of the antibiotic substances from culture broth was fractionated by ethyl acetate (EtOAc) solvent and EtOAc-soluble acidic fraction. The antibiotic substances were purified through a silica gel, Sephadex LH-20, ODS column chromatography, and high performance liquid chromatography, respectively. Four active substances were identified as phenylacetic acid, hydrocinnamic acid, 4-hydroxyphenylacetic acid and 4-hydroxyphenylacetate methyl ester by gas chromatographic-mass spectrum analysis. The minimum inhibition of concentration (MIC) of each active compound inhibited the growth of the microorganisms tested at 250 to $2500{\mu}g\;ml^{-1}$. The antimicrobial activity of crude acidic fraction at 1 mg of dry weight per 6 mm paper disc was more effective than authentic standard mixture (four active substances were mixed with the same ratio as acidic fraction) over a wide range of bacterial test.

  • Various microorganisms live in soil, of which those colonizing rhizosphere interact with nearby plants and tend to develop unique microbial communities. In this study, we isolated diverse actinomycetes from rhizosphere of rice (Oryza sativa L.) cultivated in fertilized (APK) and non-fertilized (NF) paddy soils, and investigated the diversity and antimicrobial activity of them. Using four kinds of selective media, 152 isolates were obtained from the soil samples and identified by determining 16S rRNA gene sequence. All of the isolates showed 99.0%~100.0% similarities with type strains and were classified into six genera: Dactylosporangium, Micromonospora, Kitasatospora, Promicromonospora, Streptomyces and Streptosporangium. Most of the isolates, 143 isolates, were classified into the genus Streptomyces. Additionally, many isolates had antimicrobial activity against plant pathogens, especially Magnaporthe oryzae (rice blast pathogen) in fungi. These findings demonstrated that rice rhizosphere can be a rich source of antagonistic actinomycetes producing diverse bioactive compounds.
