Redescription of Two Marine Ciliates (Ciliophora: Urostylida: Pseudokeronopsidae), Pseudokeronopsis carnea and Uroleptopsis citrina, from Korea

  • The morphology of the two marine urostyloid ciliates, Pseudokeronopsis carnea (Cohn, 1866) and Uroleptopsis citrina Kahl, 1932, in the family Pseudokeronopsidae, collected from the Yellow Sea, and the East Sea, Korea, respectively, were studied using live observation and protargol impregnation. Additionally, the small subunit ribosomal RNA (SSU rRNA) gene was sequenced. These two species are firstly recorded in Korea. The main diagnostic key is as follows. Pseudokeronopsis carnea: body outline elongate-elliptical, brown-reddish or orange-red in colour in vivo; bicorona of 16-24 frontal cirri; one buccal and two frontoterminal cirri; 7-10 transverse cirri; 5-7 dorsal kineties; two types of cortical granules (one orange-red pigment, mainly grouped around cirri and dorsal bristles, arranged in typical rubra-pattern; the other, colourless and blood-cell-shaped, and densely distributed); contractile vacuole in the posterior half of the cell on the left side, usually in posterior 1/3-2/5. Uroleptopsis citrina: body outline elongate-elliptical, lemon-yellow in colour in vivo; two types of cortical granules (one yellow pigment; the other, blood-cell-shaped, densely distributed); bicorona of 12-18 frontal cirri; 2-3 frontoterminal cirri; two midventral rows comprising 26-35 cirri (consisting of anterior paired cirri, non-paired single cirri, and posterior paired cirri); three dorsal kineties. In addition, the SSU rRNA sequences of the two species were compared with public database of these species and consequently, showed high similarity.

  • 형태학적으로 카스텔라니가시아메바 혹은 대식가시아메바로 동정된 12 분리주들과 쿨버트손가시 아메바. 힐리가시아메바(Aconthomoeba hedwi) 팔레스타인가시아메바(A. plestinenis) 별가시아메바의 small subunitribosomal RNA유전자(ssu rDNA) 중 conserved region을 PCRR 증폭하여 제한효노절단부위를 비교하떴다. 별가시아메바의 PCR증폭 산물의 크기는 1.170 bp였고 나머지 분기주들의 것은 910-930 bp 사이였다 카스텔라너가시아메바로 분류건 여섯 주간의 추정 염기치찬율의 평근은 9.BPg였고. 대식가시아떼바로 분류된 분리주간의 그 평근은 9 6U였다. 카스텔 라니가시아메바로 분류된 여섯 분리주들 사이의 최대 염기치환율은 Chang주와 Ma주 사이의 (7 3%)였고 대식가시아메바로 분류된 여섯 분리주간의 최대 염기치환율은 1A/S3주와 KA/S7주 사이의 (16.1%)였다. 이들 종내 최대 염기치환율은 Castellani주 혹은 CCAP 1501/12g주와 KA/S3 주 사이에거 나타난 카스텔라니가시아메바와 대식가시아떼바간의 종간 최소 염기치환율(2.6%)보 다 횔씬 컸나. 쿨버트손가시아메바. 힐리가시아메바 팔레스타인가시아메바 및 별가시아메바의 PCR-RFLP 양상은 카스텔라니가시아메바 또는 대식가시아메바로 동정된 분리주들의 것들과 그리 고 강호간에서도 높은 염기치환율(평균 23 6%)을 보였다. 이상의 성적으로 미루어 보아 가시아메바속의 분류는 재평가 해 보아야 할 건으로 생각된다 A. healyi와 A. palestinensis의 우리말 이름을 각각 힐리가시아메바와 팔레스타인가시아메바로 제안한다.

Caspase 활성 및 Bid의 발현 저하를 통한 단백질 생성 억제제인 anisomycin의 인체간암세포에서 TRAIL 매개 apoptosis 유발의 활성화 (Anisomycin, an Inhibitor of Protein Synthesis, Overcomes TRAIL Resistance in Human Hepatocarcinoma Cells via Caspases Activation and Bid Downregulation)

  • Anisomycin은 Streptomyces griseolus에 의하여 생성되는 항생제의 일종으로 flagecidin으로도 알려져 있으며, ribosomal 28S subunit에 결합함으로서 단백질의 생성을 억제하는 것으로 알려져 있다. TRAIL은 ligand로서의 death receptor와의 결합을 통하여 세포의 apoptosis를 유발하는 것으로 알려져 있으나, 많은 암세포에서는 이미 TRAIL에 대한 저항성을 획득하여 TRAIL 유도 apoptosis를 회피하는 능력을 가지고 있다. 본 연구에서는 TRAIL 저항성 Hep3B 간암세포를 대상으로 anisomycin이 TRAIL 매개 apoptosis를 촉진 시킬 수 있는지의 여부를 조사하였다. 본 연구의 결과에 의하면, 단독 처리 조건에서 Hep3B 세포의 증식에 유의적인 영향을 미치는 않았던 anisomycin과 TRAIL의 동시 처리는 anisomycin 처리 농도 의존적으로 세포의 증식을 시켰으며, 이는 caspase 활성화를 통한 apoptosis 유발 증가와 연관성이 있음을 확인하였다. 특히 siRNA를 이용한 Hep3B 세포의 인위적인 Bid 발현의 차단은 anisomycin과 TRAIL 동시 처리군에 비하여 apoptosis 유발능이 더욱 증대시켜 TRAIL 연관 Bid의 truncation을 통한 미토콘드리아 의존적 apoptosis 유발 과정을 anisomycin이 효과적으로 촉진시켰음을 보여주었다. 따라서 본 연구의 결과는 anisomycin과 TRAIL의 동시 처리는 TRAIL 저항성 암세포의 사멸을 촉진시킬 수 있는 효과적인 방법임을 의미한다.

Mitochondrial Genome Sequence of Echinostoma revolutum from Red-Crowned Crane (Grus japonensis)

  • Echinostoma revolutum is a zoonotic food-borne intestinal trematode that can cause intestinal bleeding, enteritis, and diarrhea in human and birds. To identify a suspected E. revolutum trematode from a red-crowned crane (Grus japonensis) and to reveal the genetic characteristics of its mitochondrial (mt) genome, the internal transcribed spacer (ITS) and complete mt genome sequence of this trematode were amplified. The results identified the trematode as E. revolutum. Its entire mt genome sequence was 15,714 bp in length, including 12 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes and one non-coding region (NCR), with 61.73% A+T base content and a significant AT preference. The length of the 22 tRNA genes ranged from 59 bp to 70 bp, and their secondary structure showed the typical cloverleaf and D-loop structure. The length of the large subunit of rRNA (rrnL) and the small subunit of rRNA (rrnS) gene was 1,011 bp and 742 bp, respectively. Phylogenetic trees showed that E. revolutum and E. miyagawai clustered together, belonging to Echinostomatidae with Hypoderaeum conoideum. This study may enrich the mitochondrial gene database of Echinostoma trematodes and provide valuable data for studying the molecular identification and phylogeny of some digenean trematodes.

Morphological and Molecular Identification of Pseudo-nitzschia sp. Strain G3 Isolated from Northern Coast of Vietnam Based on ITS Region Sequences

  • For the first time in Vietnam, morphological and molecular studies of a species belonging to Bacillariophyceae collected in Northern coast of Vietnam are presented. Observations with microscope showed that this species belong to genus: Pseudo-nitzschia and seem like P. pungens. Sequence data from the partial 18S small subunit ribosomal RNA gene (18S rDNA) and the internal transcribed spacer 1 - 5.8S - internal transcribed 2 have been used to determine clearly and generate a phylogenetic framework of the obtained sequences to previously reported sequences in GenBank. These results allowed us to highlight described species of Bacillariophyceae in Northern coast of Vietnam. Furthermore, accumulation of molecular study would be helpful for the identification of scientific name of harmful algal species and further taxonomic studies in Vietnam.

Genotyping of Giardia duodenalis Isolates from Dogs in Guangdong, China Based on Multi-Locus Sequence

  • This study aimed to identify the assemblages (or subassemblages) of Giardia duodenalis by using normal or nested PCR based on 4 genetic loci: glutamate dehydrogenase (gdh), triose phosphate isomerase (tpi), ${\beta}$-giardin (bg), and small subunit ribosomal DNA (18S rRNA) genes. For this work, a total of 216 dogs' fecal samples were collected in Guangdong, China. The phylogenetic trees were constructed with MEGA5.2 by using the neighbor-joining method. Results showed that 9.7% (21/216) samples were found to be positive; moreover, 10 samples were single infection (7 isolates assemblage A, 2 isolates assemblage C, and 1 isolate assemblage D) and 11 samples were mixed infections where assemblage A was predominant, which was potentially zoonotic. These findings showed that most of the dogs in Guangdong were infected or mixed-infected with assemblage A, and multi-locus sequence typing could be the best selection for the genotype analysis of dog-derived Giardia isolates.

Acanthamoeba sohi, n. sp., a pathogenic Korean isolate YM-4 from a freshwater fish

  • A new species of Acanthamoeba was isolated from a freshwater fish in Korea and tentatively named Acanthamoeba sp. YM-4 (Korean isolate YM-4). The trophozoites were $11.0-23.0{\;}{\mu\textrm{m}}$ in length and had hyaline filamentous projections. Cysts were similar to those of A. culbertsoni and A. royreba, which were previously designated as Acanthamoeba group III. Acanthamoeba YM-4 can survive at $40^{\circ}C$, and its generation time was 19.6 hr, which was longer than that of A. culbertsoni. In terms of the in vitro cytotoxicity of lysates, Acanthamoeba YM-4 was weaker than A. culbertsoni, but stronger than A. polyphaga. On the basis of the mortality of experimentally infected mice, Acanthamoeba YM-4 was found to be highly virulent. The isoenzymes profile of Acanthamoeba YM-4 was similar to that of A. royreba. An anti-Acanthamoeba YM-4 monoclonal antibody, McAY7, was found to react only with Acanthamoeba YM-4, and not with A. culbertsoni. Random amplified polymorphic DNA marker analysis and RFLP analysis of mitochondrial DNA and of 18S small subunit ribosomal RNA, placed Acanthamoeba YM-4 in a separate cluster on the basis of phylogenetic distances. Thus the Acanthamoeba Korean isolate YM-4 was identified as a new species, and assigned as Acanthamoeba sohi.

Cockroach as a Vector of Blastocystis sp. is Risk for Golden Monkeys in Zoo

  • Blastocystis sp. is a kind of protozoa living in the intestinal tract of human and animals, which will cause intestinal diseases such as diarrhea, abdominal distension and vomiting. This paper was aimed to understand the infection of Blastocystis sp. In golden monkeys and the transmission path in North China. Thirty-seven feces samples from golden monkeys and 116 cockroach samples from Shijiazhuang Zoo were collected from July to October 2019 for PCR analysis of Blastocystis sp. Genetic diversity analysis was further conducted on the samples with positive PCR results. The results showed that the infection rate was 48.7% (18/37) in golden monkeys and 82.8% (96/116) in cockroaches, respectively. The genetic evolution analysis based on small subunit ribosomal RNA demonstrated that three subtypes (ST) of Blastocystis sp. including ST1, ST2, and ST3 existed in the intestinal tract of golden monkeys, while only ST2 was detected in the intestinal tract of cockroaches. This paper may provide supports for the quarantine and control of Blastocystis sp. for the zoo in Northern China.

Molecular Characterization of Various Trichomonad Species Isolated from Humans and Related Mammals in Indonesia

  • Trichomonad species inhabit a variety of vertebrate hosts; however, their potential zoonotic transmission has not been clearly addressed, especially with regard to human infection. Twenty-one strains of trichomonads isolated from humans (5 isolates), pigs (6 isolates), rodents (6 isolates), a water buffalo (1 isolate), a cow (1 isolate), a goat (1 isolate), and a dog (1 isolate) were collected in Indonesia and molecularly characterized. The DNA sequences of the partial 18S small subunit ribosomal RNA (rRNA) gene or 5.8S rRNA gene locus with its flanking regions (internal transcribed spacer region, ITS1 and ITS2) were identified in various trichomonads; Simplicimonas sp., Hexamastix mitis, and Hypotrichomonas sp. from rodents, and Tetratrichomonas sp. and Trichomonas sp. from pigs. All of these species were not detected in humans, whereas Pentatrichomonas hominis was identified in humans, pigs, the dog, the water buffalo, the cow, and the goat. Even when using the high-resolution gene locus of the ITS regions, all P. hominis strains were genetically identical; thus zoonotic transmission between humans and these closely related mammals may be occurring in the area investigated. The detection of Simplicimonas sp. in rodents (Rattus exulans) and P. hominis in water buffalo in this study revealed newly recognized host adaptations and suggested the existence of remaining unrevealed ranges of hosts in the trichomonad species.

Subtype Distribution of Blastocystis in Communities along the Chao Phraya River, Thailand

  • Blastocystis is a common zoonotic enteric protozoan that has been classified into 17 distinct subtypes (STs). A cross-sectional study was conducted to determine the prevalence and subtype distributions of Blastocystis in villagers living along the Chao Phraya River, Ayutthaya Province, Thailand, and to assess the risk of zoonotic infection. In total, 220 stool samples were collected, and DNA was extracted. PCR and sequencing were performed with primers targeting the small-subunit ribosomal RNA (SSU rRNA) genes. Blastocystis was present in 5.9% (13/220) of samples, and ST3 (5.0%; 11/220) was the predominant subtype, followed by ST2 (0.45%; 1/220) and ST6 (0.45%; 1/220). Phylogenetic trees were constructed with the maximum-likelihood method based on the Hasegawa-Kishino-Yano + G + I model, neighbor-joining, and maximum parsimony methods. The percentage of bootstrapped trees in which the associated taxa clustered together was relatively high. All the sequences of the Blastocystis-positive samples (KU051524-KU051536) were closely related to those from animals (pig, cattle, and chicken), indicating a zoonotic risk. Therefore, the villagers require proper health education, especially regarding the prevention of parasitic infection, to improve their personal hygiene and community health. Further studies are required to investigate the Blastocystis STs in the animals living in these villages.
