In order to develop the enzymatic hydrolysis system concerned with taste and flavor, strains having the high hydrolyzing activity on the soy protein were selected from some traditional Mejus. Two molds and one bacterium producing enzymes which were different in character of hydrolysis were isolated and identified. Leucine and azodye enzyme activities of both M4 and M5 were relatively high among in the isolated molds. And, leucine enzyme activity of B16 was the lowest in the isolated bacteria. These strains were isolated as microorganisms having a dissimilar hydrolysis pattern on the soy protein by enzymatic reactions. Mold M4 on the culture solid media was mycelium colors of white and its sclerotia colors were changed from white to black. According to the result of slide culture, radial conidial head, subclavate vesicle, conidia of subglobose, stipes of uncolored with smooth walls and metula and phialides were existed. Because M4 was taxonomically similar to the characteristics of Aspergillus oryzae (ahlburg) species, M4 was identified and named as Aspergillus oryzae M4.Mold M5 showed white and black mycelium on the MEA medium. Mold M5 colony exhibited grayish-green color and have long(7 mm) sporangiophores at slide culture. Sporangia became brownish-gray and the wall of larger sporangia was broken to form small collars, and smaller sporangia were fomed continually from large basal membrane. Columella is globose and hyaline, and sporangiospores are ellipsoidal of small diameter$(80\;{\mu}m)$. Because M5 was taxonomically similar to the Mucor circinelloides of zygomycetes, M5 was was identified and named as Mucor circinelloides M5. Bacteria B16 colony was opaque white, circular and lobate, and had rod shaped endospore. B16 was found positive in stain, catalase, ${\beta}-glucosidse$ and V-P tests. B16 was found to utilize D-fructose, ${\alpha}-D-glucose$, maltose, D-mannose, D-raffinose, stachyose and sucrose. By the morphological and physiological results, the characteristics of B16 was thought to correspond to that of Bacillus megaterium. However, fatty acid composition was similar to Paenibacillus marcerans, requiring further study for the definite identification. Accordingly, Bacteria B16 was provisionally classified and named as Bacillus megaterium B16.

A series of drained plane strain compression tests was performed on dried samples of dense Toyoura sand and Silver Leighton Buzzard sand prepared by air-pluviation method to find out the deformation and strength characteristics on the value of confining pressure ${\sigma}{_3}^{\prime}({\sigma}{_3}^{\prime}=0.05{\sim}4.0kgf/cm^2)$. The axial and lateral strains measured in this apparatus ranged from $10^{-6}$ up to the failure of the specimen. So the stress-strain characteristics would be investigated from very small to very large strain levels. It was found that the change of the angle of internal friction ${\phi}^{\prime}{_{max}}=arcsin\{({\sigma}{_1}^{\prime}-{\sigma}{_3}^{\prime})/({\sigma}{_1}^{\prime}+{\sigma}{_3}^{\prime})\}_{max}$ with the change of ${\sigma}{_3}^{\prime}$ is very small when ${\sigma}{_3}^{\prime}$ is lower than higher. Furthermore, the effect of confining pressure on stiffness of sands was evaluated. It was also found that for the range of shear strain ${\gamma}$ from $10^{-6}$ to those at peak, the Rowe's stress-dilatancy relation seems to be a good approximation for air-dried Toyoura sand and Silver Leighton Buzzard sand, irrespective of the change of ${\sigma}{_3}^{\prime}$.
We aimed to increase the utility of Broussonetia kazinoki (paper mulberry) by using its woody parts as a substrate for Lentinula edodes cultivation. Using different mixing ratios of paper mulberry and oak tree sawdust, five types of column-type media were prepared (1.2 kg each). Two L. edodes strains (NIFoS 2462 and NIFoS 2778) were used in this study. In the first flush, numerous small mushrooms were harvested because the fruited mushrooms were densely packed on the small media. The highest productivity was obtained with a 3:1 mixture of paper mulberry and oak tree sawdust (Q. acutissima:Q. mongolica, 1:1). In particular, for NIFoS 2462, the proportion of paper mulberry in the medium positively correlated with productivity. The size of NIFoS 2778 was uniform in all media tested. By contrast, the mushroom weight of NIFoS 2462 decreased, but the other characteristics were not significantly affected. Collectively, these findings suggest that B. kazinoki sawdust could be used to grow shiitake mushrooms and that some oak sawdust substitution is also possible. Our results could increase the utility of discarded by-products, such as sawdust.

The variations of the uniaxial compressive strength, the strains and the elastic constants with respect to the angle of anisotropy are analyzed in order to investigate the characteristics of a transversely isotropic rock experimentally. Total 35 specimens of 7 different angles from a large block of rhyolite presenting the flow structure obviously are used in tests. This study is composed of two reports; the elastic constants are mainly analyzed for the every individual angle in the report No. 1 and they will be discussed synthetically in the report No. 2. From the specimens of 0 and 90 degree, 4 independent elastic constants which can directly be obtained without the help of any other suggested equations, may be referred to the true values. Data variation in the strain measurements differs on the angle is analyzed. That of small angle specimens tends higher than that of large angle specimens. The relation of apparent Young’s modulus and angle is found to be M- or U-shaped. For small angle specimens, Saint-Venant approximation cannot be applied successfully on account of showing the non-monotonous increase, and E1 is analyzed out of the true value range. In the specimen of $\phi$ = 75, the deviation of strain measurement and strength are smallest and 4 all constants are analyzed in the true value range. Therefore, specimen of the angle of around 75 may become preferable if only one specimen should be used in test of a transversely isotropic rock.
An antimicrobial bacterium to pathogenic microorganisms, strain $W5-1^T$ was isolated from Korean fermented-food Kimchi. The isolate was Gram-staining-variable, strictly aerobic, rod-shaped, endospore-forming, and motile with peritrichous flagella. It grew at $15-40^{\circ}C$, at pH 6.0-10.0, and in the presence of 0-4% NaCl. Strain $W5-1^T$ could hydrolyze esculin and xylan, and assimilate $\small{D}$-mannose, but not $\small{D}$-mannitol. Strain $W5-1^T$ showed antimicrobial activity against Listeria monocytogens, Pseudomonas aeruginosa, Staphylococcus aureus, and Salmonella typhi. The G+C content of the DNA of strains $W5-1^T$ was 52.6 mol%. The predominant respiratory quinone was menaquinone-7 (MK-7) and the major cellular fatty acids were $C_{16:0}$, antieiso-$C_{15:0}$, $C_{18:0}$, and $C_{12:0}$. The strain contained meso-diaminopimelic acid in cell-wall peptidoglycan. On the basis of 16S rRNA gene sequence and phylogenetic analysis, the strain W5-1 was shown to belong to the family Paenibacillaceae and was most closely related to Paenibacillus pinihumi $S23^T$ (98.4% similarity) and Paenibacillus tarimensis $SA-7-6^T$ (96.4%). The DNA-DNA relatedness between the isolate and Paenibacillus pinihumi $S23^T$ was 8.5%, indicating that strain $W5-1^T$ represented a species in the genus Paenibacillus. On the basis of the evidence from this polyphasic study, it is proposed that strain $W5-1^T$ is considered to represent a novel species of the genus Paenibacillus, for which the name Paenibacillus kimchicus sp. nov. is proposed. The type strain is $W5-1^T$ (=KACC $15046^T$ = $LMG 25970^T$).
Among 6 leu codons, CUG is the most frequently used codon in E. coli. It is recognized by leu-tRNA(CAG) encoded by four genes scattered on two chromosomal loci (leuT and leuPQV ). In the process of constructing a strain with no functional leu-tRNA (CAG) gene on chromosome, we made two mutant strains separately, one on leuPQV locus (${\Delta}leuPQV$), and the other on leuT locus [$leuT^*$(GAG)], where the anticodon of leuT was changed from CAG to GAG, thereby altering its recognition codon from CUG to CUC. We attempted to combine these two mutations by transduction using $leuT^*$(GAG) strain as a donor and ${\Delta}leuPQV$ strain as a recipient. Large and small colonies appeared from this transduction. From PCR and DNA sequencing, large colony was confirmed to be the reciprocal recombinant as expected, but the small colonies contained both mutant $leuT^*$(GAG) and wild type leuT (CAG) genes in the cell. This heterozygous diploid strain did not show any unusual morphology under microscopic observation, but, interestingly, it showed a linear growth curve in rich medium with much slower growth rate than wild type cell. It always formed homogenous small colonies in the selection medium, but, when there was no selection, it readily segregated into $leuT^*$(GAG) and leuT (CAG). From these observations, we suggested that the strain with both $leuT^*$(GAG) and leuT (CAG) genes was not a partial diploid (merodiploid), but a full diploid cell having two different chromosomes. We proposed a model explaining how such a heterozygous diploid cell was formed and how and why its growth showed a linear growth curve.

Conjugated linoleic acid (CLA) is a mixture of positional and geometric isomers of linoleic acid with conjugated double bonds. These conjugated dienes were found to be responsible for many biological properties related to health. The objective of this study was to evaluate the production of cis-9, trans-11 CLA by Lactobacillus acidophilus isolated from breast-fed infants. Nine different cultures were tested for their ability to produce cis-9, trans-11 CLA from free linoleic acid in MRS broth and 8% reconstituted skim milk medium supplemented with linoleic acid at $37^{\circ}C$ for 48 hr. cis-9, trans-11 CLA was not detected or detected in very small amount when cell pellets of strains grown in MRS broth and 8% reconstituted skim milk supplemented with linoleic acid of $200{\mu}g/mL$. However, free cis-9, trans-11 CLA was produced in both media. It appeared that 8% reconstituted skim milk produced more cis-9, trans-11 CLA than MRS broth. L. acidophilus NB 203 and NB 209 produced more cis-9, trans-11 CLA than other tested cultures. The inhibitory effects of supplemented linoleic acid on the growth of L. acidophilus NB 203 and NB 209 were not detected up to $3,000{\mu}g/mL$ linoleic acid addition during the growth at $37^{\circ}C$ for 48 h. The production of cis-9, trans-11 CLA by these two L. acidophilus strains increased in the logarithmic growth phase until 24 hr incubation. Under this experimental condition, the best yield of CLA isomers for L. acidophilus NB 203 and NB 209 could be obtained from medium supplemented with $500{\mu}g/mL$ linoleic acid at $37^{\circ}C$ after 24 hr of incubation. These results indicate that the use of lactic acid bacteria producing free CLA in fermented dairy products may have potential health or nutritional benefits.

This study was conducted to survey the epidemiological characteristics and the isolated strains for pathogenic E. coli which was the major causative organisms for food poisoning occurred at school food services in the Gyeonggi-do area during the past three years. We investigated 19 accidents of food-borne disease outbreaks by pathogenic E. coli at school food services from 2010 to 2012. Food-borne disease outbreaks by pathogenic E. coli were usually occurred at direct management type (18 accidents, 95%) and high schools. For the seasonal factors, 13 accidents (65%) were occurred in June to September, especially the end of August and September after the summer holidays. The first patients were occurred on Wednesday (7 accidents, 37%) and Thursday (7 accidents, 37%), and they were mainly reported on Thursday (7 accidents, 37%) and Friday (5 accidents, 26%). The exposure of risk was estimated in Monday (4 accidents, 21%), Tuesday (7 accidents, 37%) and Wednesday (4 accidents, 21%), and kimchi (5 accidents, 50%) was estimated as the food of the high risk responsible for the outbreaks. 98 isolates of pathogenic E. coli consisted of PEC (50%), ETEC (34%), EAEC (15%), and EHEC (1%). The antibiotic resistance of pathogenic E. coli showed in the descending order of ampicilline (40%), nalidixic acid (37%), trimethoprim/sulfamethoxazole (24%), and tetracycline (19%). The antibiotics of second and third generation cephalosporins, cabarpenem, aminoglycosides, and second generation quinolones had antimicrobial susceptibilities and cefalotin, ampicillin/sulbactam and chloramphenicol showed medium resistance at 29%, 25%, and 6% respectively, and 70% of isolates were resistant to more than one antibiotic. By the PFGE analysis, they were classified into nine major groups and 31 profiles with 57% pattern similarity. It was very difficult to find the correlation of antimicrobial susceptibilities and genotype in the small scale-food poisoning, but the similarity of antimicrobial resistance and PFGE patterns in the large scale-food poisoning enabled the outbreaks to estimate the same pathotype of E. coli derived from identical origins.
Lateral gene transfer (LGT) of genes from other bacteria into Vibrio cholerae is expectable because of the pronounced natural competence of the bacterium. In this study, quantitative aspects of LGT among the three species of Vibrio pathogenic to humans were characterized. Genome sequences of V. cholerae N16961, V. parahaemolyticus RIMD2210633, V. vulnificus CMCP6, and Escherichia coli K12 substrain MG1655 were analyzed to determine orthologous quartets of protein coding genes present in all four genomes. Phylogenetic analyses on the quartets were conducted to resolve vertical versus lateral patterns of gene polymorphisms based on congruence versus incongruence of phylogenetic trees. About 70% of the quartets could be resolved as either cohesive topology (75%) or LGT tree topologies (25%). The amount of LGT genes in Vibrio spp. appeared to be abnormally high for a genus and comparable to those of families. Patched distributions of LGT from different donors were observed on a chromosome. In the small chromosome of V. cholerae, physical linkages among LGT loci spanned half the length of the chromosome. Either accumulative selection for the donor alleles in LGT or presence of large-scale LGT events was hypothesized. These findings warrant further studies on the nature of donor-specificity of LGT alleles and its influence on evolution of Vibrio virulence to humans.
The cracks are developed in reinforced concrete(RC) beams at the early stage of service load because of the relatively small tensile strength of concrete. The structural strength and stiffness are decreased by reduction of tensile resistance capacity of concrete due to the developed cracks. Using the fiber reinforced concrete that is increased the flexural strength and tensile strength at tensile part can enhance the strength and stiffness of concrete structures and decrease the tensile flexural cracks and deflections. Therefore, the RC beams used of the fiber reinforced concrete at. tensile part ensure the safety and serviceability of the concrete structures. In this work, analytical model of a dual concrete beams composed of the normal strength concrete at compression part and the high tension strength concrete at tensile part is developed by using the equilibrium conditions of forces and compatibility conditions of strains. Three groups of test beams that are formed of one reinforced concrete beam and two dual concrete beams for each steel reinforcement ratio are tested to examine the flexural behavior of dual concrete beams. The comparative study of total nine test beams is shown that the ultimate load of a dual concrete beams relative to the RC beams is increased in approximately 30%. In addition, the flexural rigidity, as used here, referred to the slope of load-deflection curves is increased and the deflection is decreased.
