Effect of Epithelial Inflammation Relief through Regulation of Lipid Barrier Formation of Coptidis Rhizoma Extract-Ceramide Complex (황련추출물-ceramide 복합물의 지방장벽 형성 조절을 통한 상피 염증 완화 효과)

  • Objective The purpose of this study was to confirm the effectiveness of coptidis rhizoma extract-ceramide complex on skin barrier, transepidermal water loss (TEWL) and pH reduction, and inflammation of the skin. Methods Coptidis rhizoma extract-ceramide complex was applied in 6-week-old Balb/C mice after dermatitis was induced. To confirm the skin condition changes, TEWL and pH were observed, and filaggrin in the stratum corneum of the skin was observed. Kallikrein-related peptidase (KLK) 7, Protease activated receptor (PAR)-2, Thymic stromal lymphopoietin (TSLP), and IL-4 were observed in the stratum corneum to confirm the changes in the inflammatory response. Results Filaggrin positive reaction was increased in the experiment group compared to the control group. TEWL and pH were lower in the experiment group compared to the control group. KLK7, PAR2, TSLP, and IL-4 positive responses were decreased in the experiment group compared to the control group. Conclusions It was confirmed that the coptidis rhizoma extract-ceramide complex can relieve the inflammatory response of atopic dermatitis by restoring the skin lipid barrier damage.

Glabridin Liposome Ameliorating UVB-Induced Erythema and Lethery Skin by Suppressing Inflammatory Cytokine Production

  • Glabridin, a compound of the flavonoid, has shown outstanding skin-whitening and anti-aging properties, but its water insolubility limits its wide application. Therefore, glabridin liposome (GL) has been developed to improve its poor bioavailability, while there are few studies to evaluate its amelioration of UVB- induced photoaging. This study is performed to investigate the amelioration of GL against UVB- induced cutaneous photoaging. The prepared GL has a spheroidal morphology with an average diameter of 200 nm. The GL shows lower cytotoxicity than glabridin, but it has a more effective role in inhibition of melanin. Moreover, the application of GL can effectively relieve UV radiation induced erythema and leathery skin, associated with the down-regulated expression of inflammatory cytokines (TNF-α, IL-6 and IL-10). Taken together, these results demonstrate that GL has potentials as topical therapeutic agents against UVB radiation induced skin damage through inhibiting inflammation.

Inhibitory activity of Terminalia chebula extract against TNF-α/IFN-γ-induced chemokine increase on human keratinocyte, HaCaT cells (TNF-α/IFN-γ 유도된 각질형성세포 염증에서 가자 추출물의 케모카인 저해 효과)

  • Objectives : Terminalia chebula (TC) has been used as a traditional remedy to treat gastrointestinal infectious and inflammatory diseases. However, its protective effects and mechanisms against skin inflammation have not been well-elucidated. Thus, the aim of this study is to evaluate the protective effects of the TC water extract and also to suggest a putative mechanism of TC against skin injury on human keratinocytes, HaCaT cells. Methods : HaCaT cells were pre-treated with TC for 1 h and then stimulated with tumor necrosis factor-alpha (TNF-α) and interferon-gamma (IFN-γ) (10 ng/mL each) to induce skin inflammation and injury. After 24 h, the cells were harvested to evaluate the expression of Th2 chemokines, such as C-C motif chemokine ligand 5 (CCL5, also known as RANTES), C-C chemokine ligand 17 (CCL17, also known as TARC) and C-C chemokine ligand 22 (CCL22, also known as MDC). To investigate the regulatory mechanisms of TC, we also assessed the phosphorylation of signal transducer and activator of transcription 1 (STAT1) signaling pathways in HaCaT cells. Results : Treatment of TC decreased the mRNA levels of RANTES, TARC and MDC with a concentration dependent manner against co-stimulation of TNF-α and IFN-γ. In addition, TC significantly reduced TNF-α and IFN-γ induced phosphorylation of STAT1. Conclusions : In summary, we propose that TC may be a promising candidate for anti-inflammatory skin protector through the inhibition of chemokines via STAT1 deactivation.

Saccharina japonica Attenuates the Allergic Inflammation in vivo and in vitro

  • Saccharina japonica (SJ), a brown algae, exerts various pharmacological effects, including anti-oxidant, immunemodulating and anti-cancer properties. This study aimed to determine the pharmacological mechanism of SJ on atopic dermatitis in vivo and in vitro. We investigated the pharmacological effects of SJ on 2, 4-dinitrochlrobenzene (DNCB)- induced atopic dermatitis clinical symptoms in mice. Additionally, we evaluated the effects of SJ on the inflammatory cytokine production and nuclear factor-κB (NF-κB) activation in HaCaT cells. The findings of this study demonstrated that SJ reduced the clinical symptoms of atopic dermatitis, such as skin dryness, erythema and eczematous, and serum histamine and IgE level in DNCB-induced atopic dermatitis model. Additionally, SJ inhibited the NF-κB activation in atopic dermatitis-like skin lesion and HaCaT cells. Collectively, this result suggests that SJ could be used as a therapeutic agent for skin inflammation, including atopic dermatitis.

Effects of Danggwieumja on the Healing of Full-Thickness Skin Injury in Rat

  • The purpose of this study was to investigate the wound healing effects of Danggwieumja (DG), which is commonly used for skin inflammation, skin wound, skin pruritus, and chronic hives etc. The 1.5 cm ${\times}$ 1.5 cm full-thickness skin wound was induced to two groups, DG (n=16) and Saline (n=16) group. The DG extract and Saline were orally administrated daily for 15 days after skin wound induction. Then, the body weight of rats and the congestion indices were daily measured for 15 days after skin wound induction. The wound contractions and epithelizations were also measured. The wound contractions were daily measured for 15 days after wound induction and wound epithelizations were measured for 8 days from day 7 after wound induction. For evaluating angiogenesis, the immunoreactivities of vWF and VEGF protein were measured immunohistochemistrically on day 15. In results, although the percentage increases in mean body weight of rats in the DG and Saline groups hve no significant differences, DG extract decreased the time of wound healing and congestion around wound, and improved wound contraction and epithelization. The contraction percentage of DG group was significantly increased on day 5 (P<0.05) and day 7 (P<0.01) than that of Saline group. DG group showed significant increase of wound epithelization on day 7 (P<0.05) as compared to Saline group. Moreover, DG extract reduced the inflammation of skin dermis and promoted the growth of vascular vessels of dermis by accelerating vascular endothelial growth factor (VEGF) protein. These results suggest that DG has the beneficial effects on skin incision wound and can be the suitable wound healing agent for various surgical wounds.

Development of a Cosmetic Ingredient Containing DHA Derivatives for Anti-inflammation, Anti-wrinkle, and Improvement of Skin Barrier Function (DHA 유도체를 이용한 항염, 항노화, 피부장벽 강화용 화장품 원료의 개발)

  • It is very important to control the inflammation of the skin because it can develop into diseases such as atopy as well as scarring and aging. In this work, we recently identified the in vitro synthesis of specialized pro-resolving mediators (SPMs) known to control inflammation in the human body and the applicability of cosmetics. Using recombinant protein of lipoxygenase from Glycine max, we succeeded to prepare mixtures of mono- or di-hydroxy DHA named as S-SPMs and used them for in vitro efficacy test. To investigate anti-inflammatory effect of S-SPMs, mRNA level of TNF-α and IL-6 were analyzed. Under UVB exposed condition, expression of both were decreased by S-SPMs treatment. And we observed reduced production of nitric oxide (NO) by S-SPMs application under the condition with diesel particulate matter (DPM). At the same experimental condition, malondialdehyde (MDA) production was decreased by S-SPMs, indicating the inhibitory effect of S-SPMs in lipid peroxidation. In addition, S-SPMs treatment resulted in reduction of matrix metalloproteinases-1 (MMP-1) expression and elevation of procollagen type I synthesis. Together with this, mRNA level of filaggrin and loricrin were increased by S-SPMs, indicating enhancement of skin barrier function. These results demonstrate that S-SPMs is a good candidate to develop as a cosmetic ingredient for anti-inflammation, anti-wrinkle, and improvement of skin barrier function.

Combined Effect of Korean Red Ginseng and EGb 761 on Ultraviolet B-induced Contact Hypersensitivity (UV-B조사로 유도된 접촉 과민반응에 대한 홍삼과 홍삼+EGb 761 혼합물의 억제 효과 비교)

  • Exposure of skin to UV-B radiation can cause inflammatory response and immunosuppression. It has been reported that Korean Red Ginseng (KRG) has several pharmacological and physiological effects such as antioxidant, anticancer and improving immune function. In this study, we investigated that topical KRG and KRG + EGb 761 (Ginkgo biloba extract) combination prevented UV-B induced inflammation and inhibition of contact hypersensitivity response. Topical application of KRG, f days prior to or 5 days after exposure to 1MED and 2MED of UV-B, reduced skin thickness compared to non -treated group and resulted in protection against immunosuppression. However, KRG+EGb 761 combination has a little protection against the only 1MED UV-B. In conclusion. Topical application of KRG was more effective than combination in protection against UV-B induced inflammation and immune suppression. Also, we suggest that KRG can provide protection from inflammation and immunosuppression by UV-B radiation.

Effects of Samhwangsasim-tang Gamibang on Atopic Dermatitis in Mice (삼황사심탕가미방(三黃瀉心湯加味方)이 생쥐에 유발된 아토피 피부염에 미치는 영향)

  • Objectives : Atopic Dermatitis (AD) is chronic skin disease characterized by allergic hypersensitivity reactions. Samhwangsasim-tang Gamibang(SHSST) can treat skin disease by cooling down blood heat, clearing away congenital fever and detoxicating. This study was conducted to investigate the effects of SHSST on AD induced by oxazolone in mice. Methods : We investigated the effects of SHSST on skin tickness, skin lesions, erythema and melanin index, water content and histopathological changes in vivo. The effects on body weights and spleen body weight ratio were also investigated in mice with AD. Results : In our results, oral administration of SHSST ameliorated skin lesion such as flare, erythema, exudate and petechia and lowered skin thickness induced by oxazolone. In addition, SHSST lowered erythema index and elevated water content compared to AD control respectively. In the histopathological observation, oral administration of SHSST prevented epidermal hyperplasia and spongiotic changes which is a hallmark of skin inflammation respectively. Finally, SHSST did not affect spleen/body weight ratio. Conclusions : These data imply that SHSST can ameliorate skin lesion of AD such as flare, erythema, exudate and petechia, and be used to treat AD patients with relative safety.

Effect of Sophora flavescens Extract on Reinforcing Skin Barrier and Alleviating Inflammation (고삼 추출물의 피부장벽 강화와 염증완화 효과)

  • Atopic dermatitis (AD) is a common and multifactorial inflammatory skin disease that is characterized by skin barrier dysfunction, inflammation, and chronic pruritus. AD has a complex etiology that includes genetic, immunological, and environmental factors that cause skin barrier abnormalities and immune dysfunctions. Sophora flavescens (SF) has been used in traditional Chinese medicine, but little research has been conducted on its anti-AD efficacy. In this study, we evaluated the effect of SF extract (SFE) on improving skin barrier function and immune abnormalities, which are the main symptoms of AD. SFE has the capacity to enhance the formation of cornified envelope (CE) that plays an important role in the skin barrier function. In addition, it was confirmed that SFE increased the expression of hyaluronic acid related to skin moisture. The effect of SFE against Staphylococcus aureus (S. aureus), which increases specifically in AD lesions, confirmed that SFE inhibited the production of pro-inflammatory cytokines induced by S. aureus. Furthermore, SFE was shown to inhibit the expression of pro-inflammatory cytokines induced by substance P (SP), the cause of skin neurogenic inflammation. These results demonstrate that SFE could be one of potential candidate agent for the treatment of AD by improving the skin barrier function and immune responses.

The Alleviative Effect of White Tea Water Extract on Inflammation and Skin Barrier Damage (백차 열수추출물의 자외선 조사에 의한 피부염증 및 피부장벽손상 완화 효과)

  • To investigate the alleviative effect of white tea water extract on the inflammation and skin barrier damage, skin aging animal model was produced by the irradiation of UVB to the backs of hairless mice for 12 weeks. And then experimental materials were applied topically for 4 weeks. At the 28th day of experiment, positive control (PC, 0.01% retinoic acid treatment) and experimental groups (E1, 1% white tea water extract treatment; E2, 2% white tea water extract treatment) had significantly (p<0.001) lower values of both skin erythema index and transepidermal water loss (TEWL) than the control (C, saline treatment) group. The appearance of mast cell and the degree of its degranulation in dermal and subcutaneous layers were remarkably reduced in E1 and E2 groups compared to the C group. It is found that white tea water extract is effective in skin barrier damage and inflammation in hairless mouse.
