Anti-aging and Anti-inflammatory Activities of the Extracts of Calamagrostis arundinacea (Calamagrostis arundinacea (실새풀) 추출물의 항노화 및 항염증 활성)

  • The anti-aging and anti-inflammatory activities of hot-water (Ca-HW) and 70% ethanol (Ca-E70) whole-plant Calamagrostis arundinacea extracts, as well as their bioactive potentials, were investigated using cell-free and cell-mediated experimental systems. Use of the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical decolorization assay to evaluate the antioxidant activity of the Ca-HW and Ca-E70 extracts revealed DPPH radical scavenging activities of 27% and 48%, respectively. Neither extract caused significant cytotoxicity, and both showed cell proliferation and promotion effects using RAW 264.7, B16F10, and CCD986sk cells. B16F10 melanoma cells showed higher melanin synthesis when treated with 100 mg/ml Ca-HW or Ca-E70 than with arbutin, indicating a stronger inhibitory effect of arbutin on melanin synthesis. Ca-HW and Ca-E70 increased pro-collagen biosynthesis in the human fibroblast CCD986-SK cell line by 24.69% and 12.55%, respectively. Analysis of the anti-inflammatory effects of different concentrations of Ca-HW and Ca-E70 in RAW264.7 cells revealed that Ca-E70 appeared to inhibit the lipopolysaccharide-induced production of nitric oxide and IL-6, a proinflammatory cytokine; therefore, Ca-E70 showed an anti-inflammatory effect. These results suggested that C. arundinacea extracts could have skin anti-aging and anti-inflammatory properties.

Effects of Resveratrol and Resveratryl Triacetate on The Inflammatory Responses of Human Epidermal Keratinocytes Exposed to Airborne Particulate Matter PM10 (대기 미립자 물질 PM10에 노출된 인간 표피 각질형성세포의 염증 반응에 대한 레스베라트롤과 레스베라트릴 트라이아세테이트(RTA)의 영향)

  • Airborne pollution causes oxidative damage, inflammation, and premature aging of skin. Resveratrol is a polyphenol compound that has various biological activities such as antioxidant, anti-inflammation, and anti-melanogenic activities but it is unstable to heat and light. Resveratryl triacetate (RTA) is a new cosmetic ingredient that is more stable than resveratrol and its skin safety and whitening efficacy have been reported previously. The purpose of this study was to examine the effects of resveratrol and resveratryl triacetate (RTA) on the inflammatory responses of human epidermal keratinocytes (HEKs) exposed to airborne particulate matters with a diameter of < $10{\mu}m$ (PM10). Cultured HEKs were exposed to PM10 in the absence or presence of resveratrol and RTA. Assays were undertaken to determine cell viability, the production of reactive oxygen species (ROS), and the expression of inflammatory cytokines. PM10 treatment decreased cell viability, and increased the expression of pro-inflammatory cytokines such as tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), $interleukin-1{\beta}$ ($IL-1{\beta}$), interleukin-6 (IL-6), and interleukin-8 (IL-8). Resveratrol and RTA reduced cell death and ROS production induced by PM10. PM10-induced mRNA expression of the inflammatory cytokines was either attenuated (IL-6), or enhanced ($IL-1{\beta}$), or unaffected ($TNF-{\alpha}$ and IL-8) by resveratrol and RTA. PM10-induced IL-6 protein expression was attenuated by resveratrol and RTA. This study suggests that resveratrol and RTA have activities regulating cell damage and inflammatory responses of the skin exposed to airborne particulate matters.

Anti-aging Effects of the Extracts from Leaf. Stem, Fruit and Seed of Yew (Taxus cuspidata Sieb) by Solvent Extraction Method (용매추출법에 의한 주목의 잎, 줄기, 과실 추출물의 항 노화 효과)

  • Yew (Taxus cuspidata Sieb.) chose that grow as medicine, food, decorative plant in Korea's Kyong-Gi province surroundings. Extracts of yew extracted leaf of 250g and stems of 300g with 1,3-butylene glycol (l,3-BG), propylene glycol (PG) and water. As results, external appearance of leaf extract of yew was slightly brown clear extract. The pH was 5.3${\pm}$0.5, and specific gravity was 1.012${\pm}$0.05, and refractive index was l.375${\pm}$0.05. Also, appearance of stem's extract was slightly brown clear extract, and the pH was 5.4${\pm}$0.5, and specific gravity was 1.016${\pm}$0.05, and refractive index was 1.358${\pm}$0.05. Oil of yew separated from seeds, and extracted polysaccharide high purity from fruits. As a result, specific gravity of oil was 0.987, and obtained 40.0% of yield. Total polyphenols amount of yew extract is detected 0.563% in leaves, 0.325% in stems, whereas total tannins amount contained 0.054% and 0.037% each in leaves and stems. As effect in cosmetics, the anti-oxidative effect by DPPH method is 75.0% in leaves, and stems was 64.0%. Collagen synthesis rate was shown high activity by 54.16% in stem's extract, 33.18% in leaves' extract. Also, PPE-inhibitory activities were 13.7% and 23.5% each in leaves and stems. Anti-inflammatory effect of yew seed oil displayed superior effect of 41% than control. Polysaccharide's molecular weight that is gotten from fruits was 5${\times}$10$^4$-3${\times}$10$\^$5/ dalton, and got 20.0${\pm}$5% of yield.

Anti-oxidation, anti-inflammation, anti-wrinkle, and pore-tightening effects of phenolic compounds from Aeonium sedifolium leaves (소인제(Aeonium sedifolium) 잎 유래 phenolic 성분의 항산화, 피부주름생성 억제, 항염증 및 모공 수축 효과)

  • The succulent plant Aeonium sedifolium leaves contain several compounds that are of interest for their cosmetic uses on the skin. This study measured the inhibitory effects of enzyme production and antioxidant, astringent effects and skin wrinkles using Aeonium sedifolium leaves (ASL). The total phenolics compounds (TPC) content of ASL under optimal extraction conditions was 34.49 mg/g for hot water extract (ASLW) and 61.64 mg/g for 50% ethanol extract (ASLE). The ASLW and ASLE extracts were freeze-dried, powdered, and used as solids. TPC content, 2,2-diphenyl-1-picrylhydrazy (DPPH) radical scavenging activity, and 2,2'-azinobis (3-ethylben-zothiazoline 6-sulfonate) (ABTS) radical inhibition of the ASL phenolics were tested. The DPPH radical scavenging activities of ASLW and ASLE were tested at a TPC of 100 ㎍/mL. ABTS radical inhibition showed antioxidant activity of 100.00% in ASLW and ASLE, and the antioxidant protection factor of ASLW and ASLE was 1.07 and 1.22, respectively. The thiobarbituric acid-reactive substance (TBARS) inhibitory activity of ASLW and ASLE was 77.00%. The elastase inhibitory activity of ASLE was 69.03%, and collagenase inhibition activity for ASLW and ASLE was 29.82% and 54.76%, respectively. The astringent effect of ASLE was 89.82% at a TPC of 200 ㎍/mL. Thus, we concluded that ASL has the potential as a functional cosmetic ingredient with anti-aging effects on the skin.

Physiochemical Characteristics of Raw and Dried Jerusalem Artichoke Jangachi (생돼지감자와 건조돼지감자 장아찌의 이화학적 특성)

  • Jerusalem artichoke is often stored poorly after harvesting due to its delicate skin. For this reason, Jerusalem artichoke is mainly distributed in the market as a dried material. In order to improve utilization of dried Jerusalem artichoke, we made Jangachi and analyzed physiochemical characteristics with raw Jerusalem artichoke Jangachi during storage. Raw Jerusalem artichoke was sliced into 7 and 10 mm slices and dried in hot air. After aging, we analyzed rehydration capacity, pH, hardness, salinity, sugar content, and fructan content of both. Dried Jerusalem artichoke sliced into 10 mm slices took over twice as long (420 min) to be rehydrated over 90% than 7 mm sliced (200 min) Jerusalem artichoke. In the case of raw Jerusalem artichoke, hardness showed a tendency to decline. Relatively, hardness of dried Jerusalem artichoke was measured consistently. After 4 weeks, salinity and sugar content were 3.63% and $41.23^{\circ}Brix$ in raw Jerusalem artichoke Jangachi, 3.47% and $37.05^{\circ}Brix$ in 7 mm dried Jerusalem artichoke, and 3.77% and $39.15^{\circ}Brix$ in 10 mm dried sample, respectively. As a result of this study, the possibility of dried Jerusalem artichoke as a pickle was confirmed.

Effects of Platycodon grandiflorum on the Induction of Autophagy and Apoptosis in HCT-116 Human Colon Cancer Cells (길경 추출물에 의한 HCT-116 대장암 세포주에서의 autophagy와 apoptosis 유발 효과)

  • Platycodon grandiflorum (PG) has been known to possess many biological effects, including anti-inflammatory and anti-allergy activity and anti-obesity and hyperlipidemia effects. However, little research has been conducted regarding its anticancer effects, with the exception of its ability to stimulate apoptosis in skin cells. There has also been no study regarding PG-induced autophagy. The modulation of autophagy is recognized as one of the hallmarks of cancer cells. Depending on the type of cancer and the context, autophagy can suppress or help cancer cells to overcome metabolic stress and the cytotoxicity of chemotherapy. Therefore, the present study was designed to investigate whether or not extracts from PG-induced cell death were connected with autophagy and apoptosis in HCT-116 human colon cancer cells. PG stimulation decreased cell proliferation in a dose- and time-dependent manner and induced apoptosis, which was partially dependent on the activation of caspases. PG treatment also resulted in the formation of autophagic vacuoles simultaneously with regulation of autophagy-related genes. Interestingly, a PG-mediated apoptotic effect was further triggered by pretreatment with the autophagy inhibitors 3-methyladenin and bafilomycin A1. However, cell viability recovered quite well with bafilomycin A1 treatment. These findings show that PG treatment promotes both autophagy and apoptosis and that PG-induced autophagic response might play a role in the autophagic cell death of HCT-116 cells.

Inhibitory Effect on Melanin Formation, Collagenase and Elastase Activity by synthesized Coenzyme $Q_{10}$ Derivatives (세포내 멜라닌 생성 및 Collagenase와 Elastase에 대한 Coenzyme $Q_{10}$ 유도체들의 억제활성)

  • Coenzyme $Q_{10}$ and six derivatives of coenzyme Qn were synthesized and tested for their inhibitory effects on melanogenesis occurred in murine melanoma (B16/F1) cells and on collagenase/elastase activities as well. As the result, synthetic coenzyme Qn showed a potent inhibitory effect on melanin formation, collagenase and elastase activities in all tested concentrations. Among these synthetic compounds, coenzyme $Q_1$ and coenzyme $Q_2$ potentially inhibited melanin formation and elastase activity when compared to other coenzyme Qn derivatives. For the collagenase activities, all coenzyme Qn derivatives inhibited 80-85% of controls. As compared, coenzyme Qn derivatives exhibited strong inhibitory activities with the decrease of isoprenoid unit number of coenzyme Qn derivatives except for collagenase activity. For the inhibition of collagenase activity, moiety of benzoquinone might be considered as the active functional group. Taken together, coenzyme $Q_1$ and coenzyme $Q_2$ might be used for functional cosmetics.

The Effects of Paeonia Lactiflora Pallas on Inhibition of Oxygen Free Radical, Anti-inflammation and MMP-1 Inhibitory Activity (적작약 꽃 추출물의 활성산소 억제와 항염증 및 MMP-1 발현 억제능 효과에 관한 연구)

  • This study attempted to investigate the effects of Paeonia Lactiflora Pallas (P. lactiflora) on the inhibition of oxygen free radical, anti-inflammation and MMP-1 inhibitory activity and examine its possibility as a functional cosmetic material. For test methods, the inhibition of oxygen free radical after measuring reactive oxygen species (ROS) in the cell, cytotoxicity assessment and anti-inflammation were measured, and MMP-1 inhibitory effects in the HDF cell were measured. According to the test, the inhibition of ROS was confirmed in RAW 264.7 and HDF cells. In terms of cytotoxicity assessment, 90% or higher cell viability was detected at $5/10{\mu}g/mL$ Paeonia Lactiflora Pallas extract while it was 80% or higher at other concentration levels in both RAW 264.7 and HDF cells. In addition, NO production was inhibited in the RAW 264.7 cell while MMP-1 was significantly inhibited in the HDF cell. The above results reveal a possibility of Paeonia Lactiflora Pallas extract as a functional cosmetic material after confirming the inhibition of ROS synthesis in the cell, antioxidant and anti-inflammatory effects by inhibiting NO synthesis, low toxicity on skin cells and anti-aging effect through MMP-1 inhibition.

Antioxidative and Antiaging Effects of Sorbus commixta Twig Extracts (마가목 가지 추출물의 항산화 및 항노화에 관한 연구)

  • In this study, the antioxidative effects and inhibitory effects on tyrosinase and elastase of Sorbus commixta (S. commixta) twig extracts were investigated. The aglycone fraction of S. commixta twig extract showed the prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity($FSC_{50}$, $13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities of S. commixta twig extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated by the luminol-dependent chemiluminescence assay. The 50 % ethanol extract among extracts showed the most prominent ROS scavenging activity ($OSC_{50}$, $0.189{\mu}g/mL$). The cellular protective effects of extract/fractions of S. commixta twig on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The 50 % ethanol extract and ethyl acetate fraction showed the cellular protective effects against ROS in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The inhibitory effect of S. commixta twig extract on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate ($IC_{50}$, $113.2{\mu}g/mL$) and aglycone fraction($IC_{50}$, $105.3{\mu}g/mL$) on tyrosinase showed more remarkable inhibitory effect than arbutin($IC_{50}$, $226.88{\mu}g/mL$), known as the whitening agent. The inhibitory effect of aglycone fraction ($IC_{50}$, $6.9{\mu}g/mL$) on elastase was simillar to quercetin($IC_{50}$, $6.1{\mu}g/mL$), flavonoid known as reference compound. These results indicate that extract/fractions of S. commixta twig can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. S. commixta twig extracts can be applicable to new functional cosmetics for anti-aging products.

Cellular Protective Effects and Antioxidative Activity of Resveratrol (레스베라트롤의 세포보호 및 항산화 효과)

  • In this study, the cellular protective effect of resveratrol on oxidative damage and its antioxidative activity were investigated. The free radical-scavenging activity ($FSC_{50}$) of resveratrol was measured to be $103{\mu}M$. The reactive oxygen species-scavenging activity ($OSC_{50}$) of resveratrol on the ROS generated in a $Fe^{3+}-EDTA/H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. Resveratrol displayed $0.042{\mu}M$ ROS scavenging activity, which is 9.6-fold higher than that of L-ascorbic acid ($0.405{\mu}M$) and had a more prominent cellular protective effect than (+)-${\alpha}$-tocopherol. When HaCaT cells were exposed to $800mJ/cm^2$ of UVB or treated with $30{\mu}M$ rose bengal, resveratrol protected the cells against oxidative stress in a concentration-dependent manner; however, it was unable to protect the cells when the damage was induced by 10 mM $H_2O_2$. These results indicate that resveratrol could be employed to improve and prevent the skin aging through its antioxidative and cellular protective activities.
