풋사과 추출물의 피부 보습 효과 (Skin Moisturizing Activity of Unripe Apple(Immature Fruit of Malus pumila) in Mice)

  • Objectives : Skin aging is generally characterized by wrinkles, sagging, loss of elasticity roughness, pigmentation and dryness. This changes is caused by reducing the elements constituting the extracellular matrix contributing to the physiological properties of the skin, such as collagen fiber, elastic fiber, and hyaluronic acid. Adequate skin hydration is important to maintain normal skin function and reduce skin aging. The present study is objective to observe skin moisturizing effects of Unripe apple(UA, Immature fruit of Malus pumila Mill) in vivo and its underlying molecular mechanisms. Methods : ICR mice were orally administerd UA(100, 200 and 400mg/kg/day) for 8 weeks, and skin water contents and the expression of transforming growth factor (TGF)-𝛽1, ceramide, hyaluronan and collagen type I(COL1) were measured in dorsal back skin of the mice. Gene expression of hyaluronan synthase(HAS1, HAS2, HAS3), collagen synthase(COL1A1, COL1A2) and TGF-𝛽1 were also determined by realtime RT-PCR. Results : Skin water contents and the expression of TGF-𝛽1, ceramide, COL1 and hyaluronan were significantly increased in UA group(100, 200 and 400mg/kg/day) compared to vehicle control. The mRNA expression of HAS isoform(HAS1, HAS2, HAS3), COL1A1, COL1A2, and TGF-𝛽1 were also significantly increased by UA. Conclusions : UA has skin moisturizing effects and enhancement activities in skin function related components(COL1, hyaluronan, ceramide and TGF-𝛽1). These results suggested that UA can be a developing candidate for developing alternative skin protective agent or functional food ingredient.

각질형성세포에서 왕불유행 헥산 분획물이 Laminin-332 발현에 미치는 효과 (Hexane Fraction of Melandrium firmum Extract Induces Laminin-332 Expression in Human Keratinocyte)

  • 피부 기저막(basement membrane, BM)이란 표피와 진피 사이에 존재하는 특별한 구조물로 표피와 진피를 단단히 고정시켜 피부 구조를 유지하는 데에 중요한 역할을 수행한다. 노화 및 자외선 노출에 의한 피부 기저막의 구조적 변화와 파괴는 피부 주름 형성과 탄력 저하를 포함하는 피부노화 현상의 요인으로 여겨지고 있다. Laminin-332 (LN-332)는 피부 기저막을 구성하는 주성분으로 피부에서 표피와 진피를 단단히 고정시키는데 중요한 역할을 한다. 본 연구에서는 왕불유행 헥산 분획물(Melandrium firmum hexane fraction, MFHF)이 각질형성세포에서 LN-332 발현에 미치는 효과를 확인하였다. 정량적 real-time PCR (RT-PCR)과 단백질 발현 분석을 통해서 MFHF가 LN-332의 mRNA 발현 및 단백질 발현을 촉진시키는 것을 확인하였다. 또한 MFHF가 어떤 신호전달 경로를 통해 LN-332 발현을 조절하는지 확인하기 위하여 p38 MAPK 억제제인 SB202190과 ERK1/2 억제제인 U0126을 처리한 결과, p38 MAPK 억제제에 의해서 LN-332 발현이 완벽히 억제됨을 확인하였다. 또한, 피부 기저막을 구성하고 있는 콜라겐 타입 VII과 integrin ${\alpha}6$의 mRNA 발현 역시 MFHF에 의해 증가하는 것을 확인하였다. 우리는 본 연구를 통해 MFHF가 각질형성세포에 작용하여 피부 기저막을 구성하는 성분들의 생성을 촉진할 수 있는 소재로 작용할 수 있다는 것을 확인하였다. 이러한 결과는 기저막의 구조적, 기능적 이상에 의해 나타나는 피부노화 현상의 개선을 위해 활용할 수 있을 것이라 제안한다.

항노화 효능을 가진 천연물에 대한 최신 연구 동향에 관한 연구 (A Study on the Latest Research Trends in Natural products with Anti-Aging Effects)

  • 본 연구에서는 식약청 고시 원료인 레티놀, 레티닐팔미테이트, 아데노신, 폴리에톡실레이티드레틴아마이드 4종류와 항 노화의 효능을 가진 천연 원료인 금송, 하고초, 맨드라미, 브라질린, 여뀌, 황기, 연교, 자근, 대황, 육종용에 대한 연구 동향을 살펴보았다. 현재까지의 항 노화 연구는 주로 콜라겐 생성과 엘라스타아제 합성 억제 기전을 중심으로 이루어지고 있는 것이 확인되었다. 하지만, 피부의 노화 과정은 여러 가지 원인에 의해서 발생하는 것으로 볼 때, 다양한 기전에 맞춰 피부 항노화에 도움을 줄 수 있는 안전하고 효과적인 천연 원료를 활용한 항노화 연구가 지속적으로 이루어져야할 것으로 사료되어 진다.

Combination of Grapefruit and Rosemary Extracts Has Skin Protective Effect through MMPs, MAPKs, and the NF-κB Signaling Pathway In Vitro and In Vivo UVB-exposed Model

  • Long-term ultraviolet (UV) exposure accelerates the phenomenon of skin photo-aging by activating collagenase and elastase. In this study, we aimed to investigate the effects of a combination of grapefruit and rosemary extracts (cG&Re) on UVB-irradiated damage in HaCaT cells and dorsal mouse skin. In HaCaT cells, cG&Re recovered UVB-reduced cell viability and inhibited protein expression of mitogen-activated protein kinases (MAPKs), such as extracellular signal-regulated kinases (p-Erk), c-Jun N-terminal kinases (p-JNK), and a class of MAPKs (p-P38). Also, cG&Re suppressed UVB-induced collagen and elastin degradation by decreasing matrix metalloproteinases (MMPs) and nuclear factor kappa light chain enhancer of activated B cells (NF-κB) expression, which is a transcription factor. Similar results were observed in dorsal mouse skin. Taken together, our data indicate that cG&Re prevent UVB-induced skin photo-aging due to collagen/elastin degradation via activation of MAPKs, MMPs, and the NF-κB signaling pathway in vitro and in vivo.

  • 인공피부배양물(DA-3711)은 동아제약의 인공피부배양기술을 활용하여 개발되었으며, 노화된 피부상태를 개선시켜 줄 수 있는 천연의 세포외기질 단백질을 비롯하여 인체성장인자 등의 영양성분을 함유하고 있다 DA-3711의 항노화 효과는 in vitro와 in vivo에서 규명되었으며, 또한 인체 효능 연구 결과에서도 피부탄력을 개선시키고 주름을 감소시키는데 매우 효과적이었다. 즉, DA-3711을 이용한 새로운 주름개선제는 피부재생을 촉진시켜 항노화 및 주름개선 효능을 나타낸다.

DEVELOPMENT OF POLYETHOXYLATED RETINAMIDE AS AN ANTI-AGING AGENT

  • A novel retinol derivative, polyethoxylated retinamide(Medimin A) was synthesized, as an anti-aging agent. Collagen synthesis, skin permeation, stability, and toxicity of Medimin A were evaluated and compared with those of retinol and retinyl palmitate. In vitro collagen synthesis was evaluated by quantitative assay of $[^3H]-proline$ incorporation into collagenase sensitive protein in fibroblast cultures. For in vitro skin permeation experiments, Franz diffusion cells(effective diffusion area: 1,766 $cm^2$) and the excised skin of female hairless mouse aged 8 weeks were used, The stabilities of retinoids were evaluated at two different temperature($25^{\circ}C\;and\;40^{\circ}C$) and under UV in solubilized state and in O/W emulsion. To estimate the safety, acute oral toxicity, acute dermal toxicity, primary skin irritation, acute eye irritation and human patch test were performed. The effect of Medimin A on collagen synthesis was similar to that of retinol. The skin permeability of Medimin A was higher than those of retinol and retinyl palmitate. The Medimin A was more stable than retinol and retinyl palmitate. Medimin A was nontoxic in various toxicological tests. These results suggest that Medimin A would be a good anti-aging agent for enhancing bioavailability and stability.

큰달맞이꽃 지상부 추출물의 피부 관련 생리활성 효과 (The Skin-Related Biological Activities of Aerially Extract of Oenothera lamarckiana)

  • Skin plays important roles in protecting the internal organs from the chemical-biological risk factors and ultraviolet light. Exposure to the chemical and biological stimuli has a detrimental effect on skin's structure and physiological regulation. Therefore, much attention has been paid to natural products that show biological activities such as anti-oxidation, anti-aging and anti-bacterial activities. In this study, we investigated the skin-related biological activities of Oenothera lamarckiana aerial part extract. The extract contained 229.35 mg TAE (tannic acid equivalents)/g total polyphenolic compounds and the extract showed relative high antioxidant activity ($SC_{50}$ value: $8.52{\mu}g/mL$). The $IC_{50}$ value against tyrosinase and elastase were 307.94 and $181.51{\mu}g/mL$, respectively. This suggested that O. lamarckiana can be applied to whiten skin and slow the aging of skin. O. lamarckiana extract showed a growth inhibitory effect on Staphylococcu epidermidis (minimum inhibitory concentration: $250{\mu}g/mL$). Interestingly, O. lamarckiana extract showed no inhibitory effect on Pseudomonas aeruginosa on the paper disc assay. Yet the extract inhibited the growth of Pseudomonas aeruginosa on the broth dilution assay in a dose-dependent manners. Taken together, O. lamarckiana could have good potential for development as an additive in the cosmetic industry.

Inhibitory Effects of a Combination of Grapefruit and Rosemary Extracts for Alleviating UV-Induced Skin Ageing

  • Ultraviolet (UV) radiation is associated with the development of extrinsic skin aging. We performed in vivo assays in order to investigate the protective effect of a combination of grapefruit and rosemary extracts (cG&Re) on UVB-induced skin aging. The results indicated that cG&Re displayed elastase inhibitory activity in a dose-dependent manner. Topical application of cG&Re mitigated photo-aging related lesions such as skin erythema and thickening in photo-aged BALB/c mice dorsal skin, by preventing UVB-induced collagen degradation. Immunohistochemical analyses revealed that cG&Re stimulated SIRT-1 expression, and suppressed MMP-1 and $IL-1{\beta}$ expression. It was observed that expression of MMP-1 and -13 mRNA was downregulated in the cG&Re-treated group. Furthermore, cG&Re treatment drastically suppressed protein expression of MMP-1 and regulated the phosphorylation of p-38 kinase. As expected, oral administration of cG&Re resulted in the same SIRT-1, MMP-1, and $IL-1{\beta}$ expression patterns observed upon topical application of cG&Re in the UV-induced mice model. Overall, the current results demonstrated that cG&Re attenuated both the downregulation of MMP-1 expression and up-regulation of SIRT-1 expression, as well as decreased phosphorylation of MAPK in UVB-induced skin ageing mice model, suggesting that cG&Re might be used as an internal food ingredient for beauty-purposes as well as a functional food material.

황련 추출물을 함유한 세럼의 피부 탄력과 윤기 개선 효과 (Serum Containing Coptis Japonica Extract Improve Skin Elasticity and Radiance)

  • 약용식물 황련(Coptis Japonica)을 이용한 세럼이 피부 항노화에 효과적인지 검증하기 위해 피험자의 피부 탄력과 윤기를 측정하여 개선율을 분석하고 설문을 통해 만족도를 평가하였다. 22명의 여성 피험자 한쪽 뺨 부위에 황련 추출물을 3% 배합한 세럼을 4주 동안 사용하게 한 후 시험 부위를 측정한 결과 피부 탄력은 사용 전 0.8505 ± 0.0395 R2(mm)에서 4주 후 0.8993 ± 0.0234 R2(mm)로 5.73% 증가하였다. 피부 윤기는 55.94 ± 5.49 (a.u.)에서 58.59 ± 4.66 (a.u.)로 4.74% 증가하였으며 유의확률이 모두 p<0.001로 나타나 두 항목 모두 개선율이 유의하게 증가한 것으로 나타났다. 설문에 의한 탄력 개선은 피험자 모두 100% 만족하는 것으로 나타났고 윤기 개선은 95.5% 만족하는 것으로 나타났으며 시험 기간 내 피부 이상은 확인되지 않았다. 이와 같은 결과는 향후 황련 추출물을 이용한 항노화 기능성 화장품의 연구와 제품 개발에 본 연구가 기초 자료로 활용될 것으로 사료된다.

Anti-wrinkle Effects of Water Extracts of Teas in Hairless Mouse

  • Tea flavonoids and polyphenols are well known for their extraordinary antioxidant activity which is considered important for anti-aging processes in animals. This study evaluated the anti-wrinkle effects of three different kinds of tea (Camellia sinensis) water extracts (CSWEs) including green, white, and black teas using a photoaged hairless mouse model. Data showed that the CSWE-treatment greatly improved skin conditions of mice suffering from UVB-induced photoaging, based on the parameters including the skin erythema index, moisture capacity, and transepidermal water loss. In addition, the wrinkle measurement and image analysis of skin replicas indicated that CSWEs remarkably inhibited wrinkle formation. In histological examination, the CSWE-treated mice exhibited diminished epidermal thickness and increased collagen and elastic fiber content, key signatures for skin restoration. Furthermore, the reduced expression of MMP-3, a collagen-degradative enzyme, was observed in the skin of CSWE-treated animals. Interestingly, comparative data between green, white, and black tea indicated that the anti-wrinkle activity of white tea and black tea is equally greater than that of green tea. Taken together, these data clearly demonstrated that CSWEs could be used as an effective anti-wrinkle agent in photoaged animal skin, implying their extended uses in therapeutics.
