Synthesis of Tetrapeptide Derived from Skin Structural Protein Sequence and Identification of Skin Anti-aging Effect

  • Several structural proteins present in keratinocytes of the skin are known to play an important role in the formation of epidermal tissue and barrier function, and the absence of structural proteins in keratinocytes causes various skin diseases. In this study, 42 types of tetrapeptides derived from the sequence of Loricrin, a kind of terminally differentiating structural protein, were synthesized, and skin anti-aging properties were measured by measuring the elastase inhibition, proliferation of skin cells. The anti-aging efficacy was verified and, based on this, it succeeded in selecting one of the most excellent peptides. It is expected that the selected tetrapeptide can be used as a raw material for various cosmetics and quasi-drugs based on anti-aging and skin cell proliferation effects.

Effect of Seed of Trichosanthes kirilowii on the Skin Barrier of High Dose Corticosteroid Injected hairless Mouse (과루인(瓜蔞仁)이 장기간 고용량 corticosteroid 투여 hairless mouse의 피부장벽에 미치는 영향)

  • Objective : Prolonged use of systemic corticosteroids negatively impacts skin barrier function. Corticosteroids have a major role in the practical management of many diseases, so it is necessary to find the drug or supplement which could keep the skin healthy during the systemic corticosteroids therapy. Seed of Trichossnthcs kiiilowii was commonly used for pulmonary disease in traditional Chinese medicine. Many studies have investigated and found that seed of Trichosanthes kirilowii has anti-bacterial, anti-HIV, anti-tumor and immunoregulatory functions. This study was performed to investigate the effect of seed of Trichosmthes kirilowii on the Skin Barrier Method : Triamcinolone 0.4mg was injected male hairless mice for 5 weeks ( 2 times a week, totally 10 times), Just before the first injection, hairless mice were divided into 3 groups; Control ( normal saline medicated group ), GroupA( seed of Trichosuuhes kirilowii qd/day medicated group ) and GroupB( seed of Trichosuuhes kiiilowii bid/day medicated group ), Body weight of all the groups were checked during the experiment, After $10^{th}$ injection, TEWL (Transeidermal water loss) of 3 groups were artificially increased by 9 times tape stripping and the changes of TEWL were checked at before stripping, right after, 2h, 4h, 6h, 24h, 48 and 72h later after stripping, AST, ALT and CBC were also checked. Indeed, stratum corneum of 3 groups were also examined and compared with that of normal hairless mouse. Results : 1. GroupB showed significantly lower body weight among three groups 2, There was no statistical difference at AST, ALT and CBC among three groups 3. GroupB was showed significantly lower TEWL than TEWL of Control at 48h later. 4 GroupB recovered it's TEWL of before tape stripping at 24h later, GroupA recovered that at 48h and Control recovered that at 72h later at the same condition 5. Stratum corneum of GroupA and GroupB were looked almost normal and healthy. On the contrary, stratum corneum of Control was looked thin and unhealthy. Conclusions : Seed of Ttichossnthes kirilowii has some effects on skin barrier function, especially TEWL of high dose corticosteroids injected hairless mouse and the efficacy seemed to be related with the dosage.

Development of a Cosmetic Ingredient Containing DHA Derivatives for Anti-inflammation, Anti-wrinkle, and Improvement of Skin Barrier Function (DHA 유도체를 이용한 항염, 항노화, 피부장벽 강화용 화장품 원료의 개발)

  • It is very important to control the inflammation of the skin because it can develop into diseases such as atopy as well as scarring and aging. In this work, we recently identified the in vitro synthesis of specialized pro-resolving mediators (SPMs) known to control inflammation in the human body and the applicability of cosmetics. Using recombinant protein of lipoxygenase from Glycine max, we succeeded to prepare mixtures of mono- or di-hydroxy DHA named as S-SPMs and used them for in vitro efficacy test. To investigate anti-inflammatory effect of S-SPMs, mRNA level of TNF-α and IL-6 were analyzed. Under UVB exposed condition, expression of both were decreased by S-SPMs treatment. And we observed reduced production of nitric oxide (NO) by S-SPMs application under the condition with diesel particulate matter (DPM). At the same experimental condition, malondialdehyde (MDA) production was decreased by S-SPMs, indicating the inhibitory effect of S-SPMs in lipid peroxidation. In addition, S-SPMs treatment resulted in reduction of matrix metalloproteinases-1 (MMP-1) expression and elevation of procollagen type I synthesis. Together with this, mRNA level of filaggrin and loricrin were increased by S-SPMs, indicating enhancement of skin barrier function. These results demonstrate that S-SPMs is a good candidate to develop as a cosmetic ingredient for anti-inflammation, anti-wrinkle, and improvement of skin barrier function.

  • Lips have a unique desquamation process and moisturizing properties that are structurally different from ordinary skin. In particular, the turnover cycle of the stratum corneum is fast and the outermost stratum corneum is thin, so the amount of keratin is relatively high, and there are no skin appendages, so it is very vulnerable to maintaining moisture. In this study, we set three targets for lip care : stratum corneum, moisture, and barrier, and aimed to identify the potential of three target-specific ingredients for lips improvement. We confirmed the exfoliating, moisturizing, and barrier improvement efficacy of three target-specific ingredients which also have mild exfoliating effect. Specifically, we verified that Bacillus clausii extract improved skin exfoliation, gluconolactone improved skin moisture retention, and serine reinforced skin barrier function. To test the in vivo efficacy of the complex composed of three target-specific ingredients on the human lips, the lip balm manufactured with non-irritating range of concentration was applied, and we confirmed that it was effective in improving lip exfoliation·moisturizing·elasticity·wrinkles. Consequently, it was confirmed that the beauty and health indicators of the lips could be improved through the exfoliation-moisturization-barrier care of the lips and these ingredients were applied to LG H&H LIPCERIN products.

A Study on Anti-Inflammatory, Antioxidant Activity and Skin Barrier Improvement of Lactobacillus rhamnosus-derived Exosomes (락토바실러스 람노서스 유래 엑소좀의 항염증, 항산화 효능 및 피부 장벽 개선에 관한 연구)

  • In this study, exosomes were isolated by ultrafiltration from Lactobacillus rhamnosus J2K-821 and their various effects for skin were evaluated. Their size and concentration were identified 50 ~ 200 nm and 3.22 × 108 particles/mL, respectively through nanoparticle tracking analysis. In order to verify the inflammatory relief effect of Lactobacillus rhamnosus-derived exosomes (LRDEs), their nitric oxide (NO) production inhibitory ability in RAW 264.7 macrophages induced an inflammatory reaction with lipopolysaccharide (LPS) was confirmed. It was revealed that they inhibited NO production in a concentration-dependent manner. To evaluate the antioxidant activity and skin barrier improvement effect of LRDEs, their 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and hyaluronidase inhibitory activity were confirmed. It was also revealed that their activities were increased by concentration-dependent manner. Through these results, It is believed that LRDEs can be used as a effective natural cosmetic ingredient for anti-inflammation, antioxidation and skin barrier improvement.

The effects of the Reynoutria japonica on skin-barrier and moisturizing in HaCaT cells (인간유래각질형성세포에서 호장근 추출물이 피부장벽 보호능과 보습능에 미치는 영향)

  • Reynoutria japonica is a perennate plant belonging to Polygonaceae and grows wild in East Asia containing Korea. Roots of Reynoutria japonica (R. japonica), part of roots of Reynoutria japonica, has been used for anti-inflammation and antispasmodics and contains emodin as active compound. Epidermis of skin is crucial roles to defense our body against stimulants, harmful substance and prevent water loss. In this study, we examined the effect of R. japonica and emodin, its active compound, on skin-barrier and moisturizing on HaCaT cells. First, antioxidant effect of R. japonica was prominent by scavenging ABTS+ radicals. Next, we conducted real time PCR and expression of filaggrin mRNA which is crucial role in differentiation of keratinocyte increased by R. japonica and emodin dose-dependently. In addition, R. japonica and emodin significantly elevated the expression of HAS-2 mRNA which play a role in hyaluronic acid synthesis on HaCaT cells. Taken together, R. japonica containing emodin, as active compound has potential as a cosmetic material for enhancing the function of skin-barrier and moisturizing in epidermis.

Effects of Commercial Topical Shampoos on Skin Surface Sebum in Normal Canine Skin (개에서 피부 치료용 샴푸가 표피유분에 미치는 영향)

  • Various commercial shampoos were frequently prescribed for dermatologicstherapy in small animal practice. Skin surface sebum affected by the shampoos, however, was not evaluated routinely. In order to evaluate the skin surface sebum for the exact prescription of shampoos methods to measure skin surface sebum of shampoos are needed. This study was undertaken to evaluate the skin surface sebum effect of shampoo on canine skin using sebometry. Five healthy dogs were applied with 3 commercial shampoos: Sebocalm, Sebolytics, and Benzoyl peroxide. Sebocalm had a great effect on skin surface seoum and normal saline also affected the skin sebum. Sebolytics and benzoyl peroxide had a moderate effect of skin sebum. The lowest effect was shown in detergent. Therefore, Sebumeter was considered a good and convenient apparatus for skin sebum measurement in order to evaluate the effect of shampoos on skin sebum. None of the products tested had any negative effect on the skin barrier function.

Study on Skin pH Improvement Effect through Regulation of Na+/H+ Exchanger 1 (NHE1) Expression of Prunella vulgaris Extract and Its Active Compound, Caffeic Acid (꿀풀 추출물과 그 활성 화합물인 카페인산의 Na+/H+ exchanger 1 (NHE1) 발현 조절을 통한 피부 pH 개선 효과에 대한 연구)

  • This study was conducted to discover substances that regulate skin surface acidification using human epidermal keratinocyte cell lines, and to investigate their effects on the moisturizing ability and skin barrier function of the stratum corneum. Prunella vulgaris (P. vulgaris) is an herb widely distributed in Northwest Africa and North America that has been studied for its anti-apoptotic, antioxidant, and anti-inflammatory effects. However, research on the regulation of NHE1 expression and the restoration of skin barrier function has not been conducted. Analysis of P. vulgaris revealed the presence of rosmarinic acid and caffeic acid as active ingredients, which were tested for toxicity in human epidermal keratinocyte cell lines (HaCaT), and showed no toxic effects were observed at high concentarion (100 ㎍/mL or 100 µM). It is known that sodium-hydrogen ion exchange pumps (NHE1) decrease in expression in aging skin to maintain the acidic pH of the stratum corneum, and it is hypothesized that this decrease plays an important role in the impaired restoration of skin barrier function in aging skin. P. vulgaris extract and caffeic acid increased the expression of NHE1 in keratinocytes, increased the expression of natural moisturizing factor (NMF) precursor filaggrin and ceramide synthesis enzyme serine palmitoyl transferase (SPT). In addition, P. vulgaris and caffeic acid decreased the extracellular pH of keratinocytes, indicating a direct effect on skin pH regulation. Taken together, these results suggest that P. vulgaris and caffeic acid can regulate skin pH through NHE1 modulation, and may help to restore skin barrier function by increasing NMF and ceramide synthesis. These results show the possibility that honeysuckle and caffeic acid can have a positive effect on skin health, and can be the basis for the development of new skin protection products using them.

Effect of Oil in Water Nanoemulsion Containing a Mixture of Lactic Acid and Gluconolactone for Skin Barrier Improvement (유산 및 글루코노락톤 혼합물을 함유하는 수중유형 나노에멀젼의 피부장벽개선 효과)

  • To evaluate the effectiveness of the skin barrier improvement of lactic acid (LA) and gluconolactone (GL), the expression of filaggrin, loricrin, hyaluronic acid (HA), hyaluronan syhthase-2 (HAS2), and aquaporine-3 (AQP3) in keratinocytes, and the moisture content and transepidermal water loss (TEWL) by clinical trials were evaluated. The expression levels of filaggrin and locricrin, which are the main factors affecting the proper functioning of skin barrier function, and HA, HAS2, and AQP3, which are skin moisturizing-related proteins measured by quantitative real-time polymerase chain reaction (qRT-PCR) and western blotting. The results showed that the expression levels of the factors that decreased by H2O2 treatment were significantly increased by LA, GL, and a mixture of LA and GL at the mRNA and protein levels (p<0.05). The nanoemulsion containing a mixture of LA and GL was prepared using the emulsion inversion method, and the average particle size was 299.9 ± 0.287 nm. After measuring the TEWL of nanoemulsion using Vapometer, it was found that TEWL significantly decreased by 15.53% and 26.73% after two weeks and four weeks of product use, respectively, compared to TEWL before product use (p<0.001). Similarly, the skin moisture content of the nanoemulsion significantly increased by 15.40% and 26.59% after two weeks and four weeks of product use, respectively, compared to skin moisture content before product use (p<0.001). Therefore, the skin barrier function and moisturizing effect of a mixture of LA and GL are shown by increasing the moisture content and decreasing the TEWL by increasing the expression of filaggrin, loricrin, HA, HAS2, and AQP3. This suggests the possibility for the development of functional cosmetic ingredients in the future.

A clinical study for the effect of ointment contained Takrisodok-Eum on recovering the damaged skin barrier by Atopic dermatitis (탁리소독음(托裏消毒飮) 함유 외용제의 아토피 피부염에 대한 임상적 연구)

  • Purposes : The purpose of present study is to detect the effects of ointment contained Takrisodok-Eum on the skin barrier's recovery of the Atopic dermatitis. Methods : A total of 44 patients who visited Semyung Oriental Medical Center from April 27th, 2011 to September 20th, 2011 were included in the study. In this study, the patients were treated with Takrisodok-Eum (experimental group) and positive control group. For 8 weeks of gross examination, hematological examination and instrumental asessment were made before and after the study to examine how well the products for experimental group with those for control group in recovering the damaged skin barriers by Atopic dermatitis. Results : 1. In the primary endpoint, SCORAD Index showed a statistically significant decline in experimental group. 2. In the secondary endpoint index of skin hydration, both the experimental group and the control group showed statistical significance. 3. In the secondary endpoint index of TEWL, the experimental group showed greater statistical significance than the control group. 4. In the secondary endpoint index of skin pH, skin pH showed a statistically significant decline in both the control group and the experimental group within the normal range. 5. In the secondary endpoint index of Total IgE, Eosinophil count, IL-10, IL-13, both the experimental group and the control group showed no statistical significance. 6. To evaluate the safety of the products for the human body, hematological examination and hematological biochemical examination, vital sign check were conducted; both control group and experimental group showed no abnormal level. And There were no severe adverse events during this study. Therefore, it is suggested that the safety of the products, if used for certain period, should be safe for the human body. Conclusions : According to the above experiments, it is suggested that Takrisodok-Eum should be effective for the Atopic dermatitis.
