Effect of the Extract of Stachys Affinis on DNFB-induced Allergic Contact Dermatitis in Mice (DNFB에 의해 알레르기성 접촉성 피부염이 유도된 마우스에서 초석잠 추출물의 효과)

  • This study aims to evaluate the anti-inflammatory effect of 80% ethanol extracts of Stachys affinis (MQ) on 1-fluoro-2,4-Dinitrofluorobenzene (DNFB) induced atopic dermatitis (AD) in Balb/c mice. AD-like allergic contact dermatitis was induced by challenge of DNFB on the ear after DNFB sensitization on the back sides of mice. MQ alleivated clinical severity in AD-like skin lesions. In addition, ear thickness of epidermis and penetration of inflammatory cells in AD-like skin lesions were decreased by topical application of MQ. The the serum levels of tumor necrosis factor-α (TNF-α) and interleukin-4 (IL-4) were measured in AD mice using ELISA kits. Levels of TNF-α and IL-4 in serum were significantly decreased by topical application of MQ. Therefore, this study could give a clinical basis that MQ could be a agent to prevent AD.

Sensitive method for the detection of Apple scar skin viroid(ASSVd) by nested reverse transcription-polymerase chain reaction

  • A rapid and sensitive assay for the specific detection of plant viroids using reverse transcription-polymerase chain reaction(RT-PCR) has been developed already. The nested RT-PCR assay cloud be applied for the detection of apple scar skin viroid(ASSVd) from young leaves and other tissues. ASSVd has central conserved region(CCR), terminal left(T$\sub$L/) and terminal right(T$\sub$R/) domain. Primers were designed from these regions. Primer sets were successfully applicable for the amplification of full length or partial region of ASSVd by nested RT-PCR. Nested RT-PCR assay was more sensitive and accurate method to detect ASSVd from young trees during the early time of apple cultivation.

Therefore the art of many medicines make use of the skin as a medium of treatment. Peter mandel discovered centers, or zones, on the human body, which represent or correspond to the three primary colors(red, blue and yellow). And he came across to the four temperaments(or humors) and their respective colors, which consist of the primary colors with the addition of green.

  • Lophatheri Herba, Patriniae Radix, Alli tuberosi Herba, and Menthae Herba have long been used externally for treatment of injuries or inflammations in Korea. This study was carried out to examine the anti-dandruff effect of the mixture made with equal parts of 70%-ethanol extracts of Lophatheri Herba, Patriniae Radix, Allii tuberosi Herba, and Menthae Herba (LPAM) and of each one above, compared with 1%-zinc pyrithione (Standard) used commercially as anti-dandruff. The results showed the mixture was more effective than each crude drug. The 10%-LPAM produced no toxicity in skin reaction test and eye irritation test, and showed significant antimicrobial activity against Pityrosporum ovale, the major cause of dandruff, and also showed significant anti-dandruff and itch-improvement effects on head skin.

  • This study was performed to investigate the myxosporidian parasites from several organs of freshwater fishes caught from Dongjin River, Jeonbuk Province of Korea. Twelve myxosporidian species were dectected from five fish species out of sixteen fish species. From the gall bladder of Opsariichthys uncirostris amurensis, Zschokkella opiocephali was found; from the skin, gall bladder, liver and spleen of Pseudobagrus fulvidraco, Zschokkella opiocephali and Myxobolus cheisini were found; from the gill, skin, gall bladder and kidney of Carassius carassius, Zschokkella ctenopharyngodonis, Chloromyxum koi, Sphaerospora sinensis, Myxosoma abbottinae, Myxobolus koi and Myxobolus suturalis were found; from the kindney of Misgumus anguillicaudatus, Myxidium lieberkuhni, Neomyxobolus ophicephali and Telohanellus misgurni were found; from the gill of Cyprinus carpio, Myxosoma abbottinae and Myxobolus dispar were found. All these parasitized fishes didn't show any specific symptom.

Antibacterial Activity of Acanthoic acid Isolated from Acanthopanax koreanum against Oral and Skin Microfloras

  • The (-)-pimara-9 (11), 15-dien-19-oic acid, acanthoic acid was extracted from the roots of Acanthopanax koreanum using bioassay-guided isolation of a MeOH extract. Acanthoic acid was assayed against Streptococcus mutans and Staphylococcus epidermidis causing dental caries and opportunistic pathogen. The minimum inhibitory concentration (MIC) of acanthoic acid against Streptococcus mutans and Staphylococcus epidermidis was 2 and 4 ${\mu}g/mL$, respectively, which was much lower than those of other natural antimicrobial agents such as 8 ${\mu}g/mL$ of tanshinone IIA. Acanthoic acid also significantly inhibited the growth of other cariogenic bacteria such as Streptococcus sobrinus and Streptococcus sanguis, and Streptococcus grodenii in the MIC range of 4${\sim}$32 ${\mu}g/mL$. Our findings suggest that acanthoic acid could be employed as a potential antibacterial agent for preventing dental caries and skin infections.

  • In this study, we determined the cause of a disease outbreak in small yellow croaker(Larimichthys polyactis) in Jeju island. The major external signs in the dead fish were hemorrhage of the skin. Vibrio harveyi were isolated from a few fishes and viruses were not detected from the diseased fish. However, flukes were confirmed on the skin and we conducted molecular identification and phylogenetic analysis of the isolated parasites. The obtained 28S rRNA sequence of our specimen(Accession No. OM333244) showed the highest homology with Neobenedenia girellae, while the COI sequence of our specimen showed the highest homology with N. melleni. Further sequence analysis with other genes and morphological observation are necessary for accurate identification.

Pilot Clinical Trial to Evaluate the Efficacy of Needle-Embedding Acupuncture Therapy at Gokji(LI11) on TEWL (곡지혈 매선침 시술이 경피수분 손실량에 미치는 영향)

  • The purpose of pilot clinical trial is to evaluate the efiicacy and safety of Needle-Embedding acupuncture therapy on TEWL(Transepidermal Water Loss) and Skin hydration. A total of 24 human who visited Dongshin Oriental Medical Center from December 1st, 2016 to July 31st, 2017 were included in the pilot clinical trial. Needle-Embedding acupuncture therapy performed at Gokji(LI11). We observed change of TEWL(Transepidermal Water Loss), Skin hydration before and after Needle-Embedding acupuncture therapy. In the primary endpoint, index of TEWL showed a statistically insignificant decline ($8.5{\pm}2.12{\rightarrow}7.8{\pm}1.53g/h/m^2$). Index of skin hydration showed a statistically significant result ($38.2{\pm}6.77{\rightarrow}36.8{\pm}6.64$). To evaluate the safety, Adverse events and Vital sign check were conducted and there were no problem. And when the physical response, self-awareness, skin system, musculoskeletal pain, and other abnormal responses were assessed 60 minutes and 24 hours and 7 days after Needle-Embedding acupuncture therapy at Gokji(LI11) for stability assessment, Adverse events disappeared during a pilot clinical trial. According to the above pilot clinical trial, it is suggested Needle-Embedding acupuncture therapy were effective for Skin moisturizing and safe.

Transmission of Apple scar skin viroid by Grafting, Using Contaminated Pruning Equipment, and Planting Infected Seeds

  • Apple scar skin, one of the most destructive diseases affecting apple, is caused by Apple scar skin viroid (ASSV d). Fruit dappling appeared on several cultivars in Korea and has been distributed to major cultivated areas since 2001. ASSVd was identified from infected fruits by using nucleic acid sequence-based amplification with electrochemiluminescence (NASBA-ECL). NASBA-ECL method was faster and hundredfold more sensitive than reverse transcription-polymerase chain reaction (RT-PCR) for ASSVd detection in apple leaves/ stems. ASSVd was rapidly transmitted to the entire tree in the second year after artificial inoculation. The ASSVd could be transmitted efficiently by using contaminated pruning scissors to both lignified stems (60 to $70\%$) and green shoots (20 to $40\%$) of apple tree and young plants. Dipping of contaminated scissors in $2\%$ sodium hypochlorite solution effectively prevented viroid transmission. In the ASSV d-infected fruits, the viroid was easily detected from fruit skin, seed coat, and embryo. Moreover, embryo and endosperm separately excised from the ASSVd-infected seeds were ASSVd positive in NASBA-ECL assay. Seedlings germinated from ASSVd-positive seeds showed $7.7\%$ infection rate., which indicated that ASSVd is seed-borne.

Inhibitory Effects of Saposhnikoviae Radix Extracts on the Melanin Production and Elastase Activity in B16F10 cells (흑색종 세포주에서 멜라닌 생성과 엘라스타제 활성 억제에 미치는 방풍의 효과)

  • Saposhmikoviae Radix can treat various skin disease by anti-pruitus and anti-inflammatory effects. This study was designed to investigate effects of Saposhmikoviae Radix Extracts(SRE) on skin elasticity and whitening using B16F10 cell lines. In this experiment, We observed effect of SRE on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. In results, SRE treated group showed lowered proliferation rates significantly compared to non-treated group. More than SRE $125{\mu}g/m{\ell}$ of treated groups were lower levels of melanin synthesis respectively. SRE did not show inhibitory effect on tyrosinase activities in vitro and in B16F10 cells. Finally, SRE suppressed elastse type I and IV activities in dose-dependent manner in vitro. And SRE also slightly suppressed elastase activities in B16F10 cells. In conclusion, these results suggest that SRE can inhibit melanin synthesis and inhibt elastase activity. So, We suggest that SRE can be maintained skin elasticity or whitening.
