Structure and Expression of the Chicken Myostatin Gene

  • A new murine TGF-$\beta$ family member, myostatin(growth/differentiation factor-8) is expressed specifically in developing and adult skeletal muscle and may be a negative regulator of skeletal muscle development. This study aims at characterization and identification of genomic organization of chicken myostatin gene. In thi study, we identified the genomic organization and sequence of chicken myostatin gene. Results of RT-PCR and Northern blots from various tissues showed different mRNA expression levels in developmental stages of chick embryos and demonstrated strong expression of myostatin mRNA in skeletal muscle. These facts suggest that chicken myostatin gene would play an important role not only in skeletal muscle cell but also in other tissues.

  • Purpose : The purpose of this study is to setup a standard treatment protocol in surgical correction of skeletal malocclusion with Angle Class III patients. We asessed the soft tissue changes according to skeletal changes of patients who have undergone orthognathic surgery with bilateral split sagittal ramus osteotomy (BSSRO) and advancing genioplasty. Materials & Methods : The soft tissue change of 9 skeletal Class III patients was assessed after BSSRO and advancing genioplasty. The patient group was skeletal Class III who was surgically treated by BSSRO & advancing genioplasty. The average follow up period is 13 months with the range of 6 and 30 months. All patients have undergone preoperative and postoperative orthodontic treatment. The assessment was devided into two groups. One was antero-posterior relationship and the other was vertical relationship of dimensional changes of soft tissue after orthognathic surgery. Results : In antero-posterior dimensional changes after surgery, the percentage of soft tissue change in comparison to hard tissue was 89%. Vertical ratio after surgery, 86% soft tissue changes were assessed.

인삼이 칼슘결핍 흰쥐골격근의 소포체에 의한 칼슘 - Uptake에 미치는 영향 (The Effect of Ginseng on the Calcium Uptake by Sarcoplasmic Reticulum Fragments Isolated from Potassium Deficient Rat Skeletal Muscles)

  • The effect of orally administered ginseng ethanol extract on potassium deficient and normal rat skeletal muscles was investigated in terms of Ca uptake by sarcoplastic reticulum fragments. The ginseng ethanol extract (100mg/kg/day) was administered orally to Sprague-Dawley rats for 21 days and their changes of body weights, $K^+$ content in skeletal muscles, and calcium uptake capacity of sarcoplamic reticulurn of each groups were measured. The growth rate of rats fed with the potassium deficient diet was significantly decreased compared to that of normal rats. Ginseng components did not show any effect on the decreased growth rate of the postassium deficient rats. Potassium content in skeletal muscle from potassium deficient rats was significantly reduced compared to that of normal rats. Ginseng components showed the tendency to prevent the reduction in potassium content of potassium deficient rats, but differences were not statistically significant. Calcium uptake of SR prepared from skeletal muscles of potassium deficient rats was increased significantly compared to that of normal rats. Ginseng components prevented such increase of calcium uptake by 30%. In summary, it can be concluded that ginseng may prevent the increase in Cauptake of SR obtained from potassium deficient rats.

Skeletal cavernous hemangiomas of the frontal bone with orbital roof and rim involvement

  • Skeletal cavernous hemangiomas are rare, benign tumors that may involve the supraorbital rim and orbital roof. However, such involvement is extremely rare. We report a case of skeletal cavernous hemangioma of the frontal bone involving the orbital roof and rim. En bloc excision and reconstruction, using a calvarial bone graft for the orbital roof and rim defect, was performed. It is important not only to perform total excision of skeletal cavernous hemangiomas, but to properly reconstruct the defects after the total excision since several complications can arise from an orbital roof and rim defect.

  • Sex steroids influence the maintenance and growth of muscles. Decline in androgens, estrogens and progesterone by aging leads to the loss of muscular function and mass, sarcopenia. These steroid hormones can interact with different signaling pathways through their receptors. To date, sex steroid hormone receptors and their exact roles are not completely defined in skeletal and smooth muscles. Although numerous studies focused on the effects of sex steroid hormones on different types of cells, still many unexplained molecular mechanisms in both skeletal and smooth muscle cells remain to be investigated. In this paper, many different molecular mechanisms that are activated or inhibited by sex steroids and those that influence the growth, proliferation, and differentiation of skeletal and smooth muscle cells are reviewed. Also, the similarities of cellular and molecular pathways of androgens, estrogens and progesterone in both skeletal and smooth muscle cells are highlighted. The reviewed signaling pathways and participating molecules can be targeted in the future development of novel therapeutics.

An efficient seismic analysis of regular skeletal structures via graph product rules and canonical forms

  • In this study, graph product rules are applied to the dynamic analysis of regular skeletal structures. Graph product rules have recently been utilized in structural mechanics as a powerful tool for eigensolution of symmetric and regular skeletal structures. A structure is called regular if its model is a graph product. In the first part of this paper, the formulation of time history dynamic analysis of regular structures under seismic excitation is derived using graph product rules. This formulation can generally be utilized for efficient linear elastic dynamic analysis using vibration modes. The second part comprises of random vibration analysis of regular skeletal structures via canonical forms and closed-form eigensolution of matrices containing special patterns for symmetric structures. In this part, the formulations are developed for dynamic analysis of structures subjected to random seismic excitation in frequency domain. In all the proposed methods, eigensolution of the problems is achieved with less computational effort due to incorporating graph product rules and canonical forms for symmetric and cyclically symmetric structures.

Extraskeletal Calcifications in Children with Maintenance Peritoneal Dialysis

  • Chronic kidney disease (CKD)-mineral and bone disorder (CKD-MBD) is a common complication of CKD, often accompanied by extra-skeletal calcification in adult patients. As increased vascular calcification is predicted to increase cardiovascular mortality and morbidity, the revised Kidney Disease: Improving Global Outcomes guidelines recommend avoiding calcium-containing phosphate chelators. However, extra-skeletal calcification is less commonly noticed in pediatric patients. Here, we report our experience of such a complication in pediatric patients receiving maintenance peritoneal dialysis. Extra-skeletal calcification was noticed at the corneas, pelvic cavity, and soft tissues of the lower leg in 4 out of 32 patients on maintenance peritoneal dialysis. These patients experienced the aggravation of extra-skeletal calcifications during peritoneal dialysis, and 2 of them underwent excisional operations. It is required to monitor extra-skeletal calcifications in children on kidney replacement therapy.

Comprehensive Investigation on the Prevalence and Risk Factors of Coexistence of Age-related Loss of Skeletal Mu scle Mass and Obesity among Males in Their 40s

  • PURPOSE: This study examined the prevalence and specific risk factors in males aged 40-49 years with the coexistence of age-related loss of skeletal muscle mass and obesity (CALSMO). METHODS: The current study analyzed the data obtained from a cross-sectional study involving a sample of 1,218 men who resided in the community and fell within the age range of 40 to 49 years. Multiple risk factors were examined: age, height, weight, body mass index, waist circumference, skeletal muscle mass index, smoking and drinking habits, systolic and diastolic blood pressure, fasting glucose levels, and triglyceride and cholesterol levels. All data were analyzed via complex sampling analysis. RESULTS: The coexistence of age-related loss of skeletal muscle mass and obesity in males was 2.94% (95% CI: 2.06-4.17). The clinical risk factors were low height, high weight, body mass index, waist circumference, skeletal muscle index, systolic blood pressure, diastolic blood pressure, and fast glucose (p < .05). CONCLUSION: The study identifies the prevalence and risk factors for CALSMO among adults in the community. These findings contribute to the existing literature on CALSMO and highlight potential risk factors associated with CALSMO development in males aged 40-49 years.

돼지의 품종 및 성장 단계에 따른 등심조직의 단백질 발현 양상 비교, 분석 (Protein Expression in Pig Species Longissimus dorsi Muscles among Different Breeds and Growth Stages)

  • Landrace와 Berkshire의 longissimus dorsi muscle으로부터 단백질 발현양상의 차이를 보기 위하여 2-DE실험을 통하여 분석한 결과 Landrace 에서 특이적으로 발현 양이 증가한 단백질들은 serum albumin precursor, troponin T (TnT; slow skeletal muscle), myoglobin였다. Berkshire에서 특이적으로 발현 양이 증가한 단백질들은 heat shock 27 kDa protein 1, troponin T (fast skeletal muscle), muscle creatine kinase, phosphoglucomutase 1, triosephosphate isomerase (Tpi 1), adenylate kinase isoenzyme 1 (AK1)였다. Landrace의 longissimus dorsi muscle에서는 slow skeletal muscle과 연관된 단백질들이 발현된 반면에 Berkshire에서는 fast skeletal muscle, 물질대사경로, 에너지 생산과 관련된 단백질들이 발현되었다. Berkshire를 이용하여 성장단계별로 단백질 발현을 분석해 본 결과 growing Berkshire에서 발현이 증가한 단백질은 aldehyde dehydrogenase 1 family, member L1 (ALDHL1)와 muscle creatine kinase이고 finishing Berkshire에서 발현이 증가한 단백질은 heat shock 27 kDa protein 1, TnT (slow skeletal muscle), TnT (fast skeletal muscle), serum albumin precursor, PGM 1, AK 1, Tpi 1였다. 이 결과는 Finishing Berkshire의 등심에서는 growing Berkshire에 비교하여 골격근육, 에너지물질대사, 세포골격 등이 보다 활성화된 것으로 사료된다.

  • Evidence has suggested that an anion channel blocker, 4, 4'-diisothiocyanatostilbene-2, 2' disulfonic acid (DIDS) could trigger Ca release from skeletal sarcoplasmic reticulum (SR) by binding to a 30 kDa SR protein. Since the high molecular weight $Ca^{2+}$ release channel (CRC)/ryanodine receptor (RyR) is the main SR protein that conducts $Ca^{2+}$ efflux in skeletal muscles, the relationship between CRC and the 30kDa protein remains to be elucidated.(omitted)
