The pre-concentration and determination of ultratrace arsenic in water samples was studied by the precipitate flotation technique. The arsenic in 1.0l of water sample, in which all suspended materials were filtered out, was coprecipitated together with La(OH)$_3$ precipitates at pH 8.5${\pm}$0.1. After the precipitate was made to be hydrophobic by adding mixed surfactant of 1 : 8 mole ratio of sodium oleate and sodium dodecyl sulfate, it was floated with the aid of tiny bubbles of nitrogen gas in a flotation cell. The floated precipitate was quantitatively collected on a micropore glass filter by the suction, dissolved with small volume of 1.0M sulfuric acid, and accurately diluted to 25.00ml with a de-ionized water. Total arsenic was spectrophotometrically determinated by forming silver diethyldithiocarbamate complex of arsine generated from arsenic in the concentrated solution. The calibration curve was linear up to 20ng/ml in the original solution. Analytical results showed that contents of arsenic in a campus wastewater and a river water were 8.2ng/ml and l.0ng/ml, respectively, and their recoveries were 93${\%}$ and 90${\%}$ in water samples which a given amount of arsenic was added into. From above result, it could be concluded that this method was applicable to the determination of arsenic in various kinds of water at low ng/ml levels.
Amid the recent growing social demand for wellness culture and healing tourism, I, through a literature survey and case study, reviewed the conditions of healing tourism in Gangwon-do as a destination of healing tourism, and examined the development project of Gangwon-do type healing tourism and commercialization of the wellness perspective. According to the survey, it was found that the Yeongdong area in Gangwon Province had excellent healing-based conditions in virtue of abundant natural resources, but it was found that it was necessary for local governments to cooperate in order to prevent overlapping investments and create synergy effects by connecting with one another as development contents or business items of healing tourism projects overlap or remain in the planning stage. In the case of Goseong-gun, which was reviewed as the main site, it has high potential as a silver-healing tour and if the County seeks a wellness merchandising strategy for each healing tourism target based on a therapy program linked with local specialized resources, and effectively links and specializes regional strengths in Gangwon-do's Yeongdong region, it is expected to become a regional success story of operating the development of the local tourism for silver and health-oriented tourists.

Contents of water insoluble matter in synthetic food colors tested were not different by general test methods of various organs. Contents of chloride and sulfate salts in Food Green No.3, Food Red No.3, Food Blue No.2, and Food Yellow No.4 slightly differed in various methods, and but up to the standard, and general test methods of JECFA and USA needed far more time. This project has been worked out for isolation of EPA-producing bacteria from marine source of sea water, sea sediment and intestinal contents eviscerated from some red-muscle fish such as mackerel, horse-mackerel and spike fish. The samples were precultured on the media of PPES-II glucose broth and then pure-cultured on Nutrient agar and P-Y-M glucose. Lipids extracted from those bacterial mass collected by centrifugation were analysed in terms of lipid class and fatty acid composition. The results are resumed as follows : 1. 112 strains from sea water and 76 strains from sea sediment were tested for their EPA producing capability, but both strains of (SA-67 and SA-91) from the former and four strains(SS-35, 37, 51 and 71) from the latter have been proved to produce EPA above the level of 2% of total fatty acids. The strains such as GS-11, 29, 31, HM-9, 29, B-18, 33, 107, YL-129, 156, 203, 77, 104 and 256 which were isolated from fish intestinal contents, have also produced EPA at higher level than 2% of total fatty acids. 2. Contents of total lipids extracted from the cultures of these strains grown at $25^{\circ}C$, range from 2.8% to 6.9% (on dry weight %), and they are mainly composed of polar lipids($40.9{\sim}52.9%$) such as phosphatidyl glycerol($^{+}cardiolipin$)(?) and phosphatidyl ethanolamine ($33.8{\sim}40.0%$), with smaller amount of free fatty acid ($11.2{\sim}20.2%$). 3. EPA was isolated from a mixture of fatty acid methyl esters obtained from the lipid of each strain by HPLC in silver-ion mode and was identified by GC-Mass spectrometry. 4. The strains of SW-91, GS-11, GS-29, HM-9, B-18 and YL-203 grown at $25^{\circ}C$ have a level of 5% EPA in their total fatty acids, and the GS-11 and HM-9 strains show a tendency of increase in the EPA level with an increase of growth temperature.
The heavy metal contents and their dispersion patterns in stream water, stream sediments, land plants and aquatic larvae collected from the hydrologic system flowing via the wasted ore dump of the Moak Au-Ag mine were investigated systematically in order to evaluate the environmental impacts of the abandoned metal mine. The heavy metal content increases abruptly in the vicinity of the wasted ore dump, then attenuated with increasing distance from the mine area. Attenuating rates were stream water > stream sediments > land plants > aquatic larvae. On the other hand, the cumulative content of heavy metals was stream sediments >aquatic larvae > land plants > stream water. Each element tends to be enriched selectively according to media; Zn > Cu > Cd > Pb in stream water, Zn > Pb > Cu > Cd in stream sediments and land plants, and Zn > Cu > Pb > Cd in aquatic larvae. These results show that the degree of enrichment and dispersion of pollutant extruded from the wasted ore dump are different according to elements and media, and that the circulation system of materials of each medium is different. The heavy metals, especially Cu, Pb and Zn, of polluted downstream sediments occur in high proportions of Fe-Mn oxides and organic bounded forms, which show high potential of a secondary pollution source. The content of heavy metals and their dispersion patterns in stream sediments are different from those of ten years ago; pollution levels of heavy metals were degraded in various ranges. The Zn and Cu-polluted areas were widened whereas Fe and Pb-polluted areas were reduced. In crops collected from the farm lands in downstream area, the pepper was more concentrated in all heavy metal than rice. The pepper showed some contaminated level in Cu(9.7ppm) and Zn(149ppm), and the rice in Zn(90ppm). However, both crops showed no significant level in Cd(<0.2ppm) and Pb(<0.5ppm).
Magnolia kobus leaf extract was used for the synthesis of bimetallic Au core-Ag shell nanoparticles. Gold seeds and silver shells were formed by first treating aqueous solution of $HAuCl_4$ and then $AgNO_3$ with the plant leaf extract as reducing agent. UV-visible spectroscopy was monitored as a function of reaction time to follow the formation of bimetallic nanoparticles. The synthesized bimetallic nanoparticles were characterized with transmission electron microscopy(TEM), energy dispersive X-ray spectroscopy(EDS), and X-ray photoelectron spectroscopy(XPS). TEM images showed that the bimetallic nanoparticles are a mixture of plate(triangles, pentagons, and hexagons) and spherical structures. The atomic Ag contents of the bimetallic Au/Ag nanoparticles determined from EDS and XPS analysis were 34 and 65 wt%, respectively, suggesting the formation of bimetallic Au core-Ag shell nanostructure. This core-shell type nanostructure is expected to have potential for application in surface enhanced Raman spectroscopy and in the sensitive detection of biomolecules.

The potency of influenza vaccine is determined based on its hemagglutinin (HA) content. In general, single radial immunodiffusion (SRID) assay has been utilized as the standard method to measure HA content. However, preparation of reagents for SRID such as antigen and antibody takes approximately 2~3 months, which causes delays in the development of influenza vaccine. Therefore, quantification of HA content by other alternative methods is required. In this study, we measured HA contents of H1N1 antigen and H1N1 influenza vaccine by reverse phase-high performance liquid chromatography (RP-HPLC) methods. The presence of HA1 and HA2 was investigated by silver staining and Western blot assay. In addition, accuracy and repeatability of HA measurement by RP-HPLC were evaluated. Comparison of HA concentration by SRID and RP-HPLC revealed a precise correlation between the two methods. Our results suggest that RP-HPLC assay can replace SRID in the event of a pandemic flu outbreak for rapid vaccine development.

Calcareous corpuscles are a characteristic structure found in larval and adult stage cestddes, These corpuscles are known to contain several protein components and to possess protein-binding activity. However, the proteins bound to calcareous corpuscles in situ have not been studied. The present study was undertaken to identify the proteins on calcareous corpuscles. Calcareous corpuscles were purified from the plerocercoids (= spargana) of Spirometra erinacei, and serially dissolved using 0.1 M sulfamic acid solution. Collected supernatants were examined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and silver staining. The results showed that only the fraction remaining after the 19th dissolved fraction contained proteins. A total of 20 protein molecules were detected in gel, with major bands at 56, 53, 46, 40, 35, 29, 28, 24.5, 21, 19, 16, 13, 10 and 8 kDa. In particular, the proteins corresponding to the 21 and 16 kDa bands were most abundant. Our results demonstrated for the first time the protein contents of the calcareous corpuscles of spargana. Further studies on the functions of these proteins are required.
A variety of the porcelain fused to gold(PFG) have been developed to which porcelain can be fused. PFG alloys developed for this purposed have a high melting point and do not discolor when combined with porcelain. The design of the compositions of PFG is very important to esthetic restorative materials applying to porcelain. The purpose of this study is on the change of physical and mechanical characteristics in PFG 18K alloy by the small additional elements. Principal results are as follows. The high Au alloy containing 18Karat gold contents is respectively Au(75%), Pd(10%), Pt(4%), Ag(4%), In(2%), Sn(2%), Cu(2%), Ti(1%). These alloys are composed mainly of gold, platinum, silver and palladium with a few percent of the additional elements. By the addition of small amounts of elements such as In, Sn, Ti, the fine grain castings are produced in gold alloy and the small addition of platinum is very effective in increasing of hardness and strength. These gold alloys are representative of the changes to be expected as a result of heat treatment. These changes in strength and hardness values are sufficient to demonstrate a significant difference in performance between a as-casted and a heat-treated. These alloys have mechanical properties characteristics of Type and Type gold alloys. These alloys are useful to porcelain-metal restorations and dental laboratory. Also the porcelain fused to metal(PFM) alloys containing gold are commonly use for dental purposes in dental laboratory.
