Digitalis purpurea L. is a medicinal herb and have been used to congestive heart failure, mycocardial infarction, edema, angina etc. A protocol has been developed for in vitro propagation of adventitious shoot buds directly from leaf segments of D. purpurea Leaf explants of D. purpurea directly formed shoot buds when cultured on a MS medium supplemented with $2\;mg/l$ BA and $0.1\;mg/l$ IAA for 5 weeks. Adventitious shoots were multiplied by subculturing on the $B_5$ medium and shoot elongation was developed by subculturing on the WPM medium. Root formation from the shoot regenerated was achieved on MS basal medium containing 1 mg/ IBA.
In the present study, in vitro multiple shoot induction was achieved from cotyledonary node and stem nodal explants of cv. NARI-6 of safflower (Carthamus tinctorius L.). Among various growth regulators tested, MS salts and B5 vitamins supplemented with BA (6-Benzy-laminopurine) $17.76\;{\mu}M$ and KN (Kinetin) $6.96\;{\mu}M$ phytohormonal combination was found to be the most effective in initiating numerous shoot buds after 30 days of culture than BA ($4.44-44.39\;{\mu}M$) or KN ($2.32-46.40\;{\mu}M$) alone in the medium. In addition, 0.8% (w/v) agar (Hi-media) and 3.0% sucrose (w/v) was the optimum level for the formation of adventitious shoots. Further results showed the maximum shoot elongation occurred on MS medium with BA ($8.88\;{\mu}M$) and $GA_3$($11.56\;{\mu}M$) combinations. Efficient rooting occurred on quarter strength MS medium with NAA $10.74\;{\mu}M$. The regenerated plantlets were acclimatized and successfully transferred to the field.
This study was carried out to investigate the optimal conditions for efficient in vitro growth of Asparagus cochinchinensis (Lour.) Merr. shoot derived from shoot tip. Shoots were successfully cultured in MS medium. It was found that $23^{\circ}C{\sim}25^{\circ}C$ were suitable for shoot growth. The growth of shoot was greatly influenced by cytokinins. The shoots derived from shoot tip were well elongated on MS medium supplemented with BA and zeatin. In vitro shoots were very poor or no growing. Especially, 3.0 mg/l BA on MS medium was very effective in elongation of shoot. Root formation from in vitro growth of plant let was achieved on MS medium supplemented with 0.5 mg/l IBA. The results suggest that selection of plant growth regulator could be important factor to achieve an efficient in vitro growth.
Background: Although cetrimonium bromide is widely used for its bactericidal effects, the safety of cetrimonium bromide remains controversial. Therefore, the phytotoxicity of cetrimonium bromide was tested to evaluate its acute toxicity to plants and possible toxicity to other organisms and the ecosystem. Results: The germination rates of two test species, Lactuca sativa and Brassica campestris, were significantly decreased after cetrimonium bromide treatment. Furthermore, cetrimonium bromide treatment at over 1 mg/L concentration significantly affected root elongation immediately after germination. In pot experiments with semi-mature plants, significantly decreased shoot elongation and chlorophyll content were detected in both species following cetrimonium bromide treatment. Cetrimonium bromide treatment also significantly increased the antioxidant enzyme activities of plants. Conclusion: Our results show that cetrimonium bromide is phytotoxic, and since phytotoxicity testing can imply potential toxicity in the environment, further studies of the environmental toxicity of cetrimonium bromide should be performed.
Plant regeneration via organogenesis from leaf and stipule explants of micropropagated shoots of strawberry (Fragaria $\times$ ananassa cv. Suhong) was achieved. Leaf and stipule explants were detached from shoot-tip cultured shoots and cultured on MS medium with various combinations of BA and NAA under light or dark condition. Shoot regeneration from leaf explant was observed after 3 weeks in culture and was good at the high ratio of BA and NAA among various combination treatments. The highest shoot regeneration frequency from leaf explants was obtained with 1.0 mg/L BA and 0.1 mg/L NAA, in which 31.1% shoot regeneration frequency(1.7 shoots per leaf explant) was yielded. In case of stipule explants, shoot regeneration was largely affected by plant growth regulators during incubation under dark condition for initial 4 weeks but not under continuous light condition. The combination treatment with 2.0 mg/L BA and 0.1 mg/L NAA showed the most excellent shoot regeneration from stipule explants, where 44.4% regeneration frequency(4.0 shoots per explants) obtained. Regenerated shoots were rooted on MS medium with 0.1 mg/L NAA after shoot elongation, and the plantlets regenerated were transferred to soil mixtures with vermiculite and perlite for acclimation.
'Meyer' zoysiagrass(Zoysia japonica Steud.) is a popular turfgrass species used for transition zone golf course fairways and tees in mfd U.S.A golf courses because it is generally winter hardy while providing an excellent playing surface with minimal chemical and irrigation inputs. However, its functionality declines easily in many of the shaded areas of these courses. Reduced irradiance causes excessive shoot elongation, reduced tillering, and weak plants that are poorly suited to tolerate or recover from traffic and devoting. Trinexapac-ethyl (TE) effectively reduces gibberellic acid (GA) biosynthesis and subsequent shoot cell elongation. This study was initiated to evaluate TE effect on shoot elongation and stand persistence under two levels of shade in 'Meyer' zoysiagrass. A mature stand of 'Meyer' was treated with all combinations of three levels of shade(0%, 79%, and 92%) and three levels of monthly TE [0, 48 $g{\cdot}ha^{-1}$ a.i(0.5x) and 96 $g{\cdot}ha^{-1}$ a.i(1x)]. In full sun, the TE at 48 $g{\cdot}ha^{-1}$ a.i reduced clipping yield by 18% over a four-week period and, whereas the TE at 96 $g{\cdot}ha^{-1}$ a.i by 30% to 38%. Monthly application of TE at the 96 $g{\cdot}ha^{-1}$ a.i increased 'Meyer' tiller density in full sun and under 79% shade. Both rates of TE consistently reduced shoot growth under shade relative to the shaded control. Only the monthly applications of the TE at 96 $g{\cdot}ha^{-1}$ a.i consistently delayed loss of quality under 79% shade. Our results indicate TE can be an effective management practice to increase 'Meyer' zoysiagrass persistence in shaded environments.
The toxic effects of aluminium (Al) on growth, chlorophyll content, $\delta-aminolevulinic$ acid dehydratase (ALAD) activity and anatomy of root and shoot were investigated in 7-day-old azuki bean (Vigna angularis) seedlings. Significant depressions in root elongation was observed in the low concentrations of Al (50, 100 $\muM)$ and increasing Al concentrations caused a sharp decline of root and shoot growth. The degree of inhibition was dependent upon Al supply. Exposure to 50 $\muM$ Al or more inhibited root elongation within 1 day. In the 50 $\muM$ Al treatments, a recovery of root growth was seen after 7 days exposure. In contrast, lateral root initials was little affected by Al exposure. Al toxicity symptoms and growth responses were more well developed in the roots than in the shoots. Analysis of Al localization in root cells by hematoxylin stAlning showed that Al entered root apices and accumulated in the epidermal and cortical cells immeadiately below the epidermis. There was a good positive correlation between the level of chlorophyll and ALAD activity. Increasing Al concentrations caused a decrease in total chlorophyll contents, accompanied by proportional changes in ALAD activity, suggesting a cootr-dinated reduction of a photosynthetic machinery. Al exerted specific influence on the morphology of root ann shoot. At higher concentrations of Al the roots induced drastic anatomical changes. The epidermal cells were disorganized or destructed while the cortical cells exhibited distortion of cell shape and/or disintegration. The diameter of root and transectional area of cortical cells decreased considerably with Al treatment. In the shoot Al also enhanced reduction of diameter of shoot and cell size. Gross anatomy of leaves treated with Al did not differ significantly from the controls, except for fewer and smaller chloroplast. Our results indicate that toxic effect of Al appear to be manifested primarily in roots and secondarily on shoots, and changes in root morphology are related to changes in the root growth patterns. Results are further discussed in re181ion to the findings in other plant species, and it is concluded that Al causes morphological, structural and, presumably, functional damage to the roots of the species investigated.
Present study describes a method on the application of efficient tissue culture systems for the micro-propagation of juvenile and mature sawtooth oak(Quercus acutissima). Nodal segments with axillary buds were used as initial explant sources. WPM(Woody Plant Medium) was the best in growth and proliferation of shoot among the media tested. Although the single effect of zeatin revealed on two dorminant shoot elongation with normal growth until the elevation of levels up to 3.0mg/l, BAP($N^6$-benzyl amino purine) usually showed better response than zeatin on shoot multiplication and/or elongation. In addition, the incorporation of BAP and zeatin onto the culture media represents more effectiveness in shoot proliferation and its growth. Optimum concentrations of BAP and zeatin were 0.5 and 0.05~1.0mg/l, respectively. Ninety percent of the proliferated shoots was rooted on half-strength GD (Gresshoff and Doy, 1972) medium containing 0.5mg/l IBA(indole butyric acid) in 4 weeks after culture. More than 70% of the rooted plantlets survived after 5 months of transplanting into artificial soil mix containing equal amount of peatmoss and perlite. Among 27 plus tree clones which were grafted twice onto the juvenile rootstocks, only 4 clones revealed the possibility for shoot multiplication through tissue culture system. The capacity for the micropropagation using mature explant sources was highly depended on clonal differences compared with those of octet age. More than 90% of rooting ratio was obtained from the best responding clone. Among the 7 rooting media tested, GD medium was the best far rooting. The most effective rooting was obtained on half-strength GD medium containing 0.2 to 2.0mg/l IBA. More than 60% of rooted plantlets survived after 5 months of transplanting into the artificial soil mix.

A successful, efficient system for multiple shoot induction and plant regeneration of soybean (Glycine max) was established. Four soybean genotypes were compared for organogenic responses on various media cultured under light conditions. The adventitious shoots (98%, 2.6 shoots/cotyledon) directly from one-day-old cotyledon after germination induced by the hormone treatment and its efficiency was higher than any other conditions. The optimal medium for the induction of multiple shoots from cotyledon in Pungsannamulkong(shoot formation rate, 98%), Lx 16 (83%) and IIpumgeomjeongkong(63%) was MS medium supplemented with 2 mg/L BAP, but for Alchankong(75%), MS medium supplemented with 1mg/L zeatin and 1mg/L IAA, 3% sucrose, 4% Phytagel. Higher root induction (88%) was observed from the shoots placed on rooting medium (hormone-free MS basal). Plantlets were transferred onto the same medium supplemented with 1% activated charcoal for further development. With this treatment, regenerated plantlets were obtained within 7-8 weeks (shoot induction for 4 weeks, rooting and shoot elongation for 3-4 weeks).
This study was conducted to investigate an optimal condition for shoot proliferation and regenerate shoots from in vitro leaflet and embryogenic calli from in vitro roots in rose. The effect of BAP on shoot proliferation was somewhat different depending upon genotypes or gelling agents. Leaflets with petiole cut from donor shoots which had been cultured in MS medium supplemented with 0.1 $mg{\cdot}L^{-1}$ NAA for six weeks was effective for regeneration of adventitious buds(ABs) as well as shoot elongation of Rosa hybrida cv. Sweet Pink. Culturing seven leaflet explants per petri plate($100mm{\times}15mm$) was effective for regeneration of ABs. Embryogenesis was shown in the calli induced from roots of Rosa hybrida cv. Sweet Pink cultured in the SH medium supplemented with 11 $mg{\cdot}L^{-1}$ 2, 4-D for four weeks. Color of calli induced from roots was yellow although their color was a little different as type of basal medium.
