Effects of Variants in Proopiomelanocortin and Neuropeptide Y Genes on Growth, Carcass, and Meat Quality Traits in Rabbits

  • Appetite-related neuropeptides proopiomelanocortin (POMC) and Neuropeptide Y (NPY) are essential for regulating feeding behavior and energy homeostasis. The objective of this study was to evaluate the effects of variants in POMC and NPY genes on growth, carcass and meat quality traits in rabbits. A total of six SNPs were identified for POMC (n = 2) and NPY (n = 4) genes by direct sequencing. Three SNPs were subsequently genotyped by using MassArray system (Sequenom iPLEXassay) in 235 individuals, which belong to three meat rabbit breeds, including 93 Ira rabbits; 81 Champagne rabbits and 61 Tianfu black rabbits. The SNP c.112-12G>T was in intron-exon boundaries (intron 1) of POMC gene, and the association analysis showed that individuals with TT genotype had a greater 84 d body weight (BW84), eviscerated weight and semi-eviscerated weight than those with GT genotype (p<0.05); the TT individuals were also higher than those GG in the ripe meat ratio (RMR) (p<0.05). The g.1778G>C SNP, which was in complete linkage with other three SNPs (g.1491G>A, g.1525G>T and g.1530C>T) in intron 1 of NPY gene, was significantly correlated with eviscerated slaughter percentage and semi-eviscerated slaughter percentage in rabbits, and the individuals with CC genotype had a better performance than CG genotype (p<0.05). These findings would provide primary clues for the biological roles of POMC and NPY underlying the rabbit growth-related traits.

  • ASK 100% RF 입력신호를 이용하는 13.56MHz RFID 태그를 위한 클럭 복원회로를 제안하였다. 제안한 클럭 복원회로는, 레지스터로 조절되는 DLL을 이용하여 입력 RF 신호의 크기가 0인 구간에서도 기준 클럭 신호를 사용하지 클럭을 생성하도록 설계되었다. 제안한 회로는 TSMC 0.18um 1P6M 공정을 사용하여 설계하였으며, 제안된 회로는 DLL의 위상 잠김 시간이 6.4usec 이하이며 공급전압이 3.3V에서 43uW를 소모한다.

Intervention Effects of Nedaplatin and Cisplatin on Proliferation and Apoptosis of Human Tumour Cells in Vitro

  • Objective: To study synergistic effects of nedaplatin and cisplatin on three human carcinoma cell lines (esophageal carcinoma cell line Eca-109, ovarian carcinoma Skov-3 and cervical carcinoma Hela). Methods: Inhibition effects were evaluated by MTT assay and cell apoptosis was detected by flow cytometry. In addition, changes of Ki-67, Bax and Bcl-2 at mRNA and protein levels were quantified by RT-PCR and Western blotting. Results: Growth inhibition in each cell lines was dose-dependent after exposure to nedaplatin or cisplatin alone. The interaction of the two drugs was synergistic at higher concentrations according to the median-effect principle. The inhibition rates with nedaplatin, cisplatin and combined treatment were $41.9{\pm}4.1%$, $47.4{\pm}2.9%$, $52.5{\pm}0.9%$(Eca-109), $39.0{\pm}1.26%$, $45.0{\pm}1.45%$, $56.2{\pm}1.44%$ (Skov-3) and $44.8{\pm}2.11%$, $46.9{\pm}0.99%$, $56.6{\pm}1.83%$ (Hela) respectively, with increase in apoptosis. Compared with the nedaplatin or cisplatin alone treatment group, the combinative treatment group's Ki-67 and bcl-2 mRNA (protein) expression was decreased while that of Bax mRNA (protein) was increased. Conclusion: Compared to the effects of nedaplatin or cisplatin alone at high concentrations, combination of nedaplatin and cisplatin at low concentrations proved to be much more effective for inhibition of proliferation and the induction of apoptosis in the Eca-109, Skov-3 and Hela cell lines.

Identification and Association of SNPs in TBC1D1 Gene with Growth Traits in Two Rabbit Breeds

  • The TBC1D1 plays a key role in body energy homeostasis by regulating the insulin-stimulated glucose uptake in skeletal muscle. The present study aimed to identify the association between genetic polymorphisms of TBC1D1 and body weight (BW) in rabbits. Among the total of 12 SNPs detected in all 20 exons, only one SNP was non-synonymous (c.214G>A. p.G72R) located in exon 1. c.214G>A was subsequently genotyped among 491 individuals from two rabbit breeds by the high-resolution melting method. Allele A was the predominant allele with frequencies of 0.7780 and 0.6678 in European white rabbit (EWR, n = 205) and New Zealand White rabbit (NZW, n = 286), respectively. The moderate polymorphism information content (0.250.05). Our results implied that the c.214G>A of TBC1D1 gene might be one of the candidate loci affecting the trait of 35 d BW in the rabbit.

The Records of Origin and Transport of Sediments From the Past to the Present in the Yellow Sea

  • A total of 116 surface sediment samples were obtained on the Yellow Sea and analyzed for grain size and geochemical elements in order to interpret the present sediment transportation. Thirty-nine cores and 3,070 line-km shallow seismic profiles are analyzed for sedimentary records of Yellow Sea in the past. Results show that the boundary of sediment transport between Korean side and Chinese side is about between $123^{\circ}E$ and $124^{\circ}E$. The similar result is produced from Shi et al. (in this publication). Two cyclonic patterns of surface sediments are recognized in the northeastern and southwestern Yellow Sea, while the strong front zone of the mud patch and sandy sediments are found in the southeastern Yellow Sea (the southwestern part of Korean coasts). The formation of fine-particle sediment packages, called for Northwest Mudbelt Deposit (NWMD), Hucksan Mudbelt Deposit (HSMD) and Jeju Mudbelt Deposit (JJMD), are resulted from eddies (gyres) of water circulations in the Yellow Sea. NWMD has been formed by cyclonic (anticlockwise) eddy. NWMD is composed of thick, homogeneous, relatively semi-consolidated gray clay-dominated deposit. On the other hand, HSMD and JJMD are formed by anticyclonic (clockwise) eddies. They are thick, homogeneous, organic-rich gray, silt-dominated deposit. Both core and surface sediments show that the middle zone across Chinese and Korean side contains bimodal frequency of grain-size distribution, indicating that two different transport mechanisms exist. These mud packages are surrounded by sand deposits from both Korea and China seas, indicating that Yellow Sea, which is the shallow sea and epicontinental shelf, is formed mostly by sand deposits including relict sands. The seismic profiles show such as small erosional/non-depositional channels, sand-ridges and sand-waves, Pleistocene-channelfilled deposits, a series of channels in the N-S major channel system, and thick Holocene sediment package, indicating that more complex sedimentary history exists in the Yellow Sea.

Characterization of Vaginal Microbiota Associated with Pregnancy Outcomes of Artificial Insemination in Dairy Cows

  • The profitability of the dairy and beef industries is largely affected by the actually achieved reproductive efficiency. Although a large proportion of cows worldwide are bred by artificial insemination (AI) services, many potential factors affecting the outcome of pregnancy by AI remain to be addressed. In the present study, we investigated the vaginal microbiota by high-throughput sequencing of 16S rRNA gene and analyzed their association with differential pregnancy outcomes (i.e., pregnant vs. nonpregnant) of multiple AI services in dairy cows. Sequencing of the V3-V4 region totally produced 512,046 high-quality sequences that were computationally clustered into 2,584 operational taxonomic units (OTUs). All OTUs were taxonomically assigned to 10 bacterial phyla. There were statistically significant differences among the three AI service times (T1, T2 and T3) with respect to the Shannon index and number of observed OTUs (p < 0.05). Bray-Curtis distance-based PCoA analysis also revealed that T2 group could be significantly distinguished from T1 and T3. However, no significant difference between the pregnant and nonpregnant cows was found in confidence regarding both alpha diversity and beta diversity. These results could help us better understand the possible influence of vaginal microbial community on pregnancy outcomes of AI service in cows.

Ordinal Optimization Theory Based Planning for Clustered Wind Farms Considering the Capacity Credit

  • Wind power planning aims to locate and size wind farms optimally. Traditionally, wind power planners tend to choose the wind farms with the richest wind resources to maximize the energy benefit. However, the capacity benefit of wind power should also be considered in large-scale clustered wind farm planning because the correlation among the wind farms exerts an obvious influence on the capacity benefit brought about by the combined wind power. This paper proposes a planning model considering both the energy and the capacity benefit of the wind farms. The capacity benefit is evaluated by the wind power capacity credit. The Ordinal Optimization (OO) Theory, capable of handling problems with non-analytical forms, is applied to address the model. To verify the feasibility and advantages of the model, the proposed model is compared with a widely used genetic algorithm (GA) via a modified IEEE RTS-79 system and the real world case of Ningxia, China. The results show that the diversity of the wind farm enhances the capacity credit of wind power.

Virtual Non-Contrast Computer Tomography (CT) with Spectral CT as an Alternative to Conventional Unenhanced CT in the Assessment of Gastric Cancer

  • Objective: The purpose of this study was to evaluate computed tomography (CT) virtual non-contrast (VNC) spectral imaging for gastric carcinoma. Materials and Methods: Fifty-two patients with histologically proven gastric carcinomas underwent gemstone spectral imaging (GSI) including non-contrast and contrast-enhanced hepatic arterial, portal venous, and equilibrium phase acquisitions prior to surgery. VNC arterial phase (VNCa), VNC venous phase (VNCv), and VNC equilibrium phase (VNCe) images were obtained by subtracting iodine from iodine/water images. Images were analyzed with respect to image quality, gastric carcinoma-intragastric water contrast-to-noise ratio (CNR), gastric carcinoma-perigastric fat CNR, serosal invasion, and enlarged lymph nodes around the lesions. Results: Carcinoma-water CNR values were significantly higher in VNCa, VNCv, and VNCe images than in normal CT images (2.72, 2.60, 2.61, respectively, vs 2.35, $p{\leq}0.008$). Carcinoma-perigastric fat CNR values were significantly lower in VNCa, VNCv, and VNCe images than in normal CT images (7.63, 7.49, 7.32, respectively, vs 8.48, p< 0.001). There were no significant differences of carcinoma-water CNR and carcinoma-perigastric fat CNR among VNCa, VNCv, and VNCe images. There was no difference in the determination of invasion or enlarged lymph nodes between normal CT and VNCa images. Conclusions: VNC arterial phase images may be a surrogate for conventional non-contrast CT images in gastric carcinoma evaluation.

Epidemiological Characteristics and Prediction of Esophageal Cancer Mortality in China from 1991 to 2012

  • Background: To analyze the mortality distribution of esophageal cancer in China from 1991 to 2012, to forecast the mortality in the future five years, and to provide evidence for prevention and treatment of esophageal cancer. Materials and Methods: Mortality data for esophageal cancer in China from 1991 to 2012 were used to describe its epidemiological characteristics, such as the change of the standardized mortality rate, urban-rural differences, sex and age differences. Trend-surface analysis was used to study the geographical distribution of the mortality. Curve estimation, time series, gray modeling, and joinpoint regression were used to predict the mortality for the next five years in the future. Results: In China, the incidence rate of esophageal cancer from 2007 and the mortality rate of esophageal cancer from 2008 increased yearly, with males at $8.72/10^5$ being higher than females, and the countryside at $15.5/10^5$ being higher than in the city. The mortality rate increased from age 45. Geographical analysis showed the mortality rate increased from southern to eastern China, and from northeast to central China. Conclusions: The incidence rate and the standardized mortality rate of esophageal cancer are rising. The regional disease control for esophageal cancer should be focused on eastern, central and northern regions China, and the key targets for prevention and treatment are rural men more than 45 years old. The mortality of esophageal cancer will rise in the next five years.

Acupotomy for Scapulohumeral Periarthritis

  • Background: Scapulohumeral periarthritis causes pain and stiffness, and limits movement but it is a treatable condition. This was a clinical study of acupotomy treatment for scapulohumeral periarthritis. Methods: There were 80 patients randomly assigned to the traditional Chinese Medicine group (acupotomy) and the Western medicine group (naproxen), with 40 cases in each group. All patients had adjunct physiotherapy exercises for 14 days. Patients received acupotomy treatment 3 times for 14 days (Day 0, 7 and 14) or naproxen (0.22 g capsule; a non-steroidal anti-inflammatory drug) 3 times a day, for 14 days. The visual analogue scale (VAS) scores, range of motion (ROM) values, and the Melle scale, together with the therapeutic standard of diseases and syndromes in traditional Chinese Medicine were used for diagnosis and evaluation. Results: There were significant differences in the VAS scores, ROM, Melle scores, cure rate and total effective rate in the group which took naproxen and the acupotomy group, before and after treatment (p < 0.01). There were significant differences in the changes in VAS, ROM and Melle scores between the 2 groups (p < 0.01), and the acupotomy group was better than the naproxen group. Conclusion: Traditional Chinese Medicine and Western medicine can improve functional activity and reduce the level of pain experienced by patients suffering from scapulohumeral periarthritis. However, improvement of functional activity of the shoulder joint following acupotomy treatment was more obvious than the use of a non-steroidal anti-inflammatory drug, and the cure rate, and total effective rate of acupotomy was better.
