Esophageal Cancer and Sex Hormones (식도암과 성호르몬)

  • Globally, esophageal cancer is the seventh most common cancer, and the male-to-female ratio in esophageal adenocarcinoma (EAC) is significantly imbalanced at 4:1 to 8:1. Obesity, reflux, and smoking are known risk factors for this sex difference; however, fully explaining this remains challenging. Studies have investigated the link between exogenous sex hormones and esophageal cancer occurrence. A meta-analysis revealed a lower risk of EAC in female who had undergone hormone replacement therapy. Androgen-deprivation therapy in patients with prostate cancer was associated with a decreased risk of EAC. Tissue-based studies have reported varied results regarding the relationship between estrogen receptor expression and survival in female patients with esophageal squamous cell carcinoma (ESCC). Circulating hormone studies have suggested that higher testosterone and luteinizing hormone levels decreased EAC risk in men, and free testosterone was inversely correlated in female with ESCC. However, a high androgen-estrogen ratio in male patients with EAC was linked to increased odds of EAC. Sex hormones influence carcinogenesis, affecting cell proliferation, differentiation, metabolism, inflammation, and cell death. The studies were limited by the small sample size and varying hormone measurement methods; thus, future studies with definitive conclusions on the association between esophageal cancer and sex hormones are warranted.

Steroid Metabolism in the Blacktip Grouper Epinephelus fasciatus during Oocyte Vitellogenesis (홍바리(Epinephelus fasciatus) 난황형성기 난모세포에서의 성 스테로이드 호르몬 대사)

  • We studied oocyte steroidogenesis in blacktip grouper Epinephelus fasciatus ovarian follicles during vitellogenesis. Vitellogenic oocytes with average diameters of 0.45, 0.48 and 0.50 mm were incubated in vitro in the presence of $[^3H]17{\alpha}$-hydroxyprogesterone as a precursor. The steroid metabolites were analyzed using thin layer chromatography (TLC), high performance liquid chromatography (HPLC), and gas chromatography-mass spectrometry (GC/MS). The major metabolites in the vitellogenic oocytes were androstenedione ($A_4$), testosterone (T), estradiol-$17{\beta}$ ($E_2$), and estrone ($E_1$). The metabolites of androgen ($A_4$ and T) were higher in the 0.50-mm oocytes than in the 0.45- and 0.48-mm oocytes, while the estrogen metabolites (E2 and E1) were lower in the 0.50-mm oocytes. These results suggest that 0.50-mm oocytes are fully vitellogenic following initiation of the maturation process.

Characterization of ovarian culture in vitro and sex steroids in vivo by recombinant eel gonadotropin treatments in the eel Anguilla japonica

  • In the present study, we investigated the effects of recombinant eel gonadotropins (rec-GTHs) on maturation induction in immature ovarian culture in vitro and sex steroid hormones in vivo in the Japanese eel Anguilla japonica. To study the in vitro effects of rec-GTHs on estradiol-17β (E2) production in immature ovarian tissues, ovarian tissues were incubated with different doses of rec-follicle-stimulating hormone (rec-FSH) or rec-luteinizing hormone (rec-LH). The results revealed that the E2 levels in the rec-FSH (0.1, 0.5, or 1 ㎍/mL)- and rec-LH (0.1 or 0.5 ㎍/mL)-treated groups were significantly higher than those in the female eels from the control group. Furthermore, to investigate the in vivo effects of rec-GTHs on the gonadosomatic index (GSI) and plasma sex steroid hormone levels, the eels were injected intraperitoneally with eel's ringer (control), salmon pituitary extract (SPE; for female eels), human chorionic gonadotropin (hCG; for male eels), rec-FSH, rec-LH, and rec-FSH + rec-LH once a week. The results revealed that except for the SPE and the hCG groups, none of the groups exhibited a significant difference in GSI values. However, in vivo plasma E2 levels increased at the end of 4 weeks after rec-FSH treatment in female eels. Based on these results, it is suggested that rec-GTHs may have a positive effect on sexual maturation in female eels; however, further studies on complementary rec-protein production systems and additional glycosylation of rec-hormones are needed to elucidate hormone bioactivity in vivo and in vitro.

  • Hormone이 암발생과 밀접한 관계가 있음은 이미 1935년대에 밝혀진 바 있다. 즉 lacassagne은 estrogen이 토끼 자궁벽의 fibromyomatous transformation에 영향을 준다고 하였다. Nelson는 guinea-pig를 시험동물로 해서 estrogen을 장기투여한 결과 uterine fibroids가 수발됨을 확인한 이래, 유발됨을 확인한 이래, 이러한 hormone이 암발생의 장기요인이 됨으리 밝히게 되었다. 이러한 업적은 Lipschutz에 의해 더욱 수식되었다. 이러한 업적의 결과가 여러편의 종설로 발표되었다. 즉 Lacassagne, Zuckerman, Burrows등, Gardner는 각기 종설을 발표하여 이를 확증하기에 이르렀다. 한편 각개 hormone, 그중에서도 sex-hormone의 opposite action은 Steinach등 및 Sand에 의해 밝혀졌고 거세백서에 estrogen을 투여했을때 유기되는 vaginal mucosa의 epidermization이 testosterone의 투여에 의해 급속히 소진됨을 Currier 일파가 증명하였고 estrogen에 의해 유발된 웅백서 부속성선의 조직변성이 androgen의 동시투여의 소실됨이 homone에 의해 유발된 neoplastic에 대해 유효하리하는 것은 백명한 일이고 이러한 사실은 현재 실시로 허다히 이용되고 있는 것이다.

Hormonal regulation of uterine chemokines and immune cells

  • The ultimate function of the endometrium is to allow the implantation of a blastocyst and to support pregnancy. Cycles of tissue remodeling ensure that the endometrium is in a receptive state during the putative 'implantation window', the few days of each menstrual cycle when an appropriately developed blastocyst may be available to implant in the uterus. A successful pregnancy requires strict temporal regulation of maternal immune function to accommodate a semi-allogeneic embryo. To preparing immunological tolerance at the onset of implantation, tight temporal regulations are required between the immune and endocrine networks. This review will discuss about the action of steroid hormones on the human endometrium and particularly their role in regulating the inflammatory processes associated with endometrial receptivity.

Tributyltin-oxide (TBTO) induced Changes in Plasma Sex Steroid Hormones and Cortisol Level of Korean Rockfish, Sebastes schlegeli

  • The influence of TBTO (Tributyltin-Oxide) at a series of concentrations (0.52, 1.41 and 3.05 $\mu$g/L) for a period of 3 weeks on estradiol-17$\beta$, testosterone and cortisol levels in male Korean rockfish, Sebastes schlegeli was investigated. Compared to the stable concentration of estradiol-17$\beta$, plasma testosterone level was decreased significantly at the 3rd week after TBTO exposure in plasma of fish (>1.41 $\mu$g/L). The most profound physiological variation in TBTO exposed fish was a dramatic increase in plasma cortisol level at 3rd week. Conclusively, TBTO exposure caused increase level of cortisol as well as alteration of testosterone in the Korean rockfish. These results suggest that TBTO at environmentally relevant (nanomolar) concentrations disrupt endocrine secretions.

Combination Radiation and Steroid Therapy for Midline Granuloma (Midline Granuloma의 방사선 치료 및 Steroid병용요법)

  • Midline Granulcma(Malignant midline reticulosis) of upper respiratory tract was known as one of the fatal diseases that involve the nasal cavity, paranasal sinuses, pharynx and larynx, with progressive localized necrotizing inflammation and granuloma formation. The disease was first described in 1897 by McBride and after Stewart many clinical and histological studies have been done in 1933. But its etiology is still unclear and therapeutic methods are still under development. The authors carried out a clinical study of 15 cases which were diagnosed and treated as midline granuloma, including 7 cases which received combination radiation and steroid therapy from January 1964 to December 1980. The results are as follows: 1) Age and Sex distribution: 6 cases fell into the ages from 30 to 39 years: 13 cases (87%) were male and 2 cases (13%) were female. 2) Primary lesion sites were nasal cavity and paranasal sinuses area (8 cases: 53%), palatine tonsil (3 cases: 20%) and pharynx (2 cases: 13%) in order. 3) Common symptoms were nasal stuffiness and sore throat (6 cases: 40%), headache (5 cases: 33%), nasal discharge and facial edema (3 cases: 20%) in order. 4) Microorganisms were identified in 4 cases: staphylococcus in 2 cases, pseudomonas in 1 case and streptococcus in 1 case. 5) All 7 cases who received combination radiation and steroid therapy revealed complete regression and no recurrence. 6) Among the 5 cases, who received steroid and antibiotic therapy, 2 cases died and other 3 cases improved temporarily and then developed aggrevation of symptoms.

Sex Reverse in Teleost Culture II. Sex Reversal of Guppy and Tilapia by $17\beta$-Estradiol and $17\alpha$-Methyltestoserone (경골어류의 성전환에 관한 연구 II. $17\beta$-Estradiol 및 $17\alpha$-Methyltestosterone을 이용한 Guppy 및 Tilapia의 성전환에 관한 연구)

  • Guppy fry were treated for the first 40 days of life with 0, 20, 40, 60 & 100$\mu\textrm{g}$ of estradiol per gram of food in order to change the sex of normal males to functional females(genetic male). The present investigation deals with the effects of steroid hormones, such as $\beta$-estradiol and testosterone, on the sex differentiation in guppy and tilapia. The results obtained were summarized as follows. 1. In B (20$\mu\textrm{g}$/g diet) group 17$\beta$-estradiol-treated, 67.8% of male offsprings were produced. 2. In D (60$\mu\textrm{g}$/g diet) group 17$\beta$-treated, 67% of female offsprings were produced. 3. B, D groups of genetic male brooders had significantly different effects (P<0.01) upon sex ratios of their progeny. 4. This strongly indicates that sex direction has been achieved and that the male is the heterogametic sex. 5. The group that produced the highest percentage of male offspring(male percentage of observed number to expected number was 91%) contained only full-sibling male brooders to the sex-reversed female brooders. 6. After 7 months following treatment, the sex-reversed males had ovarian portion in the anterior region and a testicular portion in the posterior region of the same intersexual gonad, respectively. 7. At 7 months after treatment, the ovareis revealed a complete arrest of the ovarian formation, and appearances of spermatogenetic cell cysts among surviving auxocytes. 8. In most of sex-reversed fish, anterior portion of test is was devoid of sperm ducts including the seminal vesicle and vas deferens. 9. The male transferrin showed two strong bands, while the female transferrin showed a single weak band. 10. One of the two bands of male transferrin showed the same mobility with band of female transferrin.

Adverse Reproductive Effects on Plasma Vitellogenin and Sex Steroid Levels, and Gonadosomatic Index in Juvenile Common Carps (Cyprinus carpio) Exposed to 17$\beta$-Estradiol and D-2-Ethylhexyl Phthalate

  • Environmental estrogens are natural or synthetic substances present in the aquatic environment, especially in effluent from sewage treatment. However, the adverse effects of these estrogenic substances on fish reproduction are unknown. Di-2-ethylhexyl phthalate (DEHP) is the most common phthalate, which Ps used as a plasticizer in polyvinylchloride (PVC), and it is widespread in the environment and has been found in aquatic organisms and sediments. Therefore, juvenile common carps (Cyprinus carpio) were exposed to nominal concentrations of 17$\beta$-estradiol (E2) (0.5, 5, 50 $\mu\textrm{g}$/L) and DEHP (10, 100, 500 $\mu\textrm{g}$/L) for 21 days, to determine the adverse reproductive effects of these compounds on plasma vitellogenin (VTG) induction, sex steroid level, and gonad weight. Electrophoresis (SDS-PAGE) revealed that much of VTG was induced in fish exposed to 5 and 50 E$_2$ $\mu\textrm{g}$/L, but none of DEHP exposure showed induction. Enzyme-linked immunosorbent assay (ELISA) revealed that VTG was significantly induced in fish exposed to 5 and 50 E$_2$ $\mu\textrm{g}$/L, and combination of 50 E$_2$ $\mu\textrm{g}$/L with 10 and 500 DEHP $\mu\textrm{g}$/L, but none of DEHP exposure showed induction. Analysis of sex steroid levels in some fish revealed that testosterone (T) was detected in both male and female fish of the control and DEHP exposures, but none of fish exposed to 22 concentrations had detectable testosterone level. On the other hand, E$_2$ exposure induced 17$\beta$-estradiol in plasma of male fish, but there was no induction of 17$\beta$-estradiol in plasma of male fish exposed to DEHP. Comparison of gonadosomatic index (GSI) revealed that maximal E$_2$ exposure inhibited ovarian growth, but maximal DEHP exposure stimulated testicular growth. The results indicated that those comparisons can be a useful bio-indicator for determining adverse reproductive effect of endocrine disrupting chemicals (EDCs).

Changes in Plasma Levels of Thyroid and Sex Steroid Hormones in Rockfish (Sebastes schlegeli) during Maturation and Parturition Periods (조피볼락(Sebastes schlegeli)의 성숙$\cdot$출산기에 있어서 갑상선 및 성 스테로이드 호르몬의 혈중 변화)

  • Changes in plasma levels of thyroid and sex steroid Hormones were examined during maturation and parturition periods in rockfish (Sebastes schlegeli) cultured in net pens. Plasma L-thyroxine levels were $35.2{\pm}5.7\;ng/m{\ell}\;(n=\;5;\;mean\;{\pm}\;sem)$ at vitellogenesis stage and then significantly decreased to $20.5\;{\pm}\;4.2\;ng/m{\ell}$ at parturition stage (P<0.05), and rapidly returned to high level, $44.9{\pm}\;7.2\;ng/m{\ell}$ at resting stage. Plasma 3,5,3'-triiodo-L-thyronine levels were 12.9 $\pm$ 1.6 ng/ml at vitellogenesis stage, but significantly decreased to $3.7{\pm}0.7\;ng/m{\ell}$ at ovulation stage (P<0.05) and increased to $52.9{\pm}7.0\;ng/m{\ell}$ at jesting stage. Plasma estradiol-17 $\beta$ level showed the highest value ($4.3{\pm}0.9\;ng/m{\ell}$) at vitellogenesis stage, but the level significantly decreased to $0.3{\pm}0.1\;ng/m{\ell}$, during parturition stage (P<0.05). In vitellogenesis and ovulation stages, plasma testosterone levels were $1.8{\pm}0.3\;ng/m{\ell}$ and $2.1{\pm}0.7\;ng/m{\ell}$, respectively, thereafter the level significantly decreased to $0.1{\pm}0.1\;ng/m{\ell}$ at parturition stage (P<0.05). These findings suggest that thyroid hormones may have relation to maturation and parturition of mother rockfish.
