COPD 동물 모델에서 소청룡탕 흡입제형의 효과 (Effects of Inhalable Microparticles of Socheongryong-tang on Chronic Obstructive Pulmonary Disease in a Mouse Model)

  • Objectives: This study aimed to evaluate the effects of microparticles of Socheongryong-tang (SCRT) on chronic obstructive pulmonary disease (COPD) in a mouse model. Methods: The inhalable microparticles containing SCRT were produced by spray-drying with leucine as an excipient, and evaluated with respect to the aerodynamic properties of the powder by Andersen cascade impactor (ACI). Its equivalence to SCRT extract was evaluated using lipopolysaccharide (LPS) and a cigarette-smoking (CS)-induced murine COPD model. Results: SCRT microparticles provided desirable aerodynamic properties (fine particle fraction of $49.6{\pm}5.5%$ and mass median aerodynamic diameter of $4.8{\pm}0.3{\mu}m$). SCRT microparticles did not show mortality or clinical signs over 14 days. Also there were no significant differences in body weight, organ weights or serum chemical parameters between SCRT microparticle-treated and non-treated groups. After 14 days the platelet count significantly increased compared with the non-treated group, but the values were within the normal range. Inhalation of SCRT microparticles decreased the rate of neutrophils in blood, granulocytes in peripheral blood mononuclear cells (PBMC) and bronchoalveolar lavage fluid (BALF) and level of TNF-${\alpha}$ and IL-6 in BALF on COPD mouse model induced by LPS plus CS. This effect was verified by histological findings including immunofluorescence staining of elastin, collagen, and caspase 3 protein in lung tissue. Conclusions: These data demonstrate that SCRT microparticles are equivalent to SCRT extract in pharmaceutical properties for COPD. This study suggests that SCRT microparticles would be a potential agent of inhalation therapy for the treatment of COPD.

Effects of Additives on Laying Performance, Metabolic Profile, and Egg Quality of Hens Fed a High Level of Sorghum (Sorghum vulgare) during the Peak Laying Period

  • This experiment was conducted to determine the effects of supplemental methionine, lysine, choline, and sulfur on laying performance, metabolic parameters, and egg quality of hens fed diets containing sorghum (Sorghum vulgare) during the peak laying period. Lohman layers (n = 144), 30-wk of age as 6 replicate cages of 4 hens, were allocated randomly to receive basal diets containing either 22% corn (B) or 22% sorghum (BS) and diets BS plus 0.57% methionine, 0.66% lysine, 0.47% choline, or 0.05% sulfur for 98 d. Feed intake (FI) and egg production (EP) were recorded daily, egg weight (EW) was measured bi-weekly, and body weight (BW) was measured monthly. A sample of 12 eggs from each experimental group was collected every month to evaluate egg quality. At the end of the experiment, blood samples were collected for metabolite concentrations. Data were analyzed using one-way ANOVA as repeated measures and significant differences between the experimental groups were assessed using Duncan's Multiple Range test. Partial replacement of corn with sorghum in the basal diet did not affect BW, EP, and FCR but increased FI by 5.7% and EW by 2.4%. The effects of additives on laying performance were variable. Except for serum total protein (STP) concentration, other metabolic parameters were not affected by partial replacement of corn with sorghum in the basal diet. Hens fed diet BS had lower SPT concentration than hens fed diet B. Except for methionine supplementation, other supplements ameliorated depression in STP concentration. The additives did not affect other metabolic parameters. Egg quality responses to the experimental diets were also variable. Partial replacement of corn with sorghum in the basal diet did not affect eggshell characteristics (both thickness and stiffness), whereas it had variable effects on inner egg quality parameters (increased yolk index, depressed yolk color, and unaltered albumen index and Haugh unit). In conclusion, laying hen diets could include low-tannin sorghum (0.26%) up to 22% without necessitating extra supplements to overcome compromised performance.

Characterizing Milk Production Related Genes in Holstein Using RNA-seq

  • Although the chemical, physical, and nutritional properties of bovine milk have been extensively studied, only a few studies have attempted to characterize milk-synthesizing genes using RNA-seq data. RNA-seq data was collected from 21 Holstein samples, along with group information about milk production ability; milk yield; and protein, fat, and solid contents. Meta-analysis was employed in order to generally characterize genes related to milk production. In addition, we attempted to investigate the relationship between milk related traits, parity, and lactation period. We observed that milk fat is highly correlated with lactation period; this result indicates that this effect should be considered in the model in order to accurately detect milk production related genes. By employing our developed model, 271 genes were significantly (false discovery rate [FDR] adjusted p-value<0.1) detected as milk production related differentially expressed genes. Of these genes, five (albumin, nitric oxide synthase 3, RNA-binding region (RNP1, RRM) containing 3, secreted and transmembrane 1, and serine palmitoyltransferase, small subunit B) were technically validated using quantitative real-time polymerase chain reaction (qRT-PCR) in order to check the accuracy of RNA-seq analysis. Finally, 83 gene ontology biological processes including several blood vessel and mammary gland development related terms, were significantly detected using DAVID gene-set enrichment analysis. From these results, we observed that detected milk production related genes are highly enriched in the circulation system process and mammary gland related biological functions. In addition, we observed that detected genes including caveolin 1, mammary serum amyloid A3.2, lingual antimicrobial peptide, cathelicidin 4 (CATHL4), cathelicidin 6 (CATHL6) have been reported in other species as milk production related gene. For this reason, we concluded that our detected 271 genes would be strong candidates for determining milk production.

Effects of Forage:Concentrate Ratio on Growth Performance, Ruminal Fermentation and Blood Metabolites in Housing-feeding Yaks

  • The objective of this study was to determine the effect of forage: concentrate ratio (F:C) on growth performance, ruminal fermentation and blood metabolites of housing-feeding yaks. Thirty-two Maiwa male yaks (initial body weight = $207.99{\pm}3.31kg$) were randomly assigned to four dietary treatments (8 yaks per treatment). Experimental diets were: A, B, C, D which contained 70:30, 60:40, 50:50 and 40:60 F:C ratios, respectively. Dry matter intake and average daily gain in yaks fed the C and D diets were greater (p<0.05) than yaks fed the A and B diets. No differences were found in ruminal $NH_3-N$, total volatile fatty acids, acetate, butyrate, valerate, and isovalerate concentrations. The propionate concentration was increased (p<0.05) in the C and D groups compared with the A and B diets. In contrast, the acetate to propionate ratio was decreased and was lowest (p<0.05) in the C group relative to the A and B diets, but was similar with the D group. For blood metabolites, no differences were found in serum concentrations of urea-N, albumin, triglyceride, cholesterol, low density lipoprotein, alanine aminotransferase, and aspartate aminotransferase (p>0.05) among treatments. Treatment C had a higher concentration of total protein and high density lipoprotein (p<0.05) than A and B groups. In addition, there was a trend that the globulin concentration of A group was lower than other treatments (p = 0.079). Results from this study suggest that increasing the level of concentrate from 30% to 50% exerted a positive effect on growth performance, rumen fermentation and blood metabolites in yaks.

상 접촉법을 이용한 BSA의 AOT 역미셀으로 가용화: pH와 염의 영향 (Solubilization of BSA into AOT Reverse Micelles Using the Phase-Transfer Method: Effects of pH and Salts)

  • 상 접촉법을 이용하여 BSA를 음이온 계면활성제인 AOT와 isooctane으로 구성된 역미셀상으로 가용화하였다. 본 연구에서는 pH와 염의 종류 및 농도가 가용화 효율에 미치는 영향에 주목하였다. 0.1 M의 KCl, NaCl, $MgCl_2$, 그리고 $CaCl_2$와 같이 1:1 염 혹은 1:2 염을 수용액에 첨가하면 첨가한 염의 종류에 따라 pH=5~7사이에서 최대의 가용화 효율을 보였다. $CaCl_2$와 NaCl의 경우, 1 M 까지는 첨가한 염의 농도가 증가함에 따라 효율도 증가하였으나 $MgCl_2$의 경우에는 특이한 경향을 보였다. 염의 농도가 낮거나 pH가 낮은 경우에는 단백질이 광범위하게 수용액상과 유기상의 계면에서 침전되는 것은 주목할 부분이다. 역미셀의 water-pool 크기는 물에 대한 계면활성제의 몰비인 $W_0$를 측정하여 추산하였다. $MgCl_2$의 경우에는 pH변화에 관계없이 $W_0$=20의 거의 일정한 값을 보인 반면 $CaCl_2$와 KCl의 경우 염 농도가 증가함에 따라 $W_0$는 감소하였다.

핑거루트(Boesenbergia pandurata) 추출물의 고지방 식이를 섭취한 마우스의 내장 지방 축적에 대한 효능 (Fingerroot (Boesenbergia pandurata) Extract Inhibits the Accumulation of Visceral Fat in C57BL/6J Mice)

  • 본 연구는 마우스에 고지방식이로 비만을 유도하면서 핑거루트 추출물(BPE)의 비만 억제 효과를 평가하는 것이었다. C57BL/6J 마우스를 두개의 실험군으로 나누어 8주 동안 각각 고지방식이와 고지방식이에 0.5%의 BPE를 보충한 식이를 급여하였다. BPE는 식이섭취량에는 영향을 주지 않으면서도 고지방식이군과 비교하여 유의적으로 체중증가를 줄이고, 내장지방의 축적을 억제하였다. 또한 BPE를 급여한 군은 고지방식이군과 비교하여 혈중 지질, 간지질의 축적, 혈중 렙틴 농도를 감소시켰다. RT-PCR 분석 결과, 부고환지방내에서의 $C/EBP{\alpha}$$PPAR{\gamma}2$의 발현수준이 고지방식이 대비 각각 1.30배와 1.16배 증가하였다. 결론적으로 BPE는 고지방식이로 유도된 비만 마우스 모델에서 내장지방의 축적을 감소시키고, 이상지질혈증 개선을 통해 항비만 효과를 나타내는 것으로 판단된다.

Modulation of Inflammatory Cytokines and Islet Morphology as Therapeutic Mechanisms of Basella alba in Streptozotocin-Induced Diabetic Rats

  • The mechanism of the previously reported antidiabetic effect of Basella alba is unknown. This study investigated the role of B. alba aqueous leaf extract in the modulation of inflammatory cytokines and islet morphology in streptozotocin-induced diabetic rats. Forty male Wistar rats, between 8 and 10 weeks old, were randomly divided into four groups (n = 10) and administered the following treatments: Healthy control (H-c) and Diabetic control (D-c) animals received normal saline 0.5 mL/100 g body weight daily, while Healthy Treatment (H-Ba) and Diabetic Treatment (D-Ba) rats received the plant extract 200 mg/kg body weight daily. All treatments were administered by oral gavage. Diabetes was induced in D-c and D-Ba rats by a single intraperitoneal injection of streptozotocin (55 mg/kg body). The body weight and fasting blood sugar (FBS) levels were recorded every week for 4 weeks, after which the rats were euthanized and samples collected for further analysis. After the experiment, FBS level was significantly reduced (p < 0.0001) in rats in the D-Ba group, but increased (p < 0.001) in rats in the D-c group. The absolute (H-c and H-Ba vs D-c, p < 0.05) and relative (D-Ba vs H-c, p < 0.05; D-Ba vs H-Ba, p < 0.005) weights of the pancreases were significantly higher after the experiment. The rats in the D-c group had significantly higher levels of serum interleukin-$1{\beta}$ (p < 0.001 vs H-c; p < 0.05 vs H-Ba and D-Ba) and monocyte chemotactic protein-1 (p < 0.0001), but lower levels of interleukin-10 (p < 0.05) in comparison with the other groups. Histopathological examination revealed severe interstitial congestion, reduced islet area (p < 0.0001), and increased islet cell density in the D-c group compared with those in the D-Ba group. From these findings, it was concluded that the aqueous extract of B. alba stimulates the recovery of beta-islet morphology in streptozotocininduced diabetic rats by modulating the peripheral production of inflammatory cytokines.

Monascus Purpureus의 수준별 급여가 육계의 성장과 계란의 품질에 미치는 영향

  • Monascus Purpures(MP)의 수준별 급여가 육계의 성장과 계란의 품질 및 난황에 미치는 영향을 고찰하고자 사양실험을 실행하였다. 실험 1에서 1일령 육계 수컷 320수를 이용하여 사료내 MP는 0, 0.5, 1.0, 2.0, 4.0% 수준으로 처리구당 4반복 반복당 16수씩 전체 320수에 5주간 급여하였다. 증체량, 사료섭취량, 사료요구율은 주간별로 측정하였으며, 혈청의 콜레스테롤, 단백질함량은 실험 종료시에 측정하였다. 실험 2에서는 32주령 산란계 30수를 처리구당 6수씩 개체별로 수용하여 사료내 MP가 0, 0.5, 1.0, 2.0, 3.0%수준으로 4주간 급여하였다. 조사항목은 난각강도, 난각두께, 호유닛, 계란의 콜레스테롤을 주간별로 측정하였다. 실험 1에서 기초사료는 사육 전기 3주와 후기 2주간에 각각 CP 21.5, 19.0%, ME 3,100, 3,200 kca1/kg수준으로 급여하였다. 실험 2에서는 CP 16%, ME 2,800 kca1/kg 수준으로 급여하였다. 실험 1에서 증체량은 2.0%MP 처리구에서 대조구에 비하여 현저하게 개선되었으며, 사료요구율은 처리구간에 통계적인 차이는 없었지만 1.0, 2.0% MP처리구에서 대조구보다 개선되는 경향을 보였다. 혈청의 콜레스테롤은 MP 2.0% 급여구에서 낮은 경향을 보였지 만 처리구간에 통계적인 차이는 없었다. 실험 2에서 난백의 높이와 호유닛은 0.1과 0.2%MP 처리구에서 다른 처리구보다 높게 나타났지만 통계적인 차이는 없었다.

사료 내 목초액의 첨가가 토종닭의 생산성, 혈액성상, 암모니아 가스 발생량 및 가슴육의 지방산 조성에 미치는 영향 (Effect of Pyroligneous Acid Supplementation on Growth Performance, Blood Parameter, Ammonia Gas Emission and Fatty Acid Composition of Breast Meat in Korean Native Chicken)

  • 본 시험은 목초액이 토종닭 한협 3호의 생산성, 유해 가스 발생량, 혈액 성상 그리고 가슴육의 지방산 함량에 미치는 영향을 평가하기 위하여 토종닭 사료에 수준별로 첨가하여 70일 동안 급여하였다. 목초액은 0, 0.1, 0.2% 수준으로 첨가 급여하였고, 처리구당 5반복, 반복당 16수씩 총 240수를 시험에 이용하였다. 시험 결과, 목초액은 생산성에 있어서 큰 영향을 미치지 못하였으나, 암모니아 가스 발생량은 대조구에 비하여 유의하게 감소시켰다(p<0.05). 혈중 total cholesterol과 HDL-cholesterol 농도에 있어서는 목초액 처리구가 대조구에 비하여 유의하게 증가한 반면 triglyceride의 농도는 감소하였다(p<0.05). 가슴육의 포화지방산인 myristic acid와 stearic acid는 목초액을 급여함에 따라 대조구에 비하여 유의적으로 감소하였으나, 불포화지방산인 oleic acid와 linolenic acid는 유의적으로 증가하였다(P<0.05). 따라서 본 연구 결과, 목초액은 유해 가스 저감 효과가 있으며, 토종닭의 혈중 지질 수준을 개선시키고, 가슴육의 포화지방산 함량을 감소시키고 단쇄 불포화지방산의 함량은 증가시키는 것으로 판단된다.

키토산 첨가가 고지방 식이 랫드의 혈청 지질 농도의 변화에 미치는 영향 (Effect of Chitosan on Major Lipid-Related Parameters in Sera of Rats Fed High Fat Diet)

  • This study was performed to investigate the growth rate, hematological and serological changes of the rats when they were fed with the high(at diets supplemented with or without chitosan for five weeks. Twenty-four Sprague-Dawley male rats($235.7{\pm}10.7g$ of body weight) were randomly divided into three groups control group(C) and two treatment groups. Rats in the control group were fed with the high-fat diet containing 10% lard, 1% cholesterol and 0.5% sodium cholate(w/w) which was modified from the formula of the American Institute of Nutrition-76(AIN-76) diet. Rats in treatment groups were red with above diet supplemented with 2.5% of chitosan(CS-2.5) or 5.0% chitosan(CS-5) on the weight to weight basis, respectively. The supplementation of chitosan did not induce any significant difference on the final body weight, gain of body weight and amount of feed intake of rats in between control and treatment groups but the feed efficiency of rats in CS-5 was lower than that of rats in C(p<0.05). The hemoglobin concentrations and hematocrit values showed no significant differences among groups. In addition the values of glucose concentration, total protein, albumin, globulin and albumin/globulin(A/G) ratio showed no significant differences among groups. The values of total cholesterol and low density lipoprotein-cholesterol(LDL-C) in sera of rats in CS-5 were lower than those in both C and CS-2.5(p<0.01). The values of high density lipoprotein-cholesterol (HDL-C) in sera of rats in CS-5 were higher than in both in C and CS-2.5(p<0.05). The values of atherogenic index(AI) of rats in CS-5 were the lowest among groups(p<0.01). AI of CS-2.5 were lower than that or C(p<0.05). The values of triglyceride in sera of rats showed no significant differences among groups. The values of AST in sera of rats in CS-2.5 were lower than those in both C and CS-5(p<0.05). However ALT values showed no significant differences among groups. Therefore the supplementation of chitosan to high fat diet reduced effectively the serum lipid levels such as total cholesterol, LDL-C and triglycerides which were regarded as to cause the cardiovascular diseases moreover it elevated effectively HDL-C value which was regarded protect cardiovascular diseases.
