Animal cell cultures generally require a nutrient-rich medium supplemented with animal serum. Adult bovine serum contains a variety of nutrients including inorganic minerals, vitamins, salts, proteins and lipids as well as growth factors that promote animal cell growth. To evaluate the potential use of gender-specific bovine serum (GSBS) for cell culture, the biochemical properties of male serum (MS), female serum (FS) and castrated-male serum (CMS) were investigated. Overall, the chemical profile of GSBS was similar to that of bovine references except for glucose, creatine kinase, lactate dehydrogenase and potassium. FS showed elevated total protein and sodium concentrations compared to MS and CMS. Proteins present in MS, FS and CMS but absent in fetal bovine serum (FBS) were selected by two-dimensional gel electrophoresis and identified by peptide mass fingerprinting. Some of the identified proteins are known to be involved in immune responses and the others have unknown physiological roles. Moreover, it was found that some proteins such as alpha-2-macroglobulin appeared to be gender-specific with higher contents in FS. Insulin and testosterone was significantly higher in MS, and $17{\beta}$-estradiol and estrone were higher in FS, as compared to the other sera. Taken together, the results indicate that each GSBS has a different ratio of components. Differences in serum constituents may affect cell cultures in a different manner and could be beneficial, depending on the specific aim of cell cultures.
This study was designed to investigate the biochemical pollutant marker for diagnosis of marine pollutions by the changes in lipid components of the flounder (Pleuronichthys cornutus) in the Yellow Sea of Korea. Hemoglobin levels in serum of wild flounders in the yellow Sea were significantly lower $(15\~30\%)$ than those of wild flounder in Pohang (control) of the East Sea. Triglyceride (TG) contents in serum of wild flounders in the Yellow Sea were higher $(15\~50\%)$ than those of wild flounder in Pohang. Total and low density lipoprotein (LDL)-cholesterol contents in serum of flounders in the Yellow Sea were significantly higher $(20\~25\%\;and\;15\~35\%,\;respectively)$ than those of wild flounder in Pohang. Atherogenic index (Al) in serum of wild flounders in the Yellow Sea were significantly higher $(10\~60\%)$ than these of wild flounder in Pohang but T-Chol/PL ratios showed almost no change in serum of flounders in the Yellow Sea we examined. These results suggest that near-coastal water as well as neritic water of the Yellow Sea might be affected by pollutant input.
This studies were to investigate the effects of low power wavelengths Helium-Neon Infra-Red(He-Ne IR)laser on the changes of the blood biochemical components in burn rats. The thirty Spraque-Dewely adult male rats were assigned to the 5 groups; the experimental groups(3), the burn control group(1) and the control group(1). There was made three degree burn by the 250mW IR on the back of each rats, from 3 days after being, burned, the experimental laser groups were irradiated low power wavelengths (292Hz, 1168Hz, 4672Hz) He-Ne IR laser for 5 minutes every day during the 7days. The results wee as follows: The activities of aspirate aminotransferase(AST), alanine aminotransferase(ALT), and lactate dehydrogenase(LDH), on treated with the wavelengths ((292Hz, 1168Hz, 4672Hz) laser groups during 5 min. for 7 days were significantly increased to the control group respectively, but those of total cholesterol(CHOL) and alkaline phosphatase(ALP) were significantly decreased to the a control group (p<0.05). there were significantly decreased on the activity of alanine aminotransferase and alkaline phosphatase on the all experimental groups to the burn control group and those of lactate dehydronase on treated with 292Hz and 4672Hz wavelength9 groups to the burn control group but, were significantly increased on those of urea nitrogen (292Hz and 1168Hz)(p<0.05). The activity of AST, LDH, on treated with 1168Hz wavelengths were significantly decreased of the 292Hz and 4672Hz wavelengths groups that of ALT on the treated with 1168Hz group was significantly increased to the 4672 Hz wavelength group but those of CHOL and LDH on the 1168Hz and 4672Hz wavelength groups were dignificatly decreased to the MHz wavelengths group. As above results, the changes of the activities of biochemical conponents in the serum levels on the healing process have meaningful effected of the role of low power wavelengths He-Ne IR laser.
This study was designed as a part of efforts to investigate the biochemical pollutant markers for diagnosis of marine pollutions by changes in lipid components of the flounder (Paralichthys olivaceus) in the South Sea of Korea, Hemoglobin levels in serum of cultured flounders in the South Sea were lower ($4\~17\%$) than those of wild flounder in Pohang as a control group. Triglyceride (TG), Total and low density lipoprotein (LDL)-cholesterol contents in serum of cultured flounders in the South Sea were $12\~25\%,\;9\~19\%$ and $17\~25\%$ respectively which are higher than those of wild flounder in Pohang. Atherogenic index (AI) and T-Chol/PL ratios in serum of cultured flounders in the South Sea were significantly higher($26\~43\%$ and $17\~26\%$ respectively) than those of wild flounder in Pohang. These results suggest that near-coastal water as well as neritic water of the South Sea might be affected by pollutant input as well as the Yellow Sea of Korea.
This study was investigated the effects of the lower power He-Ne IR laser treatment on the changes of blood biochemical components in the rat liver damaged by the carbon tetrachloride (CCl₄). The twenty one Sprague-Dawley adult male rats weights (260±18.6 g) were designed to the three groups: one control group and two experimental groups, the experimental groups were divided into the CCl₄-treated groups and the laser therapy group (CCl₄+ Laser). The experimental groups were injected twice with CCl₄(1.0 ml/kg body weight) intraperitoneal for two days. Each group was sacrificed after two weeks irradiated with the lower power He-Ne IR laser for ten minutes per every day. The activity of alanine aminotransferase (ALT), lactic dehydrogenase (LDH), alkaline phosphatase (ALP) and the concentration of serum glucose treated with He-Ne IR laser groups was significantly decreased to the conrtol (treated with carbon CCl₄) group. The activity of aspartate aminotransferase (AST) was decreased in the laser group but not significantly, the concentration of the serum cholesterol in the laser group was significantly increased comparing with the control and case control groups. In conclusion, the effect of the lower power He-Ne IR laser treatment is believed to be a possible protective effects for CCl₄ induced acute hepatotoxicity in rats.
The effects of aloe on the MDA(malondialdehyde) and the blood biochemical components of heavy metal poisoning in SD rat were examined and the following results were obtained. In rat liver homogenate intoxicated with $CdCl_2$, lipidperoxide was increased each 2.37times(24h), 3.31times(72h) but lipidperoxide In aloe administration groups was lower each 47% , 64% than in heavy metal group. In rat kidney homo- genate intoxicated with $CdCl_2$, lipidperoxide was increased 1.85times(24h), 1.33times(72h) but lipidperoxide in groups was almost the same as that of normal group. Lipidperoxide of kidney homogenate was slightly decreased as time passed. Also heavy metal poisoning rats showed high levels(1.38-2.50times) of serum AST, ALT and BUN. However. the administration of aloe significantly inhibited the reduction of them. These results suggest that Cd-induced hepatic and renal injury, via increase llpidpero)Ode and release of AST, ALT and BUN. Aloe may be used to inhibit or prevent the hepatic and renal toxicity which results from the heavy metal.
Minerals are individual of the components of foods and are not produced in the body but essential for best possible health. Several essential metals are vital for the appropriate performance of various enzymes, transcriptional factors and proteins that are essential in various biochemical paths. Metals like zinc (Zn), magnesium (Mg), and manganese (Mn) are cofactors of hundreds of enzymes. Zn is involved in the synthesis and secretion of insulin from the pancreatic ${\beta}-cells$. Chromium (Cr) increases the insulin receptors activity on target tissues, mainly in muscle cells. Insulin hormone is required to maintain the blood glucose amount in normal range. Continual increase of blood serum glucose level leads to marked chronic hyperglycemia or diabetes mellitus. Deficiency of insulin or its resistance, blood glucose level exceeds the upper limit of the common range of 126 mg/dl. Poor glucose control and diabetes changes the levels of essential trace elements such as Zn, Mg, Mn, Cr, iron etc. by rising urinary excretion and their related decrease in the blood. The aim of this article to discusses the important roles of essential trace elements in particular perspective of type 2 diabetes.
Objectives: Cultivated wild ginseng (cWG), called SanYangSanSam, has been used clinically in patients with chronic fatigue in Korea. Little is known about effects of the ginseng distilled (volatile) components produced during evaporizaiton. Recently, we first identified one major component from cWG distilled extract, panaxydol, by using mass spectrometry. However, functional properties of cWG distilled extract and panaxydol remains elusive. Therefore, the present study evaluated the effect of cWG distilled extract or panaxydol on exercise-induced fatigue in rats. Methods: Fatigue was induced by forced swimming and the immobility time was analyzed in male Sprague-Dawley rats. The animals received intraperitoneally either vehicle, cWG distilled extract, or panaxydol 10 min prior to beginning of the forced swimming test (FST) once daily for 5 days. After the FST on day 5, we also analyzed fatigue-related biochemical levels including blood urea nitrogen (BUN), lactate acid (LAC), and lactate dehydrogenase (LDH) in serum and levels of glycogen in liver and soleus muscle. Results: The forced swimming time in cWG distilled extract (0.6 mL/kg)-treated group was significantly longer than that of control group on day 4 and 5. Panaxydol (0.1 and 0.25 mg/kg)-treated groups showed significantly enhanced performance in the forced swimming, compared to control. In addition, a significant decrease in serum LDH level was found in panaxydol-treated group, while there were no alternations in levels of serum BUN and LAC and glycogen in liver or soleus muscle. Conclusion: The present study demonstrated cWG distilled extract and its active component panaxydol have a function of anti-fatigue.
Four-d-old broiler chicks were randomly assigned to 3 groups with 9 replicates (8 birds/cage) under high ambient temperature; birds fed a basal diet (CON), a basal diet supplemented with 0.25% of probiotic complex (LPB, 1 × 106 Lactobacillus plantarum, 1 × 106 Bacillus subtilis, and 1 × 106 Saccharomyces cerevisiae) and 0.5% probiotic complex (HPB). Immediately after 28-d feeding trial, 6 birds having average body weight per group were sacrificed for evaluating the effects of probiotics on antioxidant parameters in the serum, intestine, and liver of birds. As results, serum biochemical parameters of nitrogen components including total protein, albumin, urea nitrogen, and glutathione were unaffected by dietary probiotics. In addition, serum superoxide dismutase (SOD), glutathione peroxidase (GPX), and glutathione S-transferase (GST) activities, and lipid peroxidation (MDA) were not changed by dietary probiotic supplement in birds. In the intestinal mucosa, SOD activity in the HPB group significantly (P<0.05) increased compared with that in the CON and the LPB groups. Lipid peroxidation in the HPB group significantly (P<0.05) decreased compared with that in the CON group. However, there was no statistical difference in GPX, and GST activities in the intestinal mucosa among treatment groups. In the liver, the activities of SOD, GPX, and GST, and the level of MDA were unaffected by probiotic supplement. In conclusion, 0.5% of probiotics significantly increased SOD activity and decreased lipid peroxidation in the intestinal mucosa, suggesting that probiotic complex could be potential to improve the small intestinal antioxidant capacity of bird under high ambient temperature conditions.
Objectives : Jakyakgamcho-tang has been used as analgesic and spasmolytic for muscle pain. It has reported to anti-diabetes, anti-inflammation, and neuro-protective effects. A clinical study of muscle fatigue and pain improvement effect of Jakyakgamcho-tang has been reported with increasing frequency of clinical use. However, the anti-fatigue effect of Jakyakgamcho-tang in animal model has not been studied. In this study, we compared anti-physical fatigue effects of water and 30% ethanol extract of Jakyakgamcho-tang. Methods : The amounts of components contained in water and 30% ethanol extract of Jakyakgamcho-tang were compared by ultra-performance liquid chromatography. Anti-physical fatigue effects were evaluated using weight-loaded forced swimming test. We also investigated the effects of Jakyakgamcho-tang on the change of fatigue parameters by blood biochemical analysis. Results : The relative amount of components of Jakyakgamcho-tang were about 19-53% higher in the 30% ethanol extract than in the water extract. In the swimming test, 30% ethanol extract showed a significantly greater anti-fatigue effects than conventional water extract of Jakyakgamcho-tang. The 30% ethanol extract of Jakyakgamcho-tang improved the exhausted swimming time (54%), and serum lactate dehydrogenase (48%) and lactic acid (60%) levels compared with water extract of Jakyakgamcho-tang. Conclusions : These results showed that differences in the amounts of components by different extraction were associated with differences of anti-fatigue effect of Jakyakgamcho-tang. Thus, the 30% ethanol extraction method could be applied to dose-reducing formulations of traditional herbal medicines.
