Expression of CD30 in Testis and Epididymis of Adult Mice

  • CD30 is a member of tumor necrosis factor receptor (TNFR) superfamily and has pleiotropic functions including cell activation, proliferation, differentiation, and death, depending on cell types and stage of differentiation. Although CD30 expression has been described mainly in hematopoietic tissues, several types of nonhematopoietic tumors including embryonic carcinoma and germ-cell tumors express CD30. We examined CD30 distribution in the testis and epididymis from wild type and CD30-deficient mice. In the testis, spermatogonia, spermatocytes and Sertoli cells expressed CD30, but not in spermatids. Spermatogonia and spermatocytes near the basement membrane strongly reacted to anti-CD30. In the epididymis, CD30 expression was exclusively observed in luminal epithelia and some interstitial cells. Taken together, these results show a spatio-temporal regulation of CD30 expression in mouse testis and epididymis and suggest a possible role of CD30 in spermatogonia and spermatocytes.

카드뮴이 고환에 미치는 영향에 관한 형태학적 및 생화학적 연구 (Morphological and Biochemical Studies on the Effects of Cadmium on the Rat Testis)

  • To investigate the cadmium (Cd) toxicity on the testis, male rats were treated with 1, 2, 4 and 8 mg/kg of Cd by IP. According to histochemical studies, Cd-treated testis tissue showed death of spermatozoa, death of Sertoli cells, death of all the spermatogenic cells, and finally disappearance of basal lamina of seminiferous tubules with increasing doses, and showed decreased ground substances and Leydig cells, increased inflammatory cells and fibroblasts, and fibroblasts, and finally disappearance of ground substances and all the cells except fibroblasts within interstitial tissues with increasing doses. According to biochemical studies, two kinds of proteins, 25 and 45 kDa, were dramatically disappeared from the total protein of rat testis treated with Cd comparing to normal testis. The result of electrophoresis of total protein suggests that actin (45 kDa), presumed on its mmolecular weight and amount, in the testis-cells is the primary target of Cd poisoning. Although its exact mechanism is not clear, the disappearance of two proteins when testis is exposed to Cd should give some clues to understnad the mechanism of necrosis of testis tissue crumbling by heavy metal pollutant such as Cd.

  • PURPOSE: Gene expression and apoptosis in testicular germ cells has been demonstrated in many transgenic animals. However, little is known about the transgenic pig and rates of apoptosis during spermatogenesis. METHODS : Morphological and biochemical features of apoptosis reported in other species were used to confirm that the TdT-mediated dUTP Nick end labeling (TUNEL) assay is an acceptable mothos for idendtification and quantification of apoptotic transgenic germ cells in histological tissue section from transgenic pig testis.   • The testis is composed of four cell types like supporting cells, steroid-producing cells, connective tissue cells and germ cells. Apoptosis is a common phenomenon during spormatogenesis. Apoptosis of germ cells can also be induced by exposure to radiation. Previous studies have shown that most types of germ cells are rather radiosensitive while somatic cells in testis are much more radio-resistant. The somatic cells in testis are divided to mainly Sertoli and Leydig cells. Though somatic cells are more radio-resistant than germ cells, radiation can induce the impairment of their function. This damaged function of somatic cells may accelerates degeneration of germ cell indirectly. Tn the present study, we have examined the apoptotic effect of mouse testis and irradiation effect of steroidogenesis of Leydig cells after irradiation.

돼지 Cytochrome P450 Aromatase의 새로운 기능

  • 여성호르몬으로 잘 알려진 estrogen은 난소의 granulosa 세포와 정소의 Sertoli 세포에서 주로 생성되는 것으로 알려져 있으며 최근엔 사람의 지방, 근육, 뇌, 뼈세포 등에서의 합성 가능성과 각 조직에서 생성되는 estrogen 의 생리학적인 기능에 관해 많은 관심이 모여져 왔다. 다른 steroid처럼 지방친화적인 (lipophilic) estrogen은 세포막과 핵막을 쉽게 통과하여 목표세포 (target cell)의 핵에 존재하는 estrogen receptor (ER)에 결합하여 특정 유전자를 발현에 관여하는 것으로 알려져 있다. Cytochrome P450 Aromatase (간략히, aromatase)는 steroid hormone을 합성하는 마지막 단계에서 androgens을 estrogens으로 전환시키는데 관여하는 효소이다. Aromatase의 substrate (기질)로 사용되는 androgen에는 androstenedione과 testosterone 등이 있으며, 최종 산물로써 estrone(E1)이나 17$\beta$-estradiol(E2) 등의 estrogen이 생성된다. Aromatase는 steroidogenic tissues의 세포내 골지체에 존재하는 것으로 알려져 있으며, NADPH-cytochrome P450 reductase와 함께 복합체로써 존재한다. NADPH-cytochrome P450 reductase는 다른 steroid hormone에 관여하는 효소들과도 복합체를 형성하여 다른 steroid hormone을 합성할 수 있으므로, 어떤 조직에서 여성호르몬이 만들어질 수 있느냐 하는 것은 그 조직에서 aromatase 단백질이 만들어지느냐에 따라서 결정된다.

Expression and Function of CTNNB1 in the Development of Avian Reproductive System

  • Beta-catenin (CTNNB1, catenin (cadherin-associated protein), beta 1) is involved in various biological processes, including embryogenesis, tumorigenesis, angiogenesis and progression of metastasis. CTNNB1, as a multifunctional and oncogenic protein, has important roles in adhesion between Sertoli cells through an N-cadherin-dependent manner and in various cancer types through its over-activation. In addition, CTNNB1 can interact with estrogen/estrogen receptor alpha complex, which regulates the transcription of WNT (wingless-type MMTV integration site family)/CTNNB1 target genes. Recently, we investigated the functional roles and expression pattern of CTNNB1 during the morphological changes of embryonic gonads of chickens and the estrogen-dependent regulation of CTNNB1 in oviduct development and potential functions as a biomarker of CTNNB1 in human epithelial ovarian cancer using the chicken as a biological research model. Therefore, in this review, we provide a new insight of potential role of CTNNB1 in the development of the female reproductive tract during early embryogenesis and ovarian carcinogenesis of laying hen models.

  • Testes from the allied rock wallaby(Petrogale assimilus) have been examined by the electron microscopy in thin sections in order to examine spermiogenesis and structure of spermatozoa. The spermiogenesis can be divided into nine stages: early Golgi, late Golgi, collapsing, nuclear protrusion, condensation and flattening, nuclear shaping, rotation, nuclear ring contraction, and maturation. The acrosome has been abruptly formed following the collapse of expanded acrosomal vesicle without the cap stage described in the eutheria. The flatly condensed nucleus rotates obliquely to the axis of the axial filament complex and the folded acrosome covers the anterior third of the dorsal nuclear surface forming a wide subacrosormal space as the nuclear ring has contracted. The Sertoli cell reaction and spur are prominent during the nuclear protrusion and rotation stages. A mature spermatozoon has S-shape head which has an extended part reaching to the vicinity of the middle piece.

Alkylating Agent에 의한 Mouse 정세관의 미세구조 변화 (Ultrastructural Changes of Seminiferous Tubules in the Mouse by Alkylating Agent)

  • The purpose of this study was to investigate the effect of cyclophosphamide on the mouse seminiferous tubules by transmission electron microscopy at different groups: a control group, a group treated one time a week, and a group treated two times a week. Cyclophosphamide was injected in the intraperitoneal at a dosage of level 200mg/kg. The results obtained were as follows. 1. In the group of one time a week, pyknotic body and large vesicles were observed in cytoplasm of spermatogonium of seminiferous tubules, and in the intercellular space between spermatocytes, respectively. 2. In the group of two times a week, nucleus envelope in the spermatid was disrupted partially, observed vesicles in the nucleoplasm of spermatid, and separated or disrupted inner and outer membranes of mitochondria in the Sertoli cells.

Unusual malignant neoplasms of ovary in children: two cases report

  • Sex cord tumors with annular tubules are known to originate from the sex cord of embryonic gonads that synthesize Sertoli cells, Leydig cells, granulosa cells, and theca cells of the ovarian stroma, while ovarian small cell carcinoma of the hypercalcemic type is a type of neuroendocrine tumor. Both these tumors are uncommon, potentially malignant neoplasms in children. We report the case of a sex cord tumor with annular tubules in an 11-year-old girl and a case of small cell carcinoma of the hypercalcemic type in a 10-year-old girl. We also discuss the prognosis and management of these tumors.

Sprague-Dawley 랫트에 자연발생한 고환위축의 병리조직학적 관찰 (Histopathological observation of spontaneous testicular atrophy in Sprague-Dawley rat)

  • The incidence of spontaneous testicular atrophy and its morphological changes during stage-specific spermatogenesis were investigated in male Sprague-Dawley rats at 10, 19, and 32 weeks of age. The incidence of testicular atrophy was 0.2%(2/90) 7.9%(9/114) and 10%(4/40) in 4, 13 and 26 weeks respectively. The epididymis with testicular atrophy had low sperm density. In the minimally affected tests scattered tubules showed complete depletion of germ cells without stage specificity. Testes with moderate to severe testicular atrophy showed seminiferous tubules lined with dense Sertoli cell population. While Leydig cells in the interstitium appeared hypertrophy they were immunohistochemically negative for proliferating cell nuclear antigen a marker of cell proliferation.
