  • Variations of reaction sequence of $Pb(Zn_{0.6}Mg_{0.4})_{1/3}Nb_{2/3}O_3$[PZMN] with precusor mixing methods were ex-amined using X-ray diffraction and dielectric characteristics. In the present study three different types of precursor mixing methods (oxide mixing PbO+$ZnNb_2O_6+MgNb_2O_6$[Zn+MN] and PbO+(Zn,Mg)$Nb_2O_6$[ZMN] precursor mixing) were adopted. When the oxide mixing method was used for the PZMN synthesis a Zn-rich perovskite phase and pyrochlore phase were formed. Compared with PbO+ZN+MN precursor mixing method the PbO-ZMN precursor led to a lowering of the formation temperature for perovskite sin-gle phase. These variation of composition and formation temperature of the perovskite phase were dis-cussed in terms of the difference in the solid-reaction requence between these three different types of pre-cursor mixing.

Molecular phylogeny of Indonesian Zeuzera(Lepidoptera:Cossidae) wood borer moths based on CO I gene sequence

  • Zeuzera is one of the most important wood borer pests in South East Asia. Like of most other genera of moths, the systematic of this genus is still in dispute, especially on the monophyly and the relationship within this genus due to the fact that genus is very varied. This genus was defined based on external characters only such as cross vein Sc-Rs present, humeral plate approximately triangular in shape and anal plate moderately long to moderately short. Therefore, the monophyly of this genus need to be evaluated based on more comprehensive data. To clarify the monophyly of the genus Zeuzera, to reveal the phylogenetic relationships among the Indonesian species, and to establish the genetic characters of Indonesian Zeuzera, we analyzed seven species of Indonesian Zeuzera including three other species distributed around the world based on nucleotide sequence variation across a 580-bp region in the CO I gene. The results showed that the monophyly of Zeuzera was supported by bootstrap tests at the MP and ML tree building methods (>95%). Genus Zeuzera was divided into two groups (A and B) with Z. borneana was excluded from the two groups and occupied at the basal node. Indonesian species was distributed into two different clades. CO I gene alone was able to fully resolve the relationships among species within clade B. However, further investigations were needed by including more species and other genes that the more conserved to test the validity of the phylogenetic hypothesis proposed here.

  • fMRI, functional MRI introduced recently appears based on the gradient echo technique which is sensitive to the field inhomogeneity developed due to the local susceptibility changes of blood oxygenation and deoxygenation. Common to all the gradient echo techniques is that the signal due to the susceptibility effects is generally decreased with increasing inhomogeneity due to the $T2^*$ effect or conventionally known as blood oxygenation level dependent (BOLD) effect. It is, also found that the BOLD sensitivity is also dependent on the imaging modes, namely whether the imaging is in axial, or coronal or sagittal mode as well as the directions of the vessels against the main magnetic field. We have, therefore, launched a systematic study of imaging mode dependent signal change or BOLD sensitivity as well as the signal changes due to the tilting angle of the imaging planes. Study has been made or both TRFGE sequence and CGE sequence to compare the distinctions of the each mode since each technique has different sensitivity against susceptibility effect. Method of computation and both the computer simulations and their corresponding experimental results are presented.

Identification and Phylogeny of the Human Endogenous Retrovirus HERV-W LTR Family in Cancer Cells

  • The long terminal repeats (LTRs) of human endogenous retrovirus (HERV) have been found to be coexpressed with sequences of closely located genes. It has been suggested that the LTR elements have contributed to the structural change or genetic variation of human genome connected to various diseases and evolution. We examined the HERV-W LTR elements in various cancer cells (2F7, A43l , A549, HepG2, MIA-PaCa-2, PC-3, RT4, SiHa, U-937, and UO-31). Using genomic DNA from the cancer cells, we performed PCR amplification and identified twelve new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity (88-99%) with HERV-W LTR (AF072500). A phylogenetic tree obtained by the neighbor-joining method revealed that HERV-W LTR elements could be mainly divided into two groups through evolutionary divergence. Three HERV-W LTR elements (RT4-2, A43l-1, and UO3l-2) belonged to Group 1, whereas nine LTR elements (2F7-2, A549-1, A549-3, HepG2-3, MP2-2, PC3-1, SiHa-8, SiHa-10, and U937-1) belonged to Group 11. Taken together, our new sequence data of the HERV-W LTR elements may contribute to an understanding of tissue-specific cancer by genomic instability of LTR integration.

Determination of Design Moments in Bridges Constructed by Movable Scaffolding System (MSS공법으로 시공되는 교량의 설계 모멘트 결정)

  • In this paper, a relation to calculate design moments for reinforced concrete(RC) bridges constructed by movable scaffolding system(MSS) is introduced. Through the time-dependent analysis of RC bridges considering the construction sequence, the structural responses related to the member forces and deflections are reviewed, and a governing equation for determination of the design moment, which includes the creep deformation, is derived on the basis of the displacement-force condition at every constructuion stage. By using the relation, the design moment and its variation over time can easily be obtained only with the elastic analysis results without additional time-dependent analysis. In addition, correlation studies with the results by rigorous numerical analyses are conducts to verify the applicability of the introduced relation, and a more reasonable guideline for the determination of design moments is proposed on the basis of the obtained moment envelop.

  • A turbine control system which has been operated for years in a nuclear power plant was retrofitted with a newly developed digital control system. After completion of the retrofit, turbine valve tests were performed to ensure the integrity of each valve's control function. The sequence of each valve test is composed of a closing process and a reopening process. To minimize megawatt variation which normally occurs during the test sequence, we employed a kind of compensator algorithm in the new digital control system which also have been used in the old system. There were difficulties finding optimal parameter settings for our new compensator algorithm because the power plant didn't allow us to perform necessary tuning procedures while the turbine is on load operation. Therefore an alternative measure for the compensator tuning which is independent of the turbine actual operation had to be implemented. So, a process model for the test was required to overcome this situation. We analyzed the operation data of the test and implemented the process model by use of input and output variable relations. Also we verified the process model by use of another condition's operating data. The result shows that the output of model is similar to the actual operation data.

Genetic Diversity and Metabolite Analysis of Gastrodia elata by Inter-Simple Sequence Repeats (ISSR) Markers (ISSR 표지에 의한 천마의 유전 다양성분석 및 기능성 물질분석)

  • Gastrodia elata, an achlorophyllous orchid plant, is rare medicinal plant. We investigated the genetic diversity in G. elata from 4 locations by using Inter-Simple Sequence Repeats (ISSR) markers. Shannon's information Index (S.I.) indicating genetic diversity ranged from 0.255 (Pocheon) to 0.322 (Muju) with the mean of 0.29. The level of genetic diversity was lower than other plant and most genetic diversity was allocated among individuals within populations (26.81%). The UPGMA dendrogram based on genetic distance failed in showing decisive geographic relationship. In the case of gastrodin (GA), the major components in G. elata, Sangju was highest. The ergothionine (ERG) was detected a lot of contents in Muju and Pocheon. In conclusion, our results is very important information for explaining relationship of genetic variation and functional substances without the effects of environment factors and developing genetic marker by ISSR in G. elata, which may be responsible for the development of breeds with a lot of functional substance in G. elata.

Reliability Analysis for Structure Design of Automatic Ocean Salt Collector Using Sampling Method of Monte Carlo Simulation

  • This paper presents comparative studies of reliability analysis and meta-modeling using the sampling method of Monte Carlo simulation for the structure design of an automatic ocean salt collector (AOSC). The thickness sizing variables of structure members are considered as random variables. Probabilistic performance functions are selected from strength performances evaluated via the finite element analysis of an AOSC. The sampling methods used in the comparative studies are simple random sampling and Sobol sequences with varied numbers of sampling. Approximation methods such as the Kriging model is applied to the meta-model generation. Reliability performances such as the probability failure and distribution are compared based on the variation of the sampling method of Monte Carlo simulation. The meta-modeling accuracy is evaluated for the Kriging model generated from the Monte Carlo simulation and Sobol sequence results. It is discovered that the Sobol sequence method is applicable to not only to the reliability analysis for the structural design of marine equipment such as the AOSC, but also to Kriging meta-modeling owing to its high numerical efficiency.

Development of SCAR Marker for Identification of the Perilla Species

  • This study is to generate SCARs markers for identification of Perilla species. A SCAR is a genomic DNA fragment at a single genetically defined locus that is identified by PCR amplification using a pair of specific oligonucleotide primers. We derived SCARs by sequencing and cloning the both ends of the amplified products of RAPD markers. Sixteen sequence-specific primers were synthesized from eight RAPD markers, which were completely sequenced. We developed the species-specific SCAR markers which could be used successfully in detecting genetic variation in four Perilla species. These markers could be used to verify species-origins of various forms of Perilla germplasms.

Genetic Identification of Spirometra erinaceieuropaei Spargana in Liaoning and Hubei Provinces, PR China

  • Spargana were collected from human and frogs in Liaoning and Hubei Provinces, China. PCR amplification and direct sequencing of A cox1 fragment was PCR-amplified from genomic DNA extracted from 7 specimens (5 from humans and 2 from frogs). The cox1 fragment (390 bp) showed 97-100% similarity to the reference sequence of S. erinaceieuropaei and 88-89% to the reference sequence of S. decipiens. There were 1-12 bases different between these worms, but no obvious genetic variation (0-3.3%) to the references. There was little difference of cox1 gene between sparganum samples of humans and frogs (1-3%). This study is the first report on S. erinaceieuropaei spargana from humans in Liaoning and Hubei Provinces.
