2C-methyl-D-erythritol 2, 4-cyclodiphosphate synthase (MECPS, EC: 4.6.1.12) is the fifth enzyme of the non-mevalonate terpenoid pathway for isopentenyl diphosphate biosynthesis and is involved in the methylerythritol phosphate (MEP) pathway for ginkgolide biosynthesis. The full-length mecps cDNA sequence (designated as Gbmecps) was cloned and characterized for the first time from gymnosperm plant species, Ginkgo biloba, using RACE (rapid amplification of cDNA ends) technique. The full-length cDNA of Gbmecps was 874 bp containing a 720 bp open reading frame (ORF) encoding a peptide of 239 amino acids with a calculated molecular mass of 26.03 kDa and an isoelectric point of 8.83. Comparative and bioinformatic analyses revealed that GbMECPS showed extensive homology with MECPSs from other species and contained conserved residues owned by the MECPS protein family. Phylogenetic analysis indicated that GbMECPS was more ancient than other plant MECPSs. Tissue expression pattern analysis indicated that GbMECPS expressed the highest in roots, followed by in leaves, and the lowest in seeds. The color complementation assay indicated that GbMECPS could accelerate the accumulation of $\beta$-carotene. The cloning, characterization and functional analysis of GbMECPS will be helpful to understand more about the role of MECPS involved in the ginkgolides biosynthesis at the molecular level.
Species identification using DNA sequences is the basis for DNA taxonomy. In this study, we sequenced the ribosomal large-subunit RNA gene sequences (3,037-3,061 bp) in length of 13 Chinese Theileria stocks that were infective to cattle and sheep. The complete 28S rRNA gene is relatively difficult to amplify and its conserved region is not important for phylogenetic study. Therefore, we selected the D2-D3 region from the complete 28S rRNA sequences for phylogenetic analysis. Our analyses of 28S rRNA gene sequences showed that the 28S rRNA was useful as a phylogenetic marker for analyzing the relationships among Theileria spp. in ruminants. In addition, the D2-D3 region was a short segment that could be used instead of the whole 28S rRNA sequence during the phylogenetic analysis of Theileria, and it may be an ideal DNA barcode.
Lysin motif (LysM) proteins are reported to be necessary for the virulence and immune response suppression in many herbaceous plant pathogens, while far less is documented in woody plant pathogens. In this study, we preliminarily characterized the molecular function of a LysM protein LtLysM1 in woody plant pathogen Lasiodiplodia theobromae. Transcriptional profiles revealed that LtLysM1 is highly expressed at infectious stages, especially at 36 and 48 hours post inoculation. Amino acid sequence analyses revealed that LtLysM1 was a putative glycoprotein with 10 predicted N-glycosylation sites and one LysM domain. Pathogenicity tests showed that overexpressed transformants of LtLysM1 displayed increased virulence on grapevine shoots in comparison with that of wild type CSS-01s, and RNAi transformants of LtLysM1 exhibited significantly decreased lesion length when compared with that of wild type CSS-01s. Moreover, LtLysM1 was confirmed to be a secreted protein by a yeast signal peptide trap assay. Transient expression in Nicotiana benthamiana together with protein immunoblotting confirmed that LtLysM1 was an N-glycosylated protein. In contrast to previously reported LysM protein Slp1 and OsCEBiP, LtLysM1 molecule did not interact with itself based on yeast two hybrid and co-immunoprecipitation assays. These results indicate that LtLysM1 is a secreted protein and functions as a critical virulence factor during the disease symptom development in woody plants.
Spore-forming bacteria are being used as probiotic supplements for human and animal use, due to their low pH stability and ability to survive the gastric barrier. In this study, the BCNU 9028 strain was screened from meju, a Korean fermented soybean food starter. Biochemical and physiological characteristics, as well as 16S rDNA sequence analyses, indicate that this strain belongs to the genus $Bacillus$. $Bacillus$ sp. BCNU 9028 showed a 92% survivability at pH 2.5 and could also withstand 0.3% ox bile. Furthermore, it was postulated that $Bacillus$ sp. BCNU 9028 could prevent biofilm formation and adherence of food-borne pathogens such as $Listeria$$monocytogenes$, $S.$$aureus$ and $E.$$coli$ on the basis of its autoaggregation and coaggregation capacity with food-borne pathogens. It was shown that BCNU 9028 has good abilities to adhere to the intestinal tract from its hydrophobic character (63.3%). The $Bacillus$ sp. BCNU 9028 strain especially elicited antibacterial activity against both Gram-positive and -negative pathogens. These findings suggested that the $Bacillus$ sp. BCNU 9028 strain could be used as a potential probiotic.

The Gramene database (http://www.gramene.org) is a powerful online resource for agricultural researchers, plant breeders and educators that provides easy access to reference data, visualizations and analytical tools for conducting cross-species comparisons. Learn the benefits of using Gramene to enrich your lectures, accelerate your research goals, and respond to your organismal community needs. Gramene's genomes portal hosts browsers for 44 complete reference genomes, including crops and model organisms, each displaying functional annotations, gene-trees with orthologous and paralogous gene classification, and whole-genome alignments. SNP and structural diversity data, available for 11 species, are displayed in the context of gene annotation, protein domains and functional consequences on transcript structure (e.g., missense variant). Browsers from multiple species can be viewed simultaneously with links to community-driven organismal databases. Thus, while hosting the underlying data for comparative studies, the portal also provides unified access to diverse plant community resources, and the ability for communities to upload and display private data sets in multiple standard formats. Our BioMart data mining interface enable complex queries and bulk download of sequence, annotation, homology and variation data. Gramene's pathway portal, the Plant Reactome, hosts over 240 pathways curated in rice and inferred in 66 additional plant species by orthology projection. Recent abnormal weather, especially continued rainfall during sowing season causes difficulty in proper sowing of wheat and delayed sowing after November 15 is concerned about freezing damage during winter, resulting in reduction of wheat yield. To correspond government policy of crop sufficiency improvement and produce and supply raw wheat and barley steadily, expansion of cultivation area is necessary and spring sowing of wheat is required. To obtain basic information on the improvement of spring sown wheat and barley production, comparison and path coefficients analysis was conducted for yield and yield related components from autumn and spring sown wheat and barley. Path analyses were known as very useful in clarifying the effects of yield components on grain yield formation, which were not accurately reflected in simple correlation anaylses. Most cultivated 5 wheat and 9 barley cultivars were sown on October and February at Cheon-ju province according to standard sowing method. For the spring sowing of wheat and barley, the varieties having vernalization degree I~III are seeded in the mid of February and seeding rate is 200~250kg/ha which is increased by 25% than autumn sowing. N-fertilizer of 95 kg/ha and the same amount of P, K dressed in autumn are applied at once as basal fertilizer. The magnitude of direct effect in each yield components on yield was in sequence as follows. In autumn wheat, grain number per $spike{\geq}$ the number of spike per $m^2$>1000-grain weight and in spring wheat, grain number per $ spike{\geq}the$ number of spike per $m^2$> 1000-grain weight. In autumn naked barley, 1000-grain weight> the number of spike per $m^2$, grain number per spike and in spring barely, the number of spike per $m^2$> grain number per spike > 1000-grain weight. In autumn covered barley, grain number per spike>the number of spike per $m^2$ and in spring coverd barley, the number of spike per $m^2$> grain number per spike, 1000-grain weight. In autumn malt barley, the number of spike per $m^2$>1000-grain weight and in spring malt barley, the direct effects of three yield components were similar. According to the path analysis of yield components for spring sown wheat and barley, it was suggested that adequate number of spike per $m^2$ was most important factor for yield increase.
In order to clarify intra-and inter-specific variation of Korean Rana species, the partial DNA sequences of mitochondrial 16S rDNA gene were determined from 6 Korean and 1 Japanese Rana species, DNA sequences from Korean and Japanese species were comparison-analysed within, and also with the sequences from three species of Japanese brown frogs. DNA similarities were calculated as 91.3∼97.3% among brown frog (R. amurensis coreana, R. dybowskii and R. huanrenensis), as 96.11∼97.26% among pond frogs (R. nigromaculata and R. planeyi chosenica). Genetic distance of pond frog and wrinkle fyog (R. rugosa) were near than that of pond frog and brown frog. Two clusters were formed brown frogs and the other group by neigh-bor-joining and maximum-likelihood analysis, also the populations of R. nigromaculata were well distinguished between Korean peninsula and Korean island. But result from maximum-likelihood analysis slightly differed from neighbor-joining to cluster of R. rugosa. Further analyses for their population will be necessary to study the phylogenetic status.
Mushrooms growing in the Wolchulsan National Park were surveyed from April to October 2018. As a result of this survey, 2 divisions, 9 classes, 24 orders, 71 families, 177 genera, and 407 species were found. Among them, 3 classes, 16 orders, 55 families, 150 genera, and 370 species belonged to Basidiomycota, while 6 classes, 8 orders, 16 families, 27 genera, and 37 species belonged to Ascomycota. The species richness was the highest in September at the altitudes between ~100 and 200 m. Among the investigated sites, the most diverse species occurred in the Gyeongpodae area. In this survey we found, Macrolepiota procera, Suillus bovinus, and Amanita pantherina, all of which have been known as climate-sensitive biological indicator species (CBIS). Based on the DNA sequence analyses, Craterellus parvogriseus was confirmed as a new record for Korea.

The ground around the portal of a tunnel is the most typical part showing the 3-dimensional mechanical behavior in the tunnel. The portal slope is constructed at the weathered soft rock-mass, and remains as a potential sliding mass. The slope failure around the tunnel portal may happen drastically and induce the great disaster; hence, for the permanent stability several special techniques are required. To solve this problem, the ground around the tunnel portal may be excavated in the arch shape to develop the arching effect in horizontal direction. With the arch-type portal slope, one can reduce considerably the excavation mass and the damage of environments. This approach has not been attempted yet due to the lack of understanding and the well-defined analyzing method, so the retaining wall type portal is more universal. The 3-dimensional finite element analyses were carried out to prove that the arch type is more advantageous in safety and cost than the right angle type. The influence of the tunnel construction sequence and the strength of the rock-mass on the slope stability was investigated by focusing on the maximum shear strain in the slope, and the yield zone at the tunnel face.

The experiment was carried out to analyze the inhibition effect of plant pathogenic fungi and growth promotion activity induced by the bacterial strains isolated from peatmoss. Among the isolated bacterial strains, B10-2, B10-4, B10-5 and B10-6 which showed more than 30% inhibition rate against Botrytis cinerea and Rhizoctonia solani in vitro, were further analyzed in the greenhouse for the growth promotion activity on lettuce (Lactuca sativa), pak-choi (Brassica compestris L. ssp. chinensis) and Chinese cabbage (Brassica campestris L. ssp. pekinensis). The results showed the treatment of B10-4 on lettuce showed the highest growth promotion activity with the leaf area ($169.17cm^2$), fresh weight (leaf: 40.29 g, root: 8.80 g)and dry weight (leaf: 11.24 g, root: 4.17 g), which was about two folds as compared to control. On pak-choi, the growth promotion rate was the highest with the leaf area of $112.87cm^2$, leaf fresh weight of 60.70 g, root fresh weight of 3.37 g, leaf dry weight of 14.34 g, and root dry weight of 1.90 g. As a result of treatment of B10-13 on chinese cabbage, the growth promotion rate was the highest with the leaf area ($293.56cm^2$), fresh weight (leaf: 113.67 g, root: 2.40 g) and dry weight (leaf: 6.03 g, root: 0.53 g). The production of Indole Acetic Acid (IAA) and Indole-3-Butylic Acid (IBA) were also analyzed in these bacterial isolates. The IAA and IBA analyses were carried out in all bacterial isolates each day within the 5 days of incubation period. The highest production of IAA was observed with $112.57{\mu}g/mg$ protein in B10-4 after 3 days of incubation and IBA production was the highest in B10-2 with $58.71{\mu}g/mg$ protein after 2 days of incubation. Also, phosphate solubilizing activity was expressed significantly in B10-13 in comparison to that of other bacterial isolates. Bacterial identification showed that B10-2 was Bacillaceae bacterium and B10-5 was Bacillus cereus, B10-4 and B10-6 were Bacillus sp. and B-13 was Staphylococcus sp. by ITS sequence.
