MALDI-MS-Based Quantitative Analysis of Bioactive Forms of Vitamin D in Biological Samples

  • Analyzing vitamin D levels is important for monitoring health conditions because vitamin D deficiency is associated with various diseases such as rickets, osteomalacia, cardiovascular disorders and some cancers. However, vitamin D concentration in the blood is very low with optimal level of 75 nmol/L, making quantitative analysis difficult. The objective of this study was to develop a highly sensitive analysis method for vitamin D using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-MS). 25-hydroxyvitamin D (25(OH)D), which has been used as an indicator of vitamin D metabolites in human biofluids was chemically derivatized using a secosteroid signal enhancing tag (SecoSET) with powerful dienophile and permanent positive charge. The SecoSET-derivatized 25(OH)D provided good linearity (R2 > 0.99) and sensitivity (limit of quantitation: 11.3 fmol). Chemical derivatization of deuterated 25-hydroxyvitamin D3 (d6-25(OH)D3) with SecoSET enabled absolute quantitative analysis using MALDI-MS. The highly sensitive method could be successfully applied into monitoring of quantitative changes of bioactive vitamin D metabolites after treatment with ketoconazole to inhibit 1α-hydroxylase reaction related to vitamin D metabolism in human breast cancer cells. Taken together, we developed a MALDI-MS-based platform that could quantitatively analyze vitamin D metabolites from cell products, blood and other biofluids. This platform may be applied to monitor various diseases associated with vitamin D deficiency such as rickets, osteomalacia and breast cancer.

유한 및 무한 용량 대기열을 가지는 선점 우선순위 M/G/1 대기행렬 (M/G/1 Preemptive Priority Queues With Finite and Infinite Buffers)

  • Recently, M/G/1 priority queues with a finite buffer for high-priority customers and an infinite buffer for low-priority customers have applied to the analysis of communication systems with two heterogeneous traffics : delay-sensitive traffic and loss-sensitive traffic. However, these studies are limited to M/G/1 priority queues with finite and infinite buffers under a work-conserving priority discipline such as the nonpreemptive or preemptive resume priority discipline. In many situations, if a service is preempted, then the preempted service should be completely repeated when the server is available for it. This study extends the previous studies to M/G/1 priority queues with finite and infinite buffers under the preemptive repeat-different and preemptive repeat-identical priority disciplines. We derive the loss probability of high-priority customers and the waiting time distributions of high- and low-priority customers. In order to do this, we utilize the delay cycle analysis of finite-buffer M/G/1/K queues, which has been recently developed for the analysis of M/G/1 priority queues with finite and infinite buffers, and combine it with the analysis of the service time structure of a low-priority customer for the preemptive-repeat and preemptive-identical priority disciplines. We also present numerical examples to explore the impact of the size of the finite buffer and the arrival rates and service distributions of both classes on the system performance for various preemptive priority disciplines.

확률강우량 산정을 위한 EDA 기법의 적용 (Application of EDA Techniques for Estimating Rainfall Quantiles)

  • 본 연구에서는 자료의 구조를 이용하는 통계방법인 EDA 기법을 적용하여 자료를 정량화 하고, 이를 이용하여 빈도해석을 실시하였다. 모멘트법을 이용하는 전통적 방법이 극치값에 민감하게 반응하는 통계치를 주지만, EDA 기법은 변동이 적은 안정적인 통계치를 주는 장점이 있다. 빈도해석에 EDA 기법를 적용하는 경우에는 자료의 왜곡도를 반영하기 위해 원자료의 정규화 변환 및 역변환 과정을 거쳐야 한다. 즉, 원자료를 정규화 변환하고, EDA 기법을 적용하여 변환된 자료의 통계치를 추정하며, 이를 다시 역변환하여 원자료의 통계치를 결정해야 한다. 이렇게 결정된 통계치는 주어진 확률밀도함수를 이용한 빈도해석에 적용된다. 본 연구에서는 서울 및 포항지점의 연최대치 1시간 강우자료를 대상으로 분석을 수행하였다. 그 결과 EDA 기법을 적용하는 경우 극치값에 덜 민감한 안정적인 확률강우량의 산정이 가능한 것으로 확인되었다. 이러한 방법론은 특히 기후변화 등의 원인으로 강수자체의 경년변동이 매우 큰 지점의 빈도해석에 유용하게 사용될 수 있을 것이다.

LC-MS/MS-based Quantification of Ten Neurotransmitters in Rat Limbic System and Serum: Application to Chronic Unpredictable Mild Stress-Induced Depression Rats

  • As one of the most common mood disorders, numerous studies have shown depression is the main risk factor for non-suicidal self-harm. The pathogenesis of depression is complex, and a comprehensive and rapid measurement of monoamine neurotransmitters and their metabolites will be very helpful in understanding the pathogenesis of depression. Therefore, a rapid and sensitive underivatized liquid chromatography-tandem mass spectrometry method was developed and validated for the simultaneous monitoring of the levels of ten neurotransmitters and their metabolites in rat serum and limbic system and successfully applied to quantify the changes of neurotransmitter levels in chronic unpredictable mild stress-induced rats. The analytes studied were mainly involved in tyrosine metabolism, tryptophan metabolism, and glutamate cycling pathways, which are important in the pathogenesis of depression. It had been verified the method was sensitive and effective, with satisfactory linearity, and met the requirements of biological sample determination. Levels of neurotransmitters in rat serum, hippocampus, amygdala, prefrontal cortex, striatum, and hypothalamus were determined via the method. The results showed serotonin, dopamine, norepinephrine, and their metabolites were decreased, glutamine was increased, and glutamate was disturbed in chronic unpredictable mild stress-induced depression rats. This method provides a new approach to studying the pathogenesis of depression and other neurological disorders.

애기장대의 염해 저항성 관련 유전자의 기능적 선별 (Functional Screening of Plant Genes Suppressed Salt Sensitive Phenotype of Calcineurin Deficient Mutant through Yeast Complementation Analysis)

  • 작물의 생산성 증가를 위해 염해 저항성 메커니즘을 이해하는 것이 중요하다. 식물의 염해 저항성에 관련된 유전자를 확보하기 위한 여러 가지의 선별방법이 개발되었다. 본 논문에서는 애기장대의 cDNA 라이브리를 염해감수성 효모인 cnb 돌연변이체에 삽입하여 염해 감수성 표현형을 회복하는 콜로니를 선발하였다. 이 선별방법을 통하여 34종의 cnb 돌연변이체의 염해 감수성을 회복하는 콜로니를 선별하였으며, 염기서열분석을 통하여 CaS와 AtSUMO1, AtHB-12 등 9종의 유전자임을 확인하였다. 이들 유전자 중 CaS의 발현이 염해 저항성을 증가시키는 것과 염해 처리에 의해 CaS의 유전자의 발현이 증가되는 것을 확인하였다. CaS 발현억제 형질전환체는 100 mM 염처리에 의하여 뿌리생장이 저해되었다. 또한 150 mM 염처리에 의하여 CaS 발현억제 형질전환체의 잎에서 백화현상을 나타내었다. 이러한 결과를 통하여 CaS 유전자가 효모와 식물에서 염해 저항성에 중요한 유전자임을 증명하였다.

  • Despite the increasing necessity of accurate estimation of ring-stiffened cylinders' ultimate strength, the complex structural behavior of cylinders has made their design mainly depend on empirical formulas mostly based on insufficient test data rather than theoretical background. This paper has developed the imperfection method which enables the ultimate strength analysis of buckling sensitive structures by combining two general functions of common commercial finite element softwares, linear elastic buckling analysis and nonlinear stress analysis. Developed method was applied to two of the world most renowned commercial softwares, MSC/NASTRAN and ABAQUS, for the analysis of ring-stiffened cylinders and unexpectedly big difference in their analysis results was found. This tells that many widely used commercial softwares has their different strong points and week points and the choice of commercial software should be cautiously made after thorough inspection. This paper ends with some useful information about which of the aforementioned two softwares is more appropriate respectively for linear elastic buckling analysis and ultimate strength analysis of ring-stiffened cylinders.

Bacillus sp. N32 균주가 생산하는 항균 단백질 특성 (Characterization of antimicrobial proteins produced by Bacillus sp. N32)

  • 작물 근권 토양으로부터 분리한 5,000여 길항 균주로부터 Erwinia 및 Pseudomonas등의 세균과 Trichoderma, Colletotrichum 등 곰팡이의 성장을 동시에 억제하는 Bacillus sp. N 32 균주를 선발 동정 하였다. 특히 Bacillus sp. N32 균주는 고추 탄저병균인 Colletotrichum gloeosporioides에 대하여 열에 저항성이 있는 단백질과 열에 민감한 단백질의 2종류의 항균 단백질을 동시에 생산함을 단백질 침전과 활성 검정을 통하여 확인하였다. 이 항균 단백질들을 FPLC를 이용한 gel filtration chromatography방법으로 분리한 후 SDS-PAGE와 bioautography로 항균력을 확인하였다. 또한 이 항균 단백질의 유전자들을 선발하기 위하여 기존의 알려진 그람양성 세균의 대표적인 열 저항성 항균 펩타이드 생합성 유전자 서열을 primer로 이용한 PCR 방법으로 fengycin의 생합성 유전자 단편을 분리하고 이 PCR 산물을 이용하여 Bacillus sp. N32 균주의 cosmid library로부터 fengycin의 생합성 유전자 cluster중 일부를 분리하여 염기서열을 분석하였다. 또한 열에 민감한 항균 단백질 생산 유전자는 이 항균 단백질을 SDS-PAGE 및 electroblotting으로 분리한 뒤 N-terminal 부위의 15개의 아미노산 서열을 분석하고 이를 DNA 염기배열로 치환한 다음 probe로 이용하여 ${\lambda}-ZAP$ library로부터 항균 단백질 생산 유전자가 포함된 다수의 clone을 선발하였다.

  • Factor analysis is a multivariate technique for describing the in-terrelationship among many variables in terms of a few underlying but unobservable random variables called factors. There are various approaches for this factor analysis. In particular, principal factor analysis is one of the most popular methods. This follows the mathematical algorithm of the principal component analysis based on the singular value decomposition. But it is known that the singular value decomposition is not resistant, i.e., it is very sensitive to small changes in the input data. In this article, using the resistant singular value decomposition of Choi and Huh (1994), we derive a resistant principal factor analysis relatively little influenced by notable observations.

Analysis of Types and Characteristics of Clothing Lifestyle of the New Forty Generation

  • 본 연구는 30대에서 50대 남성 직장인 394명을 대상으로 의생활 라이프스타일을 유형화 한 후 유형별 특성을 분석하고자 하였다. 자료를 처리하기 위해 PASW 18.0 통계 프로그램을 이용하여, 빈도분석, k-평균 군집분석, 일원분산분석 및 교차분석을 실시하였다. 연구결과 첫째, 의생활 라이프스타일은 '패션 리더형(22.3%)', '가격 민감형(12.2%)', '패션 무관심형(27.9%)', '놈코어 패션형(37.6%)' 등 4개의 집단으로 유형화되었다. 둘째, 의생활 라이프스타일 유형별 사회경제적 특성에서는 연령, 결혼여부, 직업, 월평균 가계소득에서만 통계적으로 유의미한 차이가 나타났다. 셋째, 의생활 라이프스타일 유형별 의복 구매 행동에서는 월평균 외모 관리 비용, 정장 소유 여부, 월평균 의복 구입 비용, 평균 정장 구입 비용에서 통계적으로 차이를 보였다.
