Functional Gene Analysis for the Protection of Male Germ Cell Injury Induced by Busulfan Treatment using cDNA Microarray Analysis

  • Male germ cell apoptosis has been extensively explored in rodent. In contrast, very little is known about their susceptibility to apoptosis stimuli of developing germ cell stages at the time when germ cell depletion after busulfan treatment occurs. Furthermore, it is still unanswered how spermatogonial stem cells are resistant to busulfan treatment. We examined the change of gene expression in detail using cDNA microarray analysis of mouse testis treated with busulfan. A subtoxic dose of busulfan (40mg/kg of body weight) transiently increased 228 mRNA levels among of the 8000 genes analyzed. TagMan analysis confirmed that the mRNA levels such as defensive protein, support protein, enzymatic protein, transport protein, and hormonal protein were rapidly increased. These results were re-confirmed by real-time PCR analysis. However, the expression levels of these genes induced by busulfan treatment were significantly reduced in control testis, indicating that both of male germ cells and somatic cells after busulfan treatment induces self-defense mechanism for protection of testicular cell death. Among them, we conclude that defense proteins play a key role in testis injury induced by busulfan.

Tumor-derived CD4+CD25+ Tregs Inhibit the Maturation and Antigen-Presenting Function of Dendritic Cells

  • CD4+CD25+regulatory T cells (Tregs) play a key role in regulation of immnue response and maintenance of self-tolerance. Studies have found Tregs could suppress tumor-specific T cell-mediated immune response and promote cancer progression. Depletion of Tregs can enhance antitumor immunity. Dendritic cells (DCs) are professional antigen-presenting cells and capable of activating antigen-specific immune responses, which make them ideal candidate for cancer immunotherapy. Now various DC vaccines are considered as effective treatment for cancers. The aim of this study was to evaluate variation of Tregs in BALB/C mice with hepatocellular carcinoma and investigate the interaction between tumor-derived Tregs, effector T cells (Teff) and splenic DCs. We found the percentages of Tregs/CD4+ in the peripheral blood of tumor-bearing mice were higher than in normal mice. Tumor-derived Tregs diminished the up-regulation of costimulatory molecule expression on splenic DCs, even in the presence of Teff cells and simultaneously inhibited IL-12 and $TNF-{\alpha}$ secretion by DCs.

Elevated level of PLRG1 is critical for the proliferation and maintenance of genome stability of tumor cells

  • Pleiotropic regulator 1 (PLRG1), a highly conserved element in the spliceosome, can form a NineTeen Complex (NTC) with Prp19, SPF27, and CDC5L. This complex plays crucial roles in both pre-mRNA splicing and DNA repair processes. Here, we provide evidence that PLRG1 has a multifaceted impact on cancer cell proliferation. Comparing its expression levels in cancer and normal cells, we observed that PLRG1 was upregulated in various tumor tissues and cell lines. Knockdown of PLRG1 resulted in tumor-specific cell death. Depletion of PLRG1 had notable effects, including mitotic arrest, microtubule instability, endoplasmic reticulum (ER) stress, and accumulation of autophagy, ultimately culminating in apoptosis. Our results also demonstrated that PLRG1 downregulation contributed to DNA damage in cancer cells, which we confirmed through experimental validation as DNA repair impairment. Interestingly, when PLRG1 was decreased in normal cells, it induced G1 arrest as a self-protective mechanism, distinguishing it from effects observed in cancer cells. These results highlight multifaceted impacts of PLRG1 in cancer and underscore its potential as a novel anti-cancer strategy by selectively targeting cancer cells.

노인복지관 사회복지사의 긍정심리자본이 직무소진에 미치는 영향 요인에 관한 연구 (Study on the Factors Effecting the Positive Psychological Capital of Social Workers at Seniors Welfare Centers on Job Burnout)

  • 본 연구는 노인복지관 사회복지사의 긍정심리자본이 직무소진에 미치는 영향관계를 검증하고자 설계되었다. 조사대상은 서울특별시와 경기도에 소재한 노인복지관에서 노인을 직접 돌보는 사회복지사 350명을 대상으로 설문조사를 실시하였다. 회수된 설문지 중 최종 344부를 SPSS 22.0 통계프로그램을 사용하여 분석 처리하였다. 자료의 분석은 신뢰도 검사, 빈도분석, t-검증, 일원배치분산분석, 상관관계 분석, 다중회귀분석을 실시하였다. 연구결과, 긍정심리자본이 직무소진의 하위변수인 감정고갈에 통계적으로 유의미한 부(-)의 영향을 미치는 것으로 나타났고, 비인격화에 통계적으로 유의미한 부(-)의 영향을 미치는 것으로 나타났다. 긍정심리자본의 하위변수가 감정고갈과 비인격화에 미치는 영향은 낙관주의>회복력>희망>자기효능감으로 나타났다. 마지막으로 긍정심리자본이 성취감저하에 통계적으로 유의미한 부(-)의 영향을 미치는 것으로 나타났다. 긍정심리자본의 하위변수가 성취감저하에 미치는 영향은 낙관주의>희망>자기효능감>회복력으로 나타났다. 이러한 연구결과를 바탕으로 사회복지사의 직무소진을 감소시키는 요인으로 확인된 긍정심리자본 향상을 위한 제언과 연구의 한계점 및 향후 연구방향을 제시하였다.

감정노동 보육교직원의 심리적 소진과 자기 돌봄의 관한 사례연구 -노래심리치료- (A Case Study on Psychological Burnout and Self-care of Childcare Teachers for Emotional Labor -Song psychotherapy-)

  • 보육교직원은 보육현장에서 요구되는 긍정적인 이미지를 연출하기 위해 자신의 감정을 억압, 과장, 통제, 조절, 규제하는 감정노동을 경험한다. 영유아 대상의 근무환경은 그 특성상 감정노동 보육교직원의 심리적 소진 원인이 되며, 나아가서 삶의 질을 저하하는 문제점을 안고 있다. 노래는 인간의 삶의 질을 향상하는 데 도움을 주므로 감정노동 보육교직원에게 노래심리치료가 심리적 소진의 회복과 자기 돌봄 과정에서 긍정적인 삶을 계획할 수 있도록 하는 연구가 필요하다. 본 연구는 첫째, 노래심리치료를 통한 감정노동 보육교직원의 심리적 소진은 어떠한가? 둘째, 노래심리치료를 통한 감정노동 보육교직원의 자기 돌봄 과정은 어떠한가?를 연구문제로 설정하였다. 연구 시기는 2017년 3월부터 2020년 5월까지였으며, 질적사례연구로 프로그램 진행과 심층인터뷰, 직접관찰, 참여관찰 등의 자료 수집을 통해 ○○○ 어린이집에서 2019년 6월부터 10월까지 구조화된 회기별로 12회기에 걸쳐 매 회기 50분간 진행되었다. 이상의 연구 결과를 토대로 다음과 같은 결론이 도출되었다. 첫째, 노래심리치료를 통한 감정노동 보육교직원은 자신 삶의 의지와 심리적으로 소진된 자신을 발견하고 이해, 인식하며 내면의 문제들을 직면하고, 소통하며 통찰할 수 있었다. 타인에게서 받은 정서적 지원은 정서적 고갈의 경험을 줄일 수 있으며 업무에서의 좌절로 인한 경험의 회복과 성취감 향상을 증명하였다. 둘째, 노래심리치료를 통한 감정노동 보육교직원의 자기 돌봄은 시간적, 공간적, 관계적, 정서적 돌봄 과정을 증명하였고, 다른 사람을 돌보는 일과 자신을 돌보는 일의 균형을 유지하면서 신체와 마음, 영성이 유기적으로 통합되며 변화가 나타났다. 본 연구는 심리적 소진과 자기 돌봄 과정은 삶의 본질적인 의미를 발견하고 자신의 내면을 탐색 및 표현할 수 있도록 기회를 제공하고, 타인과 자신의 돌봄이 균형적으로 이루어지고 전인적인 건강한 자아를 위한 통찰, 성장을 통해 삶의 질이 향상하는 기회를 제공하는 데 의의를 둔다.

하수처리장 에너지 자립화를 위한 하수 에너지 잠재력 회수 기술 (Recovering the Energy Potential of Sewage as Approach to Energy Self-Sufficient Sewage Treatment)

  • Domestic sewage treatment plants (STPs) consume about 0.5 % of total electric energy produced annually, which is equivalent to 207.7 billion Korean won per year. To minimize the energy consumption and as a way of mitigating the depletion of energy sources, the sewage treatment strategy should be improved to the level of "energy positive". The core processes for the energy positive sewage treatment include A-stage for energy recovery and B-stage for energy-efficient nitrogen removal. The integrated process is known as the A/B-process. In A-stage, chemically enhanced primary treatment (CEPT) or high rate activated sludge (HRAS) processes can be utilized by modifying the primary settling in the first stage of sewage treatment. CEPT utilizes chemical coagulation and flocculation, while HRAS applies returned activated sludge for the efficient recovery of organic contents. The two processes showed organic recovery efficiencies ranging from 60 to 70 %. At a given recovery efficiency of 80 %, 17.3 % of energy potential ($1,398kJ/m^3$) is recovered through the anaerobic digestion and combustion of methane. Besides, anaerobic membrane bioreactor (AnMBR) can recover 85% of organic contents and generate $1,580kJ/m^3$ from the sewage. The recovered energy is equal to the amount of energy consumption by sewage treatment equipped with anaerobic ammonium oxidation (ANAMMOX)-based B-stage, $810{\sim}1,620kJ/m^3$. Therefore, it is possible to upgrade STPs as efficient as energy neutral. However, additional novel technologies, such as, fuel cell and co-digestion, should be applied to achieve "energy positive" sewage treatment.

CD4+CD25+ Regulatory T Cells Selectively Diminish Systemic Autoreactivity in Arthritic K/BxN Mice

  • Although the arthritis symptoms observed in the K/BxN model have been shown to be dependent on the functions of T and B cells specific to the self antigen glucose-6-phosphate isomerase, less is known about the in vivo roles of $CD4^{+}CD25^{+}$ regulatory T($T_{reg}$) cells in the pathology of K/BxN mice. We determined the quantitative and functional characteristics of the $T_{reg}$ cells in K/BxN mice. These mice contained a higher percentage of $Foxp3^+\;T_{reg}$ cells among the $CD4^+$ T cells than their BxN littermates. These $T_{reg}$ cells were anergic and efficiently suppressed the proliferation of $na\ddot{i}ve$ $CD4^+$ T cells and cytokine production by effector $CD4^+$ T cells in vitro. Antibody-mediated depletion of $CD25^+$ cells caused K/BxN mice to develop multi-organ inflammation and autoantibody production, while the symptoms of arthritis were not affected. These results demonstrate that despite the inability of the $T_{reg}$ cells to suppress arthritis development, they play a critical role protecting the arthritic mice from systemic expansion of autoimmunity.

정상 구강 점막 소견의 구강 통증 환자의 진단 및 치료 (Treatment and diagnosis of oral pain without identifiable oral mucosa lesion)

  • Objectives : Oral pain without identifiable oral mucosa lesion is probably multifactorial origin, which include burning mouth syndrome (BMS), oral candidiasis and so on. The aim of this study was to analyze the characteristics of oral pain without identifiable oral mucosa lesion and to evaluate treatment outcome of those patients. Materials and Methods : We reviewed 50 patients without identifiable oral mucosa lesion who were complaint of oral pain. The patients were analyzed according tothe sites, associated symptoms, laboratory tests and fungus culture. The questionnaire included questions on their current diseases, smoking and alcoholic history, psychological factors, and symptoms. Results : The average age of patients was 60 years old. The most frequently involved site was tongue (92%), followed by palate, lower lip, oropharynx, and gingiva. 60% of the patients has psychological disorder as self reported. Culture for Candida was positive in 36% of patients and serum zinc deficiency was present in 60% of patients. Serum iron, vitamin B12, hemoglobin, folic acid deficiency were present in 6-2% of patients. Seventeen patients (65%) with BMS and twelve patients (66%) with oral candidiasis were improved after treatment. Conclusion : We recommend oral candida culture to oral pain patients without oral mucosa lesion. Zinc supplementation of zinc depletion patients may be helpful whereas other laboratory tests have no diagnostic values.

자기 정렬된 Mo2N/Mo 게이트 MOSFET의 제조 및 특성 (fabrication of Self-Aligned Mo2N/MO-Gate MOSFET and Its Characteristics)

  • RMOS(refractors metal oxide semiconductor)의 게이트 금속으로 사용되는 Mo2N/Mo 이중층을 N2와 Ar을 혼합하여 저온의 반응성 스펏터링법으로 제조하였다. Ar : N2=95 : 5로 혼합된 가스 분위기에서 반응성 스펏터링을 할 때 Mo2N이 잘 형성되었다. 이렇게 제조한 Mo2N 박막은 면저항이 약 1.20∼1.28Ω/□로서 다결정 실리콘의 1/10정도가 되어 반도체 소자의 동작속도를 크게 향상시킬 것으로 기대된다. 1100℃의 N2분위기에서 PSC(phosphorus silicate glass)를 불순물 확산원으로 하여 소오스와 드레인의 불순물 확산을 할때 Mo2N 박막이 Mo으로 환원되어 확산전의 면저항보다 훨씬 작은 약 0.38Ω/□정도의 면저항을 나타내었다. 본 실험에서 제작한 자기정렬된 RMOSFET의 문턱전압은 약 -1.5V이고 결핍과 증가의 두 가지 동작특성을 나타내었다.

CFD를 이용한 표면 거칠기에 따른 선박의 저항 성능 추정에 관한 연구 (A Study on Predicting Ship Resistance Performance due to Surface Roughness Using CFD)

  • In recent, shipping companies have made an enormous effort to improve the operation of vessel in various approaches, due to recession of shipping market and increasing competition among shipping companies. One of important parameters for improving the efficiency of vessel is the resistance performance that consist of friction and residual resistance. Especially, it is recognized that the friction resistance tends to be affected by conditions of vessel’s surface and occupies approximately 70~90% of the total resistance for slow speed ships. In general, the surface of vessel is covered with various type of paint to reduce fouling and corrosion. As time goes by, however, it is so hull roughness would be increased by fouling over the wetted surface that anti-fouling paints, such as CDP(Controlled Depletion Paint), Tin-Free SPC(Self Polishing Co-polymer) or Foul Release, are applied evenly on the hull surface. Nevertheless, these anti-fouling paints could not prevent fouling absolutely. A fundamental study on evaluating ship resistance performance variation due to hull roughness has been performed using a commercial software, Star-CCM+, which solves the continuity and Navier-Stokes equations for incompressible and viscous flow. The results of present simulation for plate are compared with some experimental data available and the effect of surface roughness to ship resistance performance is discussed.
