Studies on the Evaluation of the Spent Composts of Selenium-Enriched Mushrooms as a Feed Selenium Source (셀레늄강화 버섯폐배지에 대한 사료 셀레늄공급원으로의 평가 연구)

  • This study was conducted to evaluate the spent composts of selenium-enriched mushrooms as a feed selenium Source. Total selenium (Se) contents and Se profiles in the spent mushroom composts (SMC) were determined. In addtion, we also investigated the metabolism in relation to Se accumulation in the mushroom. Mushrooms used in this study were Flammulina velutipes and Se enriched mushrooms were grown for 60 days by adding 2 mg of inorganic Se (Na2SeO3) per kg of mushroom composts (MC) on as-fed basis and it was compared with mushrooms not to add Se to the MC. Total Se contents for Se-treated mushrooms were significantly increased (P<0.0001) by 20-fold (4.51 ㎍/g of dry) compared to Se-untreated (0.23 ㎍/g of dry). On the contrary, organic Se proportion was significantly lower (P<0.0001) in the Se-treated mushroom (72.3%) than Se-untreated (100%, not analytically detected of inorganic Se). Se distribution upon a length in the Se-treated mushrooms was the highest in the bottom part (6.86 ㎍/g of dry) near to MC, and top and middle parts were significantly lower (3.71 and 3.01 ㎍/g of dry, respectively) than the bottom (P<0.001). In the SMC from Se-treated mushrooms, a high concentration of Se (5.04 ㎍/g of dry) was still remained, but that from Se-untreated mushrooms was significantly low (P<0.0001) as 0.08 ㎍/g of dry. Se-treated SMC showed a high rate of organic Se (65.67%), suggesting that most of inorganic Se in the SMC was converted to organic Se by mushroom mycelia, and Se-untreated SMC showed 100% of organic Se, not being detected of inorganic Se. Prior to mycelia inoculation in the mushroom culture, the sterilization of MC brought approximately 18% of Se loss in the MC. This result is in accordance with facts generally known that Se is weak in the high temperature and it is consequently volatilized under that condition. Apparent and net accumulation rates (%) for Se into mushrooms were 14.81 and 10.14%, respectively and their difference (4.67%) is considered that it is due to the volatilization into the air via metabolic process of mushroom itself. From the result of this study, inorganic Se addition to MC for mushroom improved the Se content in the mushroom and SMC from Se-enriched mushrooms contained a high concentration of Se. Mycelium and fruiting body from mushrooms converted inorganic Se in MC to organic Se, indicating a high proportion of organic Se in the mushroom and SMC. Therefore, Se in Se-enriched mushroom and SMC was recognized as Se sources of food for human as well as feed for livestock.

  • To deal with the potential XSS vulnerabilities in the source code of the power communication network, an XSS vulnerability detection method combining the static analysis method with the dynamic testing method is proposed. The static analysis method aims to analyze the structure and content of the source code. We construct a set of feature expressions to match malignant content and set a "variable conversion" method to analyze the data flow of the code that implements interactive functions. The static analysis method explores the vulnerabilities existing in the source code structure and code content. Dynamic testing aims to simulate network attacks to reflect whether there are vulnerabilities in web pages. We construct many attack vectors and implemented the test in the Selenium tool. Due to the combination of the two analysis methods, XSS vulnerability discovery research could be conducted from two aspects: "white-box testing" and "black-box testing". Tests show that this method can effectively detect XSS vulnerabilities in the source code of the power communication network.

Characterization and Identification of Organic Selenium-enriched Bacteria Isolated from Rumen Fluid and Hot Spring Water

  • In the present study, the isolation of selenium (Se)-enriched bacteria from rumen fluid and hot spring water was carried out. Rumen fluid samples were taken from cannulated goats fed a basal diet and the water samples were collected from Selayang hot spring, Selangor-Malaysia. A total number of 140 Se-tolerant isolates were obtained aerobically using an Se-enriched medium and spread plate technique. All the isolates were initially screened for the ability to transform the Se-containing medium to a red-orange culture using a spectrophotometer. Twenty isolates of dark red-orange medium were selected for a screening of the highest Se-containing protein accumulating strains using the dialysis technique and icp.ms to measure the Se content. Four isolates, identified as Enterobacter cloacae (ADS1, ADS7, and ADS11), and Klebsiella pneumoniae (ADS2) from rumen fluid origin, as well as, one isolate from hot spring water (Stenotrophomonas maltophilia (ADS18)), were associated with the highest biomass organic Se-containing protein when grown in a medium enriched with $10{\mu}g/ml$ sodium selenite. In addition, around $50{\mu}g/100{\mu}g$ of the absorbed inorganic Se was accumulated as an organic form. Organic Se-containing protein in all the selected strains showed antioxidant properties in the range of 0.306 to 0.353 Trolox equivalent antioxidant capacity (TEAC) mg/ml. Therefore, these strains may offer a potential source of organic Se due to their Se-tolerant nature and higher biomass organic to inorganic Se ratio.

  • Copper-indium diselenide, $CuInSe_2$, thin films have been fabricated by selenizing Cu/In stacked layers with different sputtered Cu/(Cu+ln) mole ratios at 450.deg. C for 1hr on alumina substrates. The selenium source was selenium vapor. Microstructure, crystallization, and composition of the selenized $CuInSe_2$ films were examined by using scanning electron microscope, X-ray diffraction, Auger electron spectroscopy, and secondary ion mass spectrometry. Electrical resistivity and hall effects were also measured to investigate the electrical properties. As the sputtered Cu/(Cu+In) mole ratio of In/Cu layer increased, the amounts of void and CuSe phase in the selenized films increased but the composition of $CuInSe_2$ phase was the same regardless of the sputtered mole ratio. Comparing the electrical properties of $CuInSe_2$ thin film before and after the chemical etching, it was seen that the electrical resistivity, carrier concentration, and carrier mobility of the selenized films were affected by the amount of CuSe phase which seemed to increase primarily the hole concentration of the selenized films.

  • Se is essential for a number of enzymes that perform important metabolic functions necessary for good health. However, people in many countries do not appear to consume adequate amounts of Se to support the maximal expression of the selenoproteins and Se retention in the body of animals and humans is dependent on the ingested Se source such as organic and inorganic Se. Therefore, this review was discussed to explore metabolic characterization regarding intestinal absorption, bioavailability and selenoprotein synthesis according to animal species such as monogastrics including human beings and ruminants. Generally, organic Se provided to animals is more effective than inorganic Se in body retention for the animal owing to the difference of manner for intestinal absorption. But, Se absorption in ruminants depending on its chemical form still remained questioned by several microbial actions and feeding regimen in the rumen. And Se absorbed through small intestine is utilized for the synthesis of selenoproteins and/or retained as selenoamino acids in the body. Retained Se in the body may be recycled to synthesize selenoproteins as lacked of dietary Se. In conclusion, desirable forms of Se ingestion in the animal may be useful for Se fortification in animal products as well as well being for humans and animals.

Smart Synthetic Path Search System for Prevention of Hazardous Chemical Accidents and Analysis of Reaction Risk (반응 위험성분석 및 사고방지를 위한 스마트 합성경로 탐색시스템)

  • There are frequent accidents by chemicals during laboratory experiments and pilot plant and reactor operations. It is necessary to find and comprehend relevant information to prevent accidents before starting synthesis experiments. In the process design stage, reaction information is also necessary to prevent runaway reactions. Although there are various sources available for synthesis information, including the Internet, it takes long time to search and is difficult to choose the right path because the substances used in each synthesis method are different. In order to solve these problems, we propose an intelligent synthetic path search system to help researchers shorten the search time for synthetic paths and identify hazardous intermediates that may exist on paths. The system proposed in this study automatically updates the database by collecting information existing on the Internet through Web scraping and crawling using Selenium, a Python package. Based on the depth-first search, the path search performs searches based on the target substance, distinguishes hazardous chemical grades and yields, etc., and suggests all synthetic paths within a defined limit of path steps. For the benefit of each research institution, researchers can register their private data and expand the database according to the format type. The system is being released as open source for free use. The system is expected to find a safer way and help prevent accidents by supporting researchers referring to the suggested paths.

Effect of Sodium Selenite and Zinc-L-selenomethionine on Performance and Selenium Concentrations in Eggs of Laying Hens

  • The objective of this study was to determine the effect of sodium selenite and zinc-L-selenomethionine on performance and egg Se concentration in laying hens. Two hundred and twenty-four CP Browns aged 71 weeks were divided according to a $2{\times}3$ factorial in a completely randomized design. One more group without additional Se supplementation was used as a negative control. Each treatment consisted of four replicates and each replicate contained eight laying hens. The dietary treatments were T1: basal diet; T2, T3 and T4: basal diets plus 0.3, 1.0 and 3.0 mg Se from sodium selenite/kg, respectively; T5, T6 and T7: basal diets plus 0.3, 1.0 and 3.0 mg Se from zinc-L-selenomethionine/kg, respectively. The findings revealed that feed conversion rate/kg eggs, egg production, egg weight, Haugh units and eggshell thickness were not affected by source and level of Se (p>0.05). Increasing level of dietary Se significantly increased (p<0.05 the Se content of eggs. Zinc-L-selenomethionine markedly increased p<0.05 egg Se concentration as compared with sodium selenite. The results indicated that Se source did not influence performance of laying hens. However, zinc-L-selenomethionine increased p<0.05 egg Se concentration more than sodium selenite.

Recent Development of Drinking Water Quality Standard and its Application (음용수질 기준과 관리방안)

  • Recently water demand is increasing as the industry prospers. The increase of water demand is followed by the increase of wastewater discharge which pollutes rivers and ground water extensively. These rivers, reservoirs and ground water are sources for drinking water and their contamination affects the quality of water supply and other potable water. In Korea there are 776 water treatment plants which supply drinking water from main rivers or reservoirs. Rivers are the biggest water source for drinking water is being contaminated, the innovation of treatment process is needed. The construction and operation of water supply facilities is under the control of the Ministry of Construction and the water supply offices of cities and provinces. However, drinking water quality is under the control of the bureau of sanitation in the Ministry of Health and Social Affairs. There are 33 items in drinking water quality standards of Korea. Trihalomethanes, Selenium, Diazinone and other three of pesticides have been included lately, The Ministry of Health and Social Affairs is planning to enhance. the level of $VOC_S$(Vola-tile Organic Compounds) standard. Drinking water quality standard is the goal to protect the quality of supply water and ground water. In order to protect the source water from domestic or industrial water, technological improvement and adequate investment should be urgently made. The ultimate goal of drinking water quality is safety and health of consumers. The more stringent the standard are, the better the water quality will be. As the drinking water quality standards become more stringent this year, various and positive solutions by the authorities concerned must be prepared.

Studies on Selenium-fortified Functional Hanwoo-Beef by Utilizing Spent Mushroom Composts II. Effects of Spent Composts of Se-Enriched Mushrooms as the Dietary Se Source on Selenium Deposition in the Muscular Tissue and Plasma Glutathione Peroxidase Activity in the Finishing Hanwoo Steer (버섯폐배지를 이용한 셀레늄강화 기능성 한우고기 생산에 관한 연구 II. 셀레늄강화 버섯폐배지 첨가가 한우의 근육조직 내 셀레늄 축적과 혈중 glutathione peroxidase(GSH-Px)활성에 미치는 영향)

  • This study was conducted to investigate effects of spent composts of Se-enriched mushrooms as the dietary Se source on muscular Se deposition and plasma glutathione peroxidase (GSH-Px) activity in the finishing Hanwoo steer. Twenty Hanwoo steers were used in the experiment and they were divided into four groups in a randomized complete block design with five replicates. Treatments were four levels (0.1, 0.3, 0.6 and 0.9ppm as fed basis) of dietary Se from the combination with spent composts of Se-enriched mushrooms and/or Se non-enriched mushrooms, in which each treatment was formulated with corn and corn gluten meal and so forth. Treatment diets were fed to Hanwoo steers for 90 days until the slaughter. Dry matter intakes had no significant differences among treatments and there were no significant differences for performances such as total BW gain and ADG among treatments. The Se concentration in blood was linearly increased with increasing dietary selenium levels and reached a plateau level after 8 weeks (p<0.001). Plasma GSH-Px activities had the similar trends to blood Se concentrations by showing that the increased dietary Se level significantly increased plasma GSH-Px activities of both total and Se-dependent (p<0.001). Muscle Se contents of Se-supplemented groups were linearly increased by 1.35 ~ 1.68 folds compared with the control group (0.1ppm; 0.273㎍/dry g) and especially those of the hind legs for 0.9ppm treatment showed the highest Se content as shown 0.457㎍ per dry gram (p<0.01) corresponding to approximately 70% increase of the control group. Se retention rate in the muscle of dietary Se originated from spent composts of Se-enriched mushrooms was estimated of maximum approximately 30% and dietary Se content showed the significant correlation with plasma GSH-Px activities and muscle Se contents (p<0.01). Accordingly, Se present in spent composts of Se-enriched mushroom as the dietary Se source not only had great bioavailabilities showing higher blood Se concentration and plasma GSH-Px activities, but also increased Se deposition in the muscle for Hanwoo beef cattle.

Effects of High Dietary Levels of Selenium-Enriched Yeast and Sodium Selenite on Macro and Micro Mineral Metabolism in Grower-Finisher Swine

  • Thirty six barrows with an initial body weight of 28 kg were used to determine the effect of two dietary Se sources and a wide range of Se levels encompassing 0.3, 1.0, 3.0, 5.0, 7.0, and 10.0 mg/kg Se. The organic Se form was a Se-enriched yeast product, whereas the inorganic Se source was sodium selenite. The experiment was a $2{\times}6$ RCB design conducted in three replicates. Each barrow was placed in an individual metabolism crate and provided their dietary treatment and water on an ad libitum basis for a minimum 2 wk period, whereupon feed intake was adjusted to a constant intake within replicate at approximately 90% of intake for a 4 d adjustment period. Urine and feces were subsequently collected for a 7 d period and analyzed for Se and minerals. The results demonstrated that urinary Se was approximately 25% higher when pigs were fed sodium selenite (p<0.01), whereas fecal Se was lower by 25% (p<0.01). Se retention tended to be higher when organic Se was provided (p>0.15). Urinary Se increased as dietary Se level increased for both Se sources but increased more and at a high rate when sodium selenite was fed resulting in an interaction response (p<0.01). Fecal Se increased linearly as the dietary level of both Se sources increased, but the fecal Se from organic Se increased at a faster rate resulting in an interaction response (p<0.01). Se retention increased linearly (p<0.01) as dietary Se increased for both Se sources. The apparent digestibility of Se increased by Se level when pigs were fed sodium selenite, but not when the organic Se source was provided resulting in an interaction response (p<0.05). Retention of consumed Ca, Zn increased when pigs were fed organic Se (p<0.05) whereas P and Na retention were higher when the inorganic Se was provided. Mineral retention was not affected by dietary Se level except P. These results suggest that Se excretion by urine was the main route of excretion when pigs were fed sodium selenite but the fecal route when Se-enriched yeast was provided. The excretion of Fe, Zn, Mn, and Cu via urine and feces was not affected by high dietary Se level or dietary Se sources.
