Studies on Growth and Biomass Production of Abies koreana Seedlings under Different Relative Light Intensity (상대광도 차이에 따른 구상나무 유묘의 생장과 물질생산에 관한 연구)

  • This study was carried out to investigate the growth and biomass production of Abies koreana seedlings under different relative light intensity. The 5-year-old potted seedlings were grown for 24 months under different light intensity of 100%, 50%, 30%, 10% and 2% of the natural full light intensity. The results were as follows: 1. Height growth was similar in the seedlings under different light intensities except in 2% of the full sun light intensity, having shown the lowest height growth. 2. Root collar diameter growth was reduced gradually by decreasing of the relative light intensity. 3. The seedlings which were grown under relatively low light intensity showed the later starting date of the stem growth in the next spring and showed the smaller number of new needles, and the smarter number and the shorter length of new shoot of the seedlings. 4. Total dry mass including the dry mass of leaves, shoot and root was reduced by decreasing of the relative light intensity. 5. In case of the seedling grown in one year shading condition, the ratio of the dry mass of current-year needles and stem to that of the above one-year-old needles and stem was ranged from 0.99$\pm$0.25 to 1.06$\pm$0.06 and was not significally different among the seedlings grown under 100%, 50% and 30% of the relative light intensity. And the radio was ranged from 0.60$\pm$0.04 to 0.52$\pm$0.03 in the seedlings grown under 10% and 2% of the relative light intensity, respectively. In case of the seedling grown in two years shading, the ratio of the dry mass of current-year needles and stem to that of the above one-year-old needles and stem was ranged from 0.79$\pm$0.04 to 1.00$\pm$0.03 and was not significally different among the seedlings grown under 100, 50, 30% and 10% of the relative light intensity. And the ratio was 0.29$\pm$0.03 in the seedlings grown under 2% of the relative light intensity, respectively. 6. T/R ratio of the seedlings grown in one year shading condition was ranged from 3.35$\pm$0.33 to 4.61$\pm$0.25 and was not significantly different among the relative light intensities of 100%, 50%, 30% and 10%. The seedlings grown under 2% of relative light intensity showed the highest T/R ratio of 7.36$\pm$0.15. In case of the two years shadings the T/R ratio was ranged from 1.76$\pm$0.11 to 3.12$\pm$0.19 and increased as relative light intensity decreased except 2.01$\pm$0.14 in 2% of relative light intensity.

Availability of Hairy Vetch as Leguminous Cover Crops in Citrus Orchards of Volcanic Ash Soils (화산회토 감귤원에서 헤어리베치의 이용 가능성)

  • In this study we evaluated the availability of hairy vetch in citrus (Citrus unshiu Marc.) orchards of volcanic ash soils. The responses to increasing seeding rates and various growing conditions such as altitude, accumulated temperature, and soil chemical properties etc, were analyzed by means of the seedling establishment rate, weed occurrence ratio, and shoot biomass yield of hairy vetch. Field experiments were conducted at five citrus orchards by altitude from Sep. 2015 to Apr. 2016 in Jeju Island, Republic of Korea. Hairy vetch used in the study was 'Cheongpyungbora', developed by National Institute of Crop Science. Seeding rates of hairy vetch consisted of 30, 60 and $90kg{\cdot}ha^{-1}$. Results showed that the seedling establishment rates of hairy vetch were quite similar regardless of seeding rates in all fields and weed occurrence ratio at 30, 60 and $90kg{\cdot}ha^{-1}$ of seeding rates were 11.8, 3.8, and 5.1% (dry wt.), respectively. Both 60 and $90kg{\cdot}ha^{-1}$ of seeding rates, the weed occurrences were decreased by 96.2% and 94.9%. The nitrogen production of hairy vetch at 30, 60 and $90kg{\cdot}ha^{-1}$ of seeding rates were 254, 316, and $315kg{\cdot}ha^{-1}$, respectively. Both 60 and $90kg{\cdot}ha^{-1}$ of seeding rates, The nitrogen production were increased by 24%, compared to $30kg{\cdot}ha^{-1}$ of seeding rate. In these results we were considered that the cost-efficient seeding rate of vetch was $60kg{\cdot}ha^{-1}$ in citrus orchards. Also, this study showed that the shoot biomass of hairy vetch and various cultivative factors were related and The nitrogen production of hairy vetch had a little bit of positive correlation (R=0.2714) with accumulated temperature and considerable correlations with some items (EC ($R=0.4520^{**}$) and exchangeable K ($R=0.4078^{**}$)) of soil chemical properties. Therefore, we were considered that hairy vetch can be used as a leguminous cover crop in citrus orchards, the calculation formula (Y=4.4097X + 33.594 (R=0.9547)) can be suggested for nitrogen yield of hairy vetch by using the shoot fresh weight (X).

Effects of the Mixing Ratio of the Different Substrates and the Concentration of Fertigation in Nutrient Solution on the Growth of Tomato Plug Seedlings (배지의 혼합비율과 관비 양액 농도가 토마토 플러그묘의 생장에 미치는 영향)

  • This study was conducted to clarify the effects of the different mixing ratios of substrate mixtures based on peat moss and the concentration of nutrient solution on the growth of tomato (Lycopersicon esculentum Mill.) seedlings. Substrates such as peat moss, rice hull, carbonized rice hull, decomposed sawdust, perlite and granular rock wool were mixed and used. The concentration of nutrient solution were adjusted to EC $0.5{\sim}1.5mS/cm$. The volumetric moisture contents became higher as peat moss mixed were much more. Total porosities in all substrate mixtures were over 80%, and pH in substrate mixtures became lower as the volume of peat moss mixed higher. Mixing ratios of substrates suitable for the production of tomato seedlings with the higher quality were peat moss:rice hull:carbonized rice hull:decomposed sawdust:perlite=25:10:25:20:20(v/v). The plant growth was not significant among the different substrate mixtures. However, plant growth such as plant height, leaf area, and total dry weight became significantly increased as EC increasing.

Characterization of Plant Growth-Promoting Traits of Free-Living Diazotrophic Bacteria and Their Inoculation Effects on Growth and Nitrogen Uptake of Crop Plants

  • The search for diverse plant growth-promoting (PGP) diazotrophic bacteria is gaining momentum as efforts are made to exploit them as biofertilizers for various economically important crops. In the present study, 17 diazotrophic strains belonging to eight different genera isolated from rice paddy fields were screened for multiple PGP traits and evaluated for their inoculation effects on canola and rice plants. All of the strains tested positive for 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity and production of indole 3-acetic acid (IAA) and ammonia ($NH_3$). Additionally, four of the strains were able to solubilize phosphorus (P), five tested positive for zinc (Zn) solubilization and sulfur (S) oxidation, and eight strains produced siderophores. Based on the presence of multiple PGP traits, 10 strains were selected for inoculation studies. Treatment with Herbaspirillum sp. RFNB26 resulted in maximum root length (54.3%), seedling vigor, and dry biomass in canola, whereas Paenibacillus sp. RFNB4 exhibited the lowest activity under gnotobiotic conditions. However, under pot culture conditions, Paenibacillus sp. RFNB4 significantly increased plant height and dry biomass production by 42.3% and 29.5%, respectively. Canola plants and rhizosphere soils inoculated with Bacillus sp. RFNB6 exhibited significantly higher nitrogenase activity. In greenhouse experiments, Serratia sp. RFNB18 increased rice plant height by 35.1%, Xanthomonas sp. RFNB24 enhanced biomass production by 84.6%, and rice rhizosphere soils inoculated with Herbaspirillum sp. RFNB26 exhibited the highest nitrogenase activity. Our findings indicate that most of the selected strains possess multiple PGP properties that significantly improve the growth parameters of the two plants when tested under controlled conditions.

  • Fusarium proliferatum KGL0401 was previously isolated from Physalis alkekengi var. francheti plant roots and exhibited higher GA productivity than wild type Gibberella fujikuroi. The :tim of this work was to find out an optimal culture condition for GA production. Various carbon(fructose, glucose, lactose, maltose, sucrose) and nitrogen($KNO_3$, urea, glycine, $NaNO_3,\;NH_4Cl$) sources were used for this study. GAs activities were analysed by gas chromatography and mass spectrometry(GC-MS). The highest yield of $GA_3$ was found in the growth medium supplemented with sucrose as carbon source and $NH_4Cl$ as nitrogen source. The optimum carbon-nitrogen concentration for $GA_3$ production was found to be 0.5 M:0.17 M. Supernatant was prepared from the culture fluid of F. proliferatum KGL0401 cultured for 7 days at 3 0'E and the 10 ul supernatant was treated with 2 leaf-rice seedling.

Isolation and Characterization of Phosphate Solubilizing Bacteria Pantoea Species as a Plant Growth Promoting Rhizobacteria (식물 생장 촉진 활성을 가진 인산분해 미생물 Pantoea 종의 분리 및 특성 규명)

  • Plant growth-promoting rhizobacteria (PGPR) have gained worldwide importance and acceptance due to their agricultural benefits. These microorganisms are potential tools for sustainable agriculture, with effects on plant growth, biofertilization, induced systemic resistance, and biocontrol of plant pathogens. In this study, four different Pantoea species were isolated from field soil, and their plant growth-promoting characteristics were studied. Based on 16S rDNA gene sequencing analyses, the se were grouped into Pantoea ananatis, Pantoea citrea, Pantoea dispersa, Pantoea vagans and named as Pa1, Pc1, Pd1, Pv1, respectively. All of these strains have their ability for solubilization of insoluble phosphate depending on pH decrease at the range around pH 5 at 1days after inoculation and production of plant hormone indole acetic acid (IAA) with 85.3±16.3 μg/ml of Pa1, 183.9±16.8 μg/ml of Pc1, 28.8±17.3 μg/ml of Pd1 and 114.1±16.5 μg/ml of Pv1, respectively. Pa1, Pc1 and Pd1 also have high activity for production of gibberellin (GA3) hormone with 331.1±19.2 μg/ml of Pa1, 288.5±16.8 μg/ml of Pc1, 309.2±18.2 μg/ml of Pd1, but Pv1 does not. Furthermore, all these species have significantly promoted the growth of the lettuce seedling plants at the range around 32~37% for fresh weight and 10~15% for shoot length enhancement, so that these microbe could be used as a potential bio-fertilizer agents.

Induction and in vitro Proliferation of Adventitious Roots in Phyllanthus urinaria (여우구슬(Phyllanthus urinaria)의 부정근 유도 및 기내증식조건)

  • Phyllanthus urinaria was an important species in Korea and distributed in all around of Korea. The roots and stems of this plant have been used for natural medicine for the treatment of diabetes, the hepatitis B virus and disturbances of the kidney and urinary bladder. Production of adventitious roots in P. urinaria by in vitro cultures could be used as alternatives materials. Shoot and root segments from P. urinaria seedling were cultured on Murashige and Skoog (MS) medium supplemented with 3.0 mg/L IBA and 30 g/L sucrose. After 4 weeks of culture, the highest induction of adventitious roots was obtained from the shoot part. Frequency of adventitious root formation on medium with various kinds of auxins (IAA, NAA, 2,4-D, and IBA) and various concentrations of IBA (0, 0.1, 0.5, 1.0, 3.0, and 5.0 mg/L) was tested. The maximun induction of adventitious root was obtained on medium with 0.5 mg/L IBA. In liquid culture, growth of root was best on medium supplemented with 30 g/L sucrose. Adventitious roots were cultured in 5 L bioreactor containing 1/2 MS medium supplemented with 0.5 mg/L IBA and 30 g/L sucrose and mass-production of adventitious roots was successfully achieved. These results revealed the first attempt for the production of adventitious roots in P. urinaria.

Production and Transplanting of Ectomycorrhizal Pine Seedlings Using the Old Fairy Ring of Tricholoma matsutake (기존 송이 균환(菌環)을 이용한 송이균 감염 소나무의 생산 및 이식)

  • To make a new fairy ring of Tricholoma matsutake in situ, the way of production and transplanting of ectomycorrhizal seedlings of T. matsutake using Pinus densiflora was investigated after transplanting from 2000 to 2005 as well as the method to improve their survival rate for the fungus. For the production of ectomycorrhizal pine seedlings, the seedlings planted at the edge of fairy ring of T. matsutake in November showed 97% of the survival rate, while those planted in April showed 80% of the rate. For the complete infection of the T. matsutake, it required more than two years after planting. The infection rate of mycelia for the ectomycorrhizal seedlings was 17.6% when the natural seedlings were used, whereas it was relatively low when the seedlings prepared from the nursery were used. The survival of T. matsutake mycelium reached up to 22% by the transplanting in April, while the mycelium transplanted in October and November showed less than 5% of the survival. The survival of T. matsutake on the transplanted seedlings was the highest in the seedlings having 50% of infection rate before transplanting. Excavation of the ectomycorrhizal seedling to examine the vitality of ectomycorrhizal roots of T. matsutake resulted in the perishing of them. Therefore, the method using a 'rice bag triers' to check living mycelium of T. matsutake without digging of transplanted seedlings was introduced in this study. In addition, it is recommended that the examination has to be conducted at least two years after transplanting.

Study on Flowering, Pollination and Samara Characteristics of Chinese elm, Ulmus parvifolia in Wonju, Korea (참느릅나무의 개화, 수분 및 결실 특성에 관한 연구)

  • To examine the reason of empty samara production of Chinese elm, Ulmus parviflora, twenty two planted trees in Wonju-si were monitered for three years in terms of their flowering, pollinatiom system and samara characteristics. Inflorescences with bisexual flowers of Chinese elm are developed in the leaf axils on the twigs. Dichogamous flowers are varied with protogynuous and protandrous flower, and stamens in some bisexual flowers are developed in seperated time on a inflorescence or a tree. It is revealed newly that the flower of Chinese elm is out-crossed and partially insect(Apis mellifera) pollinated. The ratios of sound samara are significantly differed among years, the heighest values 65.5% were shown in 2009, lowered 42.9% in 2010, and the lowest 37.5% in 2011. This result might be affected by mean daily precipitation and number of rainy days during the flowering date, and lower temperature during the floral initiation stage, especially in 2011. These findings suggest that Chinese elm has self-incompatibility strategy and much pollination failure resulted in a production of much empty samaras. Further researches on the empty-seed production strategies and pollination system of major tree species might be needed.

Characteristics of mulberry cultivar 'palchung' (Morus Lhou(Ser.) Koidz.) for mulberry breeding as pollen parent (오디 생산용 뽕 품종 육성을 위한 교배모본 '팔청'의 특성)

  • We bred a mulberry cultivar named Palcheong, through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at four places (Suwon, Kongju, Wanju, and Jinju) for seven years from 2007. This is belonging to (Morus Lhou(Ser.) Koidz.) selected among mulberry genetic resources preserved in Suwon. Its sex expression was hermaphrodite, which can be used for the production of seedling for fruit production by artificial crossing as pollen parent Palchung showed lower yielding in fruit productivity by 16% compared to control cultivar 'Chungil (Morus alba L.)' for five years. Although Palchung was higher in sugar content of mulberry fruits, it was bigger than that of 'Chungil'. It is adaptable to every where except the places where cold damage and sclerotic disease happen frequently.
