Transmission of Apple scar skin viroid by Grafting, Using Contaminated Pruning Equipment, and Planting Infected Seeds

  • Apple scar skin, one of the most destructive diseases affecting apple, is caused by Apple scar skin viroid (ASSV d). Fruit dappling appeared on several cultivars in Korea and has been distributed to major cultivated areas since 2001. ASSVd was identified from infected fruits by using nucleic acid sequence-based amplification with electrochemiluminescence (NASBA-ECL). NASBA-ECL method was faster and hundredfold more sensitive than reverse transcription-polymerase chain reaction (RT-PCR) for ASSVd detection in apple leaves/ stems. ASSVd was rapidly transmitted to the entire tree in the second year after artificial inoculation. The ASSVd could be transmitted efficiently by using contaminated pruning scissors to both lignified stems (60 to $70\%$) and green shoots (20 to $40\%$) of apple tree and young plants. Dipping of contaminated scissors in $2\%$ sodium hypochlorite solution effectively prevented viroid transmission. In the ASSV d-infected fruits, the viroid was easily detected from fruit skin, seed coat, and embryo. Moreover, embryo and endosperm separately excised from the ASSVd-infected seeds were ASSVd positive in NASBA-ECL assay. Seedlings germinated from ASSVd-positive seeds showed $7.7\%$ infection rate., which indicated that ASSVd is seed-borne.

Identification of Fungal Pathogen Causing Seedling Rot of Lithospermum erythrorhizon and Study on the Optimum Growing Temperature for Decreasing of the Seedling Rot (지치 육묘시 발생하는 유묘썩음증상 원인균 분리·동정 및 경감 육묘환경에 관한 연구)

  • This study was conducted to isolate and identify the fungal pathogen causing seedling rot of Lithospermum erythrorhizon Siebold & Zuccarini, and to know the optimum growing temperature for decreasing seedling rot of Lithospermum erythrorhizon. On the basis of morphological characteristics, EF-1a sequence analysis, and pathogenecity to host plant, the fungi isolated from seedling rot and seeds of Lithospermum erythrorhizon were identified as Fusarium fujikuroi, indicating that disease causing fungus is seed-borne pathogen. Optimum temperature for germination of seeds of Lithospermum erythrorhizon was $15{\sim}20^{\circ}C$, but pathogenicity of Fusarium fujikuroi was shown more readily at $25{\sim}30^{\circ}C$. These results suggested that seedling culture of Lithospermum erythrorhizon between $15^{\circ}C$ and $20^{\circ}C$ might reduce seedling rot of Lithospermum erythrorhizon caused by seed-borne pathogen Fusarium fujikuroi.

Studies on the Seed Transmission of Colletotrichum spp. in Red Pepper (Capsicum annuum) (고추 탄저병균(炭疽病菌)의 종자전염(種子傳染)에 관(關)한 연구(硏究))

  • Colletotrichum acutatum, C. coccodes, C. dematium and C. gloeosporioides were detected in seed samples of red pepper (Capsicum annuum). C. dematium and C. gloeosporioides were the predominant species, maximum seed infection of the species in some samples were 84% and 28%, respectively. C. acutatum and C. coccodes were recorded only in low percentages of 1-2. The blotter method proved more suitable for detecting Colletotricum spp. than the deep freezing blotter or agar plate methods. Plating of seed components showed that C. dematium and C. gloeosporioides were recovered more frequently from seed coat, and decreasing amounts of infection were observed in the endosperm and cotyledon. Seed-borne C. dematium and C. gloeosporioides caused seed rot, damping-off, seedling blight and brown discoloration of cotyledon and hypocotyl when infected seeds were sown in agar of test tube or in soil. Inoculation experiments showed that C. acutatum was pathogenic to red fruit of red pepper and C. coccodes was highly pathogenic to red fruit and weakly pathogenic to leaf of the plant. C. dematium was highly pathogenic to leaf and green fruit and C. gloeosporioides was pathogenic to not only leaf but also green and red fruits. Host range of the four seed-borne species of Colletotrichum was also investigated.

Effects of Seed-treatment Fungicides on Bakanae Disease of Rice

  • Bakanae disease, caused by Gibberella fujikuroi (anamorph Fusarium moniliforme J. Sheldon), a typical seed-borne disease of rice occurs from nursery to paddy fields. Consequently, chemical seed disinfectants is the most efficient control method. Several seed treatment methods with various fungicides were attempted to inhibit disease. Spray and 24 hrs immersion of seeds using prochloraz emulsion reduced disease infection and the control value were 99.3 and 100%, respectively. In contrast, dressing to wet seeds thiophanate-methyl+thiram wp and benomyl+thiram wp reduced disease infection more effectively than 24 hrs immersion of seeds. However, dressing of carpropamid+imidacloprid+fludioxonil wp to wet seeds did not reduced disease as well as wettable liquid of fludioxonil. The results suggest that the bakanae disease might be disinfected effectively by 24 hrs immersion of seeds in prochloraz emulsion and seed dressing of fungicides.

  • Disease is the major problem in ginseng cultivation from seed stratification, soil preparation prior to planting, right through to drying of the roots. There are many soil-borne disease pathogen in rhizosphere soil environment, furthermore occurrence of diseases by a diverse group of fungi and related organisms are closely related to various soil condition. Observable symptoms for soil-borne diseases include wilting, leaf death and leaf fall, death of branches and limbs and in severe cases death of the whole plant. The fungus Cylindrocarpon destructans is the cause of root rot characterized by a decay of the true root system in many ginseng production areas in Korea. Some pathogens are generally confined to the juvenile roots whilst others are capable of attacking older parts of the root system. However, the relation between the soil environmental characteristics and ginseng root rot by soil-borne disease pathogen is not clearly identified in ginseng field. In this paper, we reviewed soil-borne plant pathogen causing root rot disease of ginseng with respect to soil environment.

Past and Future Epidemiological Perspectives and Integrated Management of Rice Bakanae in Korea

  • In the past, rice bakanae was considered an endemic disease that did not cause significant losses in Korea; however, the disease has recently become a serious threat due to climate change, changes in farming practices, and the emergence of fungicide-resistant strains. Since the bakanae outbreak in 2006, its incidence has gradually decreased due to the application of effective control measures such as hot water immersion methods and seed disinfectants. However, in 2013, a marked increase in bakanae incidence was observed, causing problems for rice farmers. Therefore, in this review, we present the potential risks from climate change based on an epidemiological understanding of the pathogen, host plant, and environment, which are the key elements influencing the incidence of bakanae. In addition, disease management options to reduce the disease pressure of bakanae below the economic threshold level are investigated, with a specific focus on resistant varieties, as well as chemical, biological, cultural, and physical control methods. Lastly, as more effective countermeasures to bakanae, we propose an integrated disease management option that combines different control methods, including advanced imaging technologies such as remote sensing. In this review, we revisit and examine bakanae, a traditional seed-borne fungal disease that has not gained considerable attention in the agricultural history of Korea. Based on the understanding of the present significance and anticipated risks of the disease, the findings of this study are expected to provide useful information for the establishment of an effective response strategy to bakanae in the era of climate change.

Antimicrobial Activity of Grapefruit Seed Extracts and Polylysine Mixture Against Food-borne Pathogens (자몽종자추출물과 폴리리신혼합물의 식품부패균에 대한 항균효과)

  • This study was conducted to investigate the antimicrobial activity of grapefruit extracts and polylysine mixture against food-borne pathogens. The mixture was showed a potent and quick antibacterial activity for 5 major bacteria causing food poisoning i.e. Escherichia coli, Escherichia coli O-157, Salmonella typhi, Staphylococcus aureus, Vibrio cholerae. The antibacterial effect of the mixture on the ordinary bacteria inhibiting on the surface of lettuce was lasted even 6 hrs after the treatment, however the mixture was non-effective on the color, smell and taste of lettuce. The treatment with 10% mixture solution of the foods such as fish, meat, rice and bread suppressed the growth of the bacteria and kept the foods more freshly than the untreated foods.

Development of On-line Sorting System for Detection of Infected Seed Potatoes Using Visible Near-Infrared Transmittance Spectral Technique (가시광 및 근적외선 투과분광법을 이용한 감염 씨감자 온라인 선별시스템 개발)

  • In this study, an online seed potato sorting system using a visible and near infrared (40 1100 nm) transmittance spectral technique and statistical model was evaluated for the nondestructive determination of infected and sound seed potatoes. Seed potatoes that had been artificially infected with Pectobacterium atrosepticum, which is known to cause a soil borne disease infection, were prepared for the experiments. After acquiring transmittance spectra from sound and infected seed potatoes, a determination algorithm for detecting infected seed potatoes was developed using the partial least square discriminant analysis method. The coefficient of determination($R^2_p$) of the prediction model was 0.943, and the classification accuracy was above 99% (n = 80) for discriminating diseased seed potatoes from sound ones. This online sorting system has good potential for developing a technique to detect agricultural products that are infected and contaminated by pathogens.

Application Cases of Test Construction of Hydro-Seeding Measures on the Slopes Along the National Road Between Gamchon and Yaechon (감천예천구간 국도비탈면 종자뿜어붙이기 시험시공 적용사례)

  • To protect the surface of ground-cutting slopes and ground-raised slopes and to recover environment-friendly slopes at the national road expansion construction between Gamchon and Yaechon, test-construction work was done using several modes of hydro-seeding measures to select a suitable construction method through the field survey; the results were as follows : As a result of survey on physio-chemical characteristics of soil, the average soil inclination on ground-raised slopes was from 4.73 mm to 5.37 mm; the average soil acidity was from pH 6.47 to 6.73; the average soil humidity was within the scope of 0.57~1.70 %,; the average soil inclination on ground-cutting slopes was 9.17~10.43mm; the average soil acidity was pH 6.67~6.77 and the average soil humidity was within the scope of 0.53~1.27 %; considered overall, they showed generally satisfactory base materials for plant breeding. As a result of the number of sprouting individuals, the average sprouting number of individuals on test spots executed by Coir net with Seed spray construction methods and the average number of sprouting individuals on other test spots executed by furrow-digging with Seed spray and Straw mat-mulching measures were 1,172 number of per square meter and 970 number of per square meter respectively; thus they showed higher numerical value comparing with other test spots. Likewise, the average number of sprouting individuals on test spots of the ground-cutting slopes executed by Coir net with Seed spray method and the average number of sprouting individuals on other test spot executed by furrow-digger with Seed spray and Straw mat-mulching measures were found to be 1,107 number of per square meter and 1,105 number of per square meter respectively, whose numerical value showed rather higher other test spots. As a result of a survey on living plants' breeding index [dried weight], the weight in dried state on test spots executed by Coir net with Seed spray method and other test spots executed by furrow-digger with Seed spray and Straw mat-mulching measures on both ground-raised and ground-cutting slopes was found to be higher and showed similar characteristics statistically. As a result of survey on the surface-covering degree of ground-raised slopes, nine weeks after test construction, test spots executed by Coir net with Seed spray method and other test spots executed by furrow-digger with Seed spray and Straw mat-mulching measures showed commonly more than 70% of surface-covering degree, but other test spots showed only below 35% of surface covering degree; especially in case of test spots by Verdvol seed spray measures, they showed 10% of the lowest surface-covering. Surface-covering degree on the test spot of ground-cutting slopes nine weeks after test construction showed more than 75% both executed by Coir net with Seed spray construction method and executed by furrow-digger with Seed spray and Straw mat-mulching measures. As a result of survey on appearing plants on the test spots, there dominates Eragrostis curvula both on ground-raised slopes and on ground-cutting slopes with other seeding plant life's lower breeding and there appear intruders, such as Setaria viridis Beauv, Digitaria sanguinalis Scop, and Chenopodium album var.centrorubrum Makino. As for water-borne excavation and soil's washing-away on the slopes, there happened less washing-away and water-borne excavation with good breeding of plant life on test spots executed both by Coir net with Seed spray construction method and by furrow-digger with Seed spray and Straw mat-mulching measures, but there happened much more soil's washing-away along with water-home excavation at the test spot executed by Seed spray with measures. After the research results of test construction sites are examined overall, it's presumed that furrow-digger with Seed spray and Straw mat-mulching measures [interval between lines is 40cm, 5cm in depth] will be preferred to prevent erosion and water-home excavation of slopes located within the construction sites.
