Alterations in Seed Vigour and Viability of Soybean Related with Accelerated Seed Aging and Low Dose Gamma Irradiation

  • The objective of this study was to demonstrate whether or not the deleterious effects of accelerated aging on seed vigour and viability are alleviated by interaction with gamma irradiation. Seeds of soybean (Glycine max L.) were artificially aged and subsequently irradiated with 4 and 8 Gy of gamma irradiation. Germination rate was negatively affected by accelerated aging and positively by gamma irradiation, with a positive interaction of a 3day-seed aging treatment occurring with 4 Gy, possibly suggesting that 4 Gy of gamma irradiation partially offset the adverse effects of seed aging on germination. However, 5-day aged seeds did not gain any benefits from the gamma irradiation. Electrolyte leakage from the seeds increased with the duration in days aged. Irradiation, however, did not impose any effects on the leakage. Respiration rate of the seed with hypocotyl and primary root was significantly low for the aged seeds, but not for the seeds with both irradiation and aging treatments. Accelerated aging decreased the dry weight of the hypocotyl and primary root of the seeds without any measurable effects of irradiation. $\alpha$-Amylase activity decreased with seed aging and positively responded to gamma irradiation. The data is discussed with regard to the possible roles of gamma irradiation for improving the seed vigour and viability of aged seeds.

Effects of Feeding Extruded Soybean, Ground Canola Seed and Whole Cottonseed on Ruminal Fermentation, Performance and Milk Fatty Acid Profile in Early Lactation Dairy Cows

  • Four ruminally cannulated Holstein cows averaging 43 days in milk (DIM) were used in a $4{\times}4$ Latin square to determine the effect of feeding extruded soybean, ground canola seed and whole cottonseed on ruminal fermentation and milk fatty acid profile. One hundred and twenty lactating Holstein cows, 58 (${\pm}31$) DIM, were assigned to four treatments in a completely randomized block design to study the effects of the three types of oilseeds on production parameters and milk fatty acid profile. The four diets were a control diet (CON) and three diets in which 10% extruded soybean (ESB), 5% ground canola seed (GCS) and 10% whole cottonseed (WCS) were included, respectively. Diets consisted of concentrate mix, corn silage and Chinese wild rye and were balanced to similar concentrations of CP, NDF and ADF. Ruminal fermentation results showed that ruminal fermentation parameters, dry matter intake and milk yield were not significantly affected by treatments. However, compared with the control, feeding cows with the three oilseed diets reduced C14:0 and C16:0 and elevated C18:0 and C18:1 concentrations in milk, and feeding ESB increased C18:2 and cis9, trans11 conjugated linoleic acid (CLA). Production results showed that feeding ESB tended to increase actual milk yield (30.85 kg/d vs. 29.29 kg/d) and significantly decreased milk fat percentage (3.53% vs. 4.06%) compared with CON. Milk protein (3.41%) and solid non-fat (13.27%) from cows fed WCS were significantly higher than from cows fed CON (3.24% and 12.63%, respectively). Milk urea N concentrations from cows fed the ESB (164.12 mg/L) and GCS (169.91 mg/L) were higher than cows fed CON (132.31 mg/L). However, intake of DM, 4% fat corrected milk, energy corrected milk, milk fat and protein yields, milk lactose percentage and yield, somatic cell count and body condition score were not affected by different treatments. The proportion of medium-chain fatty acid with 14 to 16 C units in milk was greatly decreased in cows fed ESB, GCS and WCS. Feeding ESB increased the concentration in milk of C18:1, C18:2, C18:3 and cis9, trans11-CLA content by 16.67%, 37.36%, 95.24%, 72.22%, respectively, feeding GCS improved C18:0 and C18:1 by 17.41% and 33.28%, respectively, and feeding WCS increased C18:0 by 31.01% compared with feeding CON. Both ruminal fermentation and production trial results indicated that supplementation of extruded soybean, ground canola seed and whole cottonseed could elevate the desirable poly- and monounsaturated fatty acid and decrease the medium chain fatty acid and saturated fatty acid content of milk fat without negative effects on ruminal fermentation and lactation performance.

수침(水浸)과 Acetic Acid 처리에 따른 콩나물의 생장과 형태 변화 (Effect of Seed Imbibition into Water and Acetic Acid Solution on its Floating Rate and Growth of Soybean Sprouts)

  • 콩나물의 생산과정에서 야기되고 있는 부패를 방지하기 위한 다양한 방법이 제시되고 있다. 본 연구는 물에 침종하는 수침(水浸)을 이용한 종자정선과 0.1% acetic acid에 2분간 실시한 종자소독이 콩나물의 발아와 생장, 형태 및 생산수율에 미치는 영향을 조사하고자 실시하였던 바 그 결과를 요약하면 다음과 같다. 1. 물에 뜨는 부유 종자의 비율은 5분간 물에 침종시킨 종자를 건져 40분간 aeration 시켜 다시 물에 침종시킬 경우 가장 높았다. 2. 상품으로 판매가 가능한 하배축 길이가 4cm이상의 비율은 은하콩에서는 水浸(水浸)과 acetic acid 처리간 차이가 없었던 반면, 풍산 나물콩과 준저리에서는 acetic acid 처리에 비하여 가결 수침 처리(水浸 處理)에서 높았다. 3. 세근형성 비율과 개체당 세근수는 은하콩과 준저리에서는 수침 처리(水浸 處理)에서, 풍산나물콩은 acetic acid 처리에서 상대적으로 높고 많았다. 4. 개체당 전체생계중은 은하콩과 풍산나물콩에서는 수침 처리(水浸 處理) 보다는 acetic acid 처리에서 많았던 반면. 준저리는 이와 반대의 결과를 보였다. 그러나 준저리를 이용한 대량생산 체계에서의 생산 수율은 수침(水浸)을 이용한 종자정선과 acetic acid를 이용한 종자소독간에 차이가 없었다.

회화나무 종자발아에 미치는 전처리의 효과 (Effects of Several Pre-treatments on Seed Germination or Sophora japonica L.)

  • 본 연구는 경립종자로서 종피가 단단하고 두꺼워 발아시 어려움이 있는 회화나무 종자를 여러 가지 전처리를 통해 발아율 및 발아지수를 향상시키고자 실시하였다. 종피 파상처리로서 황산 30, 60, 90% 용액에 30분간 침지하였고, 종자의 유근 반대편 부위를 1/8, 2/8, 3/8씩 절단하였다. 또한 종자를 젖은 수건에 싸서 밀봉 후 $4^{\circ}C$ 저온고에 각각 3, 5, 10일간 보관하였으며, $90{\sim}95^{\circ}C$ 뜨거운 물에 각각 2, 5, 10분간 침지하였다. 전처리된 종자는 각각 다른 발아특성을 나타내었다. 황산 처리된 종자는 발아율 및 발아지수에서 무처리구 보다 높게 나타났다. 특히, 황산 90%처리에서 가장 높은 효과를 보여주었다. 종자 절단 처리에서는 절단 길이가 늘어날수록 발아율 및 발아지수가 감소하였으며 종자의 냉층적 처리에서는 차이를 보이지 않았다(p=0.258). 열탕 처리시 2분 침지 종자는 무처리 종자보다 발아율 및 발아지수가 낮게 나타났으며 5, 10분 침지 종자는 발아하지 않았다. 전처리가 종자 발아뿐만 아니라 유묘활력에도 영향을 미치는지 알아보고자 전처리 종자로부터 생장한 유묘를 대상으로 상대생장율 및 T/R율을 조사한 결과 황산 90% 처리구의 유묘에서 가장 뛰어난 수고 및 근원경의 상대생장율과 T/R율을 관찰 할수 있었다. 위의 결과로서 황산 90% 용액에 30분간 침지처리를 했을 때 회화나무 종자의 발아율, 발아지수를 비롯한 유묘의 상대생장율 및 T/R율이 높게 나타남을 알 수 있었다.

Effects of Annatto (Bixa orellana L.) Seeds Powder on Physicochemical Properties, Antioxidant and Antimicrobial Activities of Pork Patties during Refrigerated Storage

  • This study investigated the effect of the powder produced by ball-milling the outer layer of annatto (Bixa orellana L.) seeds on the physicochemical properties as well as the antioxidant and antimicrobial activities of pork patties over 14 d of refrigerated storage (4±1℃). Five pork patty treatments were produced containing three different concentrations of annatto seeds, 0.1, 0.25 and 0.5% (ANT0.1, ANT0.25, ANT0.5), 0.1% ascorbic acid (AA0.1), and a control (CTL). Based on the results, annatto seed powder appeared to show antioxidant activity. The Hunter color values of pork patties were affected by the addition of annatto seed powder, which increased the redness and yellowness values, but decreased the lightness of the patties (p<0.05). To evaluate the antioxidative effects of annatto on pork patties, thiobarbituric acid reactive substances (TBARS) and peroxide values (POV) were analyzed over 14 d of refrigerated storage. Treatments containing annatto seed showed lower TBARS and POV than control (CTL) samples (p<0.05). The volatile basic nitrogen (VBN) of the pork patties containing annatto seeds were lower than that of CTL at the end of storage (p<0.05). Although no differences in total bacterial counts were observed between control and treated patties, those containing annatto seeds had lower microbial counts for Enterobacteriacease than CTL or AA 0.1%. Therefore, annatto seed powder might be a good source of natural antioxidants for the production of meat products.

Germination and Biochemical Changes in Accelerated Aged and Osmoprimed Pinus thunbergii Seeds

  • The aim of this study was to investigate relationship among seed viability and enzymes activities involved in scavenging reactive oxygen species (ROS), especially, superoxide dismutase (SOD), glutathione reductase (GR), and catalase (CAT). In other respects, osmopriming has been demonstrated to reinvigorate aged seeds. Various viabilities of seeds that were ranged from 80 to 100% of germination rate could be produced using osmopriming and accelerated aging treatments. Priming treatment of Pinus thunbergii seeds for 3 days at $15^{\circ}C$ with a polyethylene glycol solution at -1.2 MPa improved their subsequent germination at $25^{\circ}C$. Accelerated aging (3, 6, 9, and 12 days at $41^{\circ}C$ and 100% relative humidity) decreased seed germination percentage depending on aging treatment duration. Electrolyte conductivities of seeds were measured as assay of membrane integrity. The conductivity from electrolyte leakage of P. thunbergii seed was also correlated with seed germinability. Conductivity for control seeds that had 95% of germination percentage was 3.48 ${\mu}S\;g^{-1}$, but jumped as doubled (7.98 ${\mu}S\;g^{-1}$) in 12-day-aged seed that had 80% of germination percentage. Our results demonstrate that aging of P. thunbergii seeds is associated with changes in the electrolyte leakage, lipid peroxidation, and antioxidant defense system. Priming of aged seeds progressively restored the initial germinative ability and resulted in a marked decrease in the levels of MDA and conductivity of seed leachate. These effects of priming were also well recovered of GR and CAT activities in aged seed. The improved seed quality by priming treatment appears at least partly attributable to reduced lipid peroxidation, resulting from enhanced antioxidative enzyme activities that are suggesting the antioxidant defense systems play a key role in seed vigor.

Enhancement of Seed Germination and Uniformity in Triploid Watermelon (Citrullus lanatus (Thunb.) Matsum. and Nakai)

  • One of the main factors restricting production of triploid seedless watermelon is poor germination due to weak embryos, thick seed coats, and larger air spaces. This study was carried out to investigate the priming effects of different concentrations of chemicals, including hydrogen peroxide ($H_2O_2$), fusicoccin, and gibberellic acid (GA) on germination and seedling uniformity of triploid watermelon (Citrullus lanatus). Three commercial triploid cultivars, Seedless Plus, Sinus, and Sizero, were pretreated with water and different levels of $H_2O_2$ (2 and 4%), fusicoccin (FC: 1, 5, and $10{\mu}M$), and GA (1, 5, and $10{\mu}M$). The present findings helped to find optimal priming conditions for improving germination of triploid watermelon. Treatment with $5{\mu}M$ GA and hydropriming helped to break seed dormancy, enhancing the final germination percentages in all triploid cultivars and increasing the germination index in Sizero. These seed-priming treatments could be used on large scale for industrial applications. Moreover, hydropriming provides a simple, effective, and costless method to improve seed germination and seedling vigor of Sinus and Sizero varieties.

Optimization of in vitro seed germination of Taraxacum platycarpum

  • Dandelion (Taraxacum platycarpum) has been widely utilized for medicinal purposes. However, the dandelion seeds are relatively difficult to germinate under cultivation conditions, which hampers seedling propagation of dandelion plants and reduces the opportunity of usage of such a useful medicinal plant. Thus, in this study, in vitro conditions for the dandelion seed germination were optimized to enhance the germination rate. In seed washing steps, the sequential treatments with 20% of ethanol, 20% of NaOCl, and distilled water avoided microbial contamination with the highest in vitro germination rate (67.5%) from seeds sown in germination media. The media supplemented with 1.4 g/L of MS salts and 1% of sucrose significantly enhanced the germination rate compared to the media with 4.4 g/L of MS and 3% of sucrose. Sowing the seeds vertically in the optimized media supplement conditions, 1.4 g/L of MS salts and 1% of sucrose, gave the maximum in vitro germination rate (61%), which was almost three times higher than sowing seeds on a soil pot (23%). Our results indicate that the seed washing and sowing methods including germination medium supplements can be optimized to enhance in vitro seed germination of dandelion.

Changes in Functional Constituents of Grape (Vilis vinifera) Seed by Different Heat Pretreatments

  • Changes in functional constituents of grape (Vitis vinifera) seeds prepared by three different heat pretreatments were determined and compared with those of non-treated grape seed. The recovery of grape seed oils was generally increased by roasting, steaming and microwave processes, although the recovery of specific constituents varied among three heat pretreatments. The recovery of MeOH extracts of the seeds increased following the roasting process, whereas that of MeOH extracts decreased gradually with steaming and microwave treatments. Levels of four catechins in grape seeds: (+)-catechin, procyanidin B$_2$, (-)-epicatechin, and (-)-epicatechin gallate, were decreased with increased roasting and steaming time, but were unaffected by microwave treatment. During the three different heat pretreatments, levels and compositions of fatty acid did not change, whereas those of phytosterol compositions decreased greatly. These results suggest that a mild heat pretreatment, controlled for temperature and time, is needed to prevent a considerable loss in the level of valuable functional components in grape seed.

Interactive Effect of Nitrogen and Sulphur on Yield and Quality of Groundnut (Arachis hypogea L.)

  • Randomized field experiments were conducted to study the interactive effect of sulphur (S) and nitrogen (N) on seed, oil and protein yield of two cultivars of groundnut {Arachis hypogea: cv Amber $(V_{1})$: cv Kaushal, $(V_{2})$.} Two dosage levels of sulphur ($0\;and\;20kg\;ha^{-1}$) and two dosage levels of N ($23.5\;and\;43.5kg\;ha^{-1}$) in various combinations were tested as micronutrient treatments, $T_{1},\;T_{2},\;and\;T_{3}$. Results indicated significant enhancement of the yield components namely seed and oil yield as well as seed protein. Maximum response was observed with treatment $T_{3}$(having 20kg S and 43.5kg N $ha^{-1})$. Increase in seed and oil yields of 90% and 103% in $V_{1}$, and 79 and 90% in $V_{2}$, respectively were recorded as compared to the control treatment $T_{1}$(having 0kg S and 23.5kg N $ha^{-1}$). Effect of S and N interaction was observed on protein, N and S content in seeds. The results obtained by these experiments clearly suggest that judicious balanced application of N and S could improve the yield.
