Effect of Soaking and Prechilling Treatment on Seed Germination of Phellodendron amurense Rupr. (침수 및 예냉처리가 황벽나무의 종자 발아에 미치는 영향)

  • The seeds of Phellodendron amurense have difficulty to obtain because these plants show dioecism with independent distributions. This experiment was conducted to find the effects of soaking and prechilling treatment on the germination of P. amurense seeds. Seeds were soaked for 3, 5 and 10 days (soaking treatment), and placed on petri-dish at $4^{\circ}C$ for 10, 20, 30 and 50 days after soaked at room temperature for 24 hours (prechilling treatment). After the treatments, germination percentage, mean germination time, germination speed and germination performance index were analyzed. As a result of sowing, prechilled seeds for 30 days had the highest percent of germination, whereas all of the soaked seeds showed lower percent of germination than non-treatment (control). In case of mean germination time, only prechilling for 30 days was more effective than control. In germination speed, all of the soaked seeds had lower values than control whereas all prechilled seeds except prechilled seeds for 20 days showed higher values than control. Especially prechilled seeds for 30 days showed the highest rate among the prechilling treatments. Germination performance index was similar to germination speed.

A Study on the Wild Exochorda serratifolia for Landscape Horticuitural Cultivation (야생 가침박탈(Exochorda serratifolia)의 조경원예화에 관한 연구)

  • Exochorda serratifolia, broad-leaved shrub has beautiful flowers and is resistant to cold, shade and decease. New this native plant is considered to be worth being exploited as the outstanding plant for landscaping and horticulture. So this study was executed to utilize Exochorda serratifolia as the planting material for landscaping and horticulture through the survey of its habitat environment and the experimennt of its seed physiology and germination, vegetative propagation, culture and utilization, etc.. The results are as follows ; 1. The color of the flower is white, blooming in the early and middle of May and the seeds ripen late in Sep.. 2. The elevation, elevation, gradient and direction of the native habitat were 250m, 20-25$^{\circ}$and northern side respectively. 3. The soil pH of the natural habitat was 5.3 and soil fertility was poor. 4. Exochorda serratifolia appeared as indicator within P. densiflora community and its neighboring species were Q. mongolica, Q. dentata, R. mucronulatum, L. obtusiloba, E. oxyphillus, C. heterolphylla, var thunbergii, etc.. 5. The optimum temperature for seed germination was found at 20$^{\circ}C$ and the longer the stratification period at 5$^{\circ}C$ was, the lower the germination rate was. 6. The treat merits of GA and Kinetin increased the seed germination rate, especially under the dark condition but their high concentrations decreased the seed germination rate. 7. The rooted rate of the greenwood cutting was the highest at IBA 100ppm plot of vermiculite bed but its high concentration decreased the rooted rate conspicuously. 8. In the tissue culture, the each 1.0 ppd plot of NAA, Kinetin and NAA 1.0ppm + Kinetin showed the best growth. And the mixture of NAA 1.0ppm and Kinetin showed better growth than the single treatment of NAA or Kinetin did. 9. Transplanted Exochorda serratifolia showed healthy growth with shaded environmental condition(42.1% light intensity), therefore they can be cultivated as a shade tolerant landscape plant. 10. It was considered that Exochorda serratifolia was applicable to group planting at shade places or under trees in parks, homes, etc..

A Study on Amylase Activities During Germination of Maize Seeds (옥수수의 발아과정중(發芽過程中) AMYLASE활성(活性))

  • Changes of starch, soluble sugar and activities of amylase in germinating maize seeds were studied with comparison of germinating barley seeds. The starch in the endosperm was degraded slowly during the course of germination, and starch degradation of maize seeds compared with barley seed was found after 2 or 4 days of seed germination. There was a transient decline of soluble sugar come-its in ungerminated seed at early stages, followed by the rapid increase which concided with the degradation of starch. Activities of amylases were increased with the progress of germination, and maximum activities were founded after 8 days of seed germination. The higher activities of ${\alpha}$- and ${\beta}$-amylase were found at Hwang-ok maize and barley, respectively.

Autotoxicity of alfalfa flower extract and its allelopathy to Echinochloa crus-galli (알팔파 꽃 추출물의 Autotoxicity와 돌피에 대한 Allelopathy)

  • The aim of this study was to separate or purify some bioactive compounds from flowers of alfalfa(Medicago sativa L.) and to test of the isolated compounds on alfalfa for their autotoxicity and on Echinochloa crus-galli for their allelopathy for seed germination and seedling weight. Using thin layer chromatography(TLC) of $CHCl_3$ extracts, the most inhibitory band to alfalfa seed germination was determined. Germination inhibition of this extract suggested a complex chemical interaction. Separation and purification of compounds with CHCl$_3$ extract of fresh alfalfa flowers were conducted by a silica gel TLC, and microcrystalline cellulose TLC(MCTLC), followed by droplet countercurrent chromatography(DCCC) bioassay. Preliminary identification was done by high perfomance liquid chromatography(HPLC) on the most inhibitory fractions in DCCC. Ferulic acid, caffeic acid, vanillic acid, rutin, narringin were identified in fraction 5 and ferulic acid, caffeic acid, vanillic acid, rutin, coumarin in fraction 6. The phytotoxicity of their individual compound was tested on alfalfa and Echinochloa crus-galli seed germination and seedling weight. Coumarin and ferulic acid showed the most inhibitory effect on alfalfa seed germination and Echinochloa crus-galli seedling fresh and dry weight. These compounds may be, at least in part, involved in autotoxicity and allelopathy.

A Study on the Efficient Germination of Barley Seed using Electrostatic Field (정전기장을 이용한 보리종자의 효율적 발아에 관한 연구)

  • This paper discusses methods for increasing the germination rate of barley seeds using low direct and alternating current power below 110V. The experimental apparatus used here is a parallel plate, with the bottom surface of the plate designed to be wider than the top surface to increase the size of the electrostatic field. As a result, three different magnitudes of electrostatic fields were created on the plates: the first ranging from 400V/cm to 600V/cm, the second from 600V/cm to 900V/cm, and the third from 2200V/cm to 2400V/cm. The finite difference method was applied to analyze the electrostatic field inside the parallel plate. The plant seeds used in the experiments were barley seeds produced domestically. The average germination rate of barley seeds using the presented electrostatic field in this paper was 57%, while it was 65% when using a microwave of 2.45GHz, compared to a control group with a result of 31%. An important difference between using the electrostatic field and the 2.45GHz microwave is the dry method and wet method. When applying these two methods to practical seed germination, it is necessary to consider the advantages and disadvantages of each experimental approach and choose the appropriate method accordingly.

  • This experiment was carried out in order to determine the optimum harvesting time of tetraploid Italian ryegrass. Moisture content, 1000-seed weight, germination and seed shattering were investigated at six harvesting times after anthesis in 1984 at Suweon. The results are summarized as follows; 1. Seed moisture content was slowly decreased until 24 days and fastly decreased from 24 days after anthesis. The daily mean moisture loss during the ripening period was about 1.5 percentage. 2. The maximum 1000-seed weight was reached at 24 days after anthesis and 1000-seed weight of Waseking was heavier than that of Giant. 3. Shattering began at 17-20 days after anthesis and thereafter rapidly increased. 4. Germination was 50% at 10 days after anthesis and maximum germination was reached at 24 days. 5. Considering germination, 1000-seed weight and shattering, the optimum harvest time was 24 to 27 days after anthesis. At this time, moisture content was 55.0 to 43.5 percent and two to five seeds per ten spikes were shed by shaking at the vertical point.

Seed and Germination Characteristics by the Seed Coat Colors of Three Species of Genus Pinus (소나무류 3수종의 종피색깔에 따른 종자 및 발아특성 비교)

  • This study was carried out to increase germination by classifying seeds as seed coat colors of three species of genus Pinus. As result, the seeds classified as colors showed a difference. In seed length, Pinus densiflora (Pd) and P. thunbergii (Pt) had no difference while P. rigitaeda (Pr) had a difference between light yellow seeds and dark brown seeds (p<0.05). Only Pt was different in seed width. And in 1,000 seeds weight the three species showed a great difference while were not significantly different in seed moisture contents. In percent germination, all of the three species had a difference, especially that of Pr was very wide. Pd and Pt were not different in mean germination time but Pr showed a difference between light yellow seeds and dark brown seeds.

Effects of Bird Ingestion on Seed Dispersal and Germination of the Elaeagnus macrophylla (보리밥나무(Elaeagnus macrophylla)의 종자 산포와 발아율에 미치는 조류의 영향)

  • The Elaeagnus macrophylla is a stenoecious evergreen plant with nitrogen-fixing symbionts and its timing of fruit-ripening coincides with spring migration of many birds in southwestern Korea. To recognize bird species which eat fleshy fruits of the Elaeagnus macrophylla and to evaluate the effects of bird ingestion on seed germination and dispersal, we monitored birds and carried out germination experiments using its fruits and seeds from March to April 2007 at Hongdo Island, Jeonnam Province, Korea. As a result, eight species of birds including the Gray Starling (Sturnus cineraceus), the Brown-eared Bulbul (Hypsipetes amaurotis) and the Dusky Thrush (Turdus naumanni) ingested the fruits. Germination rate was enhanced but length of seed dormancy was instead shortened in ingested (collected from the feces of birds) and manually extracted seeds (obtained directly from the fruits) than in intact fruits collected directly from the plant. Moreover, the possible scale of seed dispersal by the frugivorous birds ranged up to 6.9 ha based on home ranges of starlings. Consequently, the Elaeagnus macrophylla supplies food resources for migratory birds, and the birds give the plant opportunities of new colonization. We suggest that this kind of interaction between the nitrogen-fixing plant and avian seed dispersers is applicable as a process of natural restoration in degraded coastal evergreen forests.

Change of Seed Dormancy at Different Storage Conditions in Rice (벼 저장조건에 따른 종실의 휴면성 변화)

  • This study was conducted to investigate change of seed dormancy during storage in rice. The result of germination rate observed at different storage duration after harvest indicated that two Japonica rice varieties, Sangpungbyeo and Hwaseongbyeo, tow sare type local varieties, Jakwangdo and Mongeunsare, and a Tongil type rice, Samgangbyeo, showed no dormancy in rice seed harvested at 40 days after heading, However, weak dormancy was found in matured rice seed of Sobaekbyeo and Odaebyeo, and strong seed dormancy was found in Jungwonbyeo, IR-20 and IR-42. Rice seed stored for 30 days after harvest at 2$0^{\circ}C$ constant temperature or 25/15$^{\circ}C$ of day /night temperature fluctuation showed higher germination rate above 90% in all varieties tested. But germination rate of seeds stored at 4$^{\circ}C$ for 30 to 180 days showed varietal difference, higher in rice varieties having weak seed dormancy and lower in strong dormancy varieties. Days to 80% germination and average days for germination were shortened by storage for 30 to 180 days after harvest at 2$0^{\circ}C$, 4$^{\circ}C$ with RH 40% or day /night 25/15$^{\circ}C$ temperature condition in all rice varieties tested, but the degree of acceleration was lower at the condition of 4$^{\circ}C$ with RH 40%.

Seed Yields and Germination Rates of Native Ecotype Collections for the Development of High-Yield Seeded Variety of Zoysiagrass in Korea (다수확 종자형 품종 육성을 위한 자생 한국잔디 수집계통들의 종자 수확량과 발아율)

  • Seeded variety of zoysiagrass has not been bred yet in Korea. Development of seeded zoysiagrass cultivar will be very important for the growth of turfgrass industry internationally as well as domestically. This research was conducted to investigate seed yield and germinability of 102 collected native zoysiagrass ecotypes in South Korea. Two hundred and seventy seven ecotypes were collected from various locations including coastal and mountain areas, while 102 morphologically distinct and seed producing ecotypes were selected and planted in $1m{\times}1m$ maintenance plots. Seed yield ranged from 0.1 to $32.2g\;m^{-2}$. Highest yielding line was a medium leaf type zoysiagrass of Z6011 with $32.2g\;m^{-2}$. Most collected lines showed seed germination rates of below 50%. However, Z2095 showed highest germination rate of 78%. Considering germination rate and seed yield, collected lines of Z6011, Z 6015, Z1075, ZN1008, and Z1084, which were mostly medium leaf type and Z. japonica types, showed reasonably high potential to be used as breeding lines for high yield seed varieties of zoysiagrass.
