Ecklonia cava is popular in Korea as a marine functional materials. E. cava is generally collected and used on the coast of Jeju Island. However, the continuous use of collected natural E. cava may be limited because difficult to secure throughout the year and may be exposed to environmental pollution. Jeju magma seawater (MSW) was known to be significant advantages such as safety, cleanness, stability, and functional improvement. Attempts have been reported on application of MSW to the culturing of macro- and microalgae and showed improved results. Thus, the objective of the present study was to explore the anti-melanogenesis activity of brown seaweed E. cava (E. cava cultured with MSW [MSWE]) extract cultured in tanks with MSW of Jeju Island to evaluate the possibility of cosmeceutical industrial application. MSWE extract showed the higher polyphenolic and dieckol contents than natural E. cava (NE) extract. Anti-melanogenesis activity of MSWE extract and NE extract are tested and compared using tyrosinase and dihydroxyphenylalanine (DOPA) oxidation inhibition assay. MSWE extracts evidenced more effective tyrosinase and DOPA oxidation inhibition activity than that of the NE extracts and the commercial whitening agent, arbutin. MSWE extracts also markedly inhibited melanin synthesis and decreased the expression of melanogenesis-related protein in ${\alpha}$-melanocyte stimulating hormone-stimulated B16F10 melanoma cells without cytotoxicity. These results suggest that MSW cultivation process would be more effective in releasing bioactive compounds with whitening effect from seaweed such as E. cava at an industrial scale.

In this study, we analyzed the compositional characteristics of Melosira nummuloides mass-cultured using Jeju lava seawater. M. nummuloides showed the highest growth rate when cultured for 14 days at 17-20℃ and 15,000 lx. Proximate composition of raw-material (RM) and freeze-dried M. nummuloides (FM) showed high ash content (65-72%), while ethanol-extracted M. nummuloides (EM) had low ash and high lipid contents. The predominantly occurring mineral, Si, was 334 g/kg in RM and 269 g/kg in FM, but EM contained only 1.97 g/kg. The ratio of essential amino acids was similar in RM (38.93%) and FM (36.89%) lower in EM (17.83%), but branched chain amino acids required for muscle metabolism was high (63.40%). The polyunsaturated fatty acids of EM (34.74%) was 11% more than that in RM (23.81%), and the ratio of omega-6 to omega-3 fatty acids was 1:3.6. Furthermore, the concentration of total chlorophyll was higher by 5.15 times (62.32 mg/g) and that of fucoxanthin by 7.06 times (11.02 mg/g) in EM than in RM (12.11, 1.56 mg/g, respectively). The mass cultivation and filtration of M. nummuloides using lava seawater has high prospects of commercialization as a competitive bio silica, cosmetic and healthy functional food material.

The objective of the present study was carried out to clarify the effects of deep sea water on the growth and maturation of $Porphyra$ (Rhodophyta, Bangiales). Foliose thalli for indoor culture were collected from Yeongok ($P.$$okamurae$) in Gangwon Prefecture, Tongyeong ($P.$$suborbiculata$ f. $latifolia$) and Namhae ($P.$$yezoensis$ f. $narawaensis$) in Gyongnam Prefecture respectively. Monospores were cultured at five temperatures (5, 10, 15, 20 and $25^{\circ}C$) with a photon irradiance of $80{\mu}m^{-2}s^{-1}$ under photoperiods of 14L:10D and 10L:14D in surface, deep and mixed seawater in respectively. The fast growth of foliose thalli were observed in $P.$$suborbiculata$ f. $latifolia$ cultured at deep seawater under $15^{\circ}C$ and 10L:14D. In three species, the optimum growth occurred at 10 and $15^{\circ}C$ under deep and mixed seawater and short day-length. In general, monospores from the cultured thalli were liberated within three weeks after incubation under $10-25^{\circ}C$ and both photoperiods. From the result of this study, deep seawater is considered that the natural species of the genus $Porphyra$ can be useful for the development as the new cultivars.
We used seawater temperature data, measured in the Garolim Bay, to analyze temperature variation on an hourly and daily basis. Lagrange's interpolation using before and after data was applied to restore nonconsecutive missing temperature data. The estimated error of the data restoration was 0.11℃. Spectral analyses of seawater temperature showed significant periodicities of approximately 12.4 h (semidiurnal tide) and 15.0 d (long-period tide), which is close to those of M2 and Mf partial tides. Variation in seawater temperature was correlated more with tidal height than with air temperature around the Garolim Bay. In June and December, when the seawater temperature difference between the inside and outside of the Garolim Bay was very large, the periodicities of 12.4 h and 15.0 d were highly prominent. These results indicate that the exchange of seawater between the inside and outside of the Garolim Bay induced variations in seawater temperature owing to tide. Understanding temperature variation because of tide helps to prevent abnormal mortality of cultured fish and to predict seawater temperature in the Garolim Bay.
The objective of this study was to evaluate food value and profitability of black porgy (Acanthopagrus schlegeli) cultured in freshwater. In fish flesh, muscular hardness of black porgy reared in freshwater $(9210{\pm}1215g/cm^2)$ was slightly lower than those of fish in seawater $(9987{\pm}6549g/cm^2)$, but there was no significant difference. Also, there was no difference between muscular strength of fish reared in freshwater and seawater. When the flesh qualities of black porgy reared in freshwater was compared with those of fish reared in seawater through the questionnaire, there were no significant differences between fish reared in freshwater and seawater in appearance, texture, taste and flavor. For 10 months of black porgy culture in fresh water, the gross profit in culturing from juvenile (5.5 g) to adult size (100g), and from adult to marketable size (400 g) were 24,000,000 won (30.0%) and 53,870,000 won (36.9%), respectively.

Deep seawater (DSW) refers to water extracted from the ocean, usually at depths of 200 meters or more, which is rich in inorganic materials and has attracted attention for various applications. We investigated the effects of the DSW on the synaptic maturation of cultured rat hippocampal neurons. Immunocytochemical examination of DIV21 showed that PSD-95, $\alpha$CaMKII, and synGAP$\alpha1$clusters were strengthened and coupling rates of SV2 and NR2B were significantly increased in neurons grown in the presence of H-800 and H-1000 DSW. Our results indicate that DSW promotes the formation of excitatory postsynaptic signal transduction complexes NRC/MASC and functional synapses.
Deep seawater (DSW; deep ocean water) is pure, rich in inorganic materials which have attracted attention for various applications. In this study we investigated the effects of the DSW upwelled from the East Sea, offshore Yang Yang (Korea) on the morphological differentiation of cultured rat hippocampal neurons, which were grown in the minimal essential medium containing 10% (v/v) fetal bovine serum and 25% (v/v) DSW with various hardness. DSW had no effect on initial morphological differentiation (17 hr post-plating). When observed on DIV3, 7, 14, and 17, low hardness (0 and 200) DSW reduced dendritic branching. However, dendritic branches within $80\;{\mu}m$ diameter from the center of soma nearly doubled in neurons grown in hardness 1,000 DSW-containing media. DSW with hardness 600 was more or less same as control groups. These results indicate that DSW with appropriate hardness ameliorates neuronal health.

Fish bacterial diseases have spread and caused serious problem for cultured marine fish in Korea. The important bacterial disease affecting mariculture such as olive flounder (Paralichthys olivaceus) are caused by Edwardsiella tarda, Vibrio scophthalmi and Streptococcus parauberis. For the bacterial disease protection in aquaculture industry, the water treatment is needed in aquaculture system. During the last decades atmospheric pressure non-thermal plasma in contact with liquids have received a lot of attention of environmental and medical application. In this study, we determined the disinfectant effect in seawater and three major fish bacterial disease pathogens by using low frequency plasma treatment. Three fish bacteria (E. tarda, V. schophthalmi, S. parauberis) were not detected within 16 min, 150 min and 270 min of 20 L, 500 L and 1 ton seawater post low frequency plasma treatment, respectively. Three major fish bacterial disease pathogens were not detected within 2 min after the low frequency plasma treatment, suggesting that the low frequency plasma possess disinfectant effectiveness.
The purpose of this study is to evaluate the performance of forward osmosis (FO) system for harvesting microalgae cultivated in secondary sewage effluent. Microalgae species used in this study were chlorella sp. ADE4. The drawing agents used for forward osmosis system were seawater and concentrate of sea water reverse osmosis (SWRO) system. Chlorella sp. ADE4 cultured in secondary sewage effluent illustrated moderate efficiency in removal of total nitrogen (TN) (68%) and superior performance in total phosphorus (TP) removal (99%). Comparison of seawater and SWRO concentrate as drawing agent were made in FO membrane separation of the microalgae. The result from this study depicts that SWRO concentrate is strong drawing agent in FO membrane system providing an average dewatering rate of $4.8L/(m^2{\cdot}hr)$ compared to seawater with average dewatering of $2.9L/(m^2{\cdot}hr)$. Results obtained from this study indicated that FO system could be viable option for harvesting the microalgae for further biodiesel production. SWRO concentrate as a drawing agent could be very important finding in field of membrane technology for disposal of SWRO concentrate.
Ground seawater in the east area of the volcanic Jeju Island contains abundant minerals. We investigated the metabolic activity of electrodialyzed, desalted ground seawater (EDSW) from Jeju in both cultured cells and animals. The addition of EDSW to the culture medium (up to 20%, v/v) reduced the leakage of lactate dehydrogenase and increased MTT activity in CHO-IR cells. EDSW (10%) promoted insulin-induced glucose consumption in L6 muscle cells as well as the activities of the liver ethanol-metabolizing enzymes, alcohol dehydrogenase and aldehyde dehydrogenase. Moreover, EDSW suppressed palmitate-induced intracellular fat accumulation in human hepatoma $HepG_2$ cells. Activities of AMP-stimulated protein kinase and acetyl CoA carboxylase, enzymes that modulate fat metabolism, were altered by EDSW in $HepG_2$ cells toward the suppression of intracellular lipid accumulation. EDSW also suppressed hepatic fat accumulation induced by a high-fat diet in mice. Taken together, EDSW showed beneficial metabolic effects, including the enhancement of ethanol metabolism and insulin-induced glucose consumption, and the suppression of intrahepatic fat accumulation.
