Tissue concentrations of quercitrin in spotted sea bass (Lateolabrax maculatus) after extended feeding with fish mint (Houttuynia cordata) extract (어성초 (Houttuynia cordata) 추출물을 장기간 투여한 점농어 (Lateolabrax maculatus)에서 조직내 quercitrin 잔류 농도)

  • The Houttuynia cordata has been utilized for various beneficial purposes in humans mainly because of its potent antioxidant principle quercitrin present in this plant. This study examines the possibility of producing a functional sea food commodity containing active principle quercitrin by feeding H. cordata for a extended period. Spotted sea bass (Lateolabrax maculatus) were fed a diet containing H. cordata at 0.1-1.0% levels for 1 month and tissue concentrations of quercitrin were analyzed in serum, hepatopancreas and muscle. It was observed that quercitrin was found in the ranking order of hepatopancreas>muscle>serum. After a bolus administration of quercitrin (20 mg/kg, oral) to spotted sea bass and Nile tilapia (Oreochromis niloticus), idential rank order was observed after 48 hr. In contrast, the order was liver>serum>muscle in rat and mice, indicating that higher quercitrin distribution occurs to the muscle in fishes compared with in mammals tested. High residue concentration of qeurcitrin in the edible tissue can be an advantageous property in terms of functional food production. High level H. cordata extract inclusion of 1.0% seems to have detrimental effects in spotted sea bass leading to growth retardation and hepatic damage. It was concluded that incorporation of H. cordata extract into diet can be a way of producing healthy foods. However the level of active extract needs fine tuning to avoid toxicity to fishes.

Genetic Characterization of Two Types of Sea Bass, Lateolabrax japonicus in Korea by Isozyme Analysis (Isozyme 분석에 의한 한국산 농어, Lateolabrax japonicus 2형간의 유전학적 특징)

  • Genetic characterization and identification of two types of sea bass Lateolabrax japonicus were performed by examining electrophoretic patterns of isozymes. Twenty five loci coding for thirteen enzymes were detected in two types. Among the twenty five loci, one completely divergent loci (Pt-1) was observed between two types. Nei's genetic distance between two types was 0.12036. The estimated divergence time of these two types may have about $6.2{\times}10^5$ years ago. On the other hands, the expected average heterozygosity was 0.084 in not spotted type on the body surface and 0.067 in spotted type on the body surface. These results mean that the existance of two types of sea bass was established in present study which may have had genetic divergence.

  • The stress of consecutive blood collectings resulted in evident elevation of plasma glucose level and significant lowering of chemiluminescent response of peripheral blood phagocytes in sea bass (Lateolabrax japonicus). Fish responded to the consecutive stressors in cumulative manners. The plasma glucose level in response to consecutive stressors depended on the stressor intervals. When the plasma glucose level of individual fish was compared with the chemiluminescent response, statistically significant (P<0.05) negative correlations existed.

Effect of Raw Fish-Based Moist Pellet (MP) and Commercial Red Sea Bream Feed (CF) on Growth and Body Composition of Sunshine Bass (M. saxatilis male X M. chrysops female) Reared at Various Salinity During the Winter Season

  • Possibility of raising sunshine bass during the winter season in Korea was investigated. Also, the effect of feed on growth and body composition of sunshine bass was compared at the various salinity. Twenty sunshine bass $(Mean\;weight\;\pm S.D.\: :\;53.9 \pm 0.24 g)$ were stocked into the eighteen circular flow-through tanks. A 2 (feed) $\times$ 3 (salinity) factorial design with triplicate was used for this study. Feed was prepared into the 2 groups: the raw fish-based me>ist pellet (MP) containing $59.5\%$ crude protein and $9.4\%$ crude lipid, and commercial sinking red sea bream feed (CF) containing $49.9\%$ crude protein and $9.4\%$ crude lipid, respectively. And salinity was prepared into the 3 groups: freshwater $(0\%)$, brackishwater $(15\%)$, and seawater $(32\%)$. Fish were fed to satiation twice daily for 6 days a week. Fish were all survived at the end of the 8-week feeding trial. Weight gain (WG) and specific growth rate (SGR) of sunshine bass were significantly (P<0.05) affected by both feed and salinity. WG and SGR of sunshine bass fed the MP were significantly (P<0.05) higher than those of fish fed the CF in the same salinity. Amount of dry feed fed was significantly (P<0.05) affected by both feed and salinity. Feed consumption by sunshine bass fed on the MP was significantly (P<0,05) higher than by fish fed on the CF in the same salinity. Feed efficiency ratio (FER) was significantly (P<0,05) affected by salinity, but not by feed. Protein efficiency ratio (PER) was significantly (P<0.05) affected by both feed and salinity. Moisture and protein content of the whole-body of fish was not significantly (P>0.05) affected by either feed or salinity. However, lipid and ash content of the whole-body of fish was significantly (P<0.05) affected by salinity, but not by feed. In conclusion, the MP was superior to CF for growth of sunshine bass during the winter season in Korea and no mortality occurred. And sunshine bass seemed to grow better in freshwater $(0\%)$ and brac-kishwater $(15\%)$ than seawater $(32\%)$ under these experimental conditions, in terms of improvement in FER and PER in freshwater or brackishwater.

Monthly Gonadal and Sex Hormonal Changes of Indoor-Reared Seabass, Lateolabrax japonicus during Annual Reproductive Cycle (실내사육 농어, Lateolabrax japonicus의 생식소 및 성호르몬의 주년 변화)

  • The sea bass, Lateoiabrax japonicus is a highly valued fish in aquaculture in South Korea. For establishment of seedling production of sea bass,1 japonicus, we examined change of gonadal development and plasma steroid levels of sea bass reared in indoor tank. Male matured unsimultaneously faster than females and spawning of females took place between the end of January and March. After the spawning period, and until the following January, all the females were in preyitello genesis and in some males, spermatogenetic activity restarted gradually. In October, under reducing photoperiod, cortical alveoli appeared in growing oocyte and the development of spermatogenesis greatly increased. Between October and february, vitellogenesis and spermatogenesis occurred respectively in female and male and gonadosomatic index increased from 4.31 to $24.07\%$ in female and upper 6o/o in male. Also, two sex hormones were analyzed during the course of a reproductive cycle in the sea bass: plasma levels of the gonadal steroid testosterone (T) and estradiol-l7$\beta$ (E_{2}). Variation of the plasma concentrations of T and E, appeared to depend on gonad stages. Plasma T and E, levels were high from November to January, suggesting that an sufficient gonadal stimulation by both hormones may undergoing a processes for the formation of sperm and oocyte.

Effects of Acute Change of Salinity on Physiological Response and Survival in Hybrid Striped Bass (Morone Chrysops male X Msaxatilis female) (빠른 염분변화가 Striped bass 잡종 (Morone Chrysops male X Msaxatilis female)의 생리적 반응과 생존에 미치는 영향)

  • Physiological responses and tolerance limit of hybrid striped bass $(Morone\;Chrysops\;male{\times}\;M.\;saxatilis\;female),$ were investigated on salinity changes from 33 psu to 0 psu and from 0 to psu 33 psu in two separate trials, respectively. In the acute salinity decrease from sea water (33 psu) to fresh water (0 psu), plasma level of cortisol, glucose, total protein, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were not affected, however, $Na^+,\;Cl^-$ and osmolality significantly decreased. In the acute salinity increase from fresh water to sea water (33 psu), plasma cortisol level did not change. Glucose and total protein concentrations in plasma and hematocrit (Hct) decreased, whereas AST, ALT, $Na^+,\;Cl^-$ and osmolality increased. The hybrid striped bass can overcome the acute changes of salinity, from 0 psu to 33 psu or from 33 psu to 0 psu, But salinity decrease stressed the fish much more than salinity increase.

Behavior of Juvenile Black Sea Bass, Centropristis striata (Linnaeus) on Oyster Reefs (Oyster reef에서 black sea bass, Centropristis striata 치어의 행동)

  • The substrate preferences of juvenile black sea bass Centropristis striata (Linnaeus) was tested in a circular tank (1.5 m diameter${\times}$0.4 m deep) divided into two equal areas of oyster-related (oyster reef and whole oyster shell) and sand substrates. All trials were video taped for 20 min. Tapes were viewed on a monitor and locations of all fish recorded and timed with respect to substrate. $Mean{\pm}SE$ times on oyster shell were $18.1{\pm}2.0min$ (1-fish trial-1) and $17.5{\pm}1.7min$ (5-fish trial-1). $Mean{\pm}SE$ times on sand were $2.0{\pm}1.0min$ (1-fish trial-1) and $2.5{\pm}1.7min$ (5-fish trial-1). Black sea bass juveniles showed a significant preference for oyster reef and shell over sand substrate in single-fish trials (paired t-test, P<0.05) and also in five-fish trials (paired t-test, P<0.05). $Mean{\pm}SE$ times under oyster reefs were $16.6{\pm}2.0min$ in single-fish trials and $10.7{\pm}2.3min$ in five-fish trials. Mean numbers of movements among oyster reefs were $1.1{\pm}1.0$ in single-fish trials and $11.5{\pm}3.1$ in fivefish trials. Fish spent significantly less time under oyster reefs in five-fish trials, compared to single-fish trials (paired t-test, P<0.05) and they moved more frequently in five-fish trials than in single-fish trials (paired t-test, P<0.05). Significantly higher competition for a refuge in five-fish trials may induce less time under oyster reefs as well as frequent movement of black sea bass juveniles on shell substrate.

  • Two species of digeneans, Tergestia laticollis and Biovarium cryptocotyle, were collected from the intestine of sea basses(Lateolabrax japonicus) captured at the Korean Southern Sea. T. laticollis was characterized by having cephalic lobes around oral sucker, six muscular flanges on each lateral surface at level of pharynx, and species-specific egg size. The most outstanding characteristic of B. cryptocotyle was two-divided ovary. These two digenean species were the first recording species in Korea and L. japonicus was the newly recording host for T. laticollis.

  • Korean temperate sea bass was examined for the taxonomically review. Based on the color pattern on the body sides and some morphometric characters, it was classified as two species, Lateolabrax japonicus and Lateolabrax sp.. Lateolabrax japonicus was redescribed and Lateolabrax sp. was provisionally described. Lateolabrax sp. is similar to L. japonicus, but differs from it by having some black spots on the body sides, fewer number of gill rakers, larger eye, higher position of the pored lateral line scales, and wider secondary body depth.

Selective Response of Rock bream and Sea bass to the Color Nettings (색강지에 대한 돌돔과 능성어의 선택반응)

  • The author carried out experiments to find out the response of rock bream, Oplegnathus fasciatus (TEMMINCK et SCHLEGEL) and sea bass, Epinephelus septemfasciatus (THUNBERG) to the color nettings. The experimental water tank(180L$\times$50W$\times$55Hcm) was set up n a dark room and water level was maintained 50cm high from the bottom. The tank was devided three longitudinal sections marking 60 cm interval. The illumination systems, consisted of 20 watt fluorescent lamps and filter, were suspended adove the tank. Two different color nettings selected from five colors (red, yellow, green, blue, black) were placed in each end section of the tank. Ten fish were used in each experiment and the fish were acclimatized in the dark for 60 minutes before experiment. After the light on, the number of fish in each section of the tank was counted in every 30 seconds interval for 30 minutes. The results obtained are as follows: 1. The rock bream selected the color nettings in the order of yellow, black, blue, green and red. 2. The sea bass selected the color nettings in the order of green, black, red, blue and yellow.
