Virus-like Particles and Cellular Changes in Plants Infected with Sweetpotato Viruses

  • Studies with the transmission electron microscope were used to detect and attempt to identify viruses infecting sweetpotato (Ipomoea batatas) and other Ipomoea species. Flexuous-rods, short curved-rods, and spherical virus-like particles were observed in cells of symptomatic plants. Also, various cytopathic changes such as crystals, vesicles, fibril structures, and cylindrical inclusions were observed. The present study showed that some of these cytopathic changes were associated with some viral groups, which might be helpful in diagnosis.

  • For the investigation of medicinal resources in Aconitum species, the studies were carried out to search the less toxic constituents than Aconitum alkaloid in the aerial part of Aconitum jaluense for. album (Ranunculaceae). From the BuOH fraction of MeOH extract, three flavonoid compounds, hyperoside(1), $kaempferol-3-O-{\beta}-_D-galactopyranoside(2)$, $kaempferol-3-O-{\alpha}-_L-rhamnopyranosyl-(1{\rightarrow}6)-{\beta}-_D-glucopyranoside(3)$ were isolated and identified on the basis of their physico-chemical properties, spectroscopic evidences (UV, IR, NMR and Mass etc.) and comparison with authentics, respectively.

  • Latent infections of healthy-appearing oaks of Hypoxylon atropunctatum complicates field studies by interfering with inoculation experiments to follow pathogenesis, fungal development and reproduction of this canker rot fungus. Mutants with unique and easily scorable phenotypes would be useful for inoculation studies. There is a broad range in the capacity of wild-type isolates to utilize nitrate as a sole nitrogen sources. Several types of nitrate-nonutilization mutants (nit1, Nit3, NitM) were selected from nitrate-utilizing wild-type isolates. Also, a few mutants of Hypoxylon atropunctatum were selected that could only grow poorly on basal medium supplemented with various nitrogen sources and even on yeast extract agar. These unknown mutants need to be characterized further. Nit mutants of Hypoxylon atropunctatum were readily selected, grew well and were recovered after inoculation into oak stems. These results suggest that nit mutants could be useful for inoculation studies in trees that contain latent infections.

A Seedborne Fungus Bipolaris spicifera Detected from Imported Grass Seeds

  • Seedborne fungus Bipolaris spicifera, which has not been previously reported in Korea, was detected from import-ed grass seeds in the country. The most frequently detected fungi from the seeds were Fusarium species, Ulocladium atrum, B. spicifera, Alternaria, and Cuvularia lunata among 17 different seed samples of the family Gramineae. Detection frequencies of B. spicifera were 11,8,5% in Bermuda grass, tall fescue, and mixed lawn grass imported from USA, respectively, and 9% in mixed lawn grass imported from Italy. This suggests that important seedborne pathogen could be spread between countries through seed sources. The pathogen was seed-transmitted causing damping-off of Bermuda grass seedlings and showed strong pathogenicity to vice, corn, Bermuda grass, sorghum, and tall fescue. However, it did not infect wheat and blue grass.

  • A new myxosporean species Ceratomyxa oplegnathus n. sp. was found from the gallbladder of cultured rock bream, Oplegnathus fasciatus. Mature spores were 44.14 ± 2.41 (39.04-48.41) ㎛ in length, 8.52 ± 0.36 (7.9-9.26) ㎛ in width in sutural view. Spores contained two spherical polar capsules measuring 3.04 ± 0.2 (2.71-3.37) ㎛ in diameter and a binucleated sporoplasm. Each polar capsule had a 5-6 coiled polar filament, and was situated near the sutural line. In ultrastructural observation, asynchronous divisions of generative cells without pansporoblast formation gave arise to 2 spores within a trophozoite. Sporoplasmic cell in the sporoblast was binucleated, and capsulogenic cells had large amounts of rough endoplasmic reticulum, external tubules, and capsular primordia.

Different chemical and dynamical environments in two massive star forming regions, G19.61-0.23 and G75.78+0.34

  • Complex organic molecules (COMs) are commonly detected in star forming regions and considered important species since they are seeds of prebiotic molecules. Although COMs form in ice mantles on dust grains, they are preferentially detected in the gas phase. The origin of the gaseous COMs highly depends on the circumstance of each star forming region. Therefore, the distribution of COMs emission reflects the physical and chemical conditions of the region. We present the newly detected COMs, especially methanol emission lines toward two massive star forming regions, G19.61-0.23 and G75.78+0.34 in the Atacama Large Millimeter/submillimeter Array Band 3 observations during the Cycle 2 phase. Multiple transitions of methanol are detected in both regions but show different emission morphology. The origin of the desorption (e.g. shocks or high energy photons) is discussed.

  • Dielectrophoresis has received considerable attention for separating nanotubes according to electronic types. Here we examine the effects of surface conductivity of semiconducting single-walled carbon nanotubes (SWNT), induced by ionic surfactants, on the sign of dielectrophoretic force. The crossover frequency of semiconducting SWNT increases rapidly as the conductivity ratio between the particle and medium increases, leading to an incomplete separation of ionic surfactant suspended SWNT at an electric field frequency of 10 MHz. The surface charge of SWNT is neutralized by an equimolar mixture of anionic surfactant sodium dodecyl sulfate (SDS) and cationic surfactant cetyltrimenthylammonium bromide (CTAB), resulting in negative dielectrophoresis of semiconducting species at 10 MHz. A comparative Raman spectroscopy study shows a nearly complete separation of metallic SWNT.

Floridoside suppresses pro-inflammatory responses by blocking MAPK signaling in activated microglia

  • Inflammatory conditions mediated by activated microglia lead to chronic neuro-degenerative diseases such as Alzheimer's, Parkinson's, and Huntington's diseases. This study was conducted to determine the effect of floridoside isolated from marine red algae Laurencia undulata on LPS (100 ng/ml) activated inflammatory responses in BV-2 microglia cells. The results show that floridoside has the ability to suppress pro-inflammatory responses in microglia by markedly inhibiting the production of nitric oxide (NO) and reactive oxygen species (ROS). Moreover, floridoside down-regulated the protein and gene expression levels of iNOS and COX-2 by significantly blocking the phosphorylation of p38 and ERK in BV-2 cells. Collectively, these results indicate that floridoside has the potential to be developed as an active agent for the treatment of neuro-inflammation.

  • Vertically aligned carbon nanotubes (CNTs) have been produced using various type of plasma enhanced chemical vapor deposition (PECVD). Catalysts such as Ni, Co, and Fe are used for growth of CNTs. To explain the effect of catalysts on the growth characteristics of CNTs, carbon species of $C_2H_2$ was observed in different catalysts using optical emission spectroscopy (OES) with theoretical calculation on the surface reaction in different catalysts.

  • Fuel lean reburning method is very attractive way in comparison with conventional reburning method for reducing NOX. Meanwhile, the knowledge of the how flue gas re-circulated, temperature distribution and species concentration is crucial for the design and operation of an effective fuel lean reburning system. For this reason, numerical analysis of fuel lean reburning system is a very important and challenge task. In this work, the effect of fuel lean reburn system on NOX reduction has been experimentally and numerically conducted. Experimental study has been conducted with a 15kW lab scale furnace. Liquefied Petroleum Gas is used as main fuel and reburn fuel. To carry out numerical study, the finite-volume based commercial computational fluid dynamics (CFD) code FLUENT6.3 was used to simulate the reacting flow in a given laboratory furnace. Steady state, three dimensional analysis performed for turbulent reactive flow and radiative heat transfer in the furnace.
