First Description of Petalonia zosterifolia and Scytosiphon gracilis (Scytosiphonaceae, Phaeophyceae) from Korea with Special Reference to nrDNA ITS Sequence Comparisons

  • Scytosiphonaceae is an acetocarpalean brown algal family, that is a recent focus of synstematics and marine biodiversity. We describe Petalonia zosterifolia and Scytosiphon gracilis from Korea for the first time. P. zosterifolia occurred on the East coast, and had flat, linear and solid thalli. S. gracilis was found in Jeju, and had cylindircal to flat and hol-low thalli. However, these two species are so similar that it is difficult to identify by morphology alone. In order to determine if the nuclear DNA reveals the distinctness of both species and to know their phylogenies, the ITS region sequences were newly detrmined in 22 samples of P. zosterifolia, Scytosiphon gracilis, and other three members of the genera from Korea. We found 0.12% variation among samples of P. zosterifolia from different locations, and no variation between S. gracilis samples from diferent years, but extensive interspecific divergences (13.62-22.83%) of each species to other members in Petalonia and Scytosiphon . The ITS sequence dta consistently showed a close relationship between P. zosterifolia and S. gracilis. This result is congruent with morphology and with the published data of plastid rbc and partial nrDNA large subunit gene sequences, and suggests that P. zosterifolia and S. gracilis might have diverged from the most recent common ancestor.

First Record of Scytosiphon gracilis Kogame (Scytosiphonaceae,Phaeophyceae) for the Pacific coast of Mexico

  • In this work we report the occurrence of Scytosiphon gracilis Kogame (Scytosiphonaceae, Phaeophyceae) on the Pacific coast of Mexico. This is the first report of S. gracilis outside its previously known distribution in the western North Pacific (Korea and Japan). The identification was based on a morphological revision and a comparison of the plastid-encoded RuBisCO spacer sequences determined for Korean and Mexican algal material. Thalli were collected from the intertidal zone of Saldamando Beach, Baja California, in January 2003. The vegetative structure, as well as habitat and geographic distribution of the species are described. Reproductive structures were not found in our specimens. The poor presence/absence of S. gracilis in previous floristic studies of the area could be due to its small size and low frequency.

  • 고리매는 동해안에서 식용되어온 유용 갈조류이며, 다양한 생리활성물질을 함유하여 이용이 기대되는 해조류이다. 이 연구는 2021년 1월부터 12월까지 강원도 주문진읍 소돌해안에서 고리매 개체군의 생장 및 성숙 패턴을 밝히기 위하여 수행되었다. 고리매 개체군의 정량조사를 위해 월별 형태분석, 밀도 및 현존량 분석을 실시하였다. 조사지역에서 고리매의 최대엽장은 3월(수온 10.6℃)에 16.3±3.0 cm, 평균 밀도는 32.5±2.9 ind. m-2 및 평균 현존량은 96.0±12.1 g fresh-wt. m-2 였다. 고리매 엽체의 성숙은 4월(수온 12.1℃)에 최성기를 보였으며, 성숙에 요구되는 성숙유효적산온도는 162.5 degree-days였다.

Anti-inflammatory Mechanism of Seaweeds in Murine Macrophage

  • The effect of 4 seaweed extracts (Desmarestia viridis, Dictyopteris divaricata, Scytosiphon lomentaria, and Ishige okamurae) on pro-inflammatory mediators as well as nuclear factor $(NF)-{\kappa}B$ in the stimulated Raw 264.7 cells was investigated. They reduced iNOS and interlukin $(IL)-1{\beta}$ expressions at transcription level. Of those, 3 extracts (D. divaricata, I. okamurae, and S. lomentaria) inhibited the COX-2 expression at translation level. $I{\kappa}B-{\alpha}$ degradation was inhibited by D. divaricata and S. lomentaria extracts. Therefore, we concluded that the extracts from D. divaricata and S. lomentaria could inhibit the activation of murine macrophage through the blocking of $NF-{\kappa}B$ activation.

잘피밭에 서식하는 부착해조류 군집의 종조성 및 계절변동 (Seasonal Variations in Species Composition and Biomass of Epiphytic Algal Community in an Eelgrass (Zostera marina) Bed)

  • 진동만 잘피밭에서 서식하는 대형 부착해조류는 총 3종이었으며, 출현종은 갈조류의 Scytosiphon lomentaria, Colpomenia sp., 그리고 홍조류의 Gracilaria sp., 으로 구성되어 있었다. 한편 미세 부착해조류는 Cocconeis scutellum와 Cocconeis placentula가 우점하였다. 잘피에 부착하여 서식하는 부착해조류의 현존량은 계절 변동이 뚜렷하게 나타났다. 대형 부착해조류는 2003년 11월에 3.3 g $DW/m^2$, 미세 부착해조류는 2003년 6월에 43,153 $cells/m^2$를 보이며 가장 높게 나타났다. 다른 해역의 잘피밭(광양만, 동대만 및 앵강만)과 비교해보면, 본 조사해역의 잘피밭에서 서식하는 부착해조류의 종 다양성 및 현존량이 매우 낮게 나타나 특이하였다. 이와 같은 결과는 잘피밭의 물리학적인 환경특성(예를 들면 조류 및 유속), 잘피 자체의 생물학적인 특성, 그리고 수질의 악화에 기인하였다.

Effects of Scytosiphon lomentaria on osteoblastic proliferation and differentiation of MC3T3-E1 cells

  • BACKGROUND/OBJECTIVES: Bone formation and bone resorption continuously occur in bone tissue to prevent the accumulation of old bone, this being called bone remodeling. Osteoblasts especially play a crucial role in bone formation through the differentiation and proliferation. Therefore, in this study, we investigated the effects of Scytosiphon lomentaria extract (SLE) on osteoblastic proliferation and differentiation in MC3T3-E1 cells. MATERIALS/METHODS: A cell proliferation assay, alkaline phosphatase (ALP) activity assay, alizarin red staining and protein expression analysis of osteoblastic genes were carried out to assess the osteoblastic proliferation and differentiation. RESULTS: The results indicated that treatment of SLE promoted the proliferation of MC3T3-E1 cells and improved ALP activity. And, SLE treatment significantly promoted mineralized nodule formation compared with control. In addition, cells treated with SLE significantly upregulated protein expression of ALP, type 1 collagen, bone morphogenetic protein 2, runt-related transcription factor 2, osterix, and osteoprotegerin. CONCLUSIONS: The results demonstrate that SLE promote differentiation inducement and proliferation of osteoblasts and, therefore may help to elucidate the transcriptional mechanism of bone formation and possibly lead to the development of bone-forming drugs.

한국 남서해안 하의도 갯벌의 해조상 (Algal Flora of Hauido Tidal Flats in the Southwestern Coast of Korea)

  • We performed qualitative and quantitative surveys on the distribution of seaweed species and dominant species with their standing crop and temporal variations in Hauido tidal flats in order to understand the role of seaweeds in a coastal wetland ecological system that may be used for conservation and management of coastal wetland ecological system. A total of 9 species were found at 7 stations of the study area; 4 species of Chlorophyta (Ulva compressa, U. prolifera, U. pertusa and Ulothrix flacca), 3 species of Phaeophyta (Ectocarpus arctus, Scytosiphon lomentaria and Myelophycus simplex) and 2 species of Rhodophyta (Gracilaria verrucosa and Caulacanthus okamurae). Major dominant species were U. compressa, U. prolifera and U. pertusa (Chlorophyta), Scytosiphon lomentaria (Phaeophyta) and Gracilaria verrucosa (Rhodophyta). Standing crops of U. compressa and U. prolifera were $2.94{\sim}45.25g{\cdot}dw{\cdot}m^{-2}$ and $6.19{\sim}91.9g{\cdot}dw{\cdot}m^{-2}$, respectively, in spring. Seaweed habitat was characterized by textural differences that the percentage of coarse particles and organic material were a little higher in seaweed flourishing areas than in seaweed barren areas.
