A Study on Homogeneity Analysis of the Guipi Decoction for Treatment of Insomnia Disorder -Focusing on Randomized Controlled Clinical Trials in CNKI (불면장애에 사용된 귀비탕의 동질성 연구 -CNKI에서 검색한 무작위배정 임상연구를 중심으로-)

  • Objectives: The study was designed to provide clinical evidence of homogeneity in analysis of the herbal medicine, Guipi decoction in treating insomnia, using a t-test of randomized controlled trials (RCTs). Methods: This study included RCTs that verified homogeneity of Guipi decoction in treating insomnia. Literature studies of CNKI (Chinese National Knowledge Infrastructure Database) were performed. The basic components group was made to the Guipi decoction of the Yixuerumen?s text. The excluded group was created with the number of herbs excluded from the basic components group. T-test performed that the effective rate in the basic components group and excluded groups. Results: The basic components of Guipi decoction contain Atractylodis Rhizoma Alba, Poria Sclerotium (Poria Sclerotium Cum Pini Radix), Polygalae Radix, Astragali Radix, Zizyphi Semen, Angelicae Gigantis Radix, Aucklandiae Radix, Glycyrrhiza uralensis Fischer, Ginseng Radix (Codonopsis Pilosulae Radix), Longanae Arillus. Herbs excluded from the basic components group were Longanae Arillus, Ginseng Radix (Codonopsis Pilosulae Radix), Glycyrrhiza uralensis Fischer and herbs added to the basic components group were Polygonum multiflorum Thunberg, Schisandrae Fructus, Salviae Militiorrhizae Radix, Paeoniae Radix Alba (in order from the most used to the least). T-test revealed the effective rate in the basic components group was not statistically significant compared to the excluded groups (p=0.161/p=0.162/p=0.103). Conclusions: Effectiveness in treatment of basic components of Guipi decoction and excluded groups were revealed to be homogeneous in treating insomnia. As the number of herbs in the basic components group decreased, mean value in effectiveness of treatment also decreased consequentially. However, there were no statistical significances.

Connection of the chromosome and the extent of incidence of ergot fungus in durum wheat (마카로니밀에 있어서의 맥각병의 이병정도와 염색체와의 관련성)

  • The ergot fungus requiers a rather long time for the incubation period for a disease and fungus tends to obstract the formation of the seeds. In order to do an experiment inoculated, a spore of ergot fungi into the florets was done 2-3 days before flowering. As a result, each trisomic types recognized the infection rate to be 90% to 100% so the trisomies of complete immunity was non existent in durum wheat, Triticum durum var. hordeiforme. However, the growth rate of large sclerotium than seeds differed mutually with each trisomic types. This tend to be sensitivity on the trisomies which has been the extra chromosomes of 2A, 4A and 7B in comparison with normal plant but the trisomies related to Tri-6A line was not forming the sclerotium of large size than seeds and grew outside the florets. Consequently, the resisitant gene against ergot fungi was considered to be existing on the chromosome 6A. The ergot fungi requires a long time for the incubation period until it is taken away from infection. The essential effect of resistant gene did not surmise on the chromosome 6A that gene concerned with the physiological effect to restrain the growing up of a ergot fungi in the growing process of plants.

Effect of Crop Rotation Cultivation on the Suppression of Garlic White Rot Caused by Sclerotium cepivorum (전작물 재배에 의한 마늘 흑색썩음균핵병 억제효과)

  • This study was conducted to evaluate the effect of crop rotation cultivation on the suppression of garlic white rot caused by Sclerotium cepivorum in the mini plot ($2^*1^*$ 0.5 m). Six crops, soybean, sesame, mung bean, squash, crotalaria and spring onion, were previously transplanted in the mini-plots infested with S. cepivorum before garlics were planted. After cultivation of the previous crops, garlic was sown in the mini-plot. Non-cultivation plots and non-infested plots with white rot pathogen were used as control. The effect of crop rotation cultivation on the suppression of garlic white rot was evaluated by investigating comparatively the disease incidence (the percentage of infected plants) and yields. As a results, infection rate of garlic white rot was recorded lower in the non-infested plot, crotalaria and soybean cultivation than in the plot of the other crop cultivation. Especially when squash was previously cultivated and garlics were planted in 2013, infection rate of garlic white was recorded the highest score. In 2014, the infection rate of garlic white were low in the garlic on soybean, crotalaria and spring onion treatment whereas it was high in squash treatment, as well. In 2013, garlic yield was the highest in no inoculation plot, followed by crotalaria, soybean, no crop cultivation, sesami, mungbean, squash cultivation plot. In 2014, the yield in the plot of crotalaria and soybean was much higher than that in no inoculation plot. Based on above-described results, it is considered that soybean-garlic and crotalaria-garlic cultivation system can be good crop rotation systems to control garlic white rot.

The culture conditions for mycelial growth and sclerotial formation of Polyporus umbellatus

  • Polyporus umbellatus (Syn. Grifola umbellata) is a sclerotium forming mushroom belongs to family Polyporaceae of Polyphorales, Basidiomycota. The sclerotia of P. umbellatus have long been used for traditional medicines in China, Korea and Japan. This study was initiated to obtain the basic data for artificial sclerotial production of P. umbellatus. Here, we investigated the favorable conditions for mycelial growth of P. umbellatus and its symbiotic fungus Armillaria mellea. We also evaluate the favorable carbon and nitrogen sources for sclerotial formation in dual culture between P. umbellatus and A. mellea. The favorable conditions for mycelial growth of P. umbellatus were $20^{\circ}C$ and pH 4, while optimal conditions for mycelial growth of A. mellea were $25^{\circ}C$ and pH 6. The carbon sources for optimal mycelial growth of P. umbellatus were fructose and glucose, while carbon sources for favorable mycelial growth of A. mellea were also fructose and glucose. The nitrogen sources for favorable mycelial growth P. umbellatus were peptone and yeast extract, while optimal mycelial growth of A. mellea were obtained in peptone and yeast extract. When P. umbellatus and A. mellea were dual cultured on carbon sources, sclerotia were induced on basal media supplemented with glucose, fructose and maltose at pH 4~6, while nitrogen sources inducing sclerotia were basal media supplemented with peptone and yeast extract for 60 days at $20^{\circ}C$ under dark condition.

Tyrosinase Inhibition Activity and Antioxidant Capacity by Fermented Products of Some Medicinal Plants (한방 생약재 발효액의 항산화 활성 및 tyrosinase 저해 활성)

  • The effects of fermented products from 40 medicinal herbals commonly available in Korea were examined according to concentrations of polyphenolic compound and kojic acid, and the activities of DPPH ($\alpha,\alpha$'-diphenyl-$\beta$-picrylhydrazyl) free radical scavenging and tyrosinase. The polyphenolic compound concentrations were 0.24 by Corydalis turtschaminovill ~ 11.42% (dry matter basis) by Syringa velutina in the extracts and 0.18 by Poria cocos ~ 12.27% by S. velutina in the fermented products. Kojic acid concentrations were 0.02 by Poria cocos Sclerotium ~ 9.67 mM by S. velutina in the extracts and 0.33 by P. cocos ~ 10.32 mM by S. velutina in the fermented products. Syringa velutina contained the highest polyphenolic compound and kojic acid concentrations, which were higher in the fermented product than in the extract. Higher DPPH free radical scavenging activity (>60%) was observed in the extracts of A. sessiliflorum, Citrus nobillis, and Angelica gigas and the fermented product of A. sessiliflorum compared to the other medicinal plants. Higher tyrosinase inhibition activity (>50%) was observed in the extracts of Morus alba, Glycyrrhiza glabra, and Rubus coreanus and the fermented products of G. glabra, Cnidium officinale, and S. velutina. Based on the above results, G. glabra, C. officinale, and S. velutina possessed high tyrosinase-inhibitive activities and kojic acid concentrations, which could be definitely enhanced by the fermentation of Phenillus linteus mycelium.

Chemical Composition and Biological Activities of Immunostimulants Purified from Alkali Extract of Poria cocos Sclerotium (복령 균핵의 알칼리추출물에서 정제한 면역활성 증강물질의 작용과 화학구성)

  • An one percent sodium carbonate extract prepared from sclerotia of Poria cocos activated the proliferation of the T lymphocytes as measured by mixed lymphocyte responses(MLR). The active fraction, PCSC22, was isolated from an one percent sodium carbonate extract by a combination of fractionation procedures, including ethanol precipitation and chromatographies on column of DEAE-cellulose and Sephadex G50. Carbohydrate and peptide contained in PCSC22 were 78 : 22% in ratio. On employing gel filtration high performance liquid chromatography, PCSC22 exhitited a homogeneous peak with an average molecular weight of 8 kDa. The sugar moiety of PCSC22 was composed with mannose (92%), galactose (6.2%) and arabinose (1.3%), which might be indicated as heteromannan. Fifteen amino acids were found in peptide moiety of the polysaccharide and aspartic acid, serine, and valine were major components. PCSC22 activated the primary proliferation of T lymphocytes measured by mixed lymphocyte responses, the antibody production of the B lymphocytes and the secretion of nitric oxide from macrophage cell line, RAW264.7.

Studies on Distribution and Utilization of Cordyceps militaris and C. nutans (동충하초속균의 분포 및 Cordyceps militaris와 C. nutans의 이용에 관한 연구)

  • The genus Cordyceps known as an insect parasite forms a sclerotium in insect bodies and then produces perithecia on the single or multiple stromata produced from sclerotium. Collected Cordyceps were identified into 5 species: Cordyceps militaris, C. nutans, Cordyceps sphecocephala, Isaria japonica, and Torrubiella sp. The fruit bodies of Cordyceps in petri-dish cover were fixed by tape and put the lid on water agar plates to isolate these collected Cordyceps. The germinated spores were transferred from water agar to Potato dextrose agar(PDA) after six hours. Mycelial growth of C. nutans and C. militaris was the most successful on Hamada media and was also good on Complete media and PDA. Mannose as a carbon source was good for two species and Glutamic acid as a nitrogen source was satisfactory to C. militaris and Asparagine gave a good result to C. nutans. C. militaris and C. nutans showed similar mycelial growth rate on the media that contained thiamine-HCI, biotine or nicotinic acid as a vitamine. When conidia of C. nutans were inoculated to insects, mortality was high in Artogeia napi L, Hemiptera, Plutella xylostella and 50% in Orthoptera, 12% in Acantholyda posticalise M, but not Agelastica coerulea B. in Aphididae, C. nutans was collected from only Hemiptera in nature, but killing effect on other insects was proved. Mycelial growth and fruit-body formation were good on the media that consist of rice powder 5g, wheat flour 5g, water 100ml, but formed fruit-body was not complete stromata but a mass of conidia according to results of observing microscope.

Biological Control of Soil-borne Diseases with Antagonistic Bacteria

  • Biological control has many advantages as a disease control method, particularly when compared with pesticides. One of the most important benefits is that biological control is an environmental friendly method and does not introduce pollutants into the environment. Another great advantage of this method is its selectivity. Selectivity is the important factor regarding the balance of agricultural ecosystems because a great damage to non target species can lead to the restriction of natural enemies' populations. The objective of this research was to evaluate the effects of several different bacterial isolates on the efficacy of biological control of soil borne diseases. White rot caused by Sclerotium cepivorum was reported to be severe disease of garlic and chive. The antifungal bacteria Burkholderia pyrrocinia CAB08106-4 was tested in field bioassays for its ability to suppress white rot disease. In field tests, B. pyrrocinia CAB08106-4 isolates suppressed white rot in garlic and chive, with the average control efficacies of 69.6% and 58.9%, respectively. In addition, when a culture filtrate of B. pyrrocinia CAB08106-4 was sprayed onto wounded garlic bulbs after inoculation with a Penicillium hirstum spore suspension in a cold storage room ($-2^{\circ}C$), blue mold disease on garlic bulbs was suppressed, with a control efficacy of 79.2%. These results suggested that B. pyrrocinia CAB08106-4 isolates could be used as effective biological control agents against both soil-borne and post-harvest diseases of Liliaceae. Chinese cabbage clubroot caused by Plasmodiophora brassicae was found to be highly virulent in Chinese cabbage, turnips, and cabbage. In this study, the endophytic bacterium Flavobacterium hercynium EPB-C313, which was isolated from Chinese cabbage tissues, was investigated for its antimicrobial activity by inactivating resting spores and its control effects on clubroot disease using bioassays. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivated the resting spores of P. brassicae, with the control efficacies of 90.4%, 36.8%, and 26.0%, respectively. Complex treatments greatly enhanced the control efficacy by 63.7% in a field of 50% diseased plants by incorporating pellets containing organic matter and F. hercynium EPB-C313 in soil, drenching seedlings with a culture solution of F. hercynium EPB-C313, and drenching soil for 10 days after planting. Soft rot caused by Pectobacterium carotovorum subsp. carotovorum was reported to be severe disease to Chinese cabbage in spring seasons. The antifungal bacterium, Bacillus sp. CAB12243-2 suppresses the soft rot disease on Chinese cabbage with 73.0% control efficacy in greenhouse assay. This isolate will increase the utilization of rhizobacteria species as biocontrol agents against soft rot disease of vegetable crops. Sclerotinia rot caused by Sclerotinia sclerotiorum has been reported on lettuce during winter. An antifungal isolate of Pseudomonas corrugata CAB07024-3 was tested in field bioassays for its ability to suppress scleritinia rot. This antagonistic microorganism showed four-year average effects of 63.1% of the control in the same field. Furthermore, P. corrugata CAB07024-3 has a wide antifungal spectrum against plant pathogens, including Sclerotinia sclerotiorum, Sclerotium cepivorum, Botrytis cinerea, Colletotrichum gloeosporioides, Phytophotra capsici, and Pythium myriotylum.

Antifungal Activity of Lichen-Forming Fungi Isolated from Korean and Chinese Lichen Species Against Plant Pathogenic Fungi

  • Antifungal activity of Korean and Chinese lichen-forming fungi(LFF) was evaluated against plant pathogenic fungi of Botryosphaeria dothidea, Botrytis cinerea, Diaporthe actinidiae, Pestalotiopsis longiseta, Pythium sp., Rhizoctonia solani, and Sclerotium cepivorum. The LFF were isolated from Cladonia scabriuscula, Melanelia sp., Nephromopsis asahinae, Nephromopsis pallescens, Parmelia laevior, Pertusaria sp., Ramalina conduplicans, Ramalina sinensis, Ramalina sp., Umbilicaria proboscidea and Vulpicida sp. with discharged spore method. The isolates were deposited in the herbarium of Korean Lichen Research Institute(KoLRI) in Sunchon National University. The LFF of Melanelia sp., P. laevior, Pertusaria sp., R. conduplican and Ramalina sp. exhibited strong antifungal activity against all of the pathogenic fungi examined. Among them, LFF of P. laevior showed more than 90% of inhibition in fungal hyphae growth, compared with control. The results imply that LFF can be served as a promising bioresource to develop novel biofungicides. Mass cultivation of the LFF is now under progress in laboratory conditions for chemical identification of antifungal substances.

The Current State of Clinical Studies for Herbal Medicine of Attention Deficit Hyperactivity Disorder (ADHD) -Focusing on Chinese Journals- (ADHD의 한약물 치료에 대한 최신 임상연구 동향 -2007년부터 2012년까지 중국 논문을 중심으로-)

  • Objectives : The purpose of this study was to take around herbal medicine for Attention Deficit Hyperactivity Disorder in China. Methods : A journal search was performed using China National Knowledge Infrastructure (CNKI) from 2007 to 2012. Key words searched were 'ADHD'. Results : 25 studies were selected. Case control studies showed that herbal medicine treatment reported better results than the western medicine treatment group. 12 case reports reported a significant improvement in herbal medicine treatment for patients with ADHD. Frequently used herbal materials were Rhizome of Polygalae Radix and Rhizome of Acori Gramineri Rhizoma (16 times), Root of Glycyrrhiza uralensis Fisch (14 times), Root of Rehmanniae Radix et Rhizoma Preparata and Sclerotium of Poria cocos Wolf (12 times). Conclusions : There have been many studies regarding herbal medicine for Attention Deficit Hyperactivity Disorder in China, and these studies can be applied to the clinical practices in Korea.
