Lignans from the Fruits of Schizandra chinensis and Their Inhibitory Effects on Dopamine Content in PC12 Cells

  • Five lignans including gomisin N (1), wuweizisu C (2), gomisin L1 (3), (+)-deoxyschizandrin (4), and gomisin J (5) have been isolated from the fruits of Schizandra chinensis. The structures of the isolated compounds were elucidated by analyzing MS and NMR spectra. Effects of the compounds isolated in this study on the dopamine content in PC12 cells were investigated to evaluate their inhibitory effectiveness. The gomisin N, wuweizisu C, and gomisin J showed 25.4%, 39.8%, and 35.1 %, respectively, inhibition effect on dopamine content in PC12 cells at the concentration of $50\;{\mu}g/ml$.

Gomisin J with Protective Effect Against t-BHP-Induced Oxidative Damage in HT22 Cells from Schizandra chinensis

  • Four lignan compounds including gomisin J (1), schizandrin (2), gomisin A (3), and angeloyl gomisin H (4) have been isolated from the MeOH extract of Schizandra chinensis fruits. The evaluation for protective effect of compounds 1-4 against tert-butyl hydroperoxide (t-BHP)-induced cytotoxicity in hippocampal HT22 cell line was conducted. Compound 1 showed significant protective effect with an $EC_{50}$ value of $43.3{\pm}2.3\;{\mu}M$, whereas compounds 2-4 were inactive. Trolox, one of the well-known antioxidant, used as a positive control, and also showed protective effect with an $EC_{50}$ value of $213.8{\pm}8.4\;{\mu}M$. These results suggest that compound 1 may possess the neuroprotective activity against oxidant-induced cellular injuries.

  • The heavy metal removability of four kinds of tea was investigated in the various conditions: particle size of tea(10, 35. 70, 100 mesh), concentration of heavy metal(25, 50, 100ppm) and extraction temperature(30. 50, 70. 10$0^{\circ}C$). The removabilities by the tea solids were increased as the particle size decreased, concentration of heavy metal increased, and extracting temperature increased. Of the four kinds of tea Pb, Cd and Cu removability by Eucommia ulmoides Oliv tea was the highest. In addition, heavy metal removability by Schizandra chinensis Baillon tea was better than that of other tea as extraction temperature was increased.

Analysis of Antioxidative Activity and Antimutagenic Effect of Ethanol Extract from Schizandra chinensis Baillon (오미자(Schizandra chinensis Baillon) 에탄올 추출물의 항산화 활성 및 항돌연변이 활성 분석)

  • In this study, the antioxidative activity and antimutagenic effect of ethanol extracts from Schizandra Chinensis Baillon were assessed with regard to natural antioxidants. The antioxidative activity and antimutagenicity of ethanol extracts from Schizandra chinensis Baillon were evaluated by measuring the radical-scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, and by performing the Ames test using 4-nitroquinoline-1-oxide (4-NQO) and sodium azaid as a mutagen with Salmonella typhimurium TA100, respectively. The total polyphenolic compound and flavonoid compound contents of Schisandra chinensis Baillon were also assessed. The DPPH radical-scavenging activity of ethanol extracts from Schisandra chinensis Baillon was 57.2% on the $500\;{\mu}g$/assay. The $IC_{50}$ on DPPH radical-scavenging effect of ethanol extract was $435\;{\mu}g$/assay. In addition, the inhibition rates of ethanol extract from Schisandra chinensis Baillon toward mutagenicity induced by 4-NQO and sodium azide were 82.45% and 45.3%, respectively. Furthermore, the total polyphenol and total flavonoid contents of the extract were 9.53 mg/g and 3.97 mg/g, respectively. These results indicate that the ethanol extract from Schisandra chinensis Baillon evidences a fairly good antioxidative and antimutagenic effect.

Volatile components of Schizandra Chinensis Bullion (오미자(Schizandra Chinensis Bullion)의 휘발성 성분)

  • The essential oil of Schizandrae Fructus was extracted by simultanous distillation-extraction and steam distillation and analyzed by GC and GC-MS. The oil content of Schizandrae Frutus was 0.64% when extracted by steam distillation and 0.88% in case of simultanous distillation extraction and sensory analysis of the oil indicated woody, camphorous, spicy and sour characteristic notes. The experimental results confirmed the presence 47 volatile components, the major components were ${\gamma}-terpinene$, p-cymene, ${\alpha}-ylangne$ and ${\beta}-elemene$. The oil was fractioned into ten fractions and 4 fraction of them have a good aroma character. The application of the oil showed the utilization possibility as flavoring materials for Korea tea.

Isolation of Antimicrobial Substance from Schizandra chinensis Baillon and Antimicrobial Effect (오미자로부터 항균활성 물질의 분리 및 항균효과)

  • In order to isolate antimicrobial substances from Schizandra chinensis, the dried fruits were extracted with the methanol and the extract showed a strong antimicrobial activity. Also, the methanol exract was further fractionated with hexane, dichloromethane, ethylacetate and buthanol. The ethyl acetate-soluble fraction showed the strongest antimicrobial activity. These fraction were further separated by using various chromatographic methods including thin layer chromatography, silicagel open column chromatography and prep. HPLC. A major component S-EA-5-T1 and S-EA-5-T3 from the ethyl acetate fraction, which showed a strong antimicrobial activity was identified by Mass and NMR spectrometry. Two compounds were isolated and identified as trimethylcitrate and the essential oil of Schizandra chinensis and was estimated as gomisin C, respectively. The growth of S. typhimurium was also inhibited about 1.65 to 2.86 log cycle in minced pork by the addition 1% of Schizandra chinensis extract for 12 days at $4^{\circ}C$. These results suggested that these compounds have a strong potential as a natual food preservatives.

  • Omija (Schizandra chinensis Baillon) has five lignans such as schizandrin, gomisin A, gomisin N, gomisin C and schizandrin C. Its major constituent, schizandrin, has a deterrent effect on nephrotoxicity of cisplatin, blood sugar and an ulcer of the stomach. In this study, we have investigated optimized extraction condition to make high functional beverage by quantifying schizandrin and measuring of browning index. Browning index was used as an index of color deterioration of anthocyanin. Browning index was decreased according to increment in extraction temperature and ethanol concentration. The optimum extraction time, extraction temperature and alcohol concentration levels were 3 hrs, $46^{\circ}C$ and 24% alcohol for high schizandrin concentration and desired browing index.

Characteristics of Schizandra chinensis Baillon Orchard Soils Located in Jangsu-gun, Jeollabuk-do (전라북도 장수군 오미자 재배과원 토양 특성)

  • This study was conducted to investigate the physico-chemical properties of Schizandra chinensis Baillon orchard soils located in Jangsu-gun, Jeollabuk-do. Surface (0 to 10 cm) soils were collected from 200 experimental sites located at Jangsu-eup (53 site), Gyenam-myeon (31), Chunchun-myeon (73), Janggye-myeon (12), Bunyam-myeon (31). The soil texture was mostly loamy sand, and the mean values of degree of soil aggregate and soil porosity were 33.1 and 59.9%, respectively. The pH, EC, total-N, available-P, soil organic matter, and cation exchange capacity of the soils were $5.51{\pm}0.54$, $290{\pm}139{\mu}S\;cm^{-1}$, $946.3{\pm}65.5mg\;kg^{-1}$, $319.6{\pm}29.2mg\;kg^{-1}$, $29.0{\pm}13.9g\;kg^{-1}$ and $4.11{\pm}0.34cmol_c\;kg^{-1}$, respectively. The concentrations of Pb, Cd, Cu and Zn were $3.48{\pm}0.55$, $0.09{\pm}0.04$, $6.90{\pm}0.91$ and $97.7{\pm}42.2mg\;kg^{-1}$, respectively. The presented data can be utilized in better managing Schizandra chinensis Baillon orchard soils in the studied areas.

Effect of Lignan Components from Schizandra chinensis on Beta-amyloid Aggregation Inhibition and Dissociation (오미자 리그난 화합물의 베타-아밀로이드간 결합 억제 및 해리 효과)

  • The present study tested the effect of Schizandra chinensis lignan compounds, Gomisin A and Schizandrin, on the aggregation and dissociation of beta-amyloid $(A{\beta})_{1-42}$ to explore a possible therapeutic target for Alzheimer's disease. Gomisin A significantly inhibited the $A{\beta}_{1-42}$ aggregation in a dose dependent manner, but did not induced the dissociation of aggregated $A{\beta}_{1-42}$. On the other hand, Schizandrin significantly suppressed the aggregation and dissociation of $A{\beta}_{1-42}$. These results suggest that Gomisin A and Schizandrin, which are known as biologically active ingredients from Schizandra chinensis, may be potentially useful target molecules to develop a drug for the prevention or treatment of Alzheimer's disease.
