RAPD (Random Amplified Polymorphic DNA) analysis was conducted to Schisandra nigra plants in order to select the specific markers for monoecious and dioecious individuals. RAPD results using eighty random 10-mer primers revealed that S. nigra had a different banding pattern from S. chinensis and Kadsura japonica. When DNA isolated from leaves of monoecious and dioecious plants were used as PCR template, only five primers, OPA-17, OPA-19, OPB-03, OPB-09 and OFB-16, showed polymorphic band patterns. No variation in banding profiles within male or female individuals was observed when these five primers were used whereas three monoecious plants (No 1, No 2 and No 3) showed different banding patterns one another, A 750 bp segment was amplified by primer OPB-3 from male individuals. On the other hand, two segments, 950 bp and 1690 bp, with OPA-19 and 700 bp of segment with OPB-3 were amplified in female individuals. These result indicate that the specific buds of male and female S. nigra could be used as genetic markers for the early discrimination of male and female individuals.
Schisandra nigra Max., a dioecious plant native to Jeju Island in Korea, is cultivated on a small scale for fruit production. As fruit-producing female individuals are generally considered to be more valuable than male, early identification of plant sex at the seedling stage is important. In this study, a sequence-characterized amplified region (SCAR) marker associated with a female-specific region in the genome of S. nigra was investigated. Of 120 randomly amplified polymorphic DNA (RAPD) primers, one primer (OPB-03) consistently amplified a 749 bp band in female plants. The female-specific PCR product was isolated and cloned, and the nucleotide sequences were then determined. Southern hybridization performed using the female-specific fragment as a probe produced positive signals only in genomic DNA from the female plants. This result revealed that the 749 bp segment of DNA was present in the genome of female plants but absent in the genome of male plants. A SCAR primer pair was designed based on the RAPD marker to amplify a 436 bp fragment in the genomic DNA of female plants. This primer pair amplified the expected size of DNA fragment in female plants and four monoecious individuals collected from a natural population. The SCAR marker identified in this study can be used to distinguish female-flowering individuals at the seedling stage.

It was investigated that morphological characteristics of leaf and stomata, chlorophyll contents and wood anatomy characteristics in Schisandra chinensis, Schisandra nigra and Kadsura japonica. Leaf morphological characteristics were significant at 5% levels within the species and within the individuals as a male, a female and monoecious. K. japonica was the longest in the leaf length, followed by S. chinensis and S. nigra. For the leaf length among S. nigra individuals, the female was longer than male and monoecious. As for the Petiole length, S. nigra was twice longer than S. chinensis and K. japonica. Stomata length was ranged from $92.5\;{\mu}m$ to $105.9\;{\mu}m$ for S. nigra, $79.9\;{\mu}m$ for S. chinensis, $76.2\;{\mu}m$ for K. japonica. S. nigra was longer than the others in length and width, while its stomata density was lower than the others. The size of vessel elements and xylem fiber showed a similar tendency to stomata, significant at 5% levels within S. nigra individuals (male, female and monoecious). Chlorophyll contents were also variable in inter and intra species and in general K. japonica showed higher levels of chlorophyll contents than Schisandra spp. plants.

Foliar and physiological characteristics of Schisandraceae plants distributed in Korea were investigated to elucidate differences in growth and the variations of inter and intra species of the plants. For this study, three species of Schisandraceae plants were used; i) Schisandra chinensis, decidious broad-leaved tree, ii) S. nigra, decidious broad-leaved tree, including monoecious, male and female types, ii) Kadsura japonica, evergreen broad-leaved tree. There were no significant differences in leaf length and width among three species, but leaves of Schisandra sp. plants were shorter and wider relative to those of K. japonica. The length of petiole of Schisandra sp. plants was greater than that of K. japonica and the difference was evident between female type of S. chinensis and K. japonica as longer as two times. Schisandra sp. trees showed larger size of stomata, but fewer numbers of stomata, compared with K. japonica. Although substantial changes in leaf morphology were not found in the Schisandraceae plants grown in different environmental conditions, the foliar length of K. japonica was seemed to be increased with increasing an altitude. Chlorophyll contents were also variable in inter and intra species and in, general K. japonica showed higher levels of chlorophyll contents relative to Schisandra sp. plants. K. japonica also showed higher light compensation point(LCP), light saturation point(LSP) and $CO_2$ assimilation rates at LSP, compared with Schisandra sp. trees. However, Schisandra sp. trees showed lower respiration per photosynthesis rates at LSP. The finding suggested that Schisandra sp. trees were acclimated to the growth conditions of lower light intensity with more effective photosynthetic activity. This was also confirmed by the fact that Schisandra sp. plants grown in shading places of a forest demonstrated the elevated rates of net $CO_2$ assimilation under the conditions of low light intensity.

Schisandra nigra Max. has been cultivated far a medical use as well as food. It is an endemic species which has a unique habitat at the altitude of 600-1,400 m in Cheju island. In this study, three natural populations of S. nigra were investigated by using of starch-gel electrophoresis to determine the extent and distribution of genetic diversity. Except 2 monomorphic locus (Mdh-2 and Pgi-1), 4 of the 6 isozyme locus (Idh, Mdh-2, Mnr, and Pgi-2), verified from 4 isozymes, revealed polymorphism in the three populations of S. nigra. The mean number of allele per locus was 1.7 and the percentages of polymorphism loci were 38.9% at 95% level and 50% at 99% level respectively. The observed and estimated heterozygosities were 0.141 and 0.147 respectively. Although plants which were in the face of crisis and distributed in the restricted area, have been known to the very low degree of genetic variation, S. nigra showed higher genetic variation than others. Genetic variation was mostly allocated within population and individuals than that among populations. The result of Wright's F analysis estimates of $F_{IT}$ and $F_{IT}$ showed that S. nigra population revealed Hardy-Weinberg steady state.

To propose basic information for natural resource management and conservation strategy of Schisan-dra nigra Maxim., its habitat environment and vegetation structure of forest communities growing the species was investigated. Fourteen plots (each plot size 400 $m^2$) were set up at the distributed area of Cheju island, southern part of Korea. S. nigra is the economic useful species that scatteringly grows at 850~920 m in the western part, at 660~710 m in the northern part and at 750~1,250 m in the southern part of Mt. Halla. The community of S. nigra in Mt. Halla is the mixed secondary forest of pine and broad leaved trees that are mainly composed of Pinus densif1ora, Quercus serrata, Styrax japonica, Lin-dera erythrocarpa and Carpinus laxiflora. The density in a main distribution sector of S. nigra appears in the range of 3~39 individuals per each 20$\times$20 m plot. The distribution patterns by Morisita's Index showed that S. nigra was distributed randomly in all of the stands. The association analysis showed that the following tree species were positively associated with S. nigra. In other words, the following trees were appeared to require the same ecological niche together with S. nigra: Lindera erythrocarpa, Cornus waltri, Acer mono, and Styrax japonica.

There are about 2 genera and 49 species of Schisandraceae all over the world which grow 2 genera and 5 species in korea. As'Schisandra'genus, S. chinensis, S. repanda, S. nigra and S. viridicarpa are distributed. As 'Kadsura' genus, there is K. japonica species. Although S. chinensis is often used for food and herbal medicine, 'Cheongsoon'is the only cultivar in korea. Therefore cultivar breeding is required for standardized production of Schisandra chinensis. To select superior lines, characteristics of collected varieties was investigated. These collections were from all the country including Mungyeong, Muju and Jangsu. In the case of 'Schisandra chinensis', analysis of chemical compound is very important. Because in 'Korean Pharmacopoeia', Schisandrin has to contain more than 0.7% of sum of Shizandrin, Gomisin A, Gomisin N as standard compound. According to the investigation, the weight of SC-154 was the highest one that was 99g of fresh fruit and 60g of dried fruit in the 100 granulas of it. As the control group, the weight of cultivar 'Cheongsoon'was 57g of fresh fruit and 40g of dried fruit. The chemical compounds such as Shizandrin A, Gomisin A, Gomisin N and Shizandrin C from the collections were analyzed by HPLC. As a result, all average of total content was 1.67% satisfied with the standard that is over 0.7%. Among them, SC-13, SC-22, SC-40 and SC-45 had more than 2.0% of compound. In this case, the content of Gomisin N was relatively higher than that of the other collections. It can be used for high content compound breeding. This above results indicate that these findings could be used for breeding superior Schisandra chinensis through selection of high quality resource.
Objectives : The original plant species of Schisandrae Fructus (O-mi-ja) is prescribed as Schisandra chinensis $B_{AILL.}$, in Korea, but S. chinensis $B_{AILL.}$ and S. sphenanthera $R_{EHD.}$ et $W_{ILS.}$ in China. Moreover, fruit of several other species in genus Schisandra also have been used as the same herbal medicines. To develop a reliable method for correct identification of Schisandrae Fructus and to evaluate the phylogenetic relationship of S. chinensis and its related species, we analyzed internal transcribed spacer (ITS) sequences of nuclear ribosomal DNA (nrDNA). Methods : Twenty-four plant samples of three Schisandra species and one Kadsura species, S. chinensis $B_{AILL.}$, S. spenanthera $R_{EHD.}$ et $W_{ILS.}$, S. nigra $M_{ax.}$ and Kadsura japonica $D_{UNAL}$ were collected from each different native habitate and farm in Korea and China. The nrDNA-ITS region of each samples were amplified using ITS1 and ITS4 primer and nucleotide sequences were determined after sub-cloning into the pGEM-Teasy vector. Authentic marker nucleotides were estimated by the analysis of ClastalW based on the entire nrDNA-ITS sequence. Results : In comparative analysis of the nrDNA-ITS sequences, we found specific nucleotide sequences including indels (insertions and deletions) and substitutions to distinguish C. chinensis, S. spenanthera, S. nigra, and K. japonica. These sequence differences at corresponding positions are avaliable nucleotide markers to determine the botanical origin of O-mi-ja. Moreover, we evaluated the phylogenetic relationship of four plant species by the analysis of nrDNA-ITS sequences. Conclusions : These marker nucleotides would be useful to identify the official herbal medicines by the providing of definitive information that can identify each plant species and distinguish it from unauthentic adulterants for O-mi-ja.
